N-(4-Carboxy-benzolsulfonyl)-N'-cycloheptyl- harnstoff.

Process for the production of benzenesulfonylureas It is known that certain benzenesulfonylurea derivatives have blood sugar-lowering properties and are suitable as antidiabetic agents which can be administered orally (cf., for example, Arzneimittelforschung 8 [1958], pp. 444-445). In particular, N- (4-methylbenzenesulfonyl) -N'-n-butylurea has achieved great importance in diabetes therapy because of its good blood sugar-lowering effectiveness and its good tolerability.
It is also known that said N- (4-methyl-benzenesulfonyl) -N'-n-butylurea is subject to oxidation in the human organism after its administration, the p-methyl group of the toluenesulfonyl radical initially being converted into a hydroxyme- thylgruppe and this continues to be converted into a carboxyl group.
Both the N- (4-hydroxymethylbenzenesulfonyl) -N'-n-butylurea and the corresponding carboxylic acid derivative have been shown to be ineffective when tested for blood sugar-lowering effects (see Arzneimittelforschung 8 [1958], Tab. 10, page 452) and German Medical Wo chenschrift 81 [1956], pages 887 and 888).
It has already been described that N- (4-methyl-benzenesulfonyl) -N'-cyclooctylurea and the corresponding cycloheptylurea have a strong blood sugar-lowering effect (compare Austrian Patent No. 216,521). This compound surpasses the N- (4-methylbenzenesulfonyl) -N'-n-butyl urea both in terms of potency and duration of action considerably. However, a disadvantage of the two first-mentioned compounds is their relatively poor solubility.
It has now been found that surprisingly strong and long-acting benzenesulfonylurea derivatives of the formula I
EMI0001.0016
obtained in which R is a cycloheptyl, cyclooctyl or a cyclohexyl radical optionally substituted by a low molecular weight alkyl radical and n stands for the numbers 0, 1 or 2, if Ver compounds of the formula II
EMI0001.0017
where R and n have the meaning given and R1 is hydrogen or a low molecular weight alkyl radical, reduced in the presence of lithium aluminum hydride.
It has long been known that organic acids and esters can be reduced with lithium aluminum hydride to form corresponding alcohols. However, it is also known that a carboxamide group is attacked by the same reducing agents and that, for example, carboxamides are converted into amines (see Karrer, Textbook of Organic Chemistry, 13.
Edition, pages 100/146). Thus, it could not be foreseen that the reduction of acids and esters of the benzenesulfonyl urea series according to formula II in a simple and smooth reaction, i. H. without the formation of byproducts, would lead to the desired hydroxyalkyl-benzenesulfonyl urea derivatives. However, the exemplary embodiments show that the reaction products are obtained in excellent yield and great purity.
If, according to the special process according to the invention, the stated starting materials are dissolved in a suitable solvent, for example dioxane, there is initially no reaction whatsoever after addition of excess lithium aluminum hydride. Only after heating, preferably on the steam bath, to around 80-90 ° C, suddenly a strong reaction occurs with boiling, which subsided after a short time. If the reaction solution is diluted, possibly after prior concentration, with water and acidified, the desired reaction product is obtained in practically pure form and in excellent yield.
The substance can be further purified by dissolving the reaction products in about 1% aqueous ammonia and acidifying the solution, after any previous filtration. The reaction is even smoother when tetrahydrofuran is used as the solvent. Here, a reaction occurs even at room temperature, which takes place in a very short time with self-heating. The yields achieved with simple work-up are over 90%.
It is true that hydroxyalkylbenzenesulfonylcycloalkylureas can also be obtained by using hydroxyalkylbenzenesulfonamides according to methods known for the production of other sulfonylureas with, for example, isocyanates or isocyanate formers such as urethanes, carbamic acid halides, optionally substituted ureas (compare German patent specification No. 974 062) converted into the corresponding hydroxyalkylbenzenesulfonyl-N'-cycloalkyl ureas.
However, this isocyanate process is uneconomical in comparison with the special process according to the invention, since z. B. p-Hydroxymethyl-benzenesulfonamide, which is conveniently prepared by reducing the corresponding Ben zoesäuresulfonamids or an ester derivative of this compound by reducing this connection with lithium aluminum hydride (other manufacturing processes are practically unsuitable), is relatively easily soluble in water and is difficult can be separated from the accompanying substances of the reaction such as aluminum hydroxide.
