Verfahren zur Herstellung von Sulfamylanthranilsäuren
Sulfamylanthranilsäuren und Wege zu ihrer Her stellung sind nach-den deutschen Patentschriften Nr. 1 122 541 und Nr. 1 129 501 bekannt. Die Verbindungen haben sich als gut brauchbare Diuretika und Saluretika erwiesen. Nach den genannten Patentschriften werden sie durch Umsetzen von Dihalogenosulfamylbenzoesäuren mit entsprechend substituierten Aminen bei enhöhten Temperaturen erhalten.
Als Nachteile dieses Verfahrens sind Nebenreaktionen und Verharzungserscheinungen festzustellen, die insbesondere bei der Herstellung von am Stickstoff durch Benzyl-, Furfuryl-oder Thenyl-oder Thioäthergruppen substitu ierten Verbindungen im technischen Massstab auftre ten. Denartige Verbindungen sind pharmakologisch besonders wirksam.
Es wurde nun ein Verfahren zur Herstellung von Sulfamylanthramilsäure gefunden, das dadruch gekennzeichnet ist, dass man einen Dihalogeno-sulfamylbenzoesäureester der allgemeinen Formel II zu einem Sulfamyl anthranilsäureesber der allgemeinen Formel III umsetzt

wobei X Chlor oder Brom, R einen aliphatischen oder araliphatischen Kohlenwasserstoffrest mit bis zu achtzehn Kohlenstoffatomen, Ri Wasserstoff oder die Benz ylgruppe und R2 die Bezylgruppe und, falls Ri für Wasserstoff steht, die Furfuryl-oder Thenyl-Gruppe bedeuten, und wobei die Phenylgruppe mit 1 bis 2 Kohlenstoffatomen oder die Methylendioxygruppe substitutiert sein kann,
und anschliessend den erhaltenen Ester zur freien Carbonsäure verseift.
Die als Ausgangsmaterial benötigten Ester werden aus den entsprechenden Dihalogeno-sulfamylbenzoesäuren nach bekannten Methoden dargestellt. Beispielsweise erhält man die Methylester in quantitativer Ausbeute mittels Diazomethan. Methyl-und Ä. thylester werden in guter Ausbeute erhalten, wenn man die Carbonsäure in, der 10-bis 20fachen Menge der betreffenden absoluten Alkohole löst und Chlorwasserstoff bis zur Sättigung einleitet. Allgmein anwendbar ist die Veresterung der entsprechenden Carbonsäurechloride.
Bei- spielsweise kocht man eine Mischung von Dihalogen- sulfamylbenzoesäure und Thionylchlorid (Gewichtsver hältnis : 1 : 3) einige Stunden unter Rückfluss in Dioxan, zieht dann im Vakuum Lösungsmittel und überschüssiges Thionylchlorid ab, löst den Rückstand in dem betreffenden Alkohol und versetzt mit Triäthylamin bis zur schwach alkalischen Reaktion. Nach Einengen der Lösung fällt man den Ester mit Wasser aus und kri stallisiert ihn aus wässrigem Äthanol um.
Als alkoholische Komponente der als Ausgangsstoffe verwendeten Ester kann grundsätzlich jeder einwertige aliphatische oder araliphatisch Alkohol, beispielsweise Methoanol, Äthanol, Isopropanol, n-butanol, Isobutanol, n-hexanol, Decylakohol, Stearylalkohol, allylalkohol, Cyclohexylakohol, Benzylalkohol, ss-Phenyläthylakohol oder Furfurylakohol verwendet werden, da die Beweglichkeit des zur Gruppe -COOR ortho-ständigen halogenatome von der Wahl des Restes R weitgehend unabhängig ist. iN der Praxis werden deshalb als Ausgangsmatenial vorzugswise die billigen, einfach darzu- stellenden und gut kristallisierenden Methylester verwendet.
