Patentschrift Nr. 826 539) bekanntgeworden.
Beson dere Bedeutung aber haben auf diesem Gebiete sub stituierte Sulfonylharnstoffe erlangt, von denen der N1-p-Toluolsulfonyl-N2-n-butyl-harnstoff, der N1-p- Aminobenzolsulfonyl-N2-n-butyl-harnstoff sowie der N1-p-Chlorbenzolsulfonyl-N2-n-propyl-harnstoff in die Praxis Eingang gefunden haben (z. B. brit. Patent schrift Nr. 808 071; F. Bertram und Mitarb., Dtsch. med. Wschr., 80, S. 1455, 1955; Samuel J. N. Sugar und Mitarb., Ann. of the New York Academy of Sciences, Vol. 74, S. 625, 1958/59).

Process for the preparation of new sulfonamides and their salts. Sulfonamide derivatives are known which are capable of lowering the blood sugar level after oral administration. This effect was demonstrated on 2- (p-aminobenzenesulfonamido) -5-isopropylthiodiazole by M. Janbon and colleagues. (Montpellier Méd. 21/22, 441, 1942) found. More recently, further sulfonamide derivatives of thiodiazole (British patent specification no. 822 947) and of oxadiazole (British patent specification no. 826 539) have become known.
Substituted sulfonylureas, of which N1-p-toluenesulfonyl-N2-n-butylurea, N1-p-aminobenzenesulfonyl-N2-n-butylurea and N1-p- have gained particular importance in this area. Chlorobenzenesulfonyl-N2-n-propylurea have found their way into practice (e.g. British Patent No. 808 071; F. Bertram and Mitarb., Dtsch. Med. Wschr., 80, p. 1455, 1955 ; Samuel JN Sugar et al., Ann. Of the New York Academy of Sciences, Vol. 74, pp. 625, 1958/59).
A group of new blood sugar-lowering sulfonamides, particularly suitable for the treatment of diabetes mellitus, has now been found which, when administered orally, bring about a strong and long-lasting blood sugar lowering and are characterized by low toxicity. These are compounds of the general formula:
EMI0001.0004
wherein A is a phenyl radical which is nitrogen-free on the p-carbon atom, optionally mono- or disubstituted or a saturated or unsaturated, straight-chain or branched or ring-closed hydrocarbon radical which can also be interrupted by one or more oxygen atoms, and B is the grouping -XR with X meaning a direct bond to the pyrimidine nucleus or an oxygen atom, and R denotes a saturated or unsaturated, straight-chain or branched or ring-closed hydrocarbon radical, which can also be interrupted by one or more oxygen atoms.
The salts of these compounds can also lower blood sugar levels.
The invention relates to a process for the preparation of these new compounds, which is characterized by the fact that a compound of the general formula A-SO2-NH2, where A has the meaning given above, in free form or as an alkali salt with a 2- R'-5-B-pyrimidine, in which R 'denotes a halogen atom, -N + 3 (CH3) 3, -NH 3 NO2 or NH 3 CN and B has the meaning given above.
The sulfonamides obtainable according to the invention can be used to prepare the corresponding salts, for example by reacting them with physiologically acceptable inorganic and / or organic bases.
The strong effectiveness of the group of 5-substituted 2-sulfonamidopyrimidines described above was all the less to be assumed than in the literature (Mc. Lamore et al., Ann. Of New York Acad. Of Sciences, 74, p.446 , 1958/59) it is known that 2 benzenesulfonamidopyrimidine has no effect on blood sugar.
When administered orally, the sulfonamides produced by the process according to the invention are blood-sugar-lowering agents whose strong and long-lasting effect is in some cases superior to the known agents.
A number of the compounds prepared according to the invention are also characterized by a particularly low toxicity, as is particularly desirable for a long-term therapeutic agent that has to be taken daily for years.
Table 1 below shows the mean lethal dose (LD50), tested on rats after intravenous administration, as well as the maximum blood glucose lowering given as a percentage of the initial value in fasting rabbits after an orally administered dose of 500 mg / kg Comparison of the NÚ- (4-methylbenzenesulfonyl) -Nê- n-butylurea most commonly used in medical practice was placed at the top under No. 1.

Given the importance of the possible non-toxicity of these preparations with long-term medication mentioned above, the subacute toxicity was determined in such a way that 100 mg / kg of substance were administered per os to 10 rats per day for 4 weeks, after which the liver of the killed animals was histologically was investigated.
The results of this subacute toxicity determination are compiled in Table 2 and also show a - in some cases even strong - superiority over NÚ - (4 - methylbenzenesulfonyl) - Nê-n-butylurea for the substances examined from the group prepared according to the invention Compound of the above-mentioned three sulfonylureas introduced in practice is undoubtedly the most compatible.
