Verfahren zur Herstellung von Penicillinen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung vom Pevnicilien durch Aeylje. rung von 6-Amino-penicillansÏure mit CarbonsÏurehalogeniden in einem wässrigen Reaktionsgemisch bei saurem pH Wert und in Gegenwart eines mit Wasser nicht mischbaren onganischen L¯sungsmittels.
Penicilline, die nach dem erfindunggemiassen Verfahren hergestaHt weaden können, sind z. B. solche, wie sie in den Schwerer Pabenbschriften Nr. 358 42. 0, 394 205 und 401 976 genannt sind. Bei der Herstellung in wÏssrigen Medien wird, wie in den genannten Patentschriften erwÏhnt ist, im pH-Bereich zwischen 6 und 9 gearbeitet, was. d. urch Zugabe eines Puffers, wie Natriumbicarbonat, währamd dsr Dauer der Reaktion zwischen dam Säurehalogenid mit der 6-Amino-penicillansÏure erreicht wind.
Die zur Zelit geltende Lehma fordert, daias bei der Herstellung von Amiden neutrale oder alkalische Bedingungen eingehalten werden m ssan. So lehrt z. B.
Hiokinbottom [Rfaacttion of Organic Compounds, Longmans, Green and Co., New York (1938), Seiten 262263], dass ein ganzes Mol ¯berschuss an Amin zur Umsetzung mit einem SÏurechlorid in einem wasser- freien organischen L¯sungsmittel angewendet werden muss. Wenn dagegen die Reaktion in Gagenwart einer verd nnten wÏssrigen Alkalil¯sung nach Art der Schot- ten-Baumann-Technik durchgef hrt wird, fordert Hickingbottom: ¸Es ist wesentlich, dass die wÏssrige L¯smng wÏhrend der ganzen Acylierungsreaktion alkalisch Hssibenmuss.
Nach dem erfindungsgemÏssen Verfahren k¯nnen auf vereinfachte Weise Peniciline mit verbesserten Ausbeuten hergestellt werden.
Bei den bekannten im technischen Massstab durchgef hrten Verfahren zur Acylierung von 6-AminopenicillansÏure wird die wÏssrige L¯sung der letzteren, wie sie durch PrÏ@ursor-freie Fermentation oder enzymatische Hydrolyse erhaltan worden ist, zunÏchst von ihrem nah@zu neutralen pH-Wert auf stark sauren pH-Wert, z. B. auf pH-2 durch Zugabe einer SÏure eingestellt. Hernach wird die L¯sung stehengelassen oder mit einem mit Wasser nicht mischbaren L¯sungsmittel extrahiert, um allfÏllig darin enthaltene nat rliche Penicilline einschliesslich Benzylpenicillin oder PhenylessigsÏure zu entfernen. Hernaiah wind idie Lösung durch Zugabe enier grossen Menge von Alkali auf den gew nschten pH-Bereich von 6-9, vorzugsweise 7, gebracht.
WÏhrend der Acylierung mit dem SÏurechlorid ist es dann weiterhin erforderlich, Alkali zuzugeben, um den pH-Wert bei ungefÏhr 7 zu halten. Nach Ende dsr Reaktion wenden da. nn waitechmi'gnosse Mengen Säuren zugegeben zur Einstellung des pH-Wertes von 2, worauf das entstandene Penicillin aus dem wässrigen Reaktionsgemisc h mit einem mit Wasser nicht mischbaren L¯sungsmittel zu extrahieren und aus letzterem zu iso Nerem. ist.
Dieses bekannte Verfahren erfordert demgemÏss die nacheinanderfolgende Einstellung des pH-Wertes von mitral hinab auf 2, dann wieder hinauf auf ungefÏhr 7, die Einhaltung des letzteren Wertes wÏhrend der Reaktion und schliesslich wiedar die VerÏnderung des pH WeTjts hiniab auf 2. Dias erfindungsgemÏsse Veirfahren erm¯glicht es, diese Operationen zu vereinfachen und damit Kosten einzusparen, indem die letzteren beiden pH-Regulierungen eliminisTt weitdan.
