Verfahren zur Herstellung neuer Diphenylalkanverbindungen
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Diphenylalkanverbindungen der Formel

worin jede der Gruppen Rt und R2 einen aliphatischen, cycloaliphatischen oder cycloaliphatisch-aliphatischen Kohlenwasserstoffrest bedeuten, Am eine tertiäre Aminogruppe darstellt und die Gruppe der Formel -(CnH2n)-einen Alkylenrest, der die Gruppe Am vom Sauerstoffatom durch mindestens 2 Kohlenstoffatome trennt und worin n eine ganze Zahl von 2-7 bedeutet, darstellt, und worin jede der Gruppen R3 und R4 Wasserstoff, Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxymit 1-7 Kohlenstoffatomen oder Trifluormethylreste,
Halogenatome oder Gruppen der Formel -O-(CnHOn)-Am, worin Am und die Gruppe der Formel- H2n)-die oben genannten Bedeutungen haben, darstellt, und R3 Wasserstoff, einen oder mehrere Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste oder Halogenatome oder einen, gegebenenfalls ein oder mehrere Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste oder Halo genatome enthaltenden Phenylrest darstellt, und mindestens einer der Reste R3, R4 und R-einen der von Was serstoff verschiedenen Reste bedeutet, sowie ihren Salzen.
Der Rest R3 kann einen oder mehrere gleiche oder verschiedene Substituenten der oben genannten Art bedeuten, wobei diese Substituenten sich in beliebiger Stellung des Phenylkems befinden können. Alkylreste mit 1-7 Kohlenstoffatomen sind z. B. Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-oder tert.-Butylgruppen.
Alkoxygruppen mit 1-7 Kohlenstoffatomen sind vor allem Methoxy-, Athoxy-, n-Propyloxy-, Isopropyloxy-, n-Butyloxy-oder sek.-Butyloxygruppen. Halogenatome sind Fluor, Chlor, Brom oder Jod.
Die aliphatischen Kohlenwasserstoffreste Rt und R2 sind vor allem Alkylreste mit 1-7, vorzugsweise mit 1-4 Kohlenstoffatomen, z. B. Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-oder Isobutylgruppen. Als cycloaliphatische Kohlenwasserstoffreste R, und R2 kann man vor allem Cycloalkylreste mit 3-8 Ringkohlenstoffatomen, wie Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclo- hexyl, Cycloheptyl oder Cyclooctyl, und als cycloali phatisch-aliphatische Kohlenwasserstoffreste vor allem Cycloalkyl-niederalkylreste mit 3-8 Ringkohlenstoffato- men, wie Cyclopropylmethyl, Cyclopentylmethyl, 2 Cyclopentyläthyl, Cyclohexylmethyl,
1-Cyclohexyläthyl oder Cycloheptylmethyl, erwahnen.
Die tertiäre Aminogruppe Am ist vorzugsweise eine Dialkylaminogruppe, worin Alkyl 1-7, vor allem 1-4 Kohlenstoffatome hat, besonders die Diäthylamino-, Dimethylamino-, Methyl-äthylamino-, Di-n-propylamino cder Di-isopropyl-aminogruppe, oder eine N-Cyclo alkyl-N-alkylaminogruppe, worin Cycloalkyl 3-8, vor allem 5-7 Ringkohlenstoffatome aufweist, und Alkyl vorzugsweise 1-4 Kohlenstoffatome besitzt, wie die N Cyclopentyl-N-methylaminogruppe, die N-Cyclohexyl- N-methylaminogruppe oder die N-Cyclohexyl-N-äthyl- aminogruppe. Weiter kann die tert.
Aminogruppe Am eine Alkylenaminogruppe mit 4-8 Kohlenstoffatomen, eine Azaalkylenaminogruppe, worin Alkylen 4-6 Kohlenstoffatome besitzt, oder eine Oxaalkylen-oder Thiaalkylenaminogruppe, worin Alkylen vorzugsweise 4 Koh lenstoffatome aufweist, darstellen. Zusammen mit dem Stickstoffatom stellt daher eine solche Alkylen-, Azaalkylen-, Oxaalkylen-oder Thiaalkylengruppe z.
B. eine Pyrrolidinogruppe, wie Pyrrolidino oder 2-Methylpyrrolidino, eine Piperidinogruppe, wie Piperidino, 2 Methylpiperidino oder 4-Methylpiperidino, oder eine 1, 6-Hexylenamino- oder 1, 7-Heptylenaminogruppe, sowie eine Azaalkylenaminogruppe, wie besonders N Alkyl-oder Oxyalkyl-azaalkylenaminogruppe, worin Alkylen 4-6 Kohlenstoffatome aufweist, wie Piper azino, 4-Methyl-oder-äthylpiperazino, 4-Hydroxyäthyl- piperazino, 3-Aza-1, 6-hexylenamino, 3-Aza-3-methyl- 1, 6-hexylenamino, 4-Aza-1,
7-heptylenamino, 4-Aza-4 methyl-1, 7-heptylenamino, oder eine Morpholinogruppe, wie Morpholino oder 3-Methylmorpholino, oder eine Thiamorpholinogruppe, wie Thiamorpholino, dar.
Der Alkylenrest der Fonnel-(CnH2n)-besitzt vor allem 2-3 Kohlenstoffatome und stellt deshalb vor allem einen 1, 2-Athylen-, 1-Methy11, 2-äthylen-, 2-Methyl1, 2-äthylen-oder 1, 3-Propylen-rest dar. Er kann aber auch 1, 3-Butylen, 2, 3-Butylen, 3, 4-Butylen, 1, 4-Butylen, 1, 4-Pentylen, 1, 5-Pentylen, 1, 5-Hexylen, 1, 6-Hexylen oder 1, 7-Heptylen dar.
Sofern die Reste R3 und R4 Alkyl-oder Alkoxygruppen oder Halogenatome bedeuten, so kommen vor allem die eingangs für Rg genannten Reste dieser Art in Frage. Falls R3 und/oder R4 einen Rest der Formel -0- (CnH)-Am bedeutet, so haben darin Am und n besonders die oben hervorgehobenen Bedeutungen.
Die neuen Verbindungen besitzen wertvolle anti fungus-Eigenschaften und können dementsprechend als Heil-oder Desinfektionsmittel verwendet werden. So zeigen sie eine Wirkung gegen Pilze, die oberflächliche Dermatophytosen hervorrufen, wie Trichophyton mentagrophytes, Trichophyton gallinae, Trichophyton interdigitale, Microsporum audouini, Microsporum canis, Microsporum gypseum, und können damit z. B. bei den entsprechenden Erkrankungen, wie Dermatophytosis, angewendet werden. Sie wirken auch gegen solche Pilze, welche chronische Infektionen der Haut und der darunterliegenden Gewebe verursachen, wie z.
B. Sporotrichium schenkii, oder gegen Pilze, welche tiefer sitzende systemische Mykosen verursachen, wie Candida albicans, Cryptococcus neoformans, Histoplasma capsulatum, und können dementsprechend angewendet werden. Die neuen Verbindungen sind auch wirksam gegen Actinomyceten wie Nocardia asteroides, gegen grampositive Bakterien, wie Diplococcus pneumoniae, Staphylococcus aureus, oder gramnegative Bakterien, wie Escherichia coli, Pseudomonas aeruginosa, und gegen Protozoen, wie Trichomonas vaginalis, und können entsprechend in der Heilkunst angewendet werden.
Die neuen Verbindungen wirken auch gegen Parasiten, wie z. B. Bandwürmer. Ausserdem zeigen sie gewisse stimulierende und endocrine, z. B. östrogene, Effekte.'

