Verfahren zur Herstellung von 10-Hydroxyhexadecin-(7)-säure-(1)
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von 10-Hydroxyhexadecin-(7)-säure-(1), die ein wichtiges Zwischenprodukt für die Herstellung des
10-Acetoxyhexadecen-(7cis)-ol-(1) ist, das ein hochwirksames Anlockmittel für männliche Schwammspinner (Lymantria dispar L.) darstellt.
Es ist bekannt, dass man die 10-Hydroxy- (7)-säure (1) herstellen kann, indem man Heptanal- (l) mit Prop aDgylbromid und Zink zum 4-Hydroxydecin- (l) umsetzt, dieses in den Tetrahydropyranyläther überführt, dessen Chlor-5-jodpentan zum 1-Chlor-9-tetrahydropyranyloxypentadecin-(6) knodensiert, darin das Chloratom durch die Nitrilgruppe ersetzt, das so erhaltene 10-Tetrahydropyranyloxyhexadecin-(7)-säure-(1)-nitril alkalisch zur entsprechenden Säure verseift und diese schliesslich mit sauren Mitteln behandelt (M. Jacobson, M. Beroza und W. A. Jones. J. Amer. Chem. Soc. 83, 4819 [1961]).
Dieses Verfahren'umEasst 7 Stufen, und die Gesamt- ausbeute beträgt nur etwa 0, 5 O/o der Theorie.
Es ist weiterhin bekannt, dass man Acetylcarbinole durch Umsetzung von Epoxyden mit Aaetylen-Grignard- Verbindungen erhalten kann. Schliesslich ist noch bekannt, dass man durch Umsetzung von 1, 2-Epoxyoctan mit der Lithiumverbindung von 9-Chlornonin- (l) das 1-Chlor-11-hydroxyheptadecin-(8) und daraus über die entsprechende Cyanverbindung der 12-Hydroxyocta decin-' (9)-säure- (l) herstellen kann (W. J. Gensler und C. B. Abrahams, J. Amer. Chem. Soc. 80, 4593 [1958]).
Das erfindungsgemässe Verfahren zur Herstellung der 10-Hydroxyhexadecin-(7)-säure-(1) zeichnet sich dadurch. aus, dass man das aus l-Chlor-5-jodpentan und Natriumacetylid in flüssigem Ammoniak erhältliche 7 Chlorheptin- (l) in metallorganischer Reaktion mit 1, 2 Epoxyoctan zum l-Chlor-9-hydroxypentadecin- (6) umsetzt, dieses durch Umsetzung mit Natrium-oder Kalium- cyanid in das 10-Hydroxyhexadecin-(7)-säure-(1)-nitril überführt und. dieses schliesslich alkalisch verseift.
Das erfindungsgemässe Verfahren umfasst ausgehend von l-Chlor-5-jodpentan 4 Stufen. Die Geaamtausbeute liegt bei etwa 31 /o der Theorie und beträgt somit etwa das Sechzigfache des bekannten Verfahrens, das präpa- rativ unbefriedigend ist. Man kann nach J. Amer. Chem.
Soc. 83, 4819 (1961) die nach dem erfindungsgemässen Verfahren hergestellte 10-Hydroxyhexadecin- (7)-. säure- (1) in Gegenwart von Lindtar-Katalysator partiell zur 10-Hydroxyhexadencen-(7cs)-säure-(1) hydrieren, diese mit Lithiumalanat zum hexadecen-(7cis)-diol-(1,10) reduzieren, dieses mit Acetylchlorid in das 1, IO-Diacetoxy- hexadecen-(7cis)- überführen und dieses schliesslich mit alkoholischer Lauge partiell zum 1, 10-Acetoxyhexa- , decen-, (7cis)-ol-(1) verseifen. Die Ausbeute beträgt etwa 55"/o der Theorie, bezogen auf die 10-Hydroxyhexa- decin- (7)-säure- (l).
Das für das erfindungsgemässe Verfahren als Aus gangsproidukt erforderliche 7-, Chlorheptin- (l) kann man nach J. Amer. Chem. Soc. 70, 1699 (1948) in 85%iger Ausbeute durch Umsetzung von l-Chlor-5-jodpentan mit Natriumacetylid in flüssigem Ammonialk bei-35% bis-40 C herstellen.
In der Folge wird eine bevorzugte Ausführungsform des erfindungsgemässen Verfahrens beschrieben. Hierbei wird 7-Chlorheptin- (1) mit einer ätherischen n-Butyl- lithium-Lösung in einem indifferenten Lösungsmittel, z. B. Di-n-propyläter, D-n-butylätyer, Tetrahydrofuran oder Dioxan in die entsprechende Lithiumverbindung übergeführt. Danach fügt man 1, 0 bis 1, 5 Mol 1, 2 Epoxyoctan (pro 1, 0 Mol 7-Chloriheptin-(1) hinzu, erwärmt das Reaktionsgemisch mehrere Stunden auf Tem peraturen von 60 bis 120 C und verdmmpft den gross- ten Teil des Lösungsmittels unter vermindertem Druck.