Als basische Reaktionspartner der Dih, alogeno-sulf- amylbenzoesäureester können grundsätzlich beliebig substituierte primäre und sekundäre Amine verwendet werden, vorteilhaft araliphatische und schwefelhaltige aliphatische Amine, insbesondere Benzylamin, Dibenzylamin, Furfurylamin, 2-Thenylamin auns S-Äthylcysteamin. wobei die aromatischen Gruppen durch Chlor, Alkyl- oder Alkoxygruppen mit 1 bis 2 Koblenstoffatomen substituiert sein können.
Zur Durchfüung des Verfahrens gemäss Erfin- dung setzt man die Aminbase vorteilhaft im Überschuss und ohne weiteres Lösungsmittel ein und verwendot auf ein Äquivalent des Esters bis zu 6 Äqui- valente der Base. Der Üiberschuss an Amin begünstigt die Ausbeute, ohne den Reaktionsverlauf zu beeinflussen, solange die für den Austausch des zur Estergruppe orthoständigen Halogenatoms benötigte Temperatur nicht sehr stark überschritten wird.
Bei der Verwendung technisch schwer zugänglicher Amine als Kondensa- tionspartner ist es zweckmässig, den Aminüberschuss zu verringern und zum Binden des Halogenwasserstoffs eine tertiäre organische Base, wie Triäthylamin, Pyridin oder Dimethylanilin zuzusetzen. Die günstigen Reaktionstemperaturen sind von der Basizität des als Kondensa tionspartner verwendeten Amins abhängig und liegen zwischen 80 und 200 C, vorzugsweise zweichen 90 und 160 C. Die Reaktionszeit beträgt im allgemeinen 30 bis 300 Minute.
Zur Isolierung des Esters aus der Reaktionslösung wird diese zweckmässig m Wasser eingeführt und die erhaltene Mischung unter Eiskühlung durch Zusatz von verdünnter Schwefelsäure oder Eisessig auf einen pH Wert von 4 bis 5 eingestellt. Der Sulfamylanthranil säureester scheidet sich daraufhin in den meisten Fäl- len in fester Forum aus und wird abfiltriert. Aus dem Fütrat lassen sich die im Überschuss eingesetzten Amine praktisch ohne Verluste wiedergewinnen.
Die anschliessende VerseifungdesSulfamylanthranil- säureesters zur Sulfamylanthranilsäure verläuft schnell und praktisch ohne Nebenreaktion in alkalischem Medium. Vorzugsweise behandelt man den fein verriebenen Ester unter intensive Rühren bei 40 bis 60 C mit 1, 2 bis 1, 5 Äquivalenten wässriger Kali-oder Na tronlauge, filtriert die Salzlösung und entfärbt das Filtrat mit Kohle. Die Lösung wird unter Eiskühlung und d Rühren durch Zugabe von verdunnter Schwefelsäure oder Eisessig auf einen pH-Wert von 4 bis 5 $eingestellt, wonauf sich die Sulfamylanthranilsäure abscheidet.
Zur Reinigung wird das Produkt aus Äthanol oder einem Gemisch von Äthanol und Wasser, gegebenenfalls unter Verwendung von Entfärbungskohle, umkristallisiert.
Die Verfahrenserzeugnisse können gewünschtenfalls s durch Behandlung mit anorganisshen oder organischen Basen in die entsprechenden canbonsauren Salze übergeführt werden. Als anorganische Basen kommen z. B. Ammoniak, Alkali-ader Erdalkalicarbonate,-bi- carbonate,-hydroxyde bzw.-oxyde in Betracht. Als organische Base sei z. B. Dicyclohexylamin erwähnt.
Der Verlauf des Verfahrens gemäss der Erfindung ist überraschend. Zunächst konnte nicht ohne weiteres erwartet werden, dass selektiv nur das zur Estergruppe ortho-ständige Halogenatom geigen die Aminogruppe ausgetauscht wird. Sodann wäre bei den verwendeten Temperaturen eine e aminolytische Spaltung der Esterbindung zu erwarten gewesen.
