Verfahren zur Herstellung neuer Diphenlalkanverbindungen
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Diphenylalkanverbindungen der-Formel

worin jede der Gruppen Ri und R2 einen aliphatischen, cycloaliphatischen oder cycloaliphatisch-aliphatischen Kohlenwasserstoffrest bedeuten, Am eine tertiäre Aminogruppe darstellt und die Gruppe der Formel - (C"H2n)-einen Alkylenrest, der die Gruppe Am vom Sauerstoffatom durch mindestens 2 Kohlenstoffatome trennt und worin n eine ganze Zahl von 2-7 bedeutet, darstellt, und worin jede der Gruppen Rg und R4 Wasserstoff, Alkyl-mit 1-7 Kohlenstoffatomen,
Alkoxymit 1-7 Kohlenstoffätomen oder Trifluormethylreste, Halogenatome oder Gruppen der Formel -O-(CnH2n)-Am, worin Am und die Gruppe der Formel -(CnH2n)- die oben genannnten Bedeutungen haben, darstellt, und R5 Wasserstoff, einen oder mehrere Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste oder Halogenatome oder einen, gegebenenfalls ein oder mehrere Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste oder Halogenatome enthaltenden Phenylrest darstellt, und mindestens einer der Reste R3, R4 und R5 einen der von Wasserstoff verschiedenen Reste bedeutet, sowie ihren Salzen.
Der Rest R ; kann mit Ausnahme des Arylrestes einen oder mehrere gleiche oder verschiedene Substituenten der oben genannten Art bedeuten, wobei diese Substituenten sich in beliebiger Stellung des Phenylkerns befinden können. Alkylreste mit 1-7 Kohlenstoffatomen sind z. B. Methyl-, Athyl-, n-Propyl-, Isopropyl-, n Butyl-oder tert.-Butylgruppen. Alkoxygruppen mit 1-7 Kohlenstoffatomen sind vor allem Methoxy-, ¯thoxy-, n-Propyloxy-, Isopropyloxy-, n-Butyloxy-oder sek.- Butyloxygruppen. Halogenatome sind-Fluor, Chlor ; Brom oderJod.
Die aliphatischen Kohlenwasserstoffreste Rl und Ra sind vor allem Alkylreste mit 1-7, vorzugsweise mit 1-4 Kohlenstoffatomen, z. B. Methyl-, Athyl-, n Propyl-, Isopropyl-, n-Butyl-oder-rsobutylgruppen.
Als cycloaliphatische Kohlenwasserstoffreste Ri und-R, kann man vor allem Cycloalkylreste mit 3-8 Ringkoh- lenstoffatomen, wie Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl, Cycloheptyl oder Cyclooctyl, und als cycloaliphatisch-aliphatische Kohlenwasserstoff- reste vor allem Cycloalkyl-niederalkylreste mit 3-8 Ringkohlenstoffatomen, wie Cyclopropylmethyl, Cycle ; pentylmethyl, 2-CyclopentylÏthyl, Cyclohexylmethyl, I-Cyclohexyläthyl oder Cycloheptylmethyl, erwähnen.
Die tertiäre Aminogruppe Am ist vorzugsweise eine Dialkylaminogruppe, worin Alkyl 1-7, vor allem 1-4 Kohlenstoffatome-hat, besonders die-Diäthylamino-, Dimethylamino-, Methyl-athylamino-, Di-n-propyl- amino oder Di-isopropyl-aminogruppe, oder eine N CycloaLkyl-N-alkylaminogruppe, worin Cycloalkyl 3-8, vor allem 5-7 Ringkohlenstoffatome aufweist, und Alkyl vorzugsweise 1-4 Kohlenstoffatome besitzt, wie die N-Cyclopentyl-N-methylaminogruppe, die N-Cyclohexyl-N-methylaminogruppe oder die N-Cyclohexyl-N äthylaminogruppe. Weiter kann die tert.
Aminogruppe Am eine Alkylenaminogruppe mit 4-8 Kohlenstoffato- men, eine Azaalkylenaminogruppe, worin Alkylen 4-6 Kohlenstoffatome besitzt, oder eine Oxaalkylen-oder Thiaalkylenaminogruppe, worin Alkylen vorzugsweise 4 Kohlenstoffatome aufweist, darstellen. Zusammen mit dem Stickstoffatom kann daher eine solche Alkylen-, Azaalkylen-, Oxaalkylen-oder Thiaalkylengruppe z.
