Verfahren zur Herstellung neuer Diphenylalkanverbindungen
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von quaternären Ammoniumverbindungen von Diphenylalkan-Verbindungen der Formel

worin jede der Gruppen Ri und R2 einen aliphatischen, cycloaliphatischen oder cycloaliphatischen-aliphatischen Kohlenwasserstoffrest bedeuten, Am eine tertiäre Aminogruppe darstellt und die Gruppe der Formel - (CIH")-einen Alkylenrest, der die Gruppe Am vom Sauerstoffatom durch mindestens 2 Kohlenstoffatome trennt und worin n eine ganze Zahl von 2-7 bedeutet, darstellt, worin jeder der Gruppen Rg und R4 Wasserstoff, Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste,
Halogenatome oder GruppenderFormel-O (CnH2n)-Am, worin Am und die Gruppe der Formel- (CnH2n)- die oben genannten Bedeutungen haben, darstellt, und R5 Wasserstoff, einen oder mehrere Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste oder Halogenatome oder einen, ge gebenenfalls ein oder mehere Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste oder Halogenatome enthaltenden Phenylrest darstellt, und mindestens einer der Reste R3, R4 und Ró einen der von Wasserstoff verschiedenen Reste bedeutet.
Der Rest R5 kann mit Ausnahme des Arylrestes einen oder mehrere gleiche oder verschiedene Substituenten der oben genannten Art bedeuten, wobei diese Substituenten sich in beliebiger Stellung des Phenylkern befinden können. Alkylreste mit 1-7 Kohlenstoffatomen sind z. B. Methyl-, Athyl-, n-Propyl-, Isopropyl-, n-Butyl-oder tert.-Butylgruppen. Alkoxygruppen mit 1-7 Kohlenstoffatomen sind vor allem Methoxy-, Athoxy-, n-Propyloxy-, Isopropyloxy-, n-Butyloxy-oder sek.-Butyloxygruppen. Halogenatome sind Fluor, Chlor, Brom oder Jod.
Die aliphatischen Kohlenwasserstoffreste gt und R2 sind vor allem Alkylreste mit 1-7, vorzugsweise mit 1-4 Kohlenstoffatomen, z. B. Methyl-, Athyl-, n-Propyl-, Isopropyl-, n-Butyl-oder Isobutylgruppen. Als cycloaliphatische Kohlenwasserstoffreste Rt und R2 kann man vor allem Cycloalkylreste mit 3-8 Ringkohlenstoff- atomen, wie Cyclopropyl-, Cyclobutyl, Cyclopentyl, Cyclohexyl, Cycloheptyl oder Cyclooctyl, und als cycloaliphatisch-aliphatische Kohlenwasserstoffreste vor allem Cycloalkyl-niederalkylreste mit 3-8 Ringkohlen- stoffatomen, wie Cyclopropylmethyl, Cyclopentylmethyl, 2-Cyclopentyläthyl, Cyclohexylmethyl, 1-Cyclohexyl äthyl oder Cycloheptylmethyl, erwähnen.
Die tertiäre Aminogruppe Am ist vorzugsweise eine Dialkylaminogruppe, worin Alkyl 1-7, vor allem 1-4 Kohlenstoffatome hat, besonders die Diäthylamino-, Dimethylamino-, Methyl-äthylamino-, Di-n-propyl- amino oder Di-isopropyl-aminogruppe, oder eine N- Cycloalkyl-N-niederalkylaminogruppe, worin Cycloalkyl 3-8, vor allem 5-7 Ringkohlenstoffatome aufweist, und Allcyl vorzugsweise 1-4 Kohlenstoffatome besitzt, wie die N-Cyclopentyl-N-methylaminogruppe, die N Cyclohexyl-N-methylaminogruppe oder die N-Cyclo- hexyl-N-äthylaminogruppe. Weiter kann die tert.
Aminogruppe Am eine Alkylenaminogruppe mit 4-8 Kohlenstoffatomen, eine Azaalkylenaminogruppe, worin Alkylen 4-6 Kohlenstoffatome besitzt, oder eine Oxa alkylen-oder Thiaalkylenaminogruppe, worin. Alkylen vorzugsweise 4 Kohlenstoffatome aufweist, darstellen.