Der Rückstand wird mit einer sauer reagier, end, en wäss- rigen Elektrolyt-Lösung behandelt und das Reaktions- produkt durch Anäthem isoliert. Es kanm durch Hoch- vakuumdestillation gereinigt werden oder zweckmässig als Rohprodukt weiterverarbeitet werden.
Das 1-Chlor-9-hydroxypentadein-(6) wird in Ath anol oder vorteilhaft in Dimethylsulfoxyd mit überschüs sigem Kalium-oder Natriumcyanid erwärmt und nach Entfernen das Lösungsmittels und Auswascihen von Kalium- oder Natriumchlorid und überschässigem Kalium- oder Natriumcyanid mit Wasser das 10-Hydr- oxyhexadecin-(7)-säure-(1)-nitril erhalten, das durch Hochvakuumdestillation gereinigt werden kann.
Man kann auch das 1-Chlor-9-hydroxypentadecin (6) zunächst durchEribitzenmitNatriurnjodidinAceton un des 1-Jod-9-hydroxypentadecin-(6) überführen und dieses dann als Rohprodukt in, alkoholischer Lösung mit Kalium- oder Natriumcyanid umsetzen.
Pas 10-Hydroxyhexadecin-(7)-säure-(1)-nitril wird du, rch Erhitzen mit alkoholischer Kali- oder Natronlauge bis zur Beendigung, der Ammoniakentwicklung zu 10 Hydroxyhexadecin-(7)-säure-(1) verseift. Beispiel 1
Zur Lösung von 32, 5 Gewichtsteilen 7-Chlor-l- heptin in 750 Volumteilen reinem Dioxan lässt man unter Riihren bei Raumtemperatur das Gemisch aus 82 Volumteilen einer 3,
06-molaren Lösung von n-Butyllithium in n-Hexan und 170 Volumteilen wasserfreiem Ather fliessen und erhitzt das Reaktionsgemisch l Stunde am Rüddlusskübler zum Sieden. Das Reaktionsgemisch wird dann mit 50 Gewichtsteilen 1, 2-Epoxyoctan, gelöst in 150 Volumteilen Dioxan, versetzt und 15 bis 20 Stunden am Rückflusskühler zum Sieden erhitzt. Das Lösungsmittel wird zum grössten Teil abdestilliert, der Rückstand mit Eis und verdünnter Schwefelsäure versetzt und das entstandene Gamisch mehrfach mit Äther geschüttelt.
Die vereinigten Atherauszügewerdenzu- nächst mit Wasser, dann mit wässiger 5-bis 10%iger Natriumhydrogencarbonatlösung gewaschen und über Natriumsulfat getrocknet. Der Äther wird bei Normaldruck und das nicht umjgesetzte 1, 2-Epoxyoctan unter dem Druck von 12 Torr (etwa 17, 5 Gewichtsteile vom Kp.12 = 56 bis 57 C, nD20 = 1,4215) abdestilliert. Der Rückstand wird im Hochvakuum destillierit. Man erhält 40, 5 Gewichtsteile (= 63"/o der Theorie) l-Chlor < 6 pentadecin-9-ol vom Kp. 0, o1 = 140 bis 145 C, nD20 = 1, 4750.
3\1 Gewichtsteile des erhaltenen 1-Chlor-6-penta decin-9-ols werden mit 60 Gewichtsteilen Natriumjodid in 1200 Volumteilen Aceton 24 Stunden am Rüokfluss- kuhler zum Sieden erhitzt. Dann wird das Aoeton abdestilliert und der Rückstand mit 400 bis 500 Volum teilen Äther versetzt. Die entstandene Ätherlösung wird mehrfach mit Wasser geschüttelt und dann über Natnum- sulfat getrocknet. Das nach dem Verdampfen des Äthers erhaltene rohe 1-Jod-6-pentadecin-9-ol, etwa 40 Gewichtsteile, wird mit 17 Gewiohtsteilen Kaliumcyanid in 50 Volumteilen Wasser und 300 Volumteilen Äthanol 12 bis 15 Stunden am Rückflusskühler zum Sieden er- hitzt.
Man verdampft das Athanol zum grössten Teil unter dem Dr. usk von 12 Torr, versetzt den Rückstand mit Wasser und schüttelt das Gemisch mehrfach mit Ath, er. Nach, dem Waschen der vereinigten Ätherauszüge mit Wasser und dem Trocknen über Natriumsulfat wird der Äther verdampft. Man erhält etwa 28 Gewichtsteile 10-Hydroxy-7-hexadecin-1-säurenitril vom Kp.0,001 = 1@15 C (Luftbadtemperatur), nD20 = 1,4729.