B. eine Pyrrolidinogruppe, wie Pyrrolidino oder 2-Methyl- pyrrolidino, eine Piperidinogruppe, wie Piperidino, 2 Methylpiperidino oder 4-Methylpiperidino, oder eine 1, 6-Hexylenamino- oder 1, 7-Heptylenaminogruppe, sowie eine Azaalkylenaminogruppe, wie besonders N Alkyl-oder Oxyalkyl-azaalkylenaminogruppe, worin Alkylen 4-6 Kohlenstoffatome aufweist, wie Piperazino, 4-Methyl-oder-äthylpiperazino, 4-Hydroxyäthyl-piper- azin.
o, 3-Aza.-1, 6-hexylenanino, 3-Aza 3-methyl-1, 6hexylenamino, 4-Aza-1, 7-heptylenamino, 4-Aza-4-meth yl-1, 7-heptylenamino, oder eine Morpholinogruppe, wie Morpholino oder 3-Methylmorpholino, oder eine Thiamarpbolinogruppe, wiss Thiamo'rphoimo,. darstelten.
Der Alkylenrest der Formel- (CnH2n)- besitzt vor allem 2-3 Kohlenstoffatome und stellt deshalb vor allem einen 1, 2-Äthylen-, 1-Methyl-1, 2-äthylen-, 2 Methyl-1, 2-äthylen-oder 1, 3-Propylenrest dar. Er kann aber auch 1, 3-Butylen, 2, 3-Butylen, 3, 4-Butylen, 1, 4 Butylen, 1, 4-Pentylen, 1, 5-Pentylen, 1, 5-Hexylen, 1, 6 Hexylen oder 1, 7-Heptylen darsteAen.
Sofern die Reste R3 und R4 niedere Alkyl-oder Alkoxygruppen oder Halogenatome bedeuten, so kom- men vor allem die eingangs für R. genannten Reste dieser Art in Frage. Falls Rg und/oder R4 einen Rest der Formel-0- (CnH2n)-Am bedeutet, so haben darin Am und n besonders die oben hervorgehobenen Be- deutungen.
Die neuen Verbindungen besitzen wertvolle Anti fungus-Eigenschaften und können dementsprechend als Heil-oder Desinfektionsmittel verwendet werden. So zeigen sie eine Wirkun gegen Pilze, die oberflächliche Dermatophytosen hervorrufen, wie Trichophyton mentagrophytes, Trichophyton gallinae, Trichophyton interdigitale, Microsporum audouini, Microsporum canis, Microsporum gypseum, und können damit z. B. bei den entsprechenden Erkrankungen, wie Dermatophytosis, angewendet werden. Sie wirken auch gegen solche Pilze, welche chronische Infektionen der Haut und der darunterliegenden Gewebe verursachen, wie z. B.
Sporo- trichium schenkii, oder gegen Pilze, welche tiefer sitzende systemische Mykosen verursachen, wie Candida albicans, Cryptococcus neoformans, Histoplasma capsulatum, und können dementsprechend angewendet werden. Die neuen Verbindungen sind auch wirksam gegen Actinomyceten wie Nocardia asteroides, gegen grampositive Bakterien, wie Diplococcus pneumoniae, Staphylococcus aureus, oder gramnegative Bakterien, wie Escherichia coli, Pseudomonas aeruginosa, und gegen Protozoen, wie Trichomonas vaginalis, und können entsprechend in der Heilkunst angewendet werden.
Die neuen Verbindungen wirken auch gegen Parasiten, wie z. B. Bandw rmer. Ausserdem zeigen sie gewisse stimulierende und endocrine, z. B. ¯strogene, Effekte.
Besonders wertvoll sind Verbindungen der Formel

ein oder mehrere Alkylreste mit 1-7 Kohlenstoffatomen, besonders Methylreste, Halogen, besonders Chlor, ein unsubstituierter Phenylrest, ein Alkylphenylrest, worin Alkyl 1-7 Kohlenstoffatome enthält, besonders Methylphenylrest, oder ein Halogen-, besonders Chlorphenylrest, ist, wobei mindestens eine der Gruppen R3', R4' oder R5'einen der von Wasserstoff verschiedenen Reste darstellt, sowie ihre Salze, vor allem die pharmazeutisch verwendbaren Säureadditionssalze.
