Verfahren zur Herstellung von neuen N'-ubstituierten N-Arylsulfonyl-harnstoflben
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer N'-substituierter N-Aryl sulfonyl-harnstoffe mit wertvollen pharmakologischen Eigenschaften.
Es wurde gefunden, dass N'-substituierte N-Arytsulfonyl-hamstoffe der Formel I,

in welcher R1 Halogen, die Trifluormethylgruppe, einen niederen aliphatischen Kohlenwas serstoffrest, einen niederen Alkoxy-, Alkylthio- oder Alkanoylrest oder einen unsubstituierten bzw. einen durch einen der vorgenannten Substituenten substituierten Phenyl-, Benzoyl-, Benzyl-, Phenoxy- oder Phenylthio-rest, und R9 Halogen, die Trifluormethylgruppe, einen niederen Alkoxyrest oder einen niederen, ge sättigten aliphatischen Kohlenwasserstoffrest bedeuten, und R2 zusammen mit einem benachbarten R1 auch einen unsubstituierten bzw.
durch niederes Alkyl substituierten Trimethylen- bis Pentamethylen-rest bedeuten kann, bei oraler oder parenteraler Applikation am Warmblüter bereits in sehr niedrigen Dosen eine starke hypoglykämische Wirkung aus üben. Im Gegensatz zu den für eine hypoglykämische Wirkung notwendigen Dosen liegen die toxischen Dosen der vorstehend definierten neuen Verbindungen sehr hoch, so dass sich bei der Verwendung der erfindungsgemäss herstellbaren neuen Verbindungen oder ihrer Salze als orale Antidiabetica ein äusserst günstiger therapeutischer Index ergibt.
Zur erfindungsgemässen Herstellung der neuen Verbindungen der Formel I setzt man l-Aminoadamantan mit einem reaktionsfähigen funktionellen Derivat einer N-Arylsulfonylcarbaminsäure der Formel II,

in welchen Formeln R1 und Rs die oben angegebene Bedeutung haben, gegebenenfalls in Gegenwart eines Kondensationsmittels um und hydrolysiert nötigenfalls ein zunächst erhaltenes N-Arylsulfonyl-N'-(ladamantyl)-guanidin partiell zum entsprechenden Harnstoff. Als reaktionsfähige funktionelle Derivate von Carbaminsäuren de Formel II kommen beispiels weise deren niedere Alkylester, insbesondere Methylund ethylester in Frage, sowie deren Amide, N-Methylamide, N,N-Dimethylamide und N-Acylamide, das heisst z.
B. entsprechend den Definitionen von R1 und R2 substituierte
N-Arylsulfonyt4i arnstoffe,
N-Arylsulfonyl-N'-methyl-h arnstoffe,
N-Arylsulfonyl-N',N-dimethyl-h arnstoffe,
N-Arylsulfonyl-N'-alkanoyl-harnstoffe, N-Arylsulfonyl-N'-benzoyl-harnstoffe und N, N'-Bis-arylsulfonyl-harnstoffe.
Weiter kommen als reaktionsfähige funktionelle Derivate von Carbaminsäuren der Formel II deren Nitrile in Betracht, das heisst N-Arylsulfonyl-cyanamide, bei deren Umsetzung mit l-Amino-adamantan zunächst die oben erwähnten Guanidine entstehen sowie die Halogenide, insbesondere Chloride.
Die Umsetzungen werden beispielsweise in der Wärme in inerten organischen Lösungsmitteln, wie z. B. Äther, Dioxan, Benzol, Chlorbenzol, Toluol, Xylol, ss-Methoxyäthanol oder Butanon, durchgeführt. Die Umsetzungen mit Carbaminsäureestern oder Harnstoffen können auch in Abwesenheit von Lösungs- oder Verdünnungsmitteln durchgeführt werden. Sie ! benötigen im allgemeinen auch keine Kondensationsmittel; gewünschtenfalls können aber als solche Mittel z. B. Alkalialkoholate verwendet werden.
Bei Umsetzungen mit Carbaminsäurechloriden dienen als Kondensationsmittel beispielsweise eben falls tertiäre organische Basen wie Pyridin, Triäthylamin oder Tributylamin. Bei Umsetzungen mit Carb aminsäurenitrilen (N-Arylsulfonylcyanamiden) dient als Kondensationsmittel z. B. Chlorwasserstoff, oder man verwendet anstelle des freien l-Amino-adaman- tans ein Salz, insbesondere das Hydrochlorid desselben.
In den Verbindungen der Formel I und den Ausgangsstoffen der Formel II ist Rt für sich allein z. B. durch Fluor, Chlor, Brom, den Trlfluormethyl-, Methyl-, Äthyl-, Propyl-, Isopropyl-, n-Butyl-, tert. Butyl-, Vinyl-, Allyl-, Methoxy-, Äthoxy-, n-Propoxy-, Is oprop oxy-, n-Butoxy-, Is obutoxy-, Methylthio-, Äthylthio-, Isopropylthio-, Acetyl-, Propionyl-oder Butyrylrest, durch gegebenenfalls definitionsgemäss substituierte Phenyl-, Benzoyl-, Benzyl-, Phenoxyoder Phenylthioreste verkörpert.
R2 ist für sich allein beispielsweise Fluor, Chlor oder Brom, der TriflUormethyl-, Methoxy-, Methyl-, Äthyl-, n-Propyl- oder Isopropykest, und bildet zusammen mit R1 z. B. einen Trimethylen-oder Tetramethylenrest, der sich vorzugsweise in 3,4Stellung befindet.
Die erfindungsgemäss herstellbaren neuen Verbindungen können als solche oder in Form ihrer Salze, z. B. von Ammonium- oder Alkalisalzen oder Salzen mit pharmakologisch unbedenklichen organischen Basen bzw. in Gegenwart von Stoffen, die zur Salzbildung Anlass geben, wie z. B. Alkalicarbonaten und -bicarbonaten, als orale Antidiabetica zur Behandlung des Diabetes mellitus am Menschen angewendet werden, wobei sie oral in den üblichen Doseneinheitsformen verabreicht werden.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1 26,9 Teile N - (5,6,7,8- Tetrahydro - naphthyl-2- sulfonyl)-methylurethan in 500 Volumteilen Dioxan und 15,1 Teile l-Amino-adamantan in 100 Volumteilen Dioxan ergeben zunächst das Salz, welches durch etwa 3stündiges Erhitzen unter ständigem Abdestillieren des gebildeten Methanols in N-(5,6,7,8- Tetrahydrownaphthyl-2-sulfonyl)-N'-(l'-adamantyl)- harnstoff übergeführt wird. Nach dem Einengen und Umkristallisieren aus Äthanol schmilzt die Verbindung bei 158-161 .
Beispiel 2
Zu 25,4 Teilen N- (5,6,7, 8-Tetrahydro-naphthyl- 2-sulfonyl)-harnstoff in 500 Volumteilen Dioxan werden 15,1 Teile l-Amino-adamantan in 100 Volumteilen Dioxan gegeben, worauf sofort das Salz ausfällt. Das Reaktionsgemisch wird unter gutem Rühren und Rückfluss gekocht, bis kein Ammoniak mehr entweicht, dann unter Vakuum eingeengt und der Rückstand aus Äthanol umkristallisiert. Der er haltende N-(5, 6, 7, 8-Tetrahydro-naphthyI-2-sulfonyl)- N'-(1-adamantyl)-harnstoff schmilzt bei 158-1610.
