<B>verfahren zur Herstellung von</B> N-Benzhydryl-tropylaminen Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer chemischer Verbindungen mit antispasmodischen, anti- histaminischen und mydriatischen Eigenschaf tes, und zwar ein Verfahren zur Herstellung von N -Benzhydryl-tropylaminverbindungen.

Es ist bei pharmakologischen Vemsuchen gefunden worden, dass dieses sekundäre Amin und jene Derivate desselben, welche in der 4-Stelling eines Benzolringes substituiert sind, sowie Salze dieser Amine, insbesondere die quaternären Ammoniumsalze, eine ungewöhn liche physiologische Aktivität aufweisen.
Diese Substanzen besitzen eine im Ausmass verschie dene antispasmodische Wirkung ähnlich der des Atropinsulfats, und einige von ihnen, wie beispielweise das N-Benzhydryl-tropylamin-di- niethy1suIfat, übertreffen sogar das Atropin- sulfat an Wirksamkeit, wobei sie gleichzeitig eine starke antihistaminische Aktivität ent wickeln.
Die Mono- und die Bioxalatsalze haben antispasmodische und mydxiat.iselie Wirkungen, die denen des Atropinsulfats nahekommen.
Die bevorzugten Produkte sind die Salze, da sie in fester und im allgemeinen in kri stalliner Form hergestellt werden können, während die freie Base gewöhnlich als eine ölige Flüssigkeit gewonnen wird. Das freie Amin kann auch als Halbhydrat kristallisiert werden. Das Kristallwasser wird aber leicht wieder abgegeben, beispielsweise bei der Trocknung im Vakuum, und es, bleibt das ölige freie Amin zurück.

Die genannten Benzhydrylamine werden vorzugsweise in Form von Salzen, beispiels weise als Hydroehloride, benutzt. Die erfindungsgemässe, reduzierende Kon= densation wird zweckmässig in einem wasser freien Lösungsmittel, wie absolutem Metha nol, Äthanol oder Isopropanol, vorzugsweise unter Wasserstoffdruck vorgenommen. Als Hydrierungskatalysatoren können Platin, Pla tinoxyd, Raneynickel, Kupferchromit oder dergleichen, verwendet werden.
Man kann einen Wasserstoffdruck von bis zu 3,5 kg/cm2 anwenden. Sehr gute Ergebnisse sind mit Drucken im Bereich von 1,4-2,8 kg/cm2 er halten worden. Die Kondensation kann auch bei atmosphärischem Druck ausgeführt wer den, jedoch auf Kosten der Reaktionsdauer und der Ausbeute.
Bei der Reaktion kann durch Reduktion des Tropinons als Nebenprodukt Tropinol ent stehen. Es ist daher ratsam, einen Überschuss an Tropinon anzuwenden. Ein Überschuss von etwa 30%, gibt befriedigende Resultate. Ein grösserer Überschuss ist nicht notwendig.
Er beeinträchtigt aber nicht die Ausbeute an dem sekiuidären Amin. Die Reaktionszeit ist ab hängig von der Menge und der Wirksamkeit des Katalysators. Es ist daher zweckmässig, die Ausgangsstoffe so lange aufeinander ein wirken zu lassen, bis die Wasserstoffaufnahme beendet ist. Vom Standpunkt der Aktivität pro Gewichtseinheit ist als - Katalysator das Platinoxyd vorzuziehen. Bei diesem Katalysa tor kann die Reaktionszeit bis zu 72 Stunden betragen, wenn die Menge d@e6- Katalysators, bezogen auf das Keton, 0,2 Gewichtsprozente ausmacht, sie kann aber auch nur 15 bis 20 Stunden umfassen, sofern man etwa. 2 Ge wichtsprozente Katalysator anwendet.
Es ist vorteilhaft-, unter wasserfreien Be dingungen zu arbeiten, da die Gegenwart <B>s</B> elbst kleiner Wassermengen die Reduktion des Ketons an Stelle der Kondensation zu för dern scheint. Ausserdem wird die Gewinnung der Kondensationsprodukte bei Verwendung von wasserfreien aliphatischen Alkoholen, ins besondere von wasserfreiem Äthanol, infolge ihrer Löslichkeit in diesen Medien erleichtert.
Da man der Meinung sein könnte, dass die vorliegende Kondensationsreaktion bekannten Reaktionen der reduzierenden Kondensation, cl-er reduzierenden Aminierung oder der redu zierenden Alkylierung ähnelt, muss hervor- s gehoben werden, da.ss bei der erfindungsgemä ssen Umsetzung die Ketogruppe einen Teil der als Ausgangsmaterial benutzten Tropyl- j,erbindung bilden muss.