Als eine Gruppe von besonders wertvollen Verbindungen können auch die Verbindungen der Formel

Erfindungsgemäss werden die oben genannten Verbindungen erhalten, indem man eine Verbindung der Formel

worin R. i, Rg, Rg und die Gruppe der Formel l)- die oben gegebenen Bedeutungen haben, wobei letztere X und O durch mindestens 2 Kohlenstoffatome trennt, und jede der Gruppen R6 und R7 f r Wasserstoff, Alkylmit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste, Halogenatome oder Gruppen der Formel -O-(CnH2n)-X steht, und X eine reaktionsfähige veresterte Hydroxygruppe darstellt, wobei mindestens eine der Gruppen R5, R6 und R, einen der von Wasserstoff verschiedenen Reste darstellt,

Eine reaktionsfähige veresterte Hydroxylgruppe X kann vor allem eine solche sein, die mit einer starken Mineralsäure, wie einer Halogenwasserstoffsäure, z. B.
Chlor-oder Bromwasserstoffsäure, verestert ist. X kann daher in erster Linie Chlor oder Brom darstellen. Die Hydroxylgruppe kann jedoch auch mit einer starken organischen Sulfonsäure, wie einer Niederalkansulfon- sÏure, z. B. Methan-oder Athansulfonsäure, oder einer Benzolsulfonsäure, wie p-Toluolsulfonsäure, verestert sein, sodass X für eine Sulfonyloxygruppe, z. B. eine Methyl-, Athyl-oder p-Tolylsulfonyloxygruppe steht.
Die Reaktion wird in üblicher Weise vorgenommen, z. B. in Gegenwart eines Uberschusses des Amins oder einer anderen Base und gegebenenfalls in Gegenwart eines geeigneten inerten Lösungsmittels, wenn nötig, unter Kühlen, vorzugsweise aber bei erhöhter Temperatur, gegebenenfalls in einem geschlossenen Gefäss und/oder in einer Inertgasatmosphäre, wie unter Stickstoff, durchgeführt.
Das hierbei verwendete Ausgangsmaterial ist bekannt oder lässt sich nach an sich bekannten Methoden herstellen. So kann man eine Verbindung der Formel

worin Rss und R, die oben genannte Bedeutung haben, in Gegenwart einer Lewissäure, besonders eines Friedel Crafts-Reagens, wie Aluminiumchlorid, umsetzen.
Ein Alkalimetallsalz des so erhaltenen Phenols kann dann mit einem reaktionsfähigen veresterten Hydroxyalkylhalogenid der Formel X- (CnHn) -Hal, worin die Sym- bole X und n die genannte Bedeutung haben und Hal ein Halogenatom darstellt, besonders mit einem Halo genalkylhalogenid, speziell einem solchen, in dem die Halogenatome verschieden sind, umgesetzt werden. Man kann aber auch das Phenol mit einer Verbindung der Formel Hal- (CnH2n)-OH, worin die Symbole die angegebene Bedeutung haben, umsetzen, und in der erhaltenen Verbindung die Hydroxylgruppe reaktionsfähig verestern, z. B. durch Behandlung mit einem Thionyl- halogenid, z. B.
Thionylchlorid, oder einem Phosphorhalogenid, wie Phosphortribromid, oder mit einem orga- nischen Sulfonsäurehalogenid, wie p-Toluolsulfonsäure- chlorid, gegebenenfalls in Gegenwart einer geeigneten Base, wie Pyridin.
Die neuen Verbindungen werden je nach den Reak tionsbedingungen in freier Form oder in Form ihrer Salze erhalten. Als Salze kommen Säureadditionssalze, z. B. therapeutisch verwendbare Säureadditionssalze mit anorganischen Sauren, wie Chlorwasserstoff-, Brom wasserstoff-, Schwefel-oder Phosphorsäure, oder organischen Säuren, wie organischen Carbonsäuren, z. B.
Essig-, Propion-, Pivalin-, Glykolsäure, Milchsäure, Malonsäure, Bernsteinsäure, Maleinsäure, Hydroxymalein- säure, Apfelsäure, Weinsäure, Zitronensäure, Benzoesäure, Salicylsäure, oder organischen Sulfonsäuren, wie Methan-oder Athansulfonsäure, Äthan-1, 2-disulfonsäure, 2-Hydroxyäthansulfonsäure, p-Toluol-oder Naphthalin-2-sulfonsäure in Frage. Andere Säureadditionssalze können auch als Zwischenprodukte verwend- bar sein, wie z. B. für die Herstellung von therapeutisch verwendbaren Säureadditionssalzen oder für die Reinigung der freien Basen oder zu ihrer Identifizierung.