Als eine Gruppe von besonders wertvollen Verbindungen können auch die Verbindungen der Formel

Erfindungsgemäss werden die oben genannten Verbindungen erhalten, indem man eine Verbindung der Formel

worin Rl, Rg und R5 die oben gegebenen Bedeutungen haben und jede der Gruppen R6 und R, für Wasserstoff, Alkyl-mit 1-7 Kohlenstoffatomen, Alkoxy-mit 1-7 Kohlenstoffatomen oder Trifluormethylreste, Halo genatome oder Hydroxylgruppen steht, wobei mindestens eine der Gruppen R5, R, und R, einen der von Wasserstoff verschiedene Reste bedeutet, oder ein Salz davon mit einer Verbindung der Formel X- (CnH2n)-Am, worin Am und die Gruppe der Formel- (CnHz")- die oben gegebene Bedeutung haben,
wobei letztere X und Am durch mindestens 2 Kohlenstoffatome trennt, und X für eine reaktionsfähige veresterte Hydroxylgruppe steht, umsetzt.
Ein Salz des Ausgangsmaterials ist ein Metallsalz, besonders ein Alkalimetallsalz, wie ein Lithium-, Natrium-oder Kaliumsalz. Dieses wird dann mit der Verbindung der Formel X-(CnH2n)-Am, worin die Gruppe der Formel- (C"H=n)- Am von X durch mindestens 2 Kohlenstoffatome trennt, umgesetzt. Eine reaktions fähige veresterte Hydroxylgruppe ist vor allem eine solche, die mit einer starken Mineralsäure, wie einer Halogenwasserstcffsäure, z. B. Chlor-oder Bromwasserstoffsäure verestert ist. X stellt daher in erster Linie Chlor oder Brom dar. Die Hydroxylgruppe kann jedoch auch mit einer starken organischen Sulfonsäure, wie einer Niederalkansulfonsäure, z. B.
Methan-oder Athansulfonsäure, oder einer Benzolsulfonsäure, wie p Toluolsulfonsäure verestert sein, sodass X für eine Sulf onyloxygruppe, z. B. eine Methyl-, Sithyl-oder p Tolylsulfonyloxygruppe steht. Der reaktionsfähige Ester kann auch in Form eines Salzes verwendet werden, wobei man das Ausgangsmaterial oder das Salz davon in Gegenwart eines Uberschusses des salzbildenden Reagens verwendet. Bei der genannten Reaktion ist es auch möglich, an Stelle eines Salzes der Hydroxyver- bindung von der Hydroxyverbindung selbst auszugehen und diese in Gegenwart eines metallsalzbildenden Kondensationsmittels zu benützen.
Die Reaktion wird in üblicher Weise, vorzugsweise in Gegenwart eines Verdünnungsmittels, wenn nötig, unter Kühlen, vorzugsweise aber bei erhöhter Temperatur, gegebenenfalls in einem geschlossenen Gefäss und/oder in einer Inert- gasatmosphäre, wie unter Stickstoff durchgeführt.
Das hierbei verwendete Ausgangsmaterial ist bekannt oder lässt sich nach an sich bekannten Methoden herstellen. So kann man eine Verbindung der Formel

