Verfahren zur Herstellung von neuen N'-substituierten N-Arylsulfonyl-harnstoffen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer N'-substituierter N-Aryl sulfonyl-hamstoffe mit wertvollen pharmakologischen Eigenschaften.
Es wurde gefunden, dass N'-substituierte N-Aryl.sulfonyl-harnstoffe der Formel I,

in welcher eines der Symbole R Halogen, einen niederen Alkyl-, Alkoxy- oder Alkylthiorest und das andere R Wasserstoff bedeuten, bei oraler oder parenteraler Applikation am Warmblüter bereits in sehr niedrigen Dosen eine starke hypoglykämische Wirkung ausüben. Im Gegensatz zu den für eine hypoglykämische Wirkung notwendigen Dosen liegen die toxischen Dosen der vorstehend definierten neuen Verbindungen sehr hoch, so dass sich bei der Verwendung der erfindungsgemäss herstellbaren neuen Verbindungen oder ihrer Salze als orale Antidiabetica ein äusserst günstiger therapeutischer Index ergibt.
Zur erfindungsgemässen Herstellung der neuen Verbindungen der Formel I setzt man l-Aminoadamantan mit einem reaktionsfähigen funktionellen Derivat einer N-Arylsulfonyl-carbaminsäure entsprechend der Formel II,

in welchen die beiden R die oben angegebene Bedeutung haben, gegebenenfalls in Gegenwart eines Kondensationsmittels um und hydrolysiert nötigenfalls ein zunächst erhaltenes N-Arylsulfonyl-N'-(l- adamantyl)-guanidin partiell zum entsprechenden Harnstoff. Als reaktionsfähige funktionelle Derivate von Carbaminsäuren der Formel II kommen beispielsweise deren niedere Alkylester, insbesondere Methyl- und Äthylester, in Frage, sowie deren.
Amide, N-Methylamide, N'N-Dimethylamide und N-Acylamide, das heisst z. B. entsprechend den Definitionen der beiden R substituierte
N-Arylsulfonyl-harnstoffe, N-Arylsulfonyl-N'-methyl-harnstoffe,
N-Arylsulfonyl-N', Nt-dimethyl-harnstoffe,
N-Arylsulfonyl-N'-alkanoyl-harnstoffe,
N-Arylsulfonyl-N'-benzoyl-harnstoffe und N,N'-Bis-arylsulfonyl-harnstoffe.
Weiter kommen als reaktionsfähige funktionelle Derivate von Carbaminsäuren der Formel II deren Nitrile in Betracht, das heisst N-Arylsulfonyl-cyanamide, bei deren Umsetzung mit l-Amino-adamantan zunächst die oben erwähnten Guanidine entstehen sowie die Halogenide, insbesondere Chloride.
Die Umsetzungen werden beispielsweise in der Wärme in inerten organischen Lösungsmitteln, wie z. B. Äther, Dioxan, Benzol, Chlorbenzol, Toluol, Xylol, ss-Methoxyäthanol oder Butanon, durchgeführt. Die Umsetzungen mit Carbaminsäureestern oder Harnstoffen können auch in Abwesenheit von Lösungs- oder Verdünnungsmitteln durchgeführt werden. Sie benötigen im allgemeinen auch keine Kondensationsmittel; gewünschtenfalls können aber als solche Mittel z. B. Alkalialkoholate verwendet werden.
Bei Umsetzungen mit Carbaminsäurechloriden dienen als Kondensationsmittel beispielsweise tertiäre organische Basen, wie Pyridin, Triäthylamin oder Tributylamin. Bei Umsetzungen mit Carb amins äurenitrilen (N-Arylsulfonylcyanamiden) dient als Kondensationsmittel z. B. Chlorwasserstoff, oder man verwendet anstelle des freien l-Amino-adaman- tans ein Salz, insbesondere das Hydrochlorid desselben.
In den Verbindungen der Formel I und den entsprechenden Ausgangsstoffen der Formel II ist ein R z. B. durch Fluor, Chlor, Brom, den Methyl-, 22ithyl-, Propyl-, Isopropyl-, n-Butyl-, tert.-Butyl-, Isobutyl-, Methoxy-, Sithoxy-, n-Propoxy-, Isopropoxy-, n-Butoxy-, Isobutoxy-, Methylthio-, Athylthio-oder Isopropylthio-rest verkörpert, und das andere R definitionsgemäss durch Wasserstoff.
Die erfindungsgemäss herstellbaren neuen Verbindungen können als solche oder in Form ihrer Salze, z. B. von Ammonium- oder Alkalisalzen oder Salzen mit pharmakologisch unbedenklichen organischen Basen bzw. in Gegenwart von Stoffen, die zur Salzbildung Anlass geben, wie z. B. Alkalicarbonaten und -bicarbonaten, als orale Antidiabetica zur Behandlung des Diabetes mellitus am Menschen angewendet werden, wobei sie oral in den üblichen Doseneinheftsformen verabreicht werden.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
Zu 24,3 Teilen N-m-Tolylsulfonyl-äthylurethan, in 600 Volumteilen Dioxan gelöst, werden 15,1 Teile l-Amino-adamantan, gelöst in 200 Volumteilen Dioxan gegeben, worauf sofort das Salz der beiden Reaktionskomponenten als dicker Kristallbrei ausfällt. Das Reaktionsgemisch wird zum Sieden erhitzt, wobei bald eine klare Lösung entsteht. Das freigesetzte Athanol wird ständig abdestilliert. Nach etwa 21/2 Stunden wird das Gemisch unter Vakuum eingeengt. Der Rückstand wird aus Methanol umkristallisiert, wobei man den N-m-Tolylsulfonyl-N' (l-adamantyl)-harnstoff erhält.
Beispiel 2
24,95 Teile N-(m-Chior-phenylsulfonyl)-methyl- urethan in 500 Volumteilen Dioxan und 15,1 Teile l-Amino-adamantan in 100 Volumteilen Dioxan ergeben zunächst das Salz, welches durch etwa 3stündiges Erhitzen unter ständigem Abdestillieren des gelbildeten Methanols in N-(m-Chlor-phenylsulfonyl) N'-(l-adamantyl)-harnstoff übergeführt wird. Nach dem Einengen und Umkristallisieren aus Athanol schmilzt der reine Harnstoff bei 179-182 .
Beispiel 3
Zu 23,45 Teilen m-Chlor-phenylsulfonyl-harnstoff (Smp. 170-172 ) in 500 Volumteilen Dioxan werden 15,1 Teile l-Amino-adamantan in 100 Vo lumteilen Dioxan gegeben, worauf sofort das Salz ausfällt. Das Reaktionsgemisch wird unter gutem Rühren und Rückfluss gekocht, bis kein Ammoniak mehr entweicht, dann unter Vakuum eingeengt und der Rückstand aus Äthanol umkristallisiert. Der erhaltene N-(m-Chlor-phenylsulfonyl)-N'-(l -adamantyl) harnstoff ist identisch mit demjenigen der Beispiele 1 und 3 und schmilzt bei 179-182 .
In analoger Weise erhält man bei der Umsetzung von 15,1 Teilen l-Amino-adamantan mit 23,45 Teilen o-Chlor-phenylsulfonyl-harn stoff (Smp. 169-171 ) den N-(o-Chlor-phenylsulfonyl)-N'-( 1 -adamantyl) - harnstoff vom Smp. 169-1700 aus Athyl- acetat; mit 23,0 Teilen m-Methoxy-phenylsulfonyl harnstoff den N-(m-Methoxy-phenylsulfonyl)-N'-(l- adamantyl-harnof; mit 21,4 Teilen. m-Tolylsulfonyl-harnstoff den N-(m-Tolylsulfonyl)-N'-( 1 -adamantyl)- harnstoff; mit 24,6 Teilen m-Methylthio-phenylsulfonyl harnstoff den N-(m-Methylthio-phenylsulfonyl)-N'- (l-adamantyl)-harnstoff.