Je nach der Natur der Endprodukte und den Reaktions- bedingungen lassen sich Mono"oder Polysalze erhalten.
Ein entstandenes Salz kann in die freie Base umgewandelt werden, z. B. durch Reaktion mit einem alkalischen Mittel, wie einem Metallhydroxyd, z. B. Na triumhydroxyd, einem Metallcarbonat, z. B. Natrium-, Kalium-oder Calciumcarbonat, oder einem entspre- chenden Bicarbonat oder Ammoniak, oder durch Be- handlung mit einem Hydroxylion-Austauscherharz.
Ein erhaltenes Salz, vor allem ein Salz mit einer anorganischen Säure, kann in ein anderes Salz umgewandelt werden, z. B. durch Reaktion mit einem geeigneten Metallsalz, z. B. einem Natrium-, Barium-oder Silbersalz, vorzugsweise in Gegenwart eines Verdün- nungsmittels, in welchem das entstehende anorganische Salz unlöslich ist und daher aus der Reaktion entfernt werden kann, oder durch Behandlung mit einem Anion Austauscher.
Eine freie Base kann in ihre Säureadditionssalze umgewandelt werden, z. B. durch Behandlung mit der entsprechenden Säure oder mittels eines geeigneten Anion-Austauschers. Ein Salz kann auch in Form eines Hydrates gewonnen werden, oder es kann mit Kristall- Lösungsmittel kristallisieren.
N-Oxyde der neuen Verbindungen lassen sich nach an sich bekannten Methoden herstellen, z. B. durch Behandlung der freien Base mit einem geeigneten Oxydationsmittel, wie Waserstoffperoxyd, Ozon oder einer Persäure, wie Peressigsäure, Perbenzoesäure, Mono perphthalsäure, Perschwefelsäure und ähnlichen. Die N-Oxyde lassen sich gemäss der oben genannten Umsetzung auch in ihre Salze überführen.
Erhaltene Mischungen isomerer Verbindungen lassen sich in üblicher Weise in die einzelnen Isomeren trennen. So kann man Mischungen von Racematen in die einzelnen reinen Racemate auftrennen, und/oder Racemate in die optisch aktiven Formen zerlegen.
Die Erfindung betrifft auch diejenigen Abände rungsformen des Verfahrens, wobei man einen Ausgangsstoff in Form eines Salzes verwendet.
Zweckmässig verwendet man solche Ausgangsstoffe, welche zu den eingangs besonders hervorgehobenen Endstoffen fiihren.
Die verfahrensgemäss erhaltenen Verbindungen können in der Form von pharmazeutischen Präparaten für enterale oder parenterale, vor allem aber topicale Applikation gebraucht werden.
Die Erfindung wird in den folgenden Beispielen näher, beschrieben. Die Tempar. afturen stnd im Celsius- graden angegeben.
Beispiel 1
Zu einer Lösung von 13, 9 g 2-p-Biphenylyl-2-p-oxy- phenyl-propan in einer Mischung von 50 ml Dimethyl- formamid und 25 ml Toluol gibt man in Portionen 4, 8 g einer 53 /eigen Natriushydridsuspension in Mineralöl.
Nach Beendigung der Waserstoffentwicklung fügt man unter Rühren langsam eine Lösung von 4, 1 g Athylenchlorhydrin und 25 ml Toluol zu. Dann lässt man bei Raumtemperatur während 20 Stunden stehen, filtriert vom anorganischen Niederschlag ab und konzentriert das Filtrat unter vermindertem Druck auf ungefähr 25 ml und verdiinnt es mit Wasser. Die Mischung wird mit Benzol gewaschen und eine kleine Menge Benzol abde- stilliert, um Spuren von Wasser zu entfernen. Die erhaltene Lösung der Hydroxy-äthoxy-verbindung wird in einem Eisbad gekühlt und tropfenweise mit einer L¯sung von 5 ml Thionylchlorid in 25 ml Benzol versetzt.