Versuche, Tropyl- amin mit Benzophenon umzusetzen, haben nicht zu dem gewünschten sekundären Amin geführt.
Nach Beendigung der Kondensation kann das sekLindäre Amin in roher Form durch Abfiltrieren des Katalysators und Konzen- f trierimg der alkoholischen Lösung im Vakuum unter Erwärmung, beispielsweise auf einem Dampfbad, gewonnen und der Rückstand in Wasser, das mit Salzsäure angesäuert ist, ge löst werden.
Nach dem Abfiltrieren des ab- e geschiedenen festen Körpers kann man die Lösung mit einer Base behandeln und hierauf mit Äther extrahieren. Nach dem Trocknen kann die Ätherlösung bis auf einen öligen Rückstand konzentriert werden, der aus dem ; sekundären Amin besteht und noch etwas von dem unverbrauchten primären Amin enthält. Man kann .aber auch die Ätherlösung unmit telbar zur Herstellung verschiedener Salze des sekundären Amins benutzen. Die Reinigung #, wird vorzugsweise so durchgeführt, dass man das sekundäre Amin in ein Salz umwandelt, dieses Salz so lange umkristallisiert, bis -es rein ist, und dann die reine Base daraus her stellt.
Die verschiedenen Salze werden zweck- f mässig durch Zusatz der betreffenden Säure bzw. der sauren Reagenzien zur Ätherlösung der freien Base gebildet. Beispieb <I>1</I> -V-B.e2izhydr-,yl-tropylaniiit-perchlorat Benzhydrylamin-hydroehlorid wurde aus Benzophenonoxim hergestellt. Tropinon wurde aus Petroläther umkristallisiert und unter Stickstoff im Kühlschrank bereitgestellt.
Die reduzierende Kondensation wurde aus geführt, indem man 25 mg Platinoxyd zu einer Lösung von 11 g (0,05 lI) Benzhydryl- amin-hydrochlorid und 9 g (0,065 M) Tropi- non in 50 ml absolutem Äthanol zusetzte. Die Mischung wurde mit Wasserstoff bei Raum- temperatur und 2,1 kg/em2 geschüttelt. Die Wasserstoffaufnahme war langsam.
Es wur den zwei weitere Anteile des Katalysators (50 mg und 100 mg) nach 24 Sauiden bzw. 48 Stunden zugesetzt. Am dritten Tag hörte die Wasserstoffaufnahme auf. Der Katalysa tor wurde durch Filtration entfernt und die alkoholische Lösung im Vakuum auf dem Dampfbad bis auf den verbleibenden Rück stand konzentriert. Man fügte nun Wasser hinzu und stellte die Lösung mit verdünnter Salzsäure auf kongosauer ein. Man behandelte dann die Lösung mit 50 ml Äther und machte :il daraufhin mit einem Überschuss von Ka- liumkarbonat basisch.
Es schied sich ein öli ges Produkt ab. Dieses öl wurde in Äther auf genommen, die ätherische Lösung mit Wasser gewaschen, über galitunkarbonat getrocknet und eingedampft, bis ein öliger Rückstand von N-Benzhydryl-tropylamin und Benzhydryl- ti min zurückblieb.
Das feste Perchlorat des N-Benzhydryl- tropy lamins wurde erhalten, indem man eine ätherische Lösung des öligen Rückstandes mit 1-u-alkoholiseher Perchlorsäure, die tropfen weise zugesetzt wurde, auf kongosauer ein stellte. Durch Umkristallisierumg aus Äthanol Äther wurde das gereinigte Perchlorat mit einem! Schmelzpunkt, von 242-244 C erhalten.
N-Benzhydryl-tropylamin-Oxalat Das wasserlösliche Monooxalat wurde durch Auflösen der freien Base, die man aus dem Perchlorat abgeschieden hatte, in Äther und Zusatz von alkoholischer Oxalsäure bis zur sauren Reaktion mit Kongopapier erhal ten. Es wurde aus Isopropanol unter Zusatz einer geringen Menge Methanol am Rück flusskühler umkristallisiert und hatte einen Schmelzpunkt von 120 C.
Der Zusatz eines Überschusses von alkoho lischer Oxalsäure zu einer ätherischen Lösung der freien Base ergab das Bioxalat vom Schmelzpunkt 152-154 C.
<I>Beispiel 2</I> N-Beii.zhyclrpl-tropylamin 20 g Tropinon und 25g Benzhydrylamin- bydrochlorid wurden 150 ml absolutem Ätha- nol zugesetzt und hierauf 500 mg Platinoxyd der entstandenen Suspension zugefügt. Die Kondensation wurde durch Schütteln der Mi schung bei Raumtempexatur und 2,3 kg/cm2 Wasserstoffdruck durchgeführt. Die Wasser stoffaufnahme war nach 16 Stunden nur noch sehr gering, und die Reaktion war nach 20 Stunden beendet.
