La présente invention concerne un procède de fabri cation de nouvelles N-aryloxyalcoyl-N-aralcoxyamines de formule :

Les nouvelles N-aryloxyalcoyl-N-aralcoxyamines peuvent servir d'intermédiaires de synthèse et sont efficaces aussi comme ingrédients actifs dans des compositions
servant à abaisser le taux de cholestérol dans le sang
d'animaux à sang chaud et dans le sang d'êtres humains.
Les N-aryloxyalcoyl-N-aralcoxyamines obtenues peu
vent être soumises à une hydrolyse alcaline pour rempla
cer le radical Rt-O-CO-par un atome d'hydrogène.
Les exemples suivants illustrent l'invention. Les nu
méros servant à identifier les composés correspondent
aux numéros utilisés au tableau B, donné dans la suite
de cet exposé.
Exemple 1
Fabrication du chlorhydrate de N-gamma-phénoxy-
propyl-N-m-méthylbenzyloxyamine (composé 4) :
a) On mélange en agitant à 250 C 38 g de N-hy
droxyuréthane dans 100 ml d'éthanol à 95 /o avec une
solution de 36 g d'hydroxyde de potassium dans 200 ml
d'méthanol à 95 /o. On ajoute en une fois 100 g d'alpha
bromo-méta-xylène et l'on maintient la température à
300 C environ en utilisant un refroidissement externe. On agite le mélange durant 16 heures et l'on sépare par filtration l'hydroxyde de potassium. On concentre le filtrat sous pression réduite et on le dissout dans l'éther.
On extrait à fond la solution éthérée à l'aide d'une solution aqueuse froide à 10 ouzo d'hydroxyde de sodium. On sèche 1'extrait éthéré, on en élimine le solvant par distillation, et l'on fractionne le résidu sous pression réduite.
On obtient 17g (22 /o) de carbéthoxyhydroxamate de méta-méthylbenzyle pur, distillant à 1000-103 C à 0, 05 mm.
b) A une solution d'éthylate de sodium préparée à partir de 2, 3 g de sodium métallique et de 220 ml d'étha- nol anhydre, on ajoute 20, 9 g de carbéthoxyhydroxamate de métha-méthylbenzyle obtenu comme décrit ci-dessus et 21, 5 g d'éther de phényle et de gamma-bromopropyle, wt l'on porte le mélange au reflux durant quatre heures.
c) On ajoute une solution de 8, 8 g d'hydroxyde de sodium dans 100 ml d'eau au mélange réactionnel et l'on poursuit le reflux durant deux heures de plus.
On chasse l'éthanol par distillation à la pression atmosphérique jusqu'à ce que la température de la colonne à distiller atteigne 870 C ; on refroidit le résidu et on 1'extrait avec 350 ml d'éther. On lave à 1'eau 1'extrait éthéré jusqu'à neutralité et on le sèche sur sulfate de sodium. On sépare l'éther et l'on distille le résidu sous pression réduite. La fraction bouillant à environ 1540C/0, 2mm consiste en
N-gamma-phénoxy-propyl-N-méta-méthylbenzyloxyami- ne.
La quantité obtenue est de 16, 6 g.
c) On fabrique le chlorhydrate du composé cidessus en combinant une solution de ce composé dans l'éther anhydre avec une quantité équivalente d'acide chlorhydrique éthanolique. La recristallisation du préci- pité résultant dans un mélange éthanol-éther donne le chlorhydrate voulu, qui fond à 840-860 C.
Exemple 2
Fabrication du N-bêta-phénoxyéthyl-carbéthoxyhy- droxamate de benzyle (compose 9) :
A une solution d'éthylate de sodium obtenue à partir de 5, 75 g de sodium métallique et 200 ml d'éthanol anhydre, on ajoute 48, 8 g de carbéthoxyhydroxamate de benzyle, obtenu en utilisant le processus général décrit dans l'exemple (la), et 50, 3 g de bêta-bromophénétole.
