Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Derivaten des Pyrimido- [4, 5-d]pyrimidins der Formel

in der R4 zusammen mit R5 und dem Stickstoffatom einen gegebenenfalls substituierten 5-oder 6gliedrigen heterocyclischen Ring, der gegebenenfalls durch weitere Heteroatome unterbrochen sein kann, bedeu- ten, und Ra einen Alkoxy-, Alkoxyalkoxy-oder Alkylaminoalkoxyrest bedeutet, bei welchem die Alkylgruppen mit dem N-Atom einen heterocyclischen Ring bilden.
Gemäss der Erfindung werden die neuen. Verbindungen hergestellt, indem man eine Verbindung der Formel

in der Z ein Halogenatom oder eine Hydroxylgruppe bedeutet, mit einer Alkalimetallverbindung eines entsprechenden Alkanols oder falls Z eine Hydroxylgruppe bedeutet, mit einem entsprechenden Alkylhalogenid oder Dialkylsulfat umsetzt.
Die Umsetzungen können in einem weiten Temperaturbereich erfolgen, vorzugsweise bei Temperaturen zwischen 20 und 200 C.
Die Reaktionskomponenten werden in der Regel in stöchiometrischen Mengenverhältnissen angewandt ; es kann auch ein Überschuss des Alkalialkoholats verwendet werden. Die Reaktion wird vorteilhaft in Gegenwart eines inerten Lösungsmittels und gegebenenfalls in Anwesenheit eines Reaktionsbeschleunigers und gegebenenfalls unter Druck durchgeführt.
Als Lösungsmittel eignen sich beispielsweise Wasser, Alkohole, Dioxane, Aceton, aromatische Kohlenwasserstoffe oder Dimethylformamid, als Re aktionsbeschleuniger Kupfer, Kupfersalze, Alkalijodide.
Die nachträgliche Alkylierung von Verbindungen, die in 4-Stellung eine freie Hydroxylgruppe enthalten, erfolgt vorzugsweise in wässrigem Medium und bei mässig erhöhten Temperaturen, in Gegenwart eines säurebindenden Mittels ; als solche haben sich anorganische Basen, wie Natriumhydroxyd oder Kaliumcarbonat, besonders bewährt.
Die als Ausgangsstoffe verwendeten Pyrimido [4, 5-d]-pyrimidine der Formel III sind entweder bekannt, oder können durch Kondensation entspre- chend substituierter Pyrimidine mit Estern oder Aminen der Ameisensäure, Kohlensäure oder Thio- kohlensäuren, nach an sich bekannten Methoden hergestellt werden. Halogensubstituierte Pyrimido [4, 5-d]-pyrimidine werden beispielsweise durch Kernhalogenierung oder durch Austausch von Hydroxyl gruppen gegen Halogenatome in an sich bekannter Weise erhalten.
Die erfindungsgemäss erhaltenen neuen Verbin- dungen besitzen wertvolle therapeutische Eigenschaften, insbesondere eine gute coronarerweiternde Wirksamkeit. Ausserdem weist eine Reihe von ihnen daneben eine sehr gute sedative Wirkung auf. Besonders hervorzuheben ist 2, 7-Dimorpholine-4-äthoxy pynido- [4, 5-d]-pyrimidin.
Die neuen Verbindungen können in Form ihrer freien Basen oder in Form ihrer nicht toxischen Salze mit anorganischen oder organischen Säuren verwendet werden. Die Überführung der Basen in die Säureadditionssalze kann auf bekannte Weise erfolgen. Als nicht toxische Säuren kommen vor allem folgende Säuren in Frage :
Salzsäure, Bromwasserstoffsäure, Schwefelsäure,
Phosphoräure, Essigsäure, Weinsäure,
Milchsäure, Bernsteinsäure, Sitronensäure,
Nikotinsäure, Camphersulfonsäure usw.
Beispiel 1
Verschiedene 2, 7-Diamino-4-alkoxy-pyrimido- [4, 5-d]-pyrimidine a) 2, 7-Dimorpholino-4-äthoxy-pyrimido- [4, 5-d] pyrimidin.
5, 0 g (0, 015 Mol) 2, 7-Dimorpholino-4-chlor- pyrimido- [4, 5-d]-pyrimidin werden in 500 ml absolutem Athanol gelöst. Zu dieser Lösung fügt man 0, 4 g (0, 017 Mol) Natrium in 20 ml absolutem ; Äthanol und erhitzt danach 2 Stunden zum Rückfluss. Man saugt vom ausgeschiedenen Natrumchlorid ab und dampft das Filtrat zur Trockene ein.
F. = 218-220 C (Athanol/Tierkohle).
Ausbeute : 2, 9 g (56 % der Theorie).

Die Mischung wird 5 Stunden unter Riihren zum Rückfluss erhitzt. Nach dem Abkühlen wird vom ausgefallenen Kochsalz abfiltriert und das Filtrat zur Trockne eingedampft.

7, 3 g (0, 022 Mol) 2, 7-Dimorpholino-4-chlor-py- rimido-[4, 5-d]-pyrimidin werden zu einer Lösung von 0, 5 g (0, 022 Mol) Natrium und 25 ml absolutem Dimethylaminoäthanol in 25 ml absolutem Toluol gegeben.
Die Mischung wird 50 Minuten unter Rühren zum gelinden Rückfluss erhitzt. Nach dem Abkühlen wird vom ausgeschiedenen NaCl abfiltriert und das Filtrat zweimal mit wenig 2n NaOH, anschliessend einmal mit wenig Wasser ausgeschüttelt und über Na2SO4 getrocknet. Nach dem Einengen wird aus Toluol/Petroläther umkristallisiert.

Beispiel 2
2, 7-Bis- (2-methylmorpholino)-4-methoxy- pyrimido- [4, 5-d]-pyrimidin
7, 0 g (0, 02 Mol) 2, 7-Bis- (2-methylmorpholino)- 4-hydroxy-pyrimido- [4, 5-d]-pyrimidin werden in 16 ml 2n Natronlauge gelöst.
Unter Rühren tropft man zu dieser Lösung bei 25 C 2, 5 ml (0, 025 Mol) Dimethylsulfat. Nach kurzer Zeit beginnt sich ein kristalliner Niederschlag abzuscheiden.
Etwa drei Stunden später werden weitere 2, 5 ml Dimethylsulfat zugegeben.
Man setzt das Rühren bei 25 C noch 15 Stunden fort, saugt dann die Kristalle ab und kristallisiert mehrmals aus Athanol um. Die Verbindung besitzt einen Schmelzpunkt von 198-199 C.

2,7-bis (2-methylmorpholino) -4-methoxypyrimido- [4,5-d] pyrimidine