Zur Verseifung werden 25 Gewiohtsteile des rohen 10-Hydroxy-7-hexadecin-1-säurenitrils mit der Lösung von 25 Gewichtsteilen Wasser. und 170 Volumteilen Athanol 15 Stunden am Rückflusskühler zum Sieden erhitzt. Man verdampft das Athanol weitestgehend und versetzt den Rückstand mit Wasser. Die entstandene klare Lösung wird bei 0 C mit 10%iger wässriger Phosphorsäure angesäuert und der ausgefallene Nieder- schlag mit Äther extrahiert.
Die Ätherauszüge werden nach dem Waschen mit wässriger Natriumchloridlösung über Natriumsulfat getrocknet. Nach dem Verdampfen des Ather. wird der Rückstand, etwa 25 Gewichtseile, aus Petroläther, Kp. = 30 bis 50 C, dem wenig Äther zugesetzt wurde, umkristallisiert. Man erhalt 16, 5 Ge wichtsteile 10-Hydroxy-7-hexadecin-1-säure vom F = 50 bis 51 C. Die Gesamtausbeute beträgt 36"/e der Theorie, bezogen auf 7-Chlor-l-heptin, bzw. 31 % der Theorie, bezogen auf l-Chlor-5-jadpentan.
Die bsscbdeibenen Verfahrensstufen werden unter Ausschluss von Luftsauerstoff zwecikmässig m einer Stickstoffatmosphäre durchgeführt.
Beispiel 2
30 Gewichtsteile 1-Chlor-6-pentadecin-9-ol werden mit 25 Gewichtsteilen Kaliumcyanid in 50 Volumteilen Wasser und 300 Volumteilen Äthanol 72 Stunden unter Stickstoff am Rüokflusskühler zum Sieden erhitzt. Nach der im Beispiel 1 beschriebenen Aufarbeitung erhält man etwa 24 Gewichtsteile 10-Hydroxy-7-hexadecin-1säurenitril vom Kp.0,001 = 115 C (Lufbbadtemperatur), nD20 = 1, 4730.
Process for the preparation of 10-hydroxyhexadecyn- (7) -acid- (1)
The present invention relates to a process for the production of 10-Hydroxyhexadecin- (7) -äur- (1), which is an important intermediate for the production of
10-acetoxyhexadecen- (7cis) -ol- (1), which is a highly effective attractant for male gypsy moths (Lymantria dispar L.).
It is known that 10-hydroxy (7) acid (1) can be prepared by reacting heptanal (l) with prop aDgylbromid and zinc to 4-hydroxydecyne (l), converting this into tetrahydropyranyl ether, whose chloro-5-iodopentane knodens to 1-chloro-9-tetrahydropyranyloxypentadecin- (6), in which the chlorine atom is replaced by the nitrile group, the 10-tetrahydropyranyloxyhexadecyn- (7) -acid- (1) -nitrile is alkaline to the corresponding acid saponified and these finally treated with acidic agents (M. Jacobson, M. Beroza and WA Jones. J. Amer. Chem. Soc. 83, 4819 [1961]).
This process comprises 7 stages, and the overall yield is only about 0.5% of theory.
It is also known that acetyl carbinols can be obtained by reacting epoxides with acetylene Grignard compounds. Finally, it is also known that by reacting 1,2-epoxyoctane with the lithium compound of 9-chlorononine- (1) the 1-chloro-11-hydroxyheptadecine- (8) and from this via the corresponding cyano compound the 12-hydroxyoctadecine- '(9) -acid- (I) can produce (WJ Gensler and CB Abrahams, J. Amer. Chem. Soc. 80, 4593 [1958]).
The process according to the invention for the preparation of 10-hydroxyhexadecyn- (7) acid- (1) is characterized. from the fact that the 7-chloroheptine- (l) obtainable from l-chloro-5-iodopentane and sodium acetylide in liquid ammonia is converted into l-chloro-9-hydroxypentadecine- (6) in an organometallic reaction with 1.2 epoxyoctane, this by reaction with sodium or potassium cyanide converted into 10-hydroxyhexadecyn- (7) acid- (1) nitrile and. this is finally saponified under alkaline conditions.
The process according to the invention comprises 4 stages starting from 1-chloro-5-iodopentane. The total yield is about 31 / o of theory and is therefore about sixty times the known method, which is preparatively unsatisfactory. According to J. Amer. Chem.