Da sie nicht eintritt, bieten die gut kristallisierenden Ester eine willkommene Möglichkeit zur Reinigung der Präparate. Überraschend und zugleich technisch fortschrittlich ist es, dass das Verfahren gemäss der Erfindung bei Temperaturen durchgeführt werden kann, die mindestens 20 bis 30 C unter denen liegen, die nach den Verfahren der deut- schen Patentschriften 1 122 541 und 1 129 501 genannt sind. Dadruch werden die dort auftretenden Nachteile vermieden, Nebenreaktionen und Verharzungen werden, insbesondere bei Durchführung des Verfahrens im technischen Massstabe, zurückgedrängt und eine wesent Lishe Verbesserung der Ausbeute Erzielt.
Ausserdem wird Wärmeenergie eimgespart.
Die Verfahrensprodukte sind wertvolle Diuretika und Saluretika.
Beispiel I 3-Sulfamyl-4-chlor-6-furfurylaminobenzoesäure
In 293 g Furfurylamin (3 Mol) werden bei Raum- temperatur unter Rühren 142 g 3 Sulfamyl-4, 6-dichlor- benzoesäure-methlester (0,5 Mol) vom Schmelzpunkt 2020 C innerhalb von 10 Minuten eingetragen. Man erwärmt die Mischung rasch auf 100 C und rührt sie eine Stunde lang bei dieser Tempenatur. Anschliessend wird die heisse Reaktionslosung in 3 1 Eiswasser eingerührt und die erhaltene Mischung bei 10 bis 20 C durch Zugabe von Eises, sig auf einen pH-Wert von 4, 5 Eingestellt.
Der in fester Form abgeschiedene Ester wird abfiltriert, mit Wasser gewaschen und danach mit der gleichen Ge wiehtsanenge Wasser zu einem Brei verrieben. Nach Zugabe von 1, 5 1 0, 5 n Kalilauge wird-die Mischung eine Stunde lang bei 55 C gerührt und heiss filtriert. Das alkalische Filtrat wird mit Kohle entfärbt und durch Zugabe von Eisessig unter Rühren auf einen pH-Wert von 4, 5 eingestellt. Die Oarbonsäure scheidet sich daraufhin kristallin ab, wird filtriert, mit Wasser gewaschen, bei 100 C getrocknet und aus 96 /oigem Äthanol umkri stallisiert.
Man erhält 86 g farblose Prismen (52 /o der Theorie) vom Zersetzungspunkt 206 C
Unter analogen Reaktionsbedingungen wird die gleiche Verbindung erhalten, wenn man anstelle von 142 g 3-Sulfamyl-4, 6-dichlorbenzoesäuremethylester, 149 g 3-Sulfamyl-4, 6-dichlor-benzoesäure-äthylester (Schmelzpunkt 116 C) oder 163 g 3-Sulfamyl-4, 6-dichlorbenzoesäure-n-butylester (Schmelzpunkt 92 C) oder 261 g 3-Sulfamyl-4, 6-, diahlorbenzoesäure-stearyl- ester oder 176 g 3-Sulfamyl-4, 6-dichlorbenzoesäure- cyclohexylester (Schmelzpunkt 128 C) oder 180 g 3-Sulfamyl-4,6-dichlonbenzoesäurebenzylester (Schmelzpunkt 168 C) verwendet.
Beispiel 2 3-Sulfamyl-4-chlor-6-(2-thenylamino)-benzoesäure
340 g 2-Thenylamin (3 Mol) und 142 g 3-Sulfamyl4,6-dichlorbenzoesäuremethylester (0,5 Mol) werden nach der in Beispiel 1 angegebenen Vorschrift umgesetzt, das Reaktionsprodukt wie.angegeben mit Kalilauge verseift und nach dem Fällen mit Essigsäure aus 50%igem Äthanol unter Verwendung von Entfärbungskohle umkristallisierit. Man erhält 73 g (42 /o der Theorie) des kristallinen Produktes vom Zersetzungspunkt 210 C.