worin Ra und R, die oben genannte Bedeutung haben, in Gegenwart einer Lewis-Säure, besonders eines Friedel-Crafts-Reagens, wie Aluminiumchlorid, umsetzen.
Die neuen Verbindungen werden je nach den Reaktionsbedingungen in freier Form oder in Form ihrer Salze erhalten. Als Salze kommen Säureadditionssalze, z. B. therapeutisch verwendbare Säureadditionssalze mit anorganischen Säuren, wie Chlorwasserstoff-, Bromwasserstcff-, Schwefel-cder Phosphorsäure, oder organischen Säuren, wie organischen Carbonsäuren, z. B.
Essig-, Propion-, Pivalin-, Glykolsäure, Milchsäure, Malonsäure, Bernsteinsäure, Maleinsäure, Hydroxy- maleinsäure, Apfelsäure, Weinsäure, Zitronensäure, Benzoesäure, Salicylsäure oder organischen Sulfonsäuren, wie Methan-oder Athansulfonsäure, Athan-1, 2-disulfonsäure, 2-Hydroxyäthansulfonsäure, p-Toluol-oder Naphthalin-2-sulfonsäure in Frage. Andere Säureadditionssalze können auch als Zwischenprodukte verwendbar sein, wie z. B. für die Herstellung von therapeutisch verwendbaren Säureadditionssalzen oder für die Reinigung der freien Basen oder zu ihrer Identifizierung. Je nach der Natur der Endprodukte und den Reaktionsbedingungen lassen sich Mono-oder Polysalze erhalten.
Ein entstandenes Salz kann in die freie Base umgewandelt werden, z. B. durch Reaktion mit einem alkalischen Mittel, wie einem Metallhydroxyd, z. B. Natriumhydroxyd, einem Metallcarbonat, z. B. Natrium-, Kalium-oder Calciumcarbonat, oder einem entsprechenden Bicarbonat oder Ammoniak, oder durch Behandlung mit einem Hydroxylion-Austauscherharz.
Ein erhaltenes Salz, vor allem ein Salz mit einer anorganischen Säure, kann in ein anderes Salz umgewandelt werden, z. B. durch Reaktion mit einem geeigneten Metallsalz, z. B. einem Natrium-, Barium-oder Silbersalz, vorzugsweise in Gegenwart eines Verdün- nungsmittels, in welchem das entstehende anorganische Salz unlöslich ist und daher aus der Reaktion entfernt werden kann, oder durch Behandlung mit einem Anion Austauscher.
Eine freie Base kann in ihre Säureadditionssalze umgewandelt werden, z. B. durch Behandlung mit der entsprechenden Säure oder mittels eines geeigneten Anion Austauschers. Ein Salz kann auch in Form eines Hydrates gewonnen werden, oder es kann mit Kristall Lösungsmittel kristallisieren.
N-Oxyde der neuen Verbindungen lassen sich nach an sich bekannten Methoden herstellen, z. B. durch Behandlung der freien Base mit einem geeigneten Oxydationsmittel, wie Wasserstoffperoxyd, Ozon oder einer Persäure, wie Peressigsäure, Perbenzoesäure, Monoperphthalsäure, Perschwefelsäure und ähnlichen. Die N Oxyde lassen sich gemäss der oben genannten Umsetzung auch in ihre Salze überführen.
Erhaltene Mischungen isomerer Verbindungen lassen sich in üblicher Weise in die einzelnen Isomeren trennen. So kann man Mischungen von Racematen in die einzelnen reinen Racemate auftrennen, und/oder Racemate in die optisch aktiven Formen zerlegen.
Die Erfindung betrifft auch diejenigen Abänderungs- formen des Verfahrens, wobei man einen Ausgangsstoff in Form eines Salzes verwendet.
Zweckmässig verwendet man solche Ausgangsstoffe, welche zu den eingangs besonders hervorgehobenen Endstoffen führen.
Die verfahrensgemäss erhaltenen Verbindungen kön- nen in Form von pharmazeutischen Präparaten für enterale oder parenterale, vor allem aber topicale Applikation gebraucht werden.
Die Erfindung wird in den folgenden Beispielen nä- her beschrieben. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu 3, 8 g 2-p-Biphenylyl-2-p-oxyphenyl-propan in 8 cm3 Dimethylformamid gibt man 0, 63 g einer 53 /oigen Mineralölsuspension von Natriumhydrid. Zu der erhaltenen Aufschlämmung gibt man unter Rühren eine Lösung von 1, 78 g ss-Diäthylamino-äthylchlorid in 8 cm3 Toluol. Man erwärmt die Reaktionsmischung auf ungefähr 50 , um eine klare braune Lösung zu erhalten, setzt das Rühren während 2 Stunden bei Raumtemperatur fort und lässt dann 15 Stunden ste- hen.
Der anorganische Niederschlag wird abfiltriert und das Filtrat unter vermindertem Druck konzentriert, worauf man den öligen Rückstand mit Wasser ver dünnt. Die wässrige Phase wird mehrmals mit Ather extrahiert, die vereinigten Extrakte mit 2-n. wässriger Salzsäure ausgezogen, wobei ein farbloses Kristallisat ausfällt, welches abfiltriert und luftgetrocknet wird. Man erhält so das 2-p-Biphenylyl-2-[-(ss-diäthylamino-äth- oxy)-phenyl]-propan-hydrochlorid der Formel