Zusammen mit dem Stickstoffatom stellt daher eine solche Alkylen-, Azaalkylen-, Oxaalkylen-oder Thia- alkylengruppe z. B. eine Pyrrolidinogruppe, wie Pyrrolidino oder 2-Methylpyrrolodino, eine Piperidino- gruppe, wie Piperidino, 2-Methylpiperidino oder 4 Methylpiperidino, oder eine 1, 6-Hexylenamino- oder 1, 7-Heptylenaminogruppe, sowie eine Azaalkylenamino- gruppe, wie besonders N-Alkyl-oder Oxyalkyl-aza-- alkylenaminogruppe, worin Alkylen 4-6 Kohlenstoff atome aufweist, wie Piperazino, 4-Methyl-oder-äthyl- piperazino, 4-Hydroxyäthyl-piperazino, 3-Aza-1, 6hexylenamino, 3-Aza-3-methyl-1,
6-hexylenamino, 4 Aza-1, 7-heptylenamino, 4-Aza-4-methyl-1, 7-heptylenamino, oder eine Morpholinogruppe, wie Morpholino oder 3-Methylmorpholino, oder eine Thiamorpholinogruppe, wie Thiamorpholino, dar.
Der Alkylenrest der Formel-(CnH2nf besitzt vor allem 2-3 Kohlenstoffatome und stellt deshalb vor allem einen 1, 2-Athylen-, 1-Methyl-1, 2-äthylen-, 2 Methyl1, 2-äthylen- oder 1, 3-Propylenrest dar. Er kann aber auch 1, 3-Butylen, 2, 3-Butylen, 3, 4-Butylen, 1, 4-Butylen, 1, 4-Pentylen, 1, 5-Pentylen, 1, 5-Hexylen, 1, 6-Hexylen oder 1, 7-Heptylen darstellen.
Sofern die Reste Rg und R4 Alkyl-oder Alkoxygruppen oder Halogenatome bedeuten, so kommen vorallem die eingangs fiir R5 genannten Reste dieser Art in Frage. Falls R, undoder R4 einen Rest der Formel -H-(CnH2n)-Am bedeutet, so haben darin Am und n besonders die oben hervorgehobenen Bedeutungen.
Die quatemären Ammoniumverbindungen sind solche, welche durch Reaktion der tertiären Amine mit reaktionsfähigen Estern von Alkoholen, vor allem Alkanolen mit 1-7 Kohlenstoffatomen, wie Methanol, Äthanol, n-Propanol oder Isopropanol, oder Phenylalkanolen, worin der Alkanolrest 1-7 Kohlenstoffatome enthält, wie Benzylalkohol, 1-Phenyl-oder 2-Phenyl äthanol, erhalten werden. Als reaktionsfähige Ester sind z. B. diejenigen starker anorganischer oder organischer Säuren, wie der Halogenwasserstoffsäuren, Schwefelsäuren oder Sulfonsäuren, wie Chlor-, Brom-oder Jodwasserstoffsäure, Schwefelsäure oder Alkansulfonsäuren mit 1-7 Kohlenwasserstoffatomen, z. B.
Methan-oder Athansulfonsäure, Hydroxyalkansulfonsäuren mit 1-7 Kohlenstoffatomen, wie 2-Hydroxyäthansulfonsäure, oder Benzolsulfonsäuren, wie p-Toluolsulfonsäuren, zu nennen. Die Erfindung betrifft auch die Herstellung quatemärer Ammoniumhydroxyde und ihrer Salze mit anderen Säuren, besonders den weiter unten genannten.
Die neuen Verbindungen besitzen wertvolle Antifungus-Eigenschaften und können dementsprechend als Heil-oder Desinfektionsmittel verwendet werden. So zeigen sie eine Wirkung gegen Pilze, die oberflächliche Dermatophytosen hervorrufen, wie Trichophyton mentagrophytes, Trichophyton gallinae, Trichophyton interdigitale, Microsporum audouini, Microsporum canis, Microsporum gypseum, und können damit z. B. bei den entsprechenden Erkrankungen, wie Dermatophytosis, angewendet werden. Sie wirken auch gegen solche Pilze, welche chronische Infektionen der Haut und der darunterliegenden Gewebe verursachen, wie z. B.