Bei semer Anwendung in. groasem M'assstab. arfor- dert das bekannte Verfahren ebenso eine kontinuierliche dosierte Zugabe an Alkali wÏhrend der Acylierungsreaktion, um den gew nschten pH-Wert aufrecht zuerhattan. Ziel der Erfindung ist. äs deswegen weiterhin, diesen aufwendigen, viel Arbeit erfordernden Schritt ein zusparen und damit die Herstellumgsreaktian einfacher zu gestalten. Schliesslich erforderte das im praktischen Massstab durchgef hrte bekannte Verfahren weiterhin ein in L¯sungbringen des in Wasser nicht l¯slichen SÏurechlorids im wassrigen Reaktionsgemisch vermittels seiner Zugabe in Form einer L¯sung in einem mlit Was- ser r mischbaren L¯sungsmittel, wie z. B. Aceton.
Hieraus erwuchsen h¯here Kosten f r das L¯sungsmittel und weiterhin eine Verunreinigung des Extraktionsmittels f r Penicillin mit Aceton, was eine aufwendige Tren nung erforderlich. machte, bevor das L¯sungsmittel er naut verwendet werden konnte. Ziel der vorlieigenden
Erfindiung ist deswegen weiterhin die aufwendige und st¯rende Notwendigkeit zum Gebrauch eines L¯sungs mittels f r das SÏurechlorid zu beseitigen.
Das erfindungsgemÏsse Verfahren zur Herstellung vol Penicillinen ist dadurch gekennzeichnet, dass man
6-Amino-penicillansÏure mit mindestens einem ¯qui valent eines CarbonsÏurechlorids, -bromids oder -jodids bei stars aauaiem pH-Wert und einer Temperatur von 0 bis SO C in emeni nem Reaktionsmedium mischt, das eine instabile Emulsion von Wasser und einem mit Wasser nicht mischbaren inerten organischen L¯sungsmittel dar stellt, worauf die das entsandene Penicillin enthaltende organische Phase abgetrennt und das Penicillin danaus isoliert wird.
Vorteilhafterweise arbeitet man dabei der art, dass mam ein ¯quivalent 6-Amino-penicillansÏure mit ungefÏhr 1,0 bis 4,5 ¯quivalenten des umzusetzen den SÏurechlorids, -bromids oder -jodids mischt und im wÏssrigen Reaktionsmedium iinen pH-Wert von 2 bis 4, vorizssweisle von ungefÏhr 2, einstellt. Vorteilhaft ver wendet man : als mit Wasser nicht mischbare organische
L¯sungsmittel zur Erzeugung einer instabilen Emulsion, . aliphatischer Ester und Ketone, wie insbesondere Methy1-isobutylketon und niedere Alkylacetate. Als g nstigstes Temperaturintervall f r die durchzuf hrende Reak tion empsseMtt siah 20 bis 35 C.
D'as Volumen des organischen L¯sungsmittels wird vorteilhafterweise zu ungefÏhr einem Viertel bis einem halben Teil des Volumens der wÏssrigen Phase bemessen, en, um einerseits seine Kosten zu sparen und anderseits eine kionzentniertie Lösiunjgdes.eniMandenenPeniciNins zu erhalten. Dieses VolumenverhÏltnis ist jedoch f r den erfindungsgemÏssen Zweck nicht von Bedeutung, und es kann sich in gewissen FÏllen sogar als vorteilhaft erweisen, gr¯ssere Volumina an organischem L¯sungsmittel anzuwenden als die wÏssrige Phase mit der darin enthaltenen 6-Amino-penicillansÏure. Wenn z.
B. die Kon- zentration der 6-Amino-penicillansÏure in der wÏssrigen L¯sung gr¯sser als 5 mg/ml ist, ist es von Vorteil, ein Volumen an organischem L¯sungsmittel anzuwenden, das gleich oder sogar doppelt so gross ist, wie dasjenige der wÏssrigen Phase.
