Geeignete Hydrazin-Derivate der Formel III sind die Hydrazine der aliphatischen, aromati'sc'hen oder der heterocyclischen Reihe, wie z.
B. Methyl-, Äthyl-, Propyl-, Isopropyl-, n-Butyl- oder Phenylhydrazin. Als Hydrazine der heterocyclischen Reihe sollen (1-Alkyl-4-piperidyl)-hydrazine zur Anwendung ge langen, wobei der Alkylrest vorzugsweise 1-4 Ko'h- lenstoffatome enthält, wie z.

Die praktische Ausführung des Verfahrens erfolgt in .der Weise, dass man 2a-Cyano-17a-mebhyl-17ss- @hydroxy-4-ahdrosten-3-on txnd das gewünschte Hydra zifn der Formell 11I, wie z.
B. Me'thyl!hyd@azin, Phehyl- hydrazin, (1-Methyl-4-piperidyl)-hydrazin oder auch Hyd'razin selbst, in einem organischen Lösungsmittel, beispielsweise in einem niederen alip'hatischen Alko hol wie Methanol oder Äthanol, mehrere Stunden am Rückfluss zum Sieden erhitzt. Die Isolierung und Reinigung geschieht nach bekannten Methoden, z. B.
durch Kristallisation oder durch chromatographische Adsorption.
Das als Ausgangsmaterial verwendete 2a-Cyano- 17a-methyl-17sshydroxy-4-androsten-3-on kann in bekannter Weise wie folgt hergestellt werden: 17a- Methyl-17ss-hydroxy-4-androsten-3-on wird in einem iiheeten orga1nischen Lösungsmittel, z, B.

Das Ausgangsmaterial kann auch direkt durch Erhitzen der Hydroxymethylen-Verbindung (Formel IV) mit 0,N-Bis-(trifluoracetyl)-hydroxylamin in abs. Benzol in Gegenwart von Pyridin erhalten werden.
Die erfindungsgemäss hergestellten neuen Steroid- [3,2-c]-aminopyrazol-Derivate, die auch als Cyclo- peimta[7,8]phenan'ümo[2,3-c]ahnmopyrazo@ Derc'ivatebe- nannt werden können, sind auf Grund ihrer aus gezeichneten pharmakodynamischen Eigenschaften als Heilmittel in hohem Masse geeignet.
Sie besitzen bei geringer Tokizhät eine au@igeprägte ahtiphi#o'gi- stische bzw. entzündungshemmende Wirkung und be sitzen ausserdem eine spezifische antirheumatische Wirkung. Sie zeichnen sich ferner durch eine bemer kenswerte serotoninhemmende Wirkung aus.
Die nach dem vorliegenden Verfahren hergestell ten neuen Verbindungen dienen aber auch als Zwi schenprodukte zur Herstellung von Heilmitteln.
Die neuen Verbindungen können als Arzneimittel allein oder in entsprechenden Arzneiformen für dnterale oder parenterale Verabreichung verwendet werden. Zwecks Hersgteillung geeiga,eia'r Arzneiformen werden diese mixt anorganischen oder orgafnitschen,
pharmakologisch indifferenten Hilfsstoffen verarbei'te't. A1\s Hilfsstoffe werden verwendet z. B.
für Tabletten und Dragees: Milchzucker, Stärke, Talk, Stearinsäure usw. für Injektionspräparate: Wasser, Alkohole, Glycerin, p'flanzl'iche Öle u. dg1. Zudem können die Zubereitungen geeignete Kon- servierungs-, Stabilisierungs-, Netzmittel, Lösungs vermittler, Süss- und Farbstoffe, Aromatien usw. ent- halten.
In den nachfolgenden Beispielen, welche die Aus führung des Verfahrens erläutern., den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden. Die Schmelzpunkte sind nicht korrigiert.