In contrast to the doses required for a hypoglycemic effect, the toxic doses of the new compounds defined above are very high, so that an extremely favorable therapeutic index results when the new compounds or their salts which can be prepared according to the invention are used as oral antidiabetics.

The reactions are carried out, for example, in the heat in inert organic solvents, such as. B. ether, dioxane, benzene, chlorobenzene, toluene, xylene, β-methoxyethanol or butanone carried out. The novel compounds which can be prepared according to the invention can be used as such or in the form of their salts, e.g. B. of ammonium or alkali salts or salts with pharmacologically acceptable organic bases or in the presence of substances that give rise to salt formation, such. B. alkali carbonates and bicarbonates, are used as oral antidiabetics for the treatment of diabetes mellitus in humans, where they are administered orally in the usual dosage forms.
In the following examples, parts mean parts by weight; these are related to parts of volume as g to cm3. The temperatures are given in degrees Celsius.
example 1
To 24.3 parts of N-m-tolylsulfonylethyl urethane, dissolved in 600 parts by volume of dioxane, 15.1 parts of 1-amino-adamantane, dissolved in 200 parts by volume of dioxane, are added, whereupon the salt of the two reaction components immediately precipitates as a thick crystal slurry. The reaction mixture is heated to the boil, a clear solution soon being formed. The released ethanol is constantly distilled off. After about 21/2 hours, the mixture is concentrated in vacuo. The residue is recrystallized from methanol, N-m-tolylsulfonyl-N '(l-adamantyl) urea being obtained.
Example 2
24.95 parts of N- (m-chloro-phenylsulfonyl) -methyl urethane in 500 parts by volume of dioxane and 15.1 parts of 1-amino-adamantane in 100 parts by volume of dioxane initially give the salt, which is formed by heating for about 3 hours while constantly distilling off the gel Methanol is converted into N- (m-chlorophenylsulfonyl) N '- (l-adamantyl) urea. After concentration and recrystallization from ethanol, the pure urea melts at 179-182.
Example 3
15.1 parts of 1-aminoadamantane in 100 parts by volume of dioxane are added to 23.45 parts of m-chloro-phenylsulfonylurea (melting point 170-172) in 500 parts by volume of dioxane, whereupon the salt precipitates immediately. The reaction mixture is refluxed with thorough stirring until no more ammonia escapes, then concentrated in vacuo and the residue is recrystallized from ethanol. The N- (m-chloro-phenylsulfonyl) -N '- (1-adamantyl) urea obtained is identical to that of Examples 1 and 3 and melts at 179-182.
In an analogous manner, the reaction of 15.1 parts of 1-amino-adamantane with 23.45 parts of o-chlorophenylsulfonylurea (melting point 169-171) gives the N- (o-chlorophenylsulfonyl) -N '- (1-adamantyl) - urea of m.p. 169-1700 from ethyl acetate; with 23.0 parts of m-methoxyphenylsulfonyl urea the N- (m-methoxyphenylsulfonyl) -N '- (l-adamantyl-urea; with 21.4 parts m-tolylsulfonylurea the N- (m-tolylsulfonyl ) -N '- (1 -adamantyl) - urea; with 24.6 parts of m-methylthiophenylsulfonyl urea the N- (m-methylthiophenylsulfonyl) -N'- (l-adamantyl) -urea.