Sporotrichium schenkii, oder gegen Pilze, welche tiefer sitzende systemische Mykosen verursachen, wie Candida albicans, Cryptococcus neoformans, Histoplasma capsulatum, und können dementsprechend angewendet werden. Die neuen Verbindungen sind auch wirksam gegen Actinomyceten wie Nocardia asteroides, gegen grampositive Bakterien, wie Diplococcus pneumoniae, Staphylococcus aureus, oder gramnegative Bakterien, wie Escherichia coli, Pseudomonas aeruginosa, und gegen Protozoen, wie Trichomonas vaginalis, und kön- nen entsprechend in der Heilkunst angewendet werden.
Die neuen Verbindungen wirken auch gegen Parasiten, wie z. B. Bandwürmer. Ausserdem zeigen sie gewisse stimulierende und endocrine, z. B. östrogene Effekte.
Besonders wertvoll sind die quaternärenAmmonium- derivate, in erster Linie die quaternären Alkyl-und bPhenylalkyl-ammoniumderivate, worin die Alkylreste 1-7 Kohlenstoffatome enthalten, von Verbindungen der Formel

Als eine Gruppe von besonders wertvollen Verbindungen können auch die quatemären Ammoniumderivate, in erster Linie die quatemären Alkyl-und , Phenylalkyl-ammoniumderivate, worin die Alkylreste 1-7 Kohlenstoffatome enthalten, von Verbindungen der Formel

worin Ri bis R5, Am und- (CnHn)- die eingangs gegebene Bedeutung haben, mit einem reaktionsfähigen Ester eines Alkohols umsetzt.
Reaktionsfähige Ester von Alkoholen mit einer starken Säure sind z. B. Alkylhalogenide, Dialkylsulfate, Alkylsulfonate oder Phenylalkylhalogenide, worin die Alkylreste 1-7 Kohlenstoffatome enthalten, z. B. die oben beschriebenen Verbindungen.
Die quatemären Verbindungen können in andere quaternäre Verbindungen umgewandelt werden, wie z. B. quaternäre Ammoniumhalogenide durch Behandlung mit Silberoxyd, oder quaternäre Ammoniumsulfate mit Bariumhydroxyd, oder quaternäre Ammoniumsalze mit einem Hydroxylion-Austauscher in die entsprechenden quatemären Ammoniumhydroxyde. Solche Austauschreaktionen lassen sich auch mittels Elektrodialyse durchführen. Ein quaternäres Ammoniumhydroxyd lässt sich durch Reaktion mit einer Säure in ein entsprechendes Salz verwandeln. Man kann auch ein quaternäres Ammoniumsalz direkt in ein anderes verwandeln. So lässt sich z.
B. ein quaternäres Ammoniumjodid mit frisch hergestelltem Silberoxyd oder mit Chlorwasserstoff in wasserfreiem Methanol zum Chlorid umsetzen.
Solche Umsetzungen lassen sich auch mittels Anion Austauschern durchführen.
Die im obigen Verfahren verwendeten Ausgangs- stoffe können z. B. durch Umsetzen einer Verbindung der Formel

worin Ri, R2 und Rg die oben gegebene Bedeutung haben und Ra und R, für Wasserstoff, Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste, Halogenatome oder Hydroxylgruppen steht, oder einem Salz davon mit einem reak tionsfähigen Ester einer Verbindung der Formel HO-(CnH2n)-Am, worin Am und die Gruppe der Formel- oben gegebene Bedeutung haben, wobei letztere Am und-OH durch mindestens 2 Kohlenstoffatome trennt, erhalten werden. Dabei kann man das Ausgangsmaterial in ein Metallsalz, besonders ein Alkalimetallsalz, wie ein Lithium-, Natrium-oder Kaliumsalz, umwandeln.
Dieses wird dann mit einer Verbindung der Formel X- (CnH-Am, worin Am und die Gruppe der Formel- (CnHQn)-, welche Am von X durch mindestens 2 Kohlenstoffatome trennt, die oben genannte Bedeutung haben und X für eine reaktions fähige veresterte Hydroxylgruppe steht, umgesetzt. Eine reaktionsfähige veresterte Hydroxylgruppe ist vor allem eine solche, die mit einer starken Mineralsäure, wie einer Halogenwasserstoffsäure, z. B. Chlor-oder Bromwasser stoffsäure, verestert ist ; X stellt daher in erster Linie ein Chlor-oder Bromatom dar. Die Hydroxylgruppe kann jedoch auch mit einer starken organischen Sulfonsäure, wie einer Alkansulfonsäure, z. B.