On chauffe ensuite le mélange au reflux durant huit heures. On ajuste le pH du mélange réactionnel à 7 environ au moyen d'acide chlorhydrique, et l'on élimine l'éthanol par distillation jusqu'à ce que la température du récipient atteigne 850 C. Après son refroidissement, on dilue le résidu avec 100ml d'eau et l'on extrait le mélange aqueux avec 300ml d'éther. On lave 1'extrait éthéré successivement avec une solution diluée d'hydrooxyde de sodium, de l'acide chlorhydrique dilué et puis à 1'eau jusqu'à neutralité. On sèche 1'extrait éthéré sur sulfate de sodium et l'on sépare l'éther. Le fractionnement du résidu sous pression réduite donne 61, 1 g du produit voulu, qui bout à 162 C/0, 1 mm.
Exemple 3
Fabrication du chlorhydrate de N-bêta-phénoxyéthyl-
N-benzyloxyamine (composé 1) :
On porte au reflux durant deux heures une solution de 43, 5 g de N-bêta-phénoxyéthyl-carbéthoxyhydroxa- mate de benzyle, obtenu comme décrit dans l'exemple 2, et de 11, 9g d'hydroxyde de sodium dans 120ml d'étha- nol aqueux à 50 /o. On chasse ensuite l'éthanol par distillation à la pression atmosphérique jusqu'à ce que la température de la colonne de distillation atteigne 87 C.
On refroidit le résidu, on le dilue avec 50 ml d'eau et on l'extrait avec 250 ml d'éther. On lave 1'extrait éthéré à 1'eau jusqu'à neutralité et on le sèche sur sulfate de sodium. On ajoute un excès d'acide chlorhydrique éthéré et l'on recueille par filtration le précipité résultant. La quantité de produit voulu obtenue est de 22, 5 g ; point de fusion : 121-122 C.
Exemple 4
Fabrication du chlorhydrate de N-bêta-(2, 4-dichlorophénoxy)-éthyl-N-gamma-phényl-propoxyamine (composé 8) :
a) A une solution d'éthylate de sodium obtenue à partir de 1, 35 g de sodium métallique et 100 ml d'étha- nol, on ajoute 13, 1 g de carbéthoxyhydroxamate de gamma-phénylpropyle, obtenu selon le processus décrit dans l'exemple (la), et 15, 8g d'éther de bêta-bromoéthyle et de 2, 4-dichlorophényle. On porte le mélange réactionnel au reflux pendant une période de huit heures.
b) On ajoute une solution de 5, 2 g d'hydroxyde de sodium dans 100 ml d'eau. On continue à maintenir au reflux pendant quatre heures de plus.
On sépare l'étha- nol par distillation et l'on extrait le résidu à l'éther. On lave 1'extrait éthéré à 1'eau jusqu'à neutralité et on le sèche ensuite sur sulfate de sodium. On fait passer de l'acide chlorhydrique dans la solution jusqu'à ce qu'il ne se produise plus de précipitation. On recueille le préci- pite par filtration et on le fait recristalliser dans l'éthanol pour obtenir 8, 8 g du produit voulu, dont le point de fusion est de 147-148 C.

Example 1
Manufacture of N-gamma-phenoxy- hydrochloride
propyl-N-m-methylbenzyloxyamine (compound 4):
a) Mixing with stirring at 250 C 38 g of N-hy
droxyurethane in 100 ml of 95 / o ethanol with a
solution of 36 g of potassium hydroxide in 200 ml
95% methanol. 100 g of alpha are added all at once
bromo-meta-xylene and the temperature is maintained at
300 C approximately using external cooling. The mixture is stirred for 16 hours and the potassium hydroxide is filtered off. The filtrate is concentrated under reduced pressure and dissolved in ether.
The ethereal solution was extracted thoroughly with a cold 10 oz aqueous solution of sodium hydroxide. The ethereal extract is dried, the solvent is removed therefrom by distillation, and the residue is fractionated under reduced pressure.