welches nach dem Umkristallisieren aus 95%igem Athanol bei 120-123 schmilzt.
Das Ausgangsmaterial wird wie folgt erhalten :
Unter Rühren und Erhitzen in einem Dampfbad wird eine Mischung von 8, 05 g geschmolzenem Phenol und 1, 77 g Aluminiumchlorid portionenweise mit einer geschmolzenen Mischung von 5, 7 g a, a-Dimethyl-p phenyl-benzylalkohol und 3 g Phenol versetzt. Das Rühren am Dampfbad wird für eine weitere Stunde fortgeführt, worauf man das Reaktionsprodukt mit Eis und konz. Salzsäure zersetzt. Dann wird dreimal mit Essigester extrahiert, die vereinigten Essigesterextrakte mit konz. Kochsalzlösung gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter verminder- tem Druck eingedampft. Das zurückbleibende 61 wird destilliert. Die Fraktion Kp. 0, 08 185-205 kristallisiert spontan und wird mit Pentan trituriert.
Man erhält so das 2-p-Biphenylyl-2-p-oxyphenyl-propan vom F. 119120 .
Beispiel 2
Eine Suspension von 3, 5 g 2-p-Biphenylyl-2-[p-(ss- diäthylamino-äthoxy)-phenyl]-propan-hydrochlorid in einer 2-n. wÏssrigen Sodalösung wird mit Essigester geschüttelt. Der Essigesterextrakt wird mit wässriger Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Destillation des zurückbleibenden Ols ergibt das 2-p-Biphenylyl-2-[p-(ss-diäthylamino-äth- oxy)-phenyl]-propan vom Kpo t 217-220 .
Beispiel 3
Zu einer Lösung von 12, 33 g 2-p-Chlorphenyl-2p-oxyphenyl-propan in einer Mischung von 75 cm3 Dimethylformamid und 40 cm3 Toluol gibt man unter Rühren und Kühlen portionenweise 2, 4 g einer 53% igen Mineralölsuspension von Natriumhydrid. Nach Beendigung der Wasserstoffentwicklung gibt man unter R hren und Kühlen tropfenweise eine Lösung von 7 g ¯-Di äthylamino-äthylchlorid in 35 cm3 Toluol zu. Dann rührt man bei Raumtemperatur drei Stunden weiter und lässt hierauf 15 Stunden stehen. Der anorganische Niederschlag wird abfiltriert, das Filtrat auf ein Volumen von ungefähr 50-80 cm3 konzentriert, mit Wasser verdünnt und mit 300 cml Ather extrahiert.
Der Atherextrakt wird mit 100 cm3 2-n. wÏssriger Salzsäure gewaschen und die saure Lösung abgetrennt. Nach einigen Minuten kristallisiert das 2-p-Chlorphenyl-2-[p-(diäthyl amino-äthoxy)-phenyl]-propan-hydrochlorid der Formel

Es wird abfiltriert und aus einer Mischung von Athanol und Ather umkristallisiert, F. 185-186 .
Das Ausgangsmaterial kann wie folgt erhalten werden :
Zu einem eisgekühlten Grignard-Reagens, hergestellt aus 284 g Methyljodid und 48, 6 g Magnesiumspänen in 550 cm3 Ather gibt man unter Rühren tropfenweise eine Lösung von 154, 6 g 4-Chloracetophenon in 200 cm3 Ather. Man lässt die Reaktionsmischung bei Raumtemperatur stehen und erhitzt sie dann während 2 Stunden am Rückfluss. Das Reaktionsprodukt wird nun unter Rühren und Kühlung in einem Eisbad langsam durch Zugabe von 300 cm3 gesättigter wässriger Ammonium chloridlösung und 300 cm3 Wasser zersetzt. Dann fügt man 300 cm3 Ather zu, trennt die organische Phase, wäscht sie und trocknet sie über Natriumsulfat.
Nach dem Abdampfen des Lösungsmittels wird der R ckstand destilliert. Das 2-p-Chlorphenyl-2-propanol destil- liert bei Kp. s 92-96 .
28, 2 g Phenol werden unter Rühren portionenweise mit 6, 7 g Aluminiumchlorid versetzt. Zu der so erhaltenen Mischung gibt man unter Rühren und Wasserkühlung eine Mischung von 17, 1 g 2-p-Chlorphenyl-2propanol und 9, 4 g Phenol. Nach beendeter Zugabe rührt man 2 Stunden bei Raumtemperatur, lässt 15 Stunden stehen und erhitzt unter Rühren eine Stunde auf 40-50 . Das Reaktionsprodukt wird unter Rühren zu 100 cm3 6-n. wÏssriger Salzsäure hinzugefügt. Die organische Phase wird abgetrennt und das überschüssige Phenol durch Destillation bis zu einer Temperatur von 130 (13 mm Hg) abdestilliert. Das zurückbleibende 61 wird destilliert, wobei man das 2-p-Chlorphenyl-2-poxyphenyl-propan vom F. 72-74 (aus Hexan) erhält.
Beispiel 4
Das durch Reaktion von 5, 6 g 2-p-Chlorphenyl-2 m-chlor-p-oxyphenyl-propan in 25 cm3 Dimethylform- amid und 25 cm3 Toluol mit 1 g einer 53% igen Na triumamidsuspension in Mineralöl erhaltene Natriumsalz wird mit einer Lösung von 3 g B-Diäthylamino- äthylchlorid in Toluol behandelt. Die Reaktionsmischung wird wie in Beispiel 3 aufgearbeitet. Man erhält so das 2-p-Chlorphenyl-2-[m-chlor-p-(¯-diÏthyl amino-äthoxy)-phenyl]-propan der Formel