Der Katalysator wurde ent- iernt und die Lösung im Vakuum bis auf den N ,erbleibenden Rückstand eingeengt. Diesem Rückstand wurde Wasser zugefügt und mit Salzsäure ein pH-Wert von 2-3 eingestellt. Man kühlte darauf die Lösung ab itid ent fernte den Niederschlag. Das Filtrat wurde durch Zusatz von Natriumhydroxyd basisch gemacht und mit Äther extrahiert. Der Äther extrakt wurde mit Wasser gewaschen und im Vakuum konzentriert, bis ein Rückstand von 1T-Benzhydryl-tropylamin verblieb.
Der Rück stand wurde in 20 ml Äthanol gelöst und eine Lösung von 14 g Oxalsäure in 40 ml absolutem Äthanol hinzugefügt. Beim Abkühlen der alkoholischen Lösung kristallisierte N-Benz- liydryl-tropylamin-bioxalatvom Schmelzpunkt 146-149 C aus. Das Produkt wurde aus 520 ml absolutem Äthanol umkristallisiert und zeigte dann einen Schmelzpunkt von 149 bis 153 C.
' N-Benzhydryl-tropylamin-dimethyls-Lilfat Aus 17 g N-Benzhydryl-tropylamin-bioxa- lat wurde die Base freigesetzt. Die Mischung wurde mit Äther extrahiert und der Äther im Vakuum abgedampft, worauf ein öliger Rück stand verblieb. 8 g des öls wurden in 80 m1. Äther gelöst, dann fügte man 4 ml Dimethyl- sulfat hinzu. Es entstand sofort ein volumi nöser Niederschlag.
Nach einstündigem Ste- len bei Raumtemperatur wurde das feste Salz abfiltriert. Die Umkristallisation aus 45 nil absolutem Alkohol ergab das kristalline quaternäre Salz vom Schmelzpunkt 193 bis 195 C.
N-Benzhydryl-tropylamin-methyljodid Die freie Base wurde aus dem Oxalat her gestellt. Durch Zusatz von Methyljodid zu einer ätherischen Lösung der Base erhielt man das schwerlösliche N-Benzhydryl-tropylamin- methyljodid vom Schmelzpunkt 253-258 C.
N-Benzhydryl-tropylamin-hemihydrat Eine wässrige Lösung von N-Benzhydryl- tropylamin-bioxalat wurde mit Natrium Lydroxyd basisch gemacht und das freie N- ssenzhydryl-tropylamin mit Äther extrahiert. Die ätherische Lösung wurde getrocknet, dann liess man den Äther auf einem Uhrglas ver dampfen. Das verbleibende Öl kristallisierte beim Reiben. Es wurde aus Petroläther um kristallisiert. Die Kristalle ei-weichten bei 58 C und schmolzen bei 62-64 C.
Durch Trocknung im Vakuum verlor die kristalline Substanz das Hydratwasser, und der feste Körper verwandelte sich in ein Öl.
N-Benzhydryl-tropylamin-methylnitrat Zu 1,2 g N-Benzhydryl-tropylamin in 3 ml Äther wurden 1,5 m1 Methylnitrat zugesetzt. Es entstand zunächst eine klare Lösung. Nach 5 Minuten setzte die Kristallisation ein. Man liess die Mischung über Nacht bei Raumtem peratur stehen, und dann wurde der- feste Körper durch Filtration abgetrennt. Die Um kristallisierung erfolgte aus 7 ml absolutem Äthanol, dem 1 ml Äther zugesetzt war. Eine nochmalige Kristallisation aus 15 ml absolu tem Äthanol unter Zusatz von 1 ml Äther er gab das reine quaternäre Salz mit . dein. Schmelzpunkt 203-204 C.
N-Benzhydryl-tropylamin-dihydrobromid Zu 200 g N-Benzhydryl-tropylamin in 10 ml Äther wurde eine Lösung von Bromwas serstoff in absolutem Äthanol hinzugesetzt. Es entstand sofort ein Niederschlag. Das feste Dihydrobramid wurde aus 15 ml absolutem Äthanol umkristallisiert -und ergab einen Schmelzpunkt von 252-254 C.
N-Benzhydryl-tropylamin-äthyljodid Zu 200 mg N-Benzhydryl-tropylamin, in <B>10</B> ml Äther wurden 0,5 ml Äthyljodid zuge setzt. Die Lösung wurde dann am Dampfbad zur Trockne eingedampft. Der Rüekstand wurde zweimal aus absolutem Äthanol unter Zusatz von.Äther umkristallisiert, Der Schmelz- Punkt des entstandenen Salzes betrug 249 bis 250 C.
