EMI0001.0046
où X et R répondent aux définitions données ci-dessus ;
b) avec un; dérivé S-alcoylé ou S-aralcoylé
EMI0002.0005
EMI0002.0022
Dans le cas de la réaction (a), on observe un départ de SH2 ou bien la formation d'un sulfhydrate d'ammo nium ou de l'amine mise en jeu, si l'on a utilisé un excès de ces derniers ;

On chauffe à reflux, pendant 2 heures,

Process for the preparation of new substituted pyrrolobenzothiazines The present invention relates to a process for the preparation of pyrrolbbbenzothiazines corresponding to the general formula
EMI0001.0011
in which the symbol X represents a hydrogen atom, an alkyl or hydroxyalkyl radical,
mono- or dialkyl @ aminoalkyl, pvperidinoalkyl, aralkyl or aryl.
According to the invention, a 3-thiono compound of formula II or a corresponding 3-alkyl-thio- or aralkyl-thio compound of formula III is reacted.
EMI0001.0031
where R and R 'represent an alkyl or aralkyl radical,
R may also represent hydrogen, with ammonia or a primary amine XNH2, according to one of the following reactions: a) with an ester (3-thiono-1,4-benzothiazine-2-yl) -acetic
EMI0001.0046
where X and R meet the definitions given above;
b) with a; S-alkyl or S-aralkyl derivative
EMI0002.0005
In reactions (a) and (b), the product corresponding to one of the tautomeric formulas (V) is generally formed intermediately, which cyclizes, during the reaction,
giving a product of formula I
EMI0002.0022
Preferably, the invention is carried out by using equimolecular amounts of the two reactants and by operating at a temperature which depends essentially on the amino derivative XNIiz involved.
For the most reactive compounds, such as ammonia and aliphatic or arylaliphatic amines, an can operate in methanol, ethanol or propanol at reflux. For less reactive aromatic amines, the mixture is heated to a temperature above 100, most often without solvent.
In the case of reaction (a), there is a departure of SH2 or the formation of an ammonium hydrosulphide or of the amine involved, if an excess of the latter has been used;
the reaction is absolutely not hampered by the addition, to the reaction mixture, of a compound capable of combining with SH2, such as lead acetate for example. In the case of reaction (b), the departure of the mercaptan R'-SH is observed.
In the case where X denotes hydrogen, it is possible to replace the hydrogen in position 1 of the compound obtained of formula I by a radical conforming to the definition of X by reacting an organometallic derivative of said compound with a reactive ester of an alcohol of formula X'-OH in
in which X 'represents the radical to be introduced.
The organo-metallic derivative of the compound corresponding to formula I can be obtained by forming it in situ with an alcoholate or an amide providing the metal and then reacting with the alkyl or aralkyl ester without prior isolation. .
To obtain the starting compounds II, one can react P2S5 at mflüx, in benzene, toluene, etc., with esters (1,4-benzothiazine-3 one-2-yl) - acetic type VI,
according to the reaction
EMI0002.0114
where R is in particular an alkyl radical, for example methyl or ethyl. These compounds of formula VI have been described in. the Bulletin of the Chemical Society of France (1962) p. 1709.
Such an ester VI, formed for example from ro-amino-benzene4hiol, can also be treated directly with P.S. without having been isolated.
To obtain the S-alkyl or S-aralkyl compounds (III), it is possible to react a halide or another alkyl or aralkyl ester, for example a sulfate, with an organometallic derivative of a compound (II), of preferably the soda derivative,
according to the equation
EMI0002.0150
where R and R 'correspond to the definition given above, R'Z being an alkylating agent such as alkyl or aralkyl halide or sulfate.
Finally, to prepare the acids corresponding to esters II and III, these esters can be saponified by means of a strong mineral base, preferably in an alcoholic medium.
Example 1 A. The preparation of methyl 2,3-dihydro-benzo-1,4-thiazine, 3-thiono-2-yl) acetate (starting material) can be carried out as follows
EMI0003.0022
a) A solution formed from 80 g (0.34 mol) of (2,3-dihydro-benzo- 1, is heated to reflux (on an oil bath at 1300) and while stirring for 30 minutes, 4-thiazin-3-one-2-yl)

by operating gradually, first over an hour from 1000 to 1600, then for 30 minutes at 160o, by distilling the methanol formed by means of a fractionation column. 1000 cm3 of toluene are then added to the mixture and approximately 100 cm3 is distilled from it in order to entrain the rest of the methanol.
A toluene solution of methyl (2,3-dihydro-benzo-1,4-thiazine-3-one-2-yl) acetate is thus obtained, which is treated as above with reflux and with stirring, with 222 g (1 mole) d e P2S5, by operations already. described under.

EMI0007.0001
Heated to reflux for 2 hours,
a solution of 10.1 g (0.04 mole) of (2,3-dihydro-3-thiono-1,4-benzothiazin-2-yl) methyl acetate and 4.5 g (0.044 mole) of ( 2-dimethyl-amino-propyl) -amine in 30 ml of absolute alcohol. After having removed the solvent under vacuum, the hydrochloride of the oily base obtained is formed by slow addition of a solution of HCl in ethyl alcohol, until slightly acidic pH: the hydrochloride obtained crystallizes; it is filtered off, washed with absolute alcohol and dried to yield 7.0 g (yield 54 0/0).
By recrystallization from 50 ml of methanol, 4.5 g (301% yield) of pure hydrochloride are obtained. Melting point = 2620 C (decomposition).
(2-Dimethylamino-propyl) -amine can be obtained in a yield of 62 fl / o by reduction of 2-dimethylamino-propionitrile using LIAIH4 in ether. The base, a colorless liquid, boils at 127 ° under 760 mm of mercury, its p-nitro benzamide (hydrochloride) melts at 1770 and its phthalimide at 570.
Other salts. that the hydrochlorides can be prepared as follows 1H, 3aH-dihydro-2,3- (diethylamino-2-ethyl) - 1-oxo-2-pyrrolo (3,2-b) -benzothiazine-4,9 acid fumarate
EMI0007.0039
To an aqueous solution of 0.68 g of the hydrochloride prepared according to Example 5,
50% excess of CO 3 K, 2 M is added: the base which precipitates is extracted with ether, the ethereal solution dried over SO 4 N% and dis tilled ether, which gives the oily base in quantitative yield.
The base is dissolved in 5 cm3 of acetone and the solution of 0.29 g of fumaric acid (25% excess over the theoretical quantity) is added: by trituration, the salt formed crystallizes: it is obtained. 0.70g. This salt is recrystallized from a mixture of acetone and ethanol; 0.55 g are collected; melting point 144o.
The pyrrolobenzothiazines prepared according to the invention can be used in the pharmaceutical industry, certain more especially as hypotensive drugs and as local anesthetics.