Beispiel 3
3-Sulfamyl-4-chlor-6-benzylaminobenzoesäure 321 g Benzylamin (3 Mol) und 142 g Sulfamyl-4, 6 dichlorbenzoesäuremethylester (0, 5 Mol) werden nach der in Beispiel 1 angegebenen Vorschrift umgesetzt, das Reaktionsprodukt wie angegaben mit Kalilauge verseift und nach dem Fällen mit Essigsäure aus Athanol unter Verwendung von Entfärbungskohle umkristallisiert. Man erhält 105 g (61 /o der Theorie) des kristallinen Produkts vom Zersetzungspunkt 244 C.
Beispiel-4 3-Sulf amyl-4-chlor-6-dibenzylaminobenzoesäure
395 g Dibenzylamin (2 Mol) und 142 g 3-, Sulfamyl 4, 6-dichlorbenzoesäure-methylester (0, 5 Mol) werden analog der in Beispiel 1 angegebenen Vorschrift 2 Stunden lang bei 150 C gerührt, der Ester wird wie in Beispiel 1 angegeben isoliert und mit Kalilauge verseift.
Nach Umkristallisieren aus Äthanol erhält man 115 g (53 /o der Theorie) des kristallinen Produkts vom Zersetzungspunkt 206 C.
Process for the preparation of sulfamylanthranilic acids
Sulfamylanthranilic acids and ways of preparing them are known from German patents No. 1 122 541 and No. 1 129 501. The compounds have been found to be useful diuretics and saluretics. The esters required as starting material are prepared from the corresponding dihalogenosulfamylbenzoic acids by known methods. For example, the methyl esters are obtained in quantitative yield using diazomethane. Methyl and al. ethyl esters are obtained in good yield if the carboxylic acid is dissolved in 10 to 20 times the amount of the absolute alcohols in question and hydrogen chloride is passed in until it is saturated. The esterification of the corresponding carboxylic acid chlorides is generally applicable.
For example, a mixture of dihalosulfamylbenzoic acid and thionyl chloride (weight ratio: 1: 3) is boiled under reflux in dioxane for a few hours, then solvent and excess thionyl chloride are removed in vacuo, the residue is dissolved in the alcohol in question and triethylamine is added to a weakly alkaline reaction. After concentration of the solution, the ester is precipitated with water and crystallized from aqueous ethanol.
In principle, any monohydric aliphatic or araliphatic alcohol, for example methanol, ethanol, isopropanol, n-butanol, isobutanol, n-hexanol, decyl alcohol, stearyl alcohol, allyl alcohol, cyclohexyl alcohol, benzyl alcohol, ss-phenylethyl ethyl alcohol can be used as the alcoholic component of the esters used as starting materials since the mobility of the halogen atom ortho to the group -COOR is largely independent of the choice of the radical R. In practice, the inexpensive, easy-to-prepare and readily crystallizing methyl esters are therefore preferably used as the starting material.
Primary and secondary amines with any desired substituents can be used as basic reactants of the dih, alogeno-sulfamylbenzoic acid esters, advantageously araliphatic and sulfur-containing aliphatic amines, in particular benzylamine, dibenzylamine, furfurylamine, 2-thenylamine and S-ethylcysteamine. it being possible for the aromatic groups to be substituted by chlorine, alkyl or alkoxy groups having 1 to 2 carbon atoms.
To carry out the process according to the invention, the amine base is advantageously used in excess and without any further solvent and up to 6 equivalents of the base are used for one equivalent of the ester. The excess of amine favors the yield without influencing the course of the reaction, as long as the temperature required for the exchange of the halogen atom ortho to the ester group is not exceeded very significantly.
When using technically difficult to obtain amines as condensation partners, it is expedient to reduce the amine excess and to add a tertiary organic base such as triethylamine, pyridine or dimethylaniline to bind the hydrogen halide. The favorable reaction temperatures depend on the basicity of the amine used as condensation partner and are between 80 and 200 ° C., preferably between 90 and 160 ° C. The reaction time is generally 30 to 300 minutes.
To isolate the ester from the reaction solution, it is expediently introduced into water and the mixture obtained is adjusted to a pH of 4 to 5 by adding dilute sulfuric acid or glacial acetic acid while cooling with ice. The sulfamylanthranilic acid ester then separates in most cases in a solid forum and is filtered off. The amines used in excess can be recovered from the feed with practically no losses.