In den meisten FÏllen ist es wichtig, dass das Reaktionsgemenge krÏftig und kontinuierlich ger hrt oder gesch ttelt wird, so dass wÏhrend der Dauer der Reaktion eine unstabile Emulsion besteht.
Geeignete CarbonsÏurechloride sind z. B. solche, welche mit der 6-AminopenicillansÏure schneller reagieren, als mit Wasser. Die letztere Bedingung lÏsst sich durch einen einfachen Test leicht feststellen. Beispiele derartiger Penicilline sind α-Phenoxyalkylpenicillin, z. B α-Phenoxybutylpenicillin, α-phenoxybenzylpenicil- lin, 2-Biphenylpenicillin und Isoxazolpenicilline, z. B.
5-Methyl-3-phenyl-4-isoxazolylpenicillin und 3-(o Chlor-phenyl)-5-methyl-4-isoxazolylpenicillin, sowie deren nicht-toxische Salze. Deraritige nicht-toxische Me taNs'aJzesindz.B.solchevonNatrium,Kalium,Calcmm , und AluminiMm,feEnfecrfdioAmmpniumsaJjzBundsbsti.- tuierte Ammoniumsalze, z. B. Sializo nicht-toxischer Amine, wie Trialkylamine, einschliesslich TriÏthylamin, Procain, Dibenzylamin, N-Benzyl-beta-phenetylamin, 1-Ephenamin, N,N'-DibenzylÏthylediamin, Dehydroabietylamin, N,N'-bis-DehydroabietylÏthylendiamin, niedrige N-Alkylpiperidine, z. B. N-¯thylpiperidin und andere Amine, wie sie bisher zur Bildung von Salzen mit Benzylpenicillin Verwendung gefunden haben.
Die 6-AnNn) pnicillansÏ re, wie ssie im erfindungsgemÏs sen Verfahren verwendet wird, kann als feste Substanz in Fo !rm'ainaswässrijgenKonjzentra < !asodar.ailsBestand- teil einer Fermentationsbr he hergestellt werden, z. B. nach Batchelor et al. (Nature 183, 257-258, January
24, 1959), oder sie kann als feste Substanz bzw. als wÏssrige L¯sung durch enzymatische Hydrolyse von
Penicillinen, wie Benzylpenicillin und Phenoxymethyl piciiiic hertgesteRt wanden, z. B. entsprechend Rolin son et al. (vgl. US-Patentschrift Nr. 3014845 und 3014846) (Nature 187, 236, 1960), Claridge et al. (Na tune 187, 237, 1960), Huang et al (J. Amies. Chíem. Soc.
82, 3790, 1960) und Kaufmann et al. (Naturewissenschaften 47, 474, 1960). Ein besonderer Vorteil des erfindungsgemÏssen Verfahrens ist es, dass es angewendet wenden kann zur Acylierung von 6-Amino-penicillan sÏure, wie sie aus Fermentationsbr hen oder enzymatischen Hydrolysaten ohne Reinigung oder Konzentrierung erhalten wird. Bevor derartige L¯sungen von
6-Amino-penicillansÏure, zur Umsetzung gelangen, sit es vorteilhaft, jedoch nicht erforderlich, f'este Anteile, wie Mycelien oder Zellen durch Zentrifugieren oder Filtrieren abzutrennen und allenfalls vorhandene Penicilline zu entfernen oder zu zerst¯ren, sowie SÏuren, wie PhenylessigsÏure oder PhenoxyessigsÏure abzutrennen, was in einfacher Weise durch Extraktion bei saurem pH Wert mit einem mit Wasser nicht mischbaren organischen L¯sungsmittel geschehen kann.