Beispiel <I>3</I> N-(4-Chloa--beaizhydryl)-tropylanziiz-oxalat Eine Mischung aus 16 g Tropinon, 25,4 g 4-Chlor-benzhydrylamin-hydrochlorid, 200 ml Äthanol und 0,5 g Platinoxyd wurde bei Raumtemperatur und 2,8 kg/cm2 Druck hy driert. Nach 21 Stunden war die theoretische Menge Wasserstoff absorbiert. Der Katalysa tor wurde durch Filtration entfernt und das Lösungsmittel im Vakuum verdampft. Der Rückstand wurde in 75 ml 2,5n-Salzsäure ge löst.
Nach 4 Stunden setzte die Kristallisation ein. Der feste Körper wurde abfiltriert. Er wog 16 g und bestand hauptsächlich aus un verändertem 4-Chlor-benzhydrylamin-hydro- chlorid. Das Filtrat wurde mit Natrium hydroxyd basisch gemacht und dann mehr mals mit Äther extrahiert. Die vereinigten Extrakte wurden mit Wasser gewaschen und getrocknet. Die trockene Ätherlösung (etwa 200 ml) wurde daraufhin einer Lösung von 7 g Oxalsäure in 50 ml Äthanol zugefügt. Ein gummiartiger Niederschlag entstand.
Nach dem Abgiessen der darüber stehenden Flüssig keit und dem Waschen des Rückstandes mit Äther wurde der Niederschlag fest. Der feste Körper wurde in 40 ml absolutem Äthanol gelöst, dann wurden 5 ml wasserfreier Äther hinzugesetzt. Nach 24 Stunden Stehen bei Raumtemperatur entfernte man den kristal linen Niederschlag, der aus N-(4-Chlor-benz- liydryl)-tropylamin-bioxalat bestand. Nach dem Uinkristallisieren aus absolutem Äthanol zeigte das Produkt einen Schmelzpunkt von 102-104 C.
N- (4-Chlor-benzhydryl) -tropylamin- dimethylsulfat Eine Lösung von 3 g N-(4-Chlor-benz- liydryl)-tropylamin-bioxalat in Wasser wurde durch Zusatz von Natriumhydroxyd alkalisch gemacht. Die Mischung wurde mit Äther ex trahiert, der Äther mit Wasser gewaschen und getroeknet. Man fügte nun DimethyIsul- fat hinzu. Es entstand eine ölige Abscheidung, welche beim Kratzen fest. wurde.
Der feste Körper (2 g) wurde abfiltriert und ergab beim Umkristallisieren aus Alkohol-Äther das N-(4-Chlor-benzhydryl)-tropylamin-dimethyl- sulfat vom Schmelzpunkt 193-l.94 C.
<I>Beispiel 4</I> ly-('4-1llethyl-beiazhydryZ)-tropylrxm,in- hydroclalorict Eine Mischung von 16 g (0,1.15 M) Tro.- pinon und 23,4 g (0,10 M) Phenyl-p-tolyl- methylamin-hydroehlorid wurde in 200 ml Äthanol gelöst. Die Lösung wurde 20 Stunden lang bei Raumtemperatur mit Wasserstoff in Gegenwart von 0,5 g Platinoxyd hydriert. Der berechnete Wasserstoffdruckabfall betrug 11,9 kg, der beobachtete Abfall war<B>12,7</B> kg.
Der Katalysator wurde abfiltriert und das Äthanol im Vakuum entfernt. Zu dem Rück stand fügte man 60 ml 2,5n-Salzsäiire, welche die Bildung eines Niederschlages hervorrief. Der feste Körper wurde gesammelt. Er wog 17,7 g, zeigte einen Schmelzpunkt von 240 C (Zersetzung) und bestand im wesentlichen aus Phenyl-p-tolyl-methylamin-hydroehlorid.
Beim Stehenlassen des Filtrats bildete sich eine ölige Ausscheidung, die allmählich fest wurde. Der feste Körper wurde dreimal mit Äther extrahiert, um alles vorhandene Tro- pinon oder Tropin zit entfernen. Der Rück stand wurde mehrere Male aus Wasser um kristallisiert und @eirgab das Hydrat von N-(4- Methyl - benzhydryl) - tropylamin-hydrochlorid mit dem Schmelzpunkt l75-177 C.
Durch Umkristallisieren aus einer Mischung von Äthanol und Äthyläther wurde ein wasser freies Produkt erhalten.
