Verfahren zur Herstellung von neuen N'-substituierten N-Arylsulfonyl-harnstoWen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer N'-substituierter N-Arylsulfonyl-harnstoffe mit wertvollen pharmakologischen Eigenschaften. Es wurde gefunden, dass N'-substituierte N-Arylsulfonyl-harnstoffe der Formel I,

in welcher R1 die Trifluormethylgruppe, einen niederen Alkenyl- oder Alkanoylrest oder einen unsubstituierten bzw.
einen durch Halogen, die Trifluormethylgruppe, einen niederen Alkyl-, Alkoxy-, Alkyl thio- oder Alkanoylrest substituierten Phenyl-, Benzoyl-, Benzyl-, Phenoxy- oder Phenylthiorest bedeutet, bei oraler oder parenteraler Applikation am Warmblüter bereits in sehr niedrigen Dosen eine starke hypoglykämische Wirkung ausüben. Im Gegensatz zu den für eine hypoglykämische Wirkung notwendigen Dosen liegen die toxischen Dosen der vorstehend definierten neuen Verbindungen sehr hoch, so dass sich bei der Verwendung der erfindungsgemäss herstellbaren neuen Verbindungen oder ihrer Salze als orale Antidiabetica ein äusserst günstiger therapeutischer Index ergibt.
Zur erfindungsgemässen Herstellung der neuen Verbindungen der Formel I setzt man l-Aminoadamantan mit einem reaktionsfähigen funktionellen Derivat einer N-Arylsulfonylcarbaminsäure der Formel II,

in welchen Formeln R1 die oben angegebene Bedeutung hat, gegebenenfalls in Gegenwart eines Kondensationsmittels um und hydrolysiert nötigenfalls ein zunächst erhaltenes N - Arylsulfonyl - N'-(1-adamantyi)- guanidin partiell zum entsprechenden Harnstoff. Als reaktionsfähige funktionelle Derivate von Carbaminsäuren der Formel II kommen beispielsweise deren niedere Alkylester, insbesondere Methyl- und Äthyl- ester, in Frage sowie deren Amide, N-Methyl-amide, N,N-Dimethylamide und N-Acylamide, das heisst z.
B. entsprechend der Definition von R1 substituierte N-Arylsulfonyl-harnstoffe,
N-Arylsulfonyl-N'-methyl-harnstoffe, N-Arylsulfonyl-N',N'-dimethyl-harnstoffe,
N-Arylsulfonyl-N'-alkanoyl-harnstoffe, N-Arylsulfonyl-N'-benzoyl-harnstoffe und N,N'-Bis-arylsulfonyi-harnstoffe.
Weiter kommen als reaktionsfähige funktionelle Derivate von Carbaminsäuren der Formel II deren Nitrile in Betracht, das heisst N-Arylsulfonyl-cyanamide, bei deren Umsetzung mit l-Amino-adamantan zunächst die oben erwähnten Guanidine entstehen, sowie die Halogenide, insbesondere Chloride. Die Umsetzungen werden beispielsweise in der Wärme in inerten organischen Lösungsmitteln wie z. B. Ather, Dioxan, Benzol, Chlorbenzol, Toluol, Xylol, ss-Methoxy-äthanol oder Butanon durchgeführt. Die Umsetzungen mit Carbaminsäureestern oder Harnstoffen können auch in Abwesenheit von Lösungs- oder Ver dünnungsmitteln durchgeführt werden. Sie benötigen im allgemeinen auch keine Kondensationsmittel; gewünschtenfalls können aber als solche Mittel z. B.
Alkalialkoholate verwendet werden.
Bei Umsetzungen mit Carbaminsäurechloriden dienen als Kondensationsmittel beispielsweise ebenfalls tertiäre organische Basen wie Pyridin, Triäthylamin oder Tributylamin. Bei Umsetzungen mit Carbaminsäurenitrilen (N-Arylsulfonyl-cyanamiden) dient als Kondensationsmittel z. B. Chlorwasserstoff, oder man verwendet anstelle des freien 1-Amino- adamantans ein Salz, insbesondere das Hydrochlorid desselben.
In den Verbindungen der Formel 1 und den entsprechenden Ausgangsstoffen der Formel II ist R1 z. B. durch den Trifluormethyl-, Vinyl-, Allyl-, Acetyl-, Propionyl- oder Butyrylrest oder durch einen ge gebenenfalls definitionsgemäss substituierten Phenyl-, Benzoyl-, Benzyl-, Phenoxy- oder Phenylthiorest verkörpert.
Die erfindungsgemäss herstellbaren neuen Verbindungen können als solche in Form ihrer Salze, z. B. von Ammonium- oder Alkalisalzen oder Salzen mit pharmakologisch unbedenklichen organischen Basen, bzw. in Gegenwart von Stoffen, die zur Salzbildung Anlass geben, wie z. B. Alkalicarbonaten und -bicarbonaten, als orale Antidiabetica zur Behandlung des Diabetes mellitus am Menschen angewendet werden, wobei sie oral in den üblichen Doseneinheitsformen verabreicht werden.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile, diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu 15,1 Teilen l-Amino-adamantan, gelöst in 120 Volumteilen absolutem Toluol, werden 27,3 Teile p-Benzyl-phenylsulfonylisocyanat (Kp. o ol 1460) gegeben, wobei Kristalle ausfallen. Das Gemisch wird 2 Stunden unter Rückfluss gekocht und anschliessend unter Vakuum auf das halbe Volumen eingeengt. Die erhaltenen Kristalle werden in 100 Volumteilen Aceton kalt gelöst und von wenig Unlöslichem abfiltriert.
Die Lösung wird mit 10 Volumteilen 2n Salzsäure versetzt und unter Rühren auf 500 Volumteile Eiswasser gegossen. Die Kristalle werden abgenutscht, gründlich getrocknet und aus Methanol umkristallisiert. Der so erhaltene N-(p-Benzyl-phenylsulfonyl) N'-(1-adamantyl)-harnstoff schmilzt bei 187,5 bis 188,50.
Beispiel 2
Aus 15,1 Teilen 1-Amino-adamantan und 25,9 Teilen p-Phenyl-phenylsulfonylisocyanat (Kp. oot 1600) erhält man analog zu Beispiel 1 den N-(p-Phenyl- phenylsulfonyl) - N' - (1 - adamantyl) - harnstoff vom Schmelzpunkt 177-1780 (aus Methanol).
Process for the preparation of new N'-substituted N-arylsulfonyl urines
The present invention relates to a process for the preparation of new N'-substituted N-arylsulfonylureas with valuable pharmacological properties. It has been found that N'-substituted N-arylsulfonylureas of the formula I,