Bei der Durchf hrung des erfindungsgemÏssen Verfahrens kann der pH-Wert der Aktivierungsreaktion im gew nschten stark sauren Bereich durch Zugabe von Alkali gehalten werden. Im Allgemeinen wird eine derartige Alkalizugabe notwendig, wenn reine wÏssrige L¯sungen von 6-AminopinicillansÏure oder enzymatische Hydrolysate verwendet werden. Sie ist jedoch nicht erforderlich, wenn PrÏcursor-freie Penicillin-Fermentationsbr hen verwendet werden. Eine bevorzugte Ausf hrungsform des erfindungsgemÏssen Verfahrens besteht darin, dass gleichzeitig das SÏurechlorid, das Methyl-isobutylketon und eine fil@rierte PrÏcursor-freie Penicillin-Fermentations- br he vom pH-Wert 2 gemischt werden. Dabei ergibt sich eine sehr schnelle Reaktion mit einem Reaktions abllaitTif inder GrösaeMomdnMng von z.
B. 5 Mimutem. Hiernach kann die aktivierte Emulsion in eine Zentrifuge bergef hrt werden, worin die organische Phase mit dem PeniiciMin abgeitrennt wird.
Ms inenhe'organische LösuMgsmiittel sind solche zu verstehen, welche unterhalb 50¯ mit den verwendeten SÏurechloriden, -bromiden oder -jodiden nicht reagieren, noch sich mit 6-Amino-penicillansÏure oder mit den entstandenen Penicillinen umsetzen, was durch ein- fache Teste leicht festzustellen ist.
BetrÏchtlich kleinere Ausbeuten am gew nschten Produkt in isolierter fester Form ergeben sich, wenn das L¯sungsmittel wÏhrend des Aktivierungsschritts nicht zugagen ist.
Faillus die Acylierung durchgef hrt wird unter Verwendung eines SÏurechlorides, welches ein asymmetri sches Koh@enstoffatom enthÏlt, entsteht bei Verwen- dung einer racemischen SÏure ein Penicillin in Form einer Mischung vom 2 Diastereoisomeren. Beide erfindungsgemÏss erhÏltlichen Isomere sind biologisch. aktiv.
Die einzelnen Isomeren k¯nnen in reiner Form hergestellt werden, indem man von der reinen Dextro- oder LÏvoform der SÏure ausgeht, oder indem man die aus der racemischen SÏure entstandene Mischung auf physikalischem Wege trennt.

Nachdem'das ReakttOBiagemenge 30 Miniuben lian,- ger hrt worden war, wurde die Methyl-isobutylketon phaiae,welche!dasa-PhenoxyäthylpsniciNinenthislit, abgetrennt und letzteres daraus direkt als Kaliumsalz durch Zugabevon-ungeirähreinemZwanzigstel-Volumenan koniZientriemterwässgerKalumaaetatlösung (pH 8. 2 bis 8, 7, speziirisches Gewicht 1, 32 bis 1, 34 bei 20 C) aus- gefällt.

Chlorid unter R hren hinzugef gt. Der pH-Wert verblieb bei 2 ohne Zugabe von Natriumhudrox. Nachdem die Reaktions- oder ¸Aktivierung¯-Mischung bei 35¯ C 'eine Stunde lang gehalten worden war, wurde die Emul- sion filtriert.
Das Methyl-isobutylketon mit dem darin enthaltenen 5-Methyl-3-phenyl-4-isoxazolpenicillin wurde abgetrennt und bei zimmertemoeratur mit 350 ml angenähertl,5"/oi.garNaHGOg-RuKerlösung10Mirnuten lamgb'eimpH-Waot 7-8 extnahiefnt. De, dasgewüniaahte ProduktmFomm.ainosNaitriumsalzesenthaltendeBi- carb : onat Puffeir wurde abgetrennt und hei Zimmeritem- pera;tu!rmit200mlMethyl-isojbUttyilibetssonbeiimpH-Wart 2 funf Minuten lang extrahiert. Der Methyl-isobutyl Ketonextrakt wurde abgetrennt und zur Entfernung von nicht gel¯stem Wasser zentrifugient.