Methan-oder Athansulfonsäure, oder einer Benzolsulfonsäure, wie p Toluolsulfonsäure verestert sein, so dass X für eine Sulfonyloxygruppe, z. B. eine Methyl-, Athyl-oder p Tolylsulfonyloxygruppe, steht. Der reaktionsfähige Ester des obigen tert. Aminoalkanols kann auch in Form eines Salzes verwendet werden, wobei man das Ausgangsmaterial oder das Salz davon in Gegenwart eines Überschusses des salzbildenden Reagens verwendet. Bei der genannten Reaktion ist es auch möglich, an Stelle eines Salzes der Hydroxydverbindung von der Hydroxyverbin dung selbst auszugehen und diese in Gegenwart eines entsprechenden Kondensationsmittels, vor allem eines metallsalzbildenden Kondensationsmittels, zu benützen.
Die Reaktion wird in üblicher Weise, vorzugsweise in Gegenwart eines Verdünnungsmittels, wennn nötig, unter Kühlen, vorzugsweise aber bei erhöhter Temperatur, gegebenenfalls in einem geschlossenen Gefäss und/ oder in einer Inertgasatmosphäre, wie unter Stickstoff, durchgeführt.
Ein entstandenes Salz kann in die freie Base umgewandelt werden, z. B. durch Reaktion mit einem alkalischen Mittel, wie einem Metallhydroxyd, z. B. Natriumhydroxyd, einem Metallcarbonat, z. B. Natrium-, Kalium-oder Calciumcarbonat, oder einem entsprechenden Bicarbonat oder Ammoniak, oder durch Behandlung mit einem Hydroxylion-Austauscherharz.
Zweckmässig verwendet man solche Ausgangsstoffe, welche zu den eingangs besonders hervorgehobenen Endstoffen führen.
Die verfahrensgemäss erhaltenen Verbindungen kön- nen in der Form von pharmazeutischen Präparaten für enterale oder parenterale, vor allem aber topicale Applikation gebraucht werden.
Die Erfindung wird in den folgenden Beispielen näher beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel
Zu einer Lösung von 2, 3 g 2-p-Chlorphenyl-2 [p-(ss-diäthylamino-äthoxy)-phenyl]-propan in 50 ml 95 /Oigem Athanol gibt man 3 ml Athylbromid. Die Mischung wird in einer rostfreien Stahlbombe auf einem Dampfbad 15 Stunden erhitzt. Hierauf entfernt man das Lösungsmittel unter vermindertem Druck und kristallisiert den Rückstand durch Zugabe von Ather. Das so erhaltene 2-p-Chlorphenyl-2- [p- (ss-diäthylamino- äthoxy)-phenyll-propan-äthobromid wird aus einer Mischung von Aceton und Ather umkristallisiert und im Vakuum getrocknet, F. 132-146 .
Das Ausgangsmaterial kann wie folgt erhalten werden :
Zu einem eisgekühlten Grignard-Reagens, hergestellt aus 284 g Methyljodid und 48, 6 Magnesiumspänen in 550 ml Ather gibt man unter Rühren tropfenweise eine Lösung von 154, 6 g 4-Chloracetophenon in 200 ml Ather. Man lässt die Reaktionsmischung bei Raumtemperatur stehen und erhitzt sie dann während 2 Stunden am Rückfluss. Das Reaktionsprodukt wird nun unter Rühren und Kühlung in einem Eisbad langsam durch Zugabe von 300 ml gesättigter wässriger Ammo- niumchloridlösung und 300 ml Wasser zersetzt. Dann fügt man 300 ml Ather zu, trennt die organische Phase, wäscht sie und trocknet sie über Natriumsulfat. Nach dem Abdampfen des Lösungsmittels wird der Rückstand destilliert.
Das 2-p-Chlorphenyl-2-propanol destilliert bei Kp. s 92-96 .
28, 2 g Phenol werden unter Rühren portionenweise mit 6, 7 g Aluminiumchlorid versetzt. Zu der so erhaltenen Mischung gibt man unter Rühren und Wasserkühlung eine Mischung von 17, 1 g 2-p-Chlorphenyl-2- propanol und 9, 4 g Phenol. Nach beendeter Zugabe rührt man 2 Stunden bei Raumtemperatur, lässt 15 Stunden stehen und erhitzt unter Rühren eine Stunde auf 40-50 . Das Reaktionsprodukt wird unter Rühren zu 100 ml 6-n wässriger Salzsäure hinzugefügt. Die organische Phase wird abgetrennt und das überschüssige Phenol durch Destillation bis zu einer Temperatur von 130 (13 mm Hg) abdestilliert. Das zurückbleibende Öl wird destilliert, wobei man das 2-p-Chlorphenyl-2-p-oxyphenyl-propan vom F. 72-74 (aus Hexan) erhält.