<I>Beispiel 1</I> 17a-Methyl-17ss-hydroxy-4-androsteno[3,2-c]5'- aminopyrazdl Eine Lösung von 1,0 g 2aCyano-17a-methyl- 17ss-hydroxy-4-androsten-3-on und 0,5 cm3 wasser frei- Hydrazin in 20 cm3 Äthanol wird 5 Std. am Rückfluss erhitzt. Beim Erkalten kristallisiert das 17u#-Methyl-17ss-hydroxy-4-androsteno- [3,2-c]5'-aminopyrazol teilweise aus.
Man fil triert ab, spült mit wenig Äthanol und Äther nach und erhält so nach dem Trocknen im Vakuum bei 60 die gewünschte Verbindung. Durch Eindampfen der Mutterlauge im Vakuum und Kristallisieren des Rückstandes aus Methanol wird noch eine weitere Fraktion der Verbindung erhalten. Nach Umkristalli- sieren aus Äthanol und Sublimieren im Hochvakuum bei 200-220 schmilzt die <RTI
ID="0002.0101"> Substanz bei 248-251'. UV-Spektrum (Äthanol): Max. bei 257 m,y (log E _ 4,03).
Das Hydrochlorid wird hergestellt, indem man die Lösung der Verbindung in Methanol mit methanoli- schem Chlorwasserstoff versetzt. Smp.251-252 (Zers.) nach Umkristallisieren aus Äthanol/Äther.
Das als Ausgangssubstanz verwendete 2a-Cyano- 17a-met'hyl-17ss-hydroxy-4-androsten-3-on kann nach zwei Herstellungsverfahren erhalten werden:
a) Die Suspension von 3,6 g 0,N Bis-(trifl'uorace- tyl)-hyd'roxylamin in 20 cm3 abs. Benzol versetzt man unter Rühren mit einer Lösung von 2,57 g 2-Hydroxymethylen-17a-methyl-17ss-hydroxy- 4-androsten-3-on (hergestellt aus 17a-Methyl- 17ss-hydroxy-4-androsten-3-on durch Behandeln mit Äthylformiat und Natriumhydrid) in 55 cm3 abs. Benzol,
gibt anschliessend 4,7 cm3 abs. Pyridin zu und erhitzt die entstandene klare gelbe Lösung 3i/2 Std. zum Sieden am Rückfluss. Nach Abkühlen wird die Benzoll'ösung im Scheidetrichter zweimal mit je 20 cm3 Eiswasser gewaschen,
über Magnesiumsulfat getrocknet und im Vakuum eingedampft. Die so er haltene Substanz wird an neutralem Aluminiumoxyd chromatographiert, wobei das 2a-Cyano-17a methyl- 17ss - hydroxy - 4 - androsten-3-on mit Benzol/Äther- Gemischen eduiert wird. Smp. 163-165 nach Um kristallisieren aus Methylenchlorid/Äther/Petroläther.
b) Die Lösung von 880 mg Natrium in 80 cm3 Methanol versetzt man in einer Stickstoffatmosphäre bei Raumtemperatur mit 4,17 g 17a-Methyl-17ss- hydroxy-4-androsteno[2,3-d]isoxazol (hergestellt aus der unter a genannten Hydroxymethylenverbindung durch Cycl'isierung mit Hydroxylamin), rührt so lange, bis alles gelöst ist und lässt die klare gelbe Lösung 16 Std. stehen.
Dann wird die Lösung bei einem Druck von 15 mm Hg eingedampft, der Rückstand in Eiswasser aufgenommen, filtriert und mit Eiswasser ausgewaschen. Sodann suspendiert man die feste Substanz in eiskalter 2n wässeriger Salzsäure, filtriert nach gutem Schütteln ab und wäscht mit eiskalter 2n Salzsäure und Eiswasser nach.
Anschliessend filtriert man scharf ab, löst den Filterrückstand in Aceton und dampft die Lösung nach dem Filtrieren ein. Darauf löst man den Rückstand in Methylenchlorid, trennt die Methylenchloridlösung vom restlichen Wasser ab, trocknet sie über Magnesiumsulfat und dampft sie im Vakuum ein.
Das so erhaltene 2a-Cyano-17a-methyl- 17ss - hydroxy - 4 - androsten - 3-on kristallisiert beim Aufnehmen in Äther. Es wird filtriert und im Va kuum bei 60 getrocknet. Smp. 163-165 .