Zum Extrakt wurden nunmehr 2 Mol Natrium-2-Ïthylhexanoat (als 22% igeLös,ungmtcoabetnjemMatli'yil-isohutylbetOin.hiaizu- gef gt). Im Verlauf von 2 kniden Stehens bei Zimmer .tempe.natm'jEeldasNa:trium-5-m.e)f)iyl-3-pihenyl-4-isssx- azolyl-penicillin-monohydrat in kristalliner Form aus und wurde abfiltrient. Zur Reinigung wurde es mit Mefhyl-Tohutylke'tomuodhiefmfaobzweimjalmift:Acetom gewasch'an..
Die im vorstehenden beschriebene Verfahrensweise wurde mit verschiedenon ¯nderungen wiederholt. So wunde das Methyl-isobutylketon ersetzt durch ¯thylacetat. Die verwendeten filtrierten Fermentatinsbr hizin enthielten als Ausgangsprodukt 1300 bis 2110 mcg/ml 6-Amino-penicillansÏure. Das VerhÏltinis vom L¯sungsmittel zum Br hvolumen wurde variiert von 25 bis35 C.DasmolareVerhältnisvomSämecMon.d zu 6-Amino-penicillansÏure varié'né von 1, 5 bis 4, 7.
Das molare VerhÏltnis von Natrium-2-Ïthylhexanoat zu SÏurechlorid wurde variiert von 8, 0 bis 2, 4. In jedem Fall wurde bei den derart durchgef hrten Verfahren feste Produkte hoher Reinheit erhalten in Ausbeuten, die zwischen 55 und 80 0 O/o lajgen.

WÏssrige L¯sungen, die 10000 mcg/ml kristalline 6-'Amino-pemciMansäuroenftihieiHen,wurdenmitMine- nalisäune'aufdenpH-Wefnt2!emgasteHit.unjdübeT'scihich- tet mit 0,25 bis 1,0 Volumenteilen Athylacetat oder Methyl-isobutylketon. F r jedes vorhandene Mol 6-Amino-penicillansÏure wurden hiernach 1,2 bis 2,0 Mole 5-Methyl-3-phenyl-isoxazol-4-carbonylchlorid un ?]'Rühpenhinzugefügt.DerpH-Wertwurdeduroh Zugabe von Natriumhydroxyd auf 2 gehalten.
Nachdem das Reaktionsgemenge eine Stunde lang bei 25 bis 35'C bslassenwomdenwafr,.wurde,diieEmuJisionfiltriert.Die organische Phase wunde'abgetpennt und zmr Entfernung von Wasserzefntriifugieit.He-naachwurden.zuJhr2Mole Natrium-2-Ïthylhexanoat, gel¯st im gleichen L¯sungsmittel, bei einer Konzentration von ungefÏhr 22 %hinzugef gt. Das Natrium-5-Methyl-3-phenyl-4-isoxazolylpenicillin-monohydrat fiel in kristalliner Form und hoher Reinheit in Ausbeuten ; von 75 bis 97%aus.
Die hierbei im einzelnen eingehaltenen Bedingungen und Resultate waren die folgenden:

Hierbei war die notwendige Zugabe an Natronlauge nach den ersten 30 Minuten sehr gering.
Daraus ist zu entnehmen, dass die Aktivierungsreakion praktisch innerhalb der ersten halben Stunde komplett ablief. Der pH-Wert wurde hiernach durch Zugabe von Mineralsäure auf 2 eingestellt und das Methyl-isobutyl keton, welches das 3- (o-Chlor-phenyl)-5-methyl-4-isox- azolyl-penicillin enthielt, abgetrennt, mit 2 g Aktivkohle geklärt und filtriert. Wurde gemäss der Arbeitsweise von Beispiel 6 Natrium-2-äthylhexonat in trockenem Methyl-isobutylketon zu der Methyl-isobutylketonlösung hinzugefügt, so fiel das Endprodukt in einer Ausbeute von mehr als 85 O/o der Theorie als Natriumsalz in fester Form und hoher Reinheit aus.