Zu einer Lösung von 12, 33 g 2-p-Chlorphenyl-2p-oxyphenyl-propan in einer Mischung von 75 ml Dimethylformamid und 40 ml Toluol gibt man unter Rüh- ren und Kühlen portionenweise 2, 4- g einer 53 /oigen Mineralölsuspension von Natriumhydrid. Nach Beendigung der Wasserstoffentwicklung gibt man unter Rüh-- ren und Kiihlen tropfenweise eine Lösung von 7 g S- Diäthylamino-äthylchlorid in 35 ml Toluol zu. Dann riihrt man bei Raumtemperatur drei Stunden weiter und lässt hierauf 15 Stunden stehen.
Der anorganische Niederschlag wird abfiltriert, das Filtrat auf ein Volumen von ungefähr 50-80 ml konzentriert, mit Wasser verdünnt und mit 300 ml Äther extrahiert. Der 2ither- extrakt wird mit 100 ml 2-n wässriger Salzsäure gewaschen und die saure Lösung abgetrennt. Nach einigen Minuten kristallisiert das 2-p-Chlorphenyl-2-[p-diäthyl- amino-äthoxy)-phenyl3-propan-hydrochlorid. Es wird abfiltriert und aus einer Mischung von Athanol und Ather umkristallisiert, F. 185-186 .
Die freie Verbindung erhält man durch Behandeln des Salzes mit 2-n wässriger Natriumcarbonatlösung und Extraktion der freien Base mit Essigsäureäthylester.

The new compounds have valuable antifungal properties and can accordingly be used as medicinal or disinfectant agents. 3 ml of ethyl bromide are added to a solution of 2.3 g of 2-p-chlorophenyl-2 [p- (ss-diethylamino-ethoxy) phenyl] propane in 50 ml of 95% ethanol. The mixture is heated in a stainless steel bomb on a steam bath for 15 hours. The solvent is then removed under reduced pressure and the residue is crystallized by adding ether. The 2-p-chlorophenyl-2- [p- (ss-diethylamino-ethoxy) -phenyl-propane-ethobromide obtained in this way is recrystallized from a mixture of acetone and ether and dried in vacuo, mp 132-146.
The starting material can be obtained as follows:
A solution of 154.6 g of 4-chloroacetophenone in 200 ml of ether is added dropwise with stirring to an ice-cold Grignard reagent, prepared from 284 g of methyl iodide and 48.6 magnesium turnings in 550 ml of ether. The reaction mixture is left to stand at room temperature and then refluxed for 2 hours. The reaction product is then slowly decomposed by adding 300 ml of saturated aqueous ammonium chloride solution and 300 ml of water while stirring and cooling in an ice bath. 300 ml of ether are then added, and the organic phase is separated, washed and dried over sodium sulfate. After the solvent has evaporated, the residue is distilled.
The 2-p-chlorophenyl-2-propanol distills at b.p. s 92-96.
28.2 g of phenol are mixed in portions with 6.7 g of aluminum chloride while stirring. A mixture of 17.1 g of 2-p-chlorophenyl-2-propanol and 9.4 g of phenol is added to the mixture thus obtained, with stirring and with water cooling. When the addition is complete, the mixture is stirred for 2 hours at room temperature, left to stand for 15 hours and heated to 40-50 for one hour while stirring. The reaction product is added to 100 ml of 6N aqueous hydrochloric acid with stirring. The organic phase is separated off and the excess phenol is removed by distillation up to a temperature of 130 (13 mm Hg). The remaining oil is distilled, whereby the 2-p-chlorophenyl-2-p-oxyphenyl-propane with a melting point of 72-74 (from hexane) is obtained.
To a solution of 12.33 g of 2-p-chlorophenyl-2p-oxyphenyl-propane in a mixture of 75 ml of dimethylformamide and 40 ml of toluene, 2.4 g of a 53% mineral oil suspension of is added in portions with stirring and cooling Sodium hydride. The free compound is obtained by treating the salt with 2N aqueous sodium carbonate solution and extracting the free base with ethyl acetate.