welches durch Destillation gereinigt werden kann (Kp...., 170 ).
Das Ausgangsmaterial kann wie folgt erhalten werden :
Eine Mischung von 7, 41 g 2-p-Chlorphenyl-2-p- oxyphenyl-propan und 10 g Sulfurylchlorid wird auf dem Dampfbad 3 Stunden zum Rückfluss erhitzt. Hierauf verdampft man das überschüssige Sulfurylchlorid und nimmt den Rückstand in ¯ther auf. Die Ätherlösung wird mit wässriger Natriumbicarbonatlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Der strohfarbene Rückstand wird destilliert.
Man erhält so das 2-p-Chlorphenyl-2-m-chlor-p-oxyphenyl-propan vom Kp. o ig 165-170 .
In ähnlicher Weise erhält man : 2-p-Chlorphenyl-2* [m-chlor-p-(ss-diäthylamino-äth- oxy)-m'-methyl-phenyl]-propan, dessen Hydrochlorid bei 146-148 schmilzt. Das als Ausgangsmaterial ver wendete 2-p-Chlorphenyl-2 (-m-chlor-poxy-m'-methyl- phenyl)-propan, Kp.0,05 125-126¯ wird durch Reaktion von 2-p-Chlorphenyl-2-propanol mit o-Chlor-@-methyl- phenol in Gegenwart von Aluminiumchlorid und Rei nigung g iiber das Benzoat, F. 129-130 erhalten ;
2-p-Chlorphenyl-2-[p-(ss-diäthylaminoXäthoxy)-m, m'- dimethyl-phenyl]-propan, Kp.. 171-173 , Hydro chlorid F. 173-174 . Das entsprechende Phenol, F. 47 48 , wird durch Zugabe von 17, 1 g 2-p-Chlorphenyl-2propanol gefolgt von 12, 2 g 2, 6-Xylenol zu einer Mischung von 36, 6 g 2, 6-Xylenol und 6, 7 g Aluminiumchlorid erhalten.
2-p-Biphenylyl-2-[m-chlor-p ; @1-diäthylamino-äth- oxy)-phenyl]-propan, Kp. 008 207-210 . Das als Aus gangsstoff verwendete Phenol, Kp. oo 180-185 , wird durch Reaktion von 2, 72 g 2-p-Biphenylyl-2-p-oxyphenyl-propan mit 1, 8 g Sulfurylchlorid und 10 ces Benzol erhalten.
2- [p- (33-Diäthylamino-äthoxy)-phenyl] ;- 2-p-methyl- phenyl-propan, dessen Hydrochlorid nach Umkristallisation aus Aceton bei 188-189 schmilzt. Das als Aus gangsstoff verwendete Phenol (F. 69-70 nach Kristallisation aus Pentan) wird hergestellt durch Zugabe von 15 g 2-p-Methyl-phenyl-2-propanol zu einer Mischung von 6, 7 g Aluminiumchlorid und 19 g Phenol.
Beispiel 5
Zu einer Lösung von 5 g 2-p-Chlorphenyl-2-p-oxyphenyl-propan in 100 cm3 einer 1 : 1-Mischung von Dimethylformamid und Toluol gibt man 3, 8 g frisch kristallisiertes ss-Piperidino-äthylchlorid-hydrochlorid.
Die erhaltene Mischung wird auf 5 abgekühlt und in kleinen Portionen mit insgesamt 1, 95 g einer 53 /0igen Mineralölsuspension von Natriumhydrid behandelt.
Nach beendigter Zugabe wird die Reaktionsmi- schung 5 Stunden bei Raumtemperatur gerührt und dann während 15 Stunden stehen gelassen. Der organische Niederschlag wird abfiltriert, das Filtrat auf 15 cml eingedampft und mit Wasser verdünnt. Das basische Material wird mit 1-n. Salzsäure extrahiert, die sauren Extrakte mit fester Soda alkalisch gestellt und mit Ather extrahiert. Beim Eindampfen hinter lässt die ätherische Lösung das 2-p-Chlorphenyl-2- [p (ss-piperidino-äthoxy)-phenyl]-propan der Formel