<I>Beispiel 2</I> 17a-Methyl-17ss-hydroxy-4-androsteno[3,2-c] 1'- methyl-5'-aminopyrazol Die Lösung von 2,0 g 2a-Cyano-17a-methyl-17ss- hydroxy-4-androsten-3-on und 2,0g Methylhydrazin in 30 cm3 Äthanol wird' 5 Std. am Rückfluss zum Sie den erhitzt. Nach Eindampfen des Reaktionsgemisches im Vakuum wird der Rückstand in wenig Methanol gelöst und die Lösung mit der etwa vierfachen Menge Äther und darauf mit Petroläther bis fast zur Trübung versetzt.
In der Kälte kristallisiert das 17a-Methyl-17ss-hydroxy-4-androsteno- [3,2-c] 1'-methyl=5'-amihopyrazoQ aus. Durch Einengen der Mutterlauge und erneutes Kristallisieren wird noch eine weitere Fraktion der Verbindung erhalten. Nach Umkristallisieren aus Me- thanol/Wasser und Sublimieren im Hochvakuum bei 200-220 schmilzt die Substanz bei 235-239 . UV- Spektrum (Äthanol): Max. 255,5 m,u (log e = 4,14).
<I>Beispiel 3</I> 17a-Methyl-17ss hydroxy-4-androsteno[3,2-c]1'- (1" mefhy4-4"-piposidyl)-5'-aminopyrazoll Die Lösung von 649 mg 2a-Cyano-17a-methyl- 17ss-hydroxy-4-androsten-3-on und 775 mg 1-Methyl- 4-piperidyl-hydrazin in 15 cm3 Äthanol wird 3 Std. am Rückfluss erhitzt.
Nach dem Eindampfen des Re aktionsgemisches im Vakuum wird der Rückstand in Methylenchlorid gelöst, .die Lösung dreimal mit Eis wasser ausgeschüttelt, über Magnesiumsul'fat getrock net und im Vakuum eingedampft. Der Rückstand wird an 15 g neutralem Aluminiumoxyd chromato- graphiert, wobei mit Benzol/lVlethylenchlorid-Gemi- schen, Methylenchlorid und hauptsächlich mit Me- thylench#lorid/Äthanol (9 : 1) die Verbindung eluiert wird.
UV-Spektrum (Äthanol): Max. bei 252 m,u (log e = 4,03).
Als Derivat wird das 17,17-Dimethyl-18-nor-4,13-androstadieno- [3,2-c] 1'-(1 "-methyl-4"-piperidyl)- 5'-aminopyrazol hergestellt. Es wird wie folgt erhalten:
Durch Ein leiten von Bromwasserstoff in die äthanolische Lö sung der Base erhält man das Di'hydrobromid des 17,17-Dimethyl-18-nor-4,13-androstadieno- [3,2-c] 1'-(1 "-methyl-4"-piperidyl)- 5'-aminopyrazols, das nach Umkristallisieren aus Methanol/Äther bei<B>311'</B> (Zers.) schmilzt. UV- Spektrum (Äthanol): Max. bei 255 m,u (log e = 4,2).
<I>Beispiel 4</I> 17a-Methyl-17ss hydroxy-4-androsteno[3,2-c]1'- phenyl-5'-aminopyrazöl Die Verbindung wird erhalten, indem man die Lösung von 2a-Cyano-17a-methyl-17ss=hydroxy-4- androsten-3-on (Herstellung siehe Beispiel 1) und Phenyl'hydrazin in Eisessig bei Zimmertempera tur über Nacht (etwa 15 Std.) stehenlässt und dann im Vakuum eindampft.
Das Rohprodukt wird durch Chromatographieren an der 20fachen Menge neu tralen Aluminiumoxyds (Akt. II) gereinigt, wobei die Verbindung mit Benzol und Benzol/Äther (9 :1) eluiert wird. Die Verbindung ist ölig. UV-Spektrum (Äthanol): Max. bei 280 m,u (log e = 4,29).