Further possible reactive functional derivatives of carbamic acids of the formula II are their nitriles, that is to say N-arylsulfonyl-cyanamides, whose reaction with l-amino-adamantane initially gives rise to the above-mentioned guanidines and the halides, in particular chlorides. The reactions are carried out, for example, in the heat in inert organic solvents such as. B. ether, dioxane, benzene, chlorobenzene, toluene, xylene, ss-methoxy-ethanol or butanone carried out. The reactions with carbamic acid esters or ureas can also be carried out in the absence of solvents or diluents. In general, they also do not require any condensation agents; if desired, however, as such means, for. B.
Alkali alcoholates are used.
In reactions with carbamic acid chlorides, for example, tertiary organic bases such as pyridine, triethylamine or tributylamine also serve as condensing agents. In reactions with carbamic acid nitriles (N-arylsulfonyl-cyanamides) z. B. hydrogen chloride, or a salt is used instead of the free 1-amino adamantane, in particular the hydrochloride of the same.
In the compounds of formula 1 and the corresponding starting materials of formula II, R1 is z. B. embodied by the trifluoromethyl, vinyl, allyl, acetyl, propionyl or butyryl radical or by an optionally substituted phenyl, benzoyl, benzyl, phenoxy or phenylthio radical.
The novel compounds which can be prepared according to the invention can be used as such in the form of their salts, e.g. B. of ammonium or alkali salts or salts with pharmacologically acceptable organic bases, or in the presence of substances that give rise to salt formation, such. B. alkali carbonates and bicarbonates, are used as oral antidiabetics for the treatment of diabetes mellitus in humans, where they are administered orally in the usual unit dosage forms.
In the examples below, parts mean parts by weight; these relate to parts by volume as g to cm3. The temperatures are given in degrees Celsius.
example 1
27.3 parts of p-benzylphenylsulfonyl isocyanate (boiling point 1460) are added to 15.1 parts of l-aminoadamantane dissolved in 120 parts by volume of absolute toluene, crystals precipitating out. The mixture is refluxed for 2 hours and then concentrated to half its volume in vacuo. The crystals obtained are dissolved in 100 parts by volume of cold acetone and little insoluble matter is filtered off.
The solution is mixed with 10 parts by volume of 2N hydrochloric acid and poured onto 500 parts by volume of ice water while stirring. The crystals are filtered off with suction, dried thoroughly and recrystallized from methanol. The N- (p-benzylphenylsulfonyl) N '- (1-adamantyl) urea thus obtained melts at 187.5 to 188.50.
Example 2
From 15.1 parts of 1-amino-adamantane and 25.9 parts of p-phenyl-phenylsulfonyl isocyanate (bp oot 1600), N- (p-phenyl-phenylsulfonyl) - N '- (1 - adamantyl) is obtained analogously to Example 1 ) - urea, melting point 177-1780 (from methanol).