17 g (22 / o) of pure meta-methylbenzyl carbethoxyhydroxamate are obtained, distilling at 1000-103 C at 0.05 mm.
b) To a solution of sodium ethoxide prepared from 2.3 g of metallic sodium and 220 ml of anhydrous ethanol is added 20.9 g of metha-methylbenzyl carbethoxyhydroxamate obtained as described above and 21.5 g of phenyl and gamma-bromopropyl ether, the mixture being refluxed for four hours.
c) A solution of 8.8 g of sodium hydroxide in 100 ml of water is added to the reaction mixture and the reflux is continued for two more hours.
Ethanol is removed by distillation at atmospheric pressure until the temperature of the distillation column reaches 870 ° C .; the residue is cooled and extracted with 350 ml of ether. The ethereal extract is washed with water until neutral and dried over sodium sulfate. The ether is separated off and the residue is distilled off under reduced pressure. The fraction boiling at about 1540C / 0.2mm consists of
N-gamma-phenoxy-propyl-N-meta-methylbenzyloxyamine.
The amount obtained is 16.6 g.
c) The hydrochloride of the above compound is made by combining a solution of this compound in anhydrous ether with an equivalent amount of ethanolic hydrochloric acid. Recrystallization of the resulting precipitate from ethanol-ether gives the desired hydrochloride, which melts at 840-860 ° C.
Example 2
Manufacture of benzyl N-beta-phenoxyethyl-carbethoxyhy-droxamate (compound 9):
To a sodium ethoxide solution obtained from 5.75 g of metallic sodium and 200 ml of anhydrous ethanol is added 48.8 g of benzyl carbethoxyhydroxamate, obtained using the general process described in Example ( la), and 50.3 g of beta-bromophenetole.
The mixture is then heated under reflux for eight hours. The pH of the reaction mixture is adjusted to approximately 7 by means of hydrochloric acid, and the ethanol is removed by distillation until the temperature of the vessel reaches 850 C. After cooling, the residue is diluted with 100 ml. of water and the aqueous mixture is extracted with 300 ml of ether. The ethereal extract is washed successively with dilute sodium hydroxide solution, dilute hydrochloric acid and then with water until neutral. The ethereal extract is dried over sodium sulfate and the ether is separated. Fractionation of the residue under reduced pressure gives 61.1 g of the desired product, which boils at 162 C / 0.1 mm.
Example 3
Manufacture of N-beta-phenoxyethyl- hydrochloride
N-Benzyloxyamine (compound 1):
A solution of 43.5 g of benzyl N-beta-phenoxyethyl-carbethoxyhydroxamate, obtained as described in Example 2, and 11.9 g of sodium hydroxide in 120 ml of sodium hydroxide is refluxed for two hours. 50% aqueous ethanol. Ethanol is then removed by distillation at atmospheric pressure until the temperature of the distillation column reaches 87 ° C.
The residue is cooled, diluted with 50 ml of water and extracted with 250 ml of ether. The ethereal extract is washed with water until neutral and dried over sodium sulfate. An excess of ethereal hydrochloric acid is added and the resulting precipitate is collected by filtration. The desired amount of product obtained is 22.5 g; melting point: 121-122 C.
Example 4
Manufacture of N-beta (2,4-dichlorophenoxy) -ethyl-N-gamma-phenyl-propoxyamine hydrochloride (compound 8):
a) To a solution of sodium ethoxide obtained from 1.35 g of metallic sodium and 100 ml of ethanol, 13.1 g of gamma-phenylpropyl carbethoxyhydroxamate, obtained according to the process described in l Example (la), and 15.8g of beta-bromoethyl ether of 2, 4-dichlorophenyl. The reaction mixture is refluxed for a period of eight hours.
b) A solution of 5.2 g of sodium hydroxide in 100 ml of water is added. Maintain at reflux for four more hours.
The ethanol is separated off by distillation and the residue is extracted with ether. The ethereal extract is washed with water until neutral and then dried over sodium sulfate. Hydrochloric acid is passed through the solution until no further precipitation occurs. The precipitate is collected by filtration and recrystallized from ethanol to give 8.8 g of the desired product, which has a melting point of 147-148 C.