Das Ausgangsmaterial wird wie folgt erhalten :
Zu einer Mischung von 13, 4 g Aluminiumchlorid und 22 g Katechol gibt man unter Rühren in Portionen 22 g 2-p-Methyl-phenyl-2-propanol. Die erhaltene Suspension wird mit 150 cm Hexan verdünnt, die Reaktionsmischung auf 80 erhitzt, durch Zugabe von 100 cm3 2-n. Salzsäure zersetzt und unter Rühren auf dem Dampfbad während 15 Minuten erhitzt. Die wäss- rige Phase wird abgetrennt und verworfen, während die organische Phase mit 100 cm3 warmem Wasser gewaschen, hierauf mit 100 cm3 Ather verdünnt und dann mit 100 cm3 2'voliger Natronlauge extrahiert wird.
Der wässrige Extrakt ergibt durch Ansäuern das 2-m, p Dioxy-phenyl-2-p-methyl-phenyl-propan vom F. 84 85 .
Das 2-p-Biphenylyl-2-[m, p-bis-(ss-diäthylamino-äth- oxy)-phenyl]-propan vom Kp. 005 219-223 wird durch Reaktion des Natriumsalzes von 2-p-Biphenylyl-2-m, p dioxy-phenyl-propan mit 2, 4 g ss-Diäthylamino-äthyl- chlorid in 60 cm3 einer 1 : 1-Mischung von Dimethyl- formamid und Toluol erhalten ; es siedet bei 0, 05 mm Hg bei 219-223 .
Der Ausgangsstoff wird erhalten durch Behandlung von 11 g Katechol und 7 g Aluminiumchlorid mit 10, 6 g 2-p-Biphenylyl-2-propanol in 150 cml Hexan und Reaktion von 3 g des erhaltenen 2-p-Biphenylyl-2-m, p-dioxy-phenyl-propans (F. 101-102 nach Umkristallisation aus Benzol/Pentan) mit 1, 04 g einer 53"/oigen Mineralölsuspension von Natriumhydrid.
Beispiel 7
Eine Lösung von 2, 7 g 2-[p-Chlorphenyl-2-[p-(¯ piperidino-äthoxy)-phenyl]-propan-hydrochlorid in 25 cm3 Wasser wird mit Natriumcarbonat basisch gestellt.
Die freie Base wird mehrmals mit Ather extrahiert und die vereinigten Extrakte mit einer wässrigen Natrium chloridlösung gewaschen, über Natriumsulfat getrocknet und zur Trockene eingedampft. Der zurückbleibende ölige Rückstand wird in 15 cm3 Methanol gelöst und mit 2 cm3 einer 30 /oigen wässrigen Wasserstoffperoxyd- Lösung versetzt. Hierauf lässt man die Lösung bei Raumtemperatur 90 Stunden stehen und zersetzt dann den Cberschuss an Wasserstoffperoxyd durch Zugabe von 0, 05 g eines Palladiumkatalysators (10 /o Palladium auf Kohle).
Nun filtriert man vom Katalysator ab und dampft das farblose Filtrat zur Trockene, wobei das N-Oxyd des 2-p-Chlorphenyl-2 [-p-(, ss-piperidino-äthoxy- phenyl]-propan als farbloses 51 zurückbleibt. Wenn man eine äthanolische Lösung des N-Oxyds mit Pikrinsäure behandelt, so erhält man das Pikrat vom F. 118-120 .
Beispiel 8
Zu einer Lösung von 11, 5 g 2-p-Brom-phenyl-2-p- oxy-phenyl-propan (F. 89-90 ) in 100 ml einer 1 : 1 Mischung von N, N-Dimethyl-formamid und Toluol gibt man 7, 2 g ss-Chloräthyl-piperidin-hydrochlorid. Man rührt die Reaktionsmischung, kühlt sie in einem Eisbad und gibt 3, 6 g einer 53"/cigen Suspension von Natrumhydrid in einem Mineralöl portionenweise zu. Nach dem Abklingen der Wasserstoffentwicklung rührt man 6 Stunden bei Raumtemperatur, lässt dannn weitere 15 Stunden bei Raumtemperatur stehen und dampft unter vermindertem Druck ein. Der Rückstand wird in Wasser aufgenommen, 2 mal mit Diäthyläther extrahiert und die vereinigten Extrakte mit 2-n. Salzsäure gewaschen.
Man stellt unter Kühlen im Eisbad die Salzsäurephase mit 50 /oiger wässriger Natriumhydroxydlö- sung und Natriumcarbonat auf ein pH von ungefähr 8 ein, extrahiert mit Diäthyläther, trocknet die ätheri- sche Lösung über Natriumsulfat und dampft zurTrockne ein. Das so erhaltene 2-p-Brom-phenyl-2-[p-(ss-piper- idino-äthoxy)-phenyl]-propan der Formel

Example 1
To 3.8 g of 2-p-biphenylyl-2-p-oxyphenyl-propane in 8 cm3 of dimethylformamide, 0.63 g of a 53% mineral oil suspension of sodium hydride is added. A solution of 1.78 g of ß-diethylaminoethyl chloride in 8 cm3 of toluene is added to the resulting suspension with stirring. The reaction mixture is warmed to about 50 in order to obtain a clear brown solution, stirring is continued for 2 hours at room temperature and then left to stand for 15 hours.

The starting material is obtained as follows:
While stirring and heating in a steam bath, a mixture of 8.05 g of molten phenol and 1.77 g of aluminum chloride is added in portions with a molten mixture of 5.7 g of a, a-dimethyl-p-phenylbenzyl alcohol and 3 g of phenol. Stirring on the steam bath is continued for a further hour, whereupon the reaction product with ice and conc. Hydrochloric acid decomposes. Then it is extracted three times with ethyl acetate, the combined ethyl acetate extracts with conc. Washed brine, dried over anhydrous sodium sulfate and evaporated under reduced pressure. The remaining 61 is distilled. The fraction bp. 08 185-205 crystallizes spontaneously and is triturated with pentane.
This gives 2-p-biphenylyl-2-p-oxyphenyl-propane of F. 119120.
Example 2
A suspension of 3.5 g of 2-p-biphenylyl-2- [p- (ss- diethylamino-ethoxy) phenyl] propane hydrochloride in a 2-n. aqueous soda solution is shaken with ethyl acetate. The ethyl acetate extract is washed with aqueous sodium chloride solution, dried over sodium sulfate and evaporated. Distillation of the remaining oil gives the 2-p-biphenylyl-2- [p- (ss-diethylamino-ethoxy) -phenyl] propane of Kpo t 217-220.
Example 3
To a solution of 12.33 g of 2-p-chlorophenyl-2p-oxyphenylpropane in a mixture of 75 cm3 of dimethylformamide and 40 cm3 of toluene, 2.4 g of a 53% mineral oil suspension of sodium hydride are added in portions with stirring and cooling. After the evolution of hydrogen has ceased, a solution of 7 g of ¯-diethylaminoethyl chloride in 35 cm3 of toluene is added dropwise with stirring and cooling. The mixture is then stirred for a further three hours at room temperature and then left to stand for 15 hours. The inorganic precipitate is filtered off, the filtrate is concentrated to a volume of approximately 50-80 cm3, diluted with water and extracted with 300 cml of ether.
The ether extract is 100 cm3 2-n. Washed aqueous hydrochloric acid and separated the acidic solution. After a few minutes, the 2-p-chlorophenyl-2- [p- (diethylamino-ethoxy) phenyl] propane hydrochloride of the formula crystallizes