Nun lässt man auf Raumtemperatur kommen und erhitzt die Mischung auf ungefähr 50 , bis die Entwicklung von Chlorwasserstoff und Schwefeldioxyd beendet ist. Hierauf konzentriert man unter vermindertem Druck auf ein Volum von ungefähr 30 ml um Spuren von Thionylchlor- id zu entfemen. Die so erhaltene Benzollösung der entsprechenden Chloräthoxyverbindung wird mit 20 ml Di Ïthylamin behandelt. Die Reaktionsmischung verd nnt man mit 50 ml Benzol und erhitzt unter R ckfluss wÏhrend 2 Stunden, setzt das Rühren während 2 Stunden bei Raumtemperatur fort und lässt dann 15 Stunden site. hen.
Nach dem Filtrieren wird das Filtrat unter vermindertem Druck-konzentriert, worauf man den ¯ligen Rückstand mit Wasser verdünnt. Die wassmge Phase wird mehrmals mit ¯ther extrahiert, die vereinigten Extrakte mit 2-n. wässriger Salzsäure ausgezogen, wobei ein farbloses Kristallisat ausfällt, welches abfiltriert und luftgetrocknet wird. Man erhalt so das 2-p-Biphenylyl-2-[p-[¯ diäthylamino-äthoxy)-phenyl]-propan-hydrochlorid der Formel

welches nach dem Umkristallisieren aus 95%igem Äthanol l20-123 schmilzt.
Das Ausgangsmaterial wird wie folgt erhalten :
Unter Ruhren und Erhitzen in einem Dampfbad wird eine Mischung von 8, 05 g geschmolzenem Phenol und 1, 77 g AlumimumcMond portionswaiae mit einer ex geschmolzenen Mischung von 5, 7 g a, a-Dimethyl-p-phen- yl-benzylalkohol und 3 g Phenol versetzt. Das Rühren am Dampfbad wird für eine weitere Stunde fortgeführt, worauf man das Reaktionsprodukt mit Eis und konz.
Salzsäure zersetzt. Dann wird dreimal mit Essigester extrahiert, die vereinigten Essigesterextrakte-mit konz.
Kochsalzlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Das zurückbleibende Öl wird destilliert. Die Fraktion Kp. 0.08 185-205¯ kristallisiert spontan und wird mit Pentan trituriert. Man erhält so das 2-p-Bi phenylyl-2-p-oxyphenyl-propan vom F. 119-120 .
In gleicher Weise erhält man durch Auswahl der geeigneten Ausgangsstoffe folgende Verbindungen : 2-p- Chlorphenyl-2-[p-(diÏthylamino-Ïthoxy)-phenyl]-propan, dessen Hydrochlorid nach Umkristallisieren aus einer Mischung von ¯thanol und Ather bei 185-186 schmilzt ; das als Ausgangsmaterial verwendete 2-p Chlorphenyl-2-p-oxyphenyl-propan hat einen Kp von 130 /13 mm Hg und schmilzt nach Umkristallisieren aus Hexan bei 7274 2-p-Chlorphenyl-2-[m-chlor-p-(ss- diäthylamino-äthoxy)-phenyl]-propan, Kp. 170 /0, 05 mm Hg. ;
das als Ausgangsmaterial verwendete 2-p- CMorphenyl-2-m-chlor-p-oxyphenyl-propan hat einen Kp. von 165-170 /0, 15 m Hg.
