sowie von Säureadditionssalzen davon.
In Formel I bedeutet R eine Alkylengruppe mit vorzugsweise höch stens 5 C-Atomen, R, ist Wasserstoff oder Alkyl, und R2 und Ra sind gleich oder verschieden und stellen Wasserstoff, Alkyl oder gemeinsam eine Äthylengruppe dar, in welch letzterem Falle R1 neben den genannten Bedeutungen auch die Bedeutung von Hydroxyalkyl oder Alkoxyalkyl haben kann.
R4 und R5 sind gleich oder verschieden und bedeuten Wasserstoff oder Halo gen. Vorzugsweise ist der basische Substituent in 6- Stellung eine 4-Methyl-l-piperazinyl-Gruppe. Allfällige Substituenten in den Benzolkernen befinden sich vor zugsweise in 3- oder 8-Stellung.
Verbindungen gemäss Formel I werden erfindungs gemäss erhalten, indem man Amine der Formel:

und wobei die Reaktionsprodukte in Form der freien Basen oder von Säureadditionssalzen gewonnen werden. worin R4 und R5 die oben genannten Bedeutungen haben, in die Thiolactame, gewünschtenfalls unter nach folgender Alkylierung der letzteren, oder durch Um- setzen der Lactame (VI) mit einem Halogenierungs- mittel, wie Phosphoroxychlorid oder Phosphorpenta- chlorid,
vorzugsweise in Gegenwart katalytischer Mengen von Dimethylanilin oder Dimethylformamid. Die Lac- tame (VI) sind ihrerseits z. B. durch Ringschluss ent sprechender o-Isocyanatodiphenylmethane mit Alu miniumchlorid erhältlich.
Die in der beschriebenen Weise erhaltenen Basen sind in den meisten Fällen kristallisierbar, sonst im Hochvakuum unzersetzt destillierbar, und bilden mit anorganischen und organischen Säuren, beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Sal petersäure, Phosphorsäure, Essigsäure, Oxalsäure, Wein säure, Toluolsülfonsäure und dergleichen, in Wasser beständige Additionssalze, in welcher Form die Pro dukte ebenfalls verwendet werden können.
Die in der beschriebenen Weise erhaltenen Basen und ihre Säureadditionssalze sind neue Verbindungen, die als Wirkstoffe in Arzneimitteln oder als Zwischen produkte zur Herstellung von solchen Verwendung fin den. Insbesondere fallen die Produkte als Neurople- gika, Neuroleptika und Analgetika in Betracht. Einzelne davon eignen sich zur Behandlung psychotischer Zu stände.
Diese Wirksamkeit äussert sich pharmakologisch in starker Motilitätsdämpfung bei Mäusen, die mit kata- leptischer Wirkung einhergehen kann. Die Motilitäts- dämpfung wird durch Messung der Laufaktivität nach der Methode von Caviezel und Baillod [Pharm. Acta Helv. 33, 469 (1958)] erfasst.
Die Laufaktivität einiger erfindungsgemässer Produkte sowie deren Toxizität wer den in der folgenden Tabelle I mit den entsprechenden Zahlen für Chlorpromazin verglichen.

Unter fortgesetztem Rückfluss wird innert 30 Minuten eine Lösung von 8,0 g Methyl-bis-f-chloräthylamin in 25 ml absolutem Dioxan zugetropft. Nach weiteren 15 Stunden Rückfhiss wird im Vakuum zur Trockne eingeengt und der Rückstand zwischen Äther und Wasser verteilt. Nach zweimaligem Waschen der Atherphase mit Wasser werden die ba- sischen Anteile durch erschöpfende Extraktion mit ver dünnter Essigsäure abgetrennt.
Der essigsaure Auszug wird mit Ammoniakwasser versetzt und dann mit Äther extrahiert. Die ätherische Lösung wird mit Wasser gewaschen, über Natriumsulfat getrocknet, durch Ton erde filtriert und eingedampft. Der Rückstand wird aus Äther/Petroläther zur Kristallisation gebracht und aus Aceton/Petroläther umkristallisiert. Man erhält 7,1 g (49 % der Theorie) 6-(4-Methyl-l-piperazinyl)-morphan- thridin vom Schmelzpunkt 137,5-138,5 C.
In analoger Weise wie im vorerwähnten Beispiel erhält man aus entsprechenden Ausgangsstoffen die in der nachfolgenden Tabelle II genannten Produkte. Darin haben R, R1, R2, R8, R4 und R5 die früher angegebene Bedeutung.
In der rechten Kolonne bedeutet Ae Aceton, Ä Äther, Ch Chloroform, Me Methanol und Pe Petrol- äther.

With continued reflux, a solution of 8.0 g of methyl-bis-f-chloroethylamine in 25 ml of absolute dioxane is added dropwise over a period of 30 minutes. After a further 15 hours of reflux, the mixture is concentrated to dryness in vacuo and the residue is partitioned between ether and water. After the ether phase has been washed twice with water, the basic components are separated off by exhaustive extraction with dilute acetic acid.
The acetic acid extract is mixed with ammonia water and then extracted with ether. The ethereal solution is washed with water, dried over sodium sulfate, filtered through clay and evaporated. The residue is crystallized from ether / petroleum ether and recrystallized from acetone / petroleum ether. 7.1 g (49% of theory) 6- (4-methyl-1-piperazinyl) -morphanthridine with a melting point of 137.5-138.5 ° C. are obtained.