The hydroxyalkyl benzenesulfonylureas obtained by the claimed process are, in contrast to hydroxymethyl benzenesulfonamide, relatively sparingly soluble in water and can therefore be easily separated from the reaction solutions and from the substances accompanying the reaction.
The starting materials used for the process according to the invention are generally N-carboxy-benzene- or N-carboxyalkyl-benzene-sulfonyl-N'-cyclohexyl-, -heptyl- and -octyl ureas and their esters with low molecular weight alkyl alcohols .
Examples include: N- (4-carboxy-benzenesulfonyl) -N '- (4- or 3- or 2-methyl-cyclohexyl) -urea, N- (4-carboxy-benzenesulfonyl) -N' - (4- or 3- or 2-ethylcyclohexyl) urea, N- (4-carboxy-benzenesulfonyl) -N '- (4- or 3- or 2-propyl-cyclohexyl) -urea, N- (4-carboxy-benzenesulfonyl) -N '- (4- or 3- or 2-isopropyl-cyclohexyl) -urea N- (4-carboxy-benzenesulfonyl) -N' - (4- or .3- or 2-butyl-cyclohexyl) -urea, N- ( 4-carboxy-benzenesulfonyl) -N '- (4- or 3- or 2-isobutyl-cyclohexyl) -urea, N- (4-carboxy-benzenesulfonyl) -N' - (4- or 3- or 2-sec. -butyl-cyclohexyl) -urea, N- (4-carboxy-benzenesulfonyl) -N '- (4- or 3- or 2-tert-butyl-cyclohexyl) -urea, N- (4-carboxy-benzenesulfonyl) - N'-cyclooctyl urea,
N- (4-carboxy-benzenesulfonyl) -N'-cycloheptyl urea.
Instead of the free acids, their esters with lower alcohols, for example methanol, ethanol, propanol, can also be used. The carboxyl or ester groups of the starting materials mentioned can furthermore be bonded to the benzene nucleus via a methylene or ethylene group.
As starting materials, for example, compounds also come into consideration in which the carboxy group contained in the abovementioned starting materials is replaced by a carbomethoxy, carbethoxy, carbonpropoxy, carboxymethyl, carbomethoxymethyl, carbethoxymethyl, Car - bopropoxymethyl, carboxy-ethyl, carbomethoxy-ethyl, carbethoxyethyl, carbopropoxyethyl it is set.
The embodiments of the method according to the invention can be varied with regard to the reaction conditions insofar as, instead of dioxane, other suitable higher-boiling ethers, such as tetrahydrofuran, diethylene glycol dimethyl ether, etc., can be used, which have a certain dissolving power for alkali aluminum hydrides. Temperatures between 20 and 100 ° C. are preferably used as reaction temperatures. In order to obtain the process products in as pure a form as possible, they can conveniently be purified by dissolving them in about 1% ammonia and then precipitating them with organic or inorganic acids.
The sulfonyl urea derivatives obtained by the method according to the invention are valuable medicaments which are characterized in particular by a strong blood sugar-lowering effectiveness. While the starting products, in particular the ester compounds, have an - albeit lesser - effectiveness in this direction, this is of an order of magnitude in the case of the process products that makes practical therapeutic use probable. The blood sugar lowering effect of the process products could z. B. in the test on dogs and rabbits who found that the process products in a dosage of 400 mg / kg or
50 mg / kg fed to rabbits in the form of the sodium salt and determined the blood sugar value in the usual way according to Hagedorn / Jensen. For example, the N- (4-hydroxy-methyl-benzenesulfonyl) -N'- (4-isopropyl-cyclohexyl) -urea and the N- (4-hydroxymethyl-benzenesulfonyl) -N'-cyclooctyl urea when applied 400 mg / kg in rabbits the lowering of blood sugar given in the table below,
the values being determined in comparison with measurements on untreated control animals. In comparison, the corresponding data for the known N (4-methyl-benzenesulfonyl) -N'-n-butyl urea are also given. The longer duration of action is particularly noteworthy.