In ähnlicher Weise erhalt man :
2-p-Chlorphenyl-2-[m-chlor-p-(¯-diÏthylamino-Ïth oxy)-m'-methyl-phenyl]-propan, dessen Hydrochlorid bei 146-148 schmilzt ; das als Ausgangsmaterial verwen dete 2-p-Chlorphenyl-2-(-m-chlor-p-oxy-m'-methyl-phen- yl)-propan, Kp. o s 125-126 wird durch Reaktion von 2-p-Chlorphenyl-2-propanol mit o-Chlor-o'-methyl-phenol in Gegenwart von Aluminiumchlorid und Reinigung ber das Benzoat, F. 129-130¯ erhalten;
2-p-Chlorphenyl-2-[p-(¯-diÏthylamino-Ïthoxy)-m,m' -dimethyl-phenyl]-propan, Kp. 0,05 171-173¯, Hydrochlorid F. 173-174¯;
das entsprechende Phenol, F. 4748¯, wird durch Zugabe von 17,1 g 2-p-Chlorphenyl-2 -propanol gefolgt von 12,2. g 2,6-Xylenol zu einer Mischung von 36, 6 g 2, 6-Xylenol und 6, 7 g-Aluminiumchlorid erhalten ;
2-p-Biphenylyl-2- [m-chlor-p- (¯-diÏthylamino-Ïth oxy)-phenyl]-propan, Kp. 0,08 207-210¯; das als Ausgangsstoff verwendete Phenol, Kp. 0,05 180-185¯, wird durch Reaktion von 2,72 g 2-p-Biphenylyl-2-p-oxy phenyl-propan mit 1, 8 g Sulfurylchlorid und 10 ml Benzol erhalten ;
2- [p- (ss-Diäthylamino-äthoxy)-phenyl]-2-p-methyl- phenyl-propan, dessen Hydrochlorid nach Umkristalli- sation aus Aceton bei 188-189 schmilzt ;
das als Aus gangsstoff verwendete Phenol (F. 69-70¯ nach Kristalli sation aus Pentan) wird hergestellt durch Zugabe von

Beispiel 2
Zu einer Lösung von 7, 0 g 2-p-Brom-phenyl-2- (p- oxyphenyl)-propan, F. 89-90 , in einer Mischung von 25 ml N, N-Dimethylformamid und 15 ml Toluol gibt man portionenweise 2, 4 g einer 53%igen Suspension von Natrium in einem Mineralöl. Nachdem die Wasserstoffentwicklung beendet ist, fügt man unter Rühren 2, 1 g Athylenchlorid in 20 ml Toluol langsam zu, lässt die Reaktionsmischung 20 Stunden bei Raumtemperatur stehen und filtriert. Das Filtrat wird unter vermindertem Druck auf 25-ml eingeengt und mit Wasser verdünnt.
Man extrahiert mit Benzol, wäscht die Benzollösung mit einer gesättigten wässrigen Kochsalzlösung und destilliert wenig Benzol ab, um Spuren von Wasser azeotrop zu entfernen. Die erhaltene getrocknete Benzollösung des 2-p-Brom-phenyl [p- (jss-hydroxy-äthoxy)-phenyl]-prop- ans wird im Eisbad gekühlt und tropfenweise eine Lösung von 5 ml Thionylchlorid in 25 ml Benzol zugegeben. Man lässt die Reaktionsmischung erwärmen und erhitzt sie auf ungefähr 50 , bis die Chlorwasserstoff- und Schwefeldioxydentwicklung beendet ist.
Die Reak tionsmischung wird unter vermindertem Druck auf unge- fähr 20 ml eingeengt, mit 50 ml Benzol verdünnt und erneut auf ungefähr 30 ml eingeengt. Zur so erhaltenen Benzollösung gibt man 15 mI Piperidin, verdünnt die Reaktionsmischung mit 50 ml Benzol und erhitzt am Rückflusskühler 2 Stunden und dampft zur Trockne ein.
Der Rückstand wird in Wasser aufgenommen, die wÏss rige L¯sung mit Diäthyläther extrahiert und der aber rische Extrakt mit 2-n. Salzsäure gewaschen. Die Salz säurephase wimd umter KüMon im Eisbad mit einer 50"/o- igen wÏssrigen Natriumhydroxydl¯sung und mit Natriumcarbonat auf ein pH von ungefähr 8 eingestellt. Man extrahiert mit Diäthyläther, trocknet die ätherische Lösung über Natriumsulfat und dampft zur Trockne ein.
Das so erhaltene 2-p-Brom-phenyl-2-[p-(¯-piperidino äthoxy)-phenyl]-propan der Formel

Der Essigesterextrakt wird mit wässriger Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Destillation des zurückbleibenden Öls ergibt das 2-p-Biphenylyl-2-1 [p-(ss-diäthylamino-äthoxy)- phenyl]-propan vom Kp. o, t 217-220 .
Beispiel 4
Eine L¯sung von 2, 7 g 2-p-Chlorphenyl-2-[p-(4- piperidino-äthoxy)-phenyl]-propan-hydrochlorid in 25 ml Wasser wird mit Natriumcarbonat basisch gestellt. Die freie Base wird mehrmals mit Ather extrahiert und die vereinigten Extrakte mit einer wässrigen Natriumchlorid- lösung gewaschen, über Natriumsulfat getrocknet und zur Trockene eingedampft. Der zurückbleibende ölige Rückstand wird in 15 ml Methanol gelöst und mit 2 ml einer 30 /igen wässrigen Wasserstoffperoxyd-Lösung versetzt.