It is filtered off and recrystallized from a mixture of ethanol and ether, F. 185-186.
The starting material can be obtained as follows:
A solution of 154.6 g of 4-chloroacetophenone in 200 cm3 of ether is added dropwise with stirring to an ice-cold Grignard reagent, prepared from 284 g of methyl iodide and 48.6 g of magnesium turnings in 550 cm3 of ether. The reaction mixture is left to stand at room temperature and then refluxed for 2 hours. The reaction product is then slowly decomposed by adding 300 cm3 of saturated aqueous ammonium chloride solution and 300 cm3 of water while stirring and cooling in an ice bath. 300 cm3 of ether are then added, and the organic phase is separated, washed and dried over sodium sulfate.
After the solvent has evaporated, the residue is distilled. The 2-p-chlorophenyl-2-propanol distills at bp s 92-96.
28.2 g of phenol are mixed in portions with 6.7 g of aluminum chloride while stirring. A mixture of 17.1 g of 2-p-chlorophenyl-2propanol and 9.4 g of phenol is added to the mixture thus obtained, with stirring and with water cooling. When the addition is complete, the mixture is stirred for 2 hours at room temperature, left to stand for 15 hours and heated to 40-50 for one hour while stirring. The reaction product becomes 100 cm3 6-n with stirring. aqueous hydrochloric acid added. The organic phase is separated off and the excess phenol is removed by distillation up to a temperature of 130 (13 mm Hg). The remaining 61 is distilled, whereby 2-p-chlorophenyl-2-poxyphenyl-propane with a melting point of 72-74 (from hexane) is obtained.
Example 4
The sodium salt obtained by reacting 5.6 g of 2-p-chlorophenyl-2-chloro-p-oxyphenylpropane in 25 cm3 of dimethylformamide and 25 cm3 of toluene with 1 g of a 53% strength sodium amide suspension in mineral oil is treated with a Treated solution of 3 g of B-diethylamino ethyl chloride in toluene. The reaction mixture is worked up as in Example 3. The 2-p-chlorophenyl-2- [m-chloro-p- (¯-diÏthylamino-ethoxy) -phenyl] -propane of the formula is obtained in this way

The starting material can be obtained as follows:
A mixture of 7.41 g of 2-p-chlorophenyl-2-p-oxyphenyl-propane and 10 g of sulfuryl chloride is refluxed on the steam bath for 3 hours. The excess sulfuryl chloride is then evaporated and the residue is taken up in ether. The ether solution is washed with aqueous sodium bicarbonate solution, dried over sodium sulfate and evaporated. The straw-colored residue is distilled.
This gives 2-p-chlorophenyl-2-m-chloro-p-oxyphenyl-propane with the bp 165-170.
The following is obtained in a similar manner: 2-p-chlorophenyl-2 * [m-chloro-p- (ss-diethylamino-ethoxy) -m'-methyl-phenyl] propane, the hydrochloride of which melts at 146-148. The 2-p-chlorophenyl-2 (-m-chloro-poxy-m'-methyl-phenyl) -propane, boiling point 0.05 125-126¯, is used as starting material by reaction of 2-p-chlorophenyl-2 -propanol with o-chloro - @ - methylphenol in the presence of aluminum chloride and purification via the benzoate, mp 129-130;
2-p-Chlorophenyl-2- [p- (ss-diethylaminoXethoxy) -m, m'-dimethyl-phenyl] -propane, bp. 171-173, hydrochloride F. 173-174. The corresponding phenol, m.p. 47 48, is made by adding 17.1 g of 2-p-chlorophenyl-2propanol followed by 12.2 g of 2,6-xylenol to a mixture of 36.6 g of 2,6-xylenol and 6 , 7 g of aluminum chloride obtained.
2-p-biphenylyl-2- [m -chloro-p; 1-diethylamino-ethoxy) phenyl] propane, bp 008 207-210. The phenol used as starting material, bp. Oo 180-185, is obtained by reacting 2.72 g of 2-p-biphenylyl-2-p-oxyphenyl-propane with 1.8 g of sulfuryl chloride and 10 ces benzene.
2- [p- (33-Diethylamino-ethoxy) -phenyl]; -2-p-methyl-phenyl-propane, the hydrochloride of which melts at 188-189 after recrystallization from acetone. The phenol used as starting material (F. 69-70 after crystallization from pentane) is prepared by adding 15 g of 2-p-methyl-phenyl-2-propanol to a mixture of 6.7 g of aluminum chloride and 19 g of phenol.
Example 5
3.8 g of freshly crystallized ß-piperidinoethyl chloride hydrochloride are added to a solution of 5 g of 2-p-chlorophenyl-2-p-oxyphenylpropane in 100 cm3 of a 1: 1 mixture of dimethylformamide and toluene.
The mixture obtained is cooled to 5 and treated in small portions with a total of 1.95 g of a 53/0 mineral oil suspension of sodium hydride.
When the addition is complete, the reaction mixture is stirred for 5 hours at room temperature and then left to stand for 15 hours. The organic precipitate is filtered off, the filtrate is evaporated to 15 cml and diluted with water. The basic material is denoted by 1-n. Hydrochloric acid extracted, the acidic extracts made alkaline with solid soda and extracted with ether. When evaporating, the ethereal solution leaves the 2-p-chlorophenyl-2- [p (ss-piperidino-ethoxy) -phenyl] -propane of the formula