Soc. 83, 4819 (1961) the 10-hydroxyhexadecine- (7) - prepared by the process according to the invention. acid- (1) partially hydrogenate in the presence of Lindtar catalyst to 10-hydroxyhexadencen- (7cs) acid- (1), reduce this with lithium alanate to hexadecen- (7cis) -diol- (1,10), this with acetyl chloride in the 1, IO-diacetoxyhexadecen- (7cis) - and finally saponify this with alcoholic liquor partially to 1, 10-acetoxyhexa-, decen-, (7cis) -ol- (1). The yield is about 55% of theory, based on the 10-hydroxyhexadecyn- (7) acid- (l).
The 7-, chlorheptine- (l) required as starting material for the process according to the invention can be found according to J. Amer. Chem. Soc. 70, 1699 (1948) in 85% yield by reacting 1-chloro-5-iodopentane with sodium acetylide in liquid ammonia at -35% to -40.degree.
A preferred embodiment of the method according to the invention is described below. Here 7-chlorheptine- (1) with an ethereal n-butyllithium solution in an inert solvent, eg. B. Di-n-propylätyer, D-n-butylätyer, tetrahydrofuran or dioxane converted into the corresponding lithium compound. 1.0 to 1.5 moles of 1.2 epoxyoctane (per 1.0 mole of 7-chloriheptin- (1)) are then added, the reaction mixture is heated to temperatures of 60 to 120 ° C. for several hours and the majority of it is evaporated of the solvent under reduced pressure.
The residue is treated with an acidic, final, aqueous electrolyte solution and the reaction product is isolated by anetheme. It can be purified by high vacuum distillation or further processed as a crude product.
The 1-chloro-9-hydroxypentadein- (6) is heated in ethanol or advantageously in dimethylsulfoxide with excess potassium or sodium cyanide and, after removing the solvent and washing out potassium or sodium chloride and excess potassium or sodium cyanide with water, the 10 -Hydr- oxyhexadecyn- (7) acid- (1) nitrile obtained, which can be purified by high vacuum distillation.
The 1-chloro-9-hydroxypentadecin (6) can also be converted first by eribitzmitnatriurniodidineacetone and the 1-iodo-9-hydroxypentadecin- (6) and then converted as a crude product in an alcoholic solution with potassium or sodium cyanide.
Pas 10-hydroxyhexadecyn- (7) -acid- (1) -nitrile is saponified to 10 hydroxyhexadecyn- (7) -acid- (1) by heating with alcoholic potash or sodium hydroxide until the ammonia development is complete. This embodiment of the method according to the invention is illustrated using the following formula:

Example 1
To dissolve 32.5 parts by weight of 7-chloro-l-heptyne in 750 parts by volume of pure dioxane, the mixture of 82 parts by volume of a 3,
06 molar solution of n-butyllithium in n-hexane and 170 parts by volume of anhydrous ether flow and the reaction mixture is heated to boiling in a Rüddluss bucket for one hour. The combined ether extracts are washed first with water, then with aqueous 5 to 10% sodium hydrogen carbonate solution and dried over sodium sulfate. The ether is distilled off at normal pressure and the unconverted 1,2-epoxyoctane under a pressure of 12 Torr (about 17.5 parts by weight, boiling point 12 = 56 to 57 ° C., nD20 = 1.4215). The residue is distilled in a high vacuum. 40.5 parts by weight (= 63 "/ o of theory) of 1-chlorine <6 pentadecyn-9-ol with a boiling point of 0.1 = 140 to 145 ° C., nD20 = 1.4750 are obtained.

For saponification, 25 parts by weight of the crude 10-hydroxy-7-hexadecyn-1-acid nitrile are mixed with the solution of 25 parts by weight of water. and 170 parts by volume of ethanol heated to boiling on a reflux condenser for 15 hours. Most of the ethanol is evaporated and water is added to the residue. The resulting clear solution is acidified at 0 C with 10% aqueous phosphoric acid and the precipitate which has separated out is extracted with ether.
The ether extracts are washed with aqueous sodium chloride solution and dried over sodium sulphate. After evaporation of the ether. the residue, about 25 parts by weight, is recrystallized from petroleum ether, boiling point = 30 to 50 ° C., to which a little ether has been added. 16.5 parts by weight of 10-hydroxy-7-hexadecyn-1-acid with an F = 50 to 51 C. The total yield is 36 "/ e of theory, based on 7-chloro-1-heptyne, or 31% of theory, based on l-chloro-5-jadpentane.
The bsscbdeibenen process steps are carried out with exclusion of atmospheric oxygen in a nitrogen atmosphere.
Example 2
30 parts by weight of 1-chloro-6-pentadecin-9-ol are heated to boiling with 25 parts by weight of potassium cyanide in 50 parts by volume of water and 300 parts by volume of ethanol for 72 hours under nitrogen on a reflux condenser. After the work-up described in Example 1, about 24 parts by weight of 10-hydroxy-7-hexadecyn-1-acid nitrile with a boiling point of 0.001 = 115 ° C. (air bath temperature), nD20 = 1.4730 are obtained.