The subsequent saponification of the sulfamylanthranilic acid ester to sulfamylanthranilic acid proceeds quickly and with practically no side reactions in an alkaline medium. The finely ground ester is preferably treated with 1.2 to 1.5 equivalents of aqueous potassium or sodium hydroxide solution at 40 to 60 ° C. with intensive stirring, the salt solution is filtered and the filtrate is decolorized with charcoal. While cooling with ice and stirring, the solution is adjusted to a pH of 4 to 5 by adding dilute sulfuric acid or glacial acetic acid, whereupon the sulfamylanthranilic acid separates out.
For cleaning, the product is recrystallized from ethanol or a mixture of ethanol and water, if necessary using decolorizing charcoal.
If desired, the products of the process can be converted into the corresponding carboxylic acid salts by treatment with inorganic or organic bases. As inorganic bases such. B. ammonia, alkali metal or alkaline earth metal carbonates, bicarbonates, hydroxides or oxides into consideration. The organic base is e.g. B. dicyclohexylamine mentioned.
The course of the method according to the invention is surprising. At first it could not be expected without further ado that only the halogen atom ortho to the ester group would be selectively exchanged for the amino group. Then an aminolytic cleavage of the ester bond would have been expected at the temperatures used.
Since it does not occur, the esters, which crystallize well, offer a welcome option for cleaning the preparations. It is surprising and at the same time technically advanced that the process according to the invention can be carried out at temperatures which are at least 20 to 30 ° C. below those specified according to the processes of German patents 1 122 541 and 1 129 501. The disadvantages occurring there are avoided, side reactions and resinification are suppressed, especially when the process is carried out on an industrial scale, and a significant improvement in the yield is achieved.
In addition, thermal energy is saved.
The products of the process are valuable diuretics and saluretics.
Example I 3-sulfamyl-4-chloro-6-furfurylaminobenzoic acid
In 293 g of furfurylamine (3 mol), 142 g of 3 sulfamyl-4,6-dichlorobenzoic acid methyl ester (0.5 mol) with a melting point of 2020 ° C. are introduced within 10 minutes at room temperature with stirring. The mixture is heated rapidly to 100 ° C. and stirred for one hour at this temperature. The hot reaction solution is then stirred into 3 l of ice water and the mixture obtained is adjusted to a pH of 4.5 at 10 to 20 ° C. by adding ice-cream.
The ester which has separated out in solid form is filtered off, washed with water and then ground to a pulp with the same amount of water. After the addition of 1.5 1 0.5 N potassium hydroxide solution, the mixture is stirred for one hour at 55 ° C. and filtered while hot. The alkaline filtrate is decolorized with charcoal and adjusted to a pH of 4.5 by adding glacial acetic acid while stirring. The carboxylic acid then separates out in crystalline form, is filtered, washed with water, dried at 100 ° C. and recrystallized from 96% ethanol.

The same compound is obtained under analogous reaction conditions if, instead of 142 g of methyl 3-sulfamyl-4, 6-dichlorobenzoate, 149 g of ethyl 3-sulfamyl-4,6-dichloro-benzoate (melting point 116 C) or 163 g of 3- Sulfamyl-4,6-dichlorobenzoic acid n-butyl ester (melting point 92 ° C.) or 261 g 3-sulfamyl-4,6, diahlobenzoic acid stearyl ester or 176 g 3-sulfamyl-4,6-dichlorobenzoic acid cyclohexyl ester (melting point 128 ° C.) 395 g of dibenzylamine (2 mol) and 142 g of methyl 3-, sulfamyl 4,6-dichlorobenzoate (0.5 mol) are stirred for 2 hours at 150.degree. C. analogously to the instructions given in Example 1; the ester is as in Example 1 indicated isolated and saponified with potassium hydroxide.
After recrystallization from ethanol, 115 g (53 / o of theory) of the crystalline product with a decomposition point of 206 ° C. are obtained.