which can be converted into its hydrochloride by treatment with a hydrochloric acid ether solution.
After recrystallization from a 5: 1: 5 mixture of acetone, ethanol and ether, this melts at 183 185.
Example 6
To a solution of 4 g of 2-m, p-dioxyphenyl-2-pmethyl-phenyl-propane in dimethylformamide, 1.72 g of a 53/0 mineral oil suspension of sodium hydride and then 4 g of s-diethylamino-ethyl chloride dissolved in 80 cm3 of a 1: 1 mixture of dimethylformamide and toluene. The reaction is carried out as in Example 1 and gives 2- [m, p-bis (ss-diethylamino-ethoxy) phenyl] -2-p-methylphenylpropane of the formula

which is distilled at 0.05 mm 175-180 and distilled a second time at 0.05 mm 178-180.
The starting material is obtained as follows:
22 g of 2-p-methylphenyl-2-propanol are added in portions to a mixture of 13.4 g of aluminum chloride and 22 g of catechol, while stirring. The suspension obtained is diluted with 150 cm of hexane, the reaction mixture is heated to 80 by adding 100 cm3 of 2-n. Hydrochloric acid decomposes and heated on the steam bath for 15 minutes while stirring. The aqueous phase is separated off and discarded, while the organic phase is washed with 100 cm3 of warm water, then diluted with 100 cm3 of ether and then extracted with 100 cm3 of 2 'volume sodium hydroxide solution.
Acidification of the aqueous extract gives 2-m, p-dioxy-phenyl-2-p-methyl-phenyl-propane with a melting point of 84.85.
The 2-p-biphenylyl-2- [m, p-bis- (ss-diethylamino-ethoxy) -phenyl] -propane of bp. 005 219-223 is produced by reaction of the sodium salt of 2-p-biphenylyl-2 -m, p dioxy-phenyl-propane with 2.4 g of ss-diethylamino-ethyl chloride in 60 cm3 of a 1: 1 mixture of dimethylformamide and toluene; it boils at 0.05 mm Hg at 219-223.
The starting material is obtained by treating 11 g of catechol and 7 g of aluminum chloride with 10.6 g of 2-p-biphenylyl-2-propanol in 150 cml of hexane and reacting 3 g of the 2-p-biphenylyl-2-m, p obtained -dioxy-phenyl-propane (F. 101-102 after recrystallization from benzene / pentane) with 1.04 g of a 53 "per cent mineral oil suspension of sodium hydride.
Example 7
A solution of 2.7 g of 2- [p-chlorophenyl-2- [p- (¯ piperidino-ethoxy) -phenyl] -propane hydrochloride in 25 cm3 of water is made basic with sodium carbonate.
The free base is extracted several times with ether and the combined extracts are washed with an aqueous sodium chloride solution, dried over sodium sulfate and evaporated to dryness. The oily residue that remains is dissolved in 15 cm3 of methanol, and 2 cm3 of a 30% aqueous hydrogen peroxide solution are added. The solution is then left to stand at room temperature for 90 hours and the excess hydrogen peroxide is then decomposed by adding 0.05 g of a palladium catalyst (10% palladium on carbon).
The catalyst is then filtered off and the colorless filtrate is evaporated to dryness, the N-oxide of 2-p-chlorophenyl-2 [-p- (, ß-piperidino-ethoxyphenyl] -propane remaining as colorless 51 Treating an ethanolic solution of the N-oxide with picric acid, the picrate of F. 118-120 is obtained.
Example 8
To a solution of 11.5 g of 2-p-bromo-phenyl-2-p-oxy-phenyl-propane (F. 89-90) in 100 ml of a 1: 1 mixture of N, N-dimethylformamide and toluene are 7.2 g of ss-chloroethyl piperidine hydrochloride. The reaction mixture is stirred, cooled in an ice bath and 3.6 g of a 53 "/ cigen suspension of sodium hydride in mineral oil are added in portions. After the evolution of hydrogen has subsided, the mixture is stirred for 6 hours at room temperature, then left to stand for a further 15 hours at room temperature and evaporated under reduced pressure, the residue is taken up in water, extracted twice with diethyl ether and the combined extracts are washed with 2N hydrochloric acid.
The hydrochloric acid phase is adjusted to a pH of approximately 8 with 50% aqueous sodium hydroxide solution and sodium carbonate, while cooling in an ice bath, extracted with diethyl ether, the ethereal solution is dried over sodium sulfate and evaporated to dryness. The 2-p-bromo-phenyl-2- [p- (ss-piper- idino-ethoxy) -phenyl] -propane of the formula obtained in this way