Hierauf lässt man die L¯sung bei Raumtemperatur 90 Stunden stehen und zersetzt dann den Überschuss an Wasserstoffperoxyd durch Zugabe von 0, 05 g eines Palladiumkatalysators (10 /o PaRadium auf Kohle).
Nun filtriert man vom Katalysator ab und dampft das farblose Filtrat zur Trockene, wobei das N-Oxyd des 2p-Chlorphenyl-2 [p-(ss-piperidino-äthoxy-phenyl]-prop- an als farbloses ¯l zurückbleibt. Wenn man eine Ïthanolische L¯sung des N-Oxyds mit Pikrinsäure behandelt, so erhält man das Pikrat vom F. 118-120¯.

15 g of 2-p-methylphenyl-2-propanol to a mixture of 6.7 g of aluminum chloride and 19 g of phenol; 2-p-Chlorophenyl-2 - [(ss-piperidmo-ethoxy) -phenyl] -propane, the hydrochloride of which, after recrystallization from a 5: 1: 5 mixture of acetone, ethanol and ether, melts at 183-185;

Example 2
To a solution of 7.0 g of 2-p-bromo-phenyl-2- (p-oxyphenyl) propane, mp 89-90, in a mixture of 25 ml of N, N-dimethylformamide and 15 ml of toluene is added in portions 2.4 g of a 53% suspension of sodium in a mineral oil. After the evolution of hydrogen has ceased, 2.1 g of ethylene chloride in 20 ml of toluene are slowly added with stirring, the reaction mixture is left to stand for 20 hours at room temperature and filtered. The filtrate is concentrated to 25 ml under reduced pressure and diluted with water.
It is extracted with benzene, the benzene solution is washed with a saturated aqueous sodium chloride solution and a little benzene is distilled off in order to azeotropically remove traces of water. The dried benzene solution of 2-p-bromo-phenyl [p- (jss-hydroxy-ethoxy) -phenyl] -propane obtained is cooled in an ice bath and a solution of 5 ml of thionyl chloride in 25 ml of benzene is added dropwise. The reaction mixture is allowed to warm and is heated to about 50 until the evolution of hydrogen chloride and sulfur dioxide has ceased.
The reaction mixture is concentrated to approximately 20 ml under reduced pressure, diluted with 50 ml of benzene and concentrated again to approximately 30 ml. 15 ml of piperidine are added to the benzene solution obtained in this way, the reaction mixture is diluted with 50 ml of benzene and heated on a reflux condenser for 2 hours and evaporated to dryness.
The residue is taken up in water, the aqueous solution is extracted with diethyl ether and the fresh extract with 2-n. Washed hydrochloric acid. The hydrochloric acid phase was changed over in an ice bath with a 50% aqueous sodium hydroxide solution and adjusted with sodium carbonate to a pH of about 8. Extraction is carried out with diethyl ether, the ethereal solution is dried over sodium sulfate and evaporated to dryness.

Example 3
A suspension of 3.5 g of 2-p-biphenylyl-2- [p- (¯ diethylamino-ethoxy) phenyl] propane hydrochloride in a 2-n. aqueous soda solution is shaken with ethyl acetate. The ethyl acetate extract is washed with aqueous sodium chloride solution, dried over sodium sulfate and evaporated. Distillation of the remaining oil gives 2-p-biphenylyl-2-1 [p- (ss-diethylamino-ethoxy) phenyl] propane of bp o, t 217-220.
Example 4
A solution of 2.7 g of 2-p-chlorophenyl-2- [p- (4-piperidino-ethoxy) -phenyl] -propane hydrochloride in 25 ml of water is made basic with sodium carbonate. The free base is extracted several times with ether and the combined extracts are washed with an aqueous sodium chloride solution, dried over sodium sulfate and evaporated to dryness. The remaining oily residue is dissolved in 15 ml of methanol and treated with 2 ml of a 30% aqueous hydrogen peroxide solution.
The solution is then left to stand at room temperature for 90 hours and the excess hydrogen peroxide is then decomposed by adding 0.05 g of a palladium catalyst (10 / o PaRadium on carbon).
The catalyst is then filtered off and the colorless filtrate is evaporated to dryness, the N-oxide of 2p-chlorophenyl-2 [p- (ss-piperidino-ethoxy-phenyl] propane) remaining as a colorless oil If an ethanolic solution of the N-oxide is treated with picric acid, the picrate is obtained from F. 118-120¯.
