Verfahren zur Herstellung von neuen Sulfonamiden und von deren Salzen Es sind Sulfonamidderivate bekannt, die imstande sind, nach oraler Verabreichung den Blutzuckerspie gel zu senken. So wurde diese Wirkung am 2-(p- Aminobenzolsulfonamido)-5-isopropylthiodiazol von M. Janbon und Mitarb. (Monpellier Méd. 2l/22, 441, 1942) aufgefunden. In neuerer Zeit sind weitere Sulfonamidderivate des Thiodiazols (brit. Patent schrift Nr. 822 947) sowie des Oxadiazols (brit. Patentschrift Nr. 826 539) bekanntgeworden.
Beson dere Bedeutung aber haben auf diesem Gebiet sub stituierte Sulfonylharnstoffe erlangt, von denen der N1-p-Toluolsulfonyl-N2-n-butyl-harnstoff, der N1-p- Aminobenzolsulfonyl-N2-n-butyl-harnstoff sowie der N1-p-Chlorbenzolsulfonyl-N2-n-propyl-harnstoff in die Praxis Eingang gefunden haben (z. B. britische Pa tentschrift Nr. 808 071; F. Bertram und Mitarb., Dtsch. med. Wschr., 80, S. 1455, 1955; Samuel J. N. Sugar und Mitarb., Ann. of the New York Academy of Sciences, Vol. 74, S. 625, 1959/59).
Es wurde nun eine Gruppe neuer, insbesondere zur Behandlung des Diabetes mellitus geeigneter blut zuckersenkender Sulfonamide gefunden, die bei oraler Verabreichung eine starke und lang anhaltende Blut zuckersenkung bewirken und sich durch geringe Toxi- zität auszeichnen.
Es handelt sich dabei um Verbin dungen der allgemeinen Formel

worin A einen am p-Kohlenstoffatom stickstofffreien, gegebenenfalls ein- oder zweifach substituierten Phe- nylrest oder einen gesättigten oder ungesättigten, ge- radkettigen oder verzweigten oder ringgeschlossenen Kohlenwasserstoffrest, der auch durch ein oder meh rere Sauerstoffatome unterbrochen sein kann, und B die Gruppierung -X-R mit X in der Bedeutung einer direkten Bindung zum Pyrimidinkern oder eines Sauerstoffatoms, und R eines gesättigten oder un gesättigten, geradkettigen oder verzweigten oder ring geschlossenen Kohlenwasserstoffrestes, der auch durch ein oder mehrere Sauerstoffatome unterbrochen sein kann, bezeichnen.
Auch die Salze dieser Ver bindungen können blutzuckersenkend wirken.
Gegenstand der Erfindung ist ein Verfahren zur Herstellung dieser Verbindungen, das sich dadurch auszeichnet, dass man Verbindungen der allgemeinen Formel: A-S(O)nY oder (A-SO2-)2O, worin A die angegebene Bedeutung hat, n die Zahlen worin A 0 bis 2 darstellt und Y, falls n = 2 ist, Hydroxyl oder Halogen, und, falls n = 0 oder 1 ist, Halogen bedeutet, mit 2-Amino-5-B-pyrimidin oder 2-Acyl- amino-5-B-pyrimidin, wobei B die oben angegebene Bedeutung hat, umsetzt und danach die erhaltenen Verbindungen erforderlichenfalls zu den Sulfonsäure- derivaten oxydiert.
Die erfindungsgemäss erhältlichen Sulfonamide können zur Herstellung der entsprechenden Salze ver wendet werden, indem man sie beispielsweise mit physiologisch unbedenklichen anorganischen und bzw. oder organischen Basen umsetzt.
Die starke Wirksamkeit der oben beschriebenen Gruppe von 5-substituierten 2-Sulfonamidopyrimidi- neu war um so weniger zu vermuten, als in der Lite ratur (Mc. Lamore und Mitarb., Ann. of New York Acad. of Sciences, 74, S. 446, 1958/59) bekannt ist, dass das 2-Benzolsulfonamidopyrimidin keine Wir kung auf Blutzucker hat.
Die nach dem erfindungsgemässen Verfahren her gestellten Sulfonamide sind bei oraler Applikation blutzuckersenkende Mittel, deren starke und lang anhaltende Wirkung den bekannten Mitteln zum Teil überlegen ist.
Eine Reihe der erfindungsgemäss hergestellten Verbindungen zeichnet sich dazu noch durch eine ganz besonders niedrige Toxizität aus, wie sie für ein Dauertherapeutikum, das jahrelang täglich eingenom men werden muss, besonders wünschenswert ist.
In der nachfolgenden Tabelle 1 ist die mittlere tödliche Dosis (LD5o), getestet an Ratten nach intra venöser Applikation, sowie die in Prozenten des Ausgangswertes angegebene maximale Blutzucker senkung am nüchternen Kaninchen nach einer oral applizierten Dosis von 500 mg/kg angeführt, wobei zum Vergleich der in der medizinischen Praxis am meisten verwendete N1-(4-Methylbenzolsulfonyl)-N2- n-butylharnstoff unter Nr. 1 an die Spitze gestellt wurde.

Bei der bereits erwähnten Bedeutung möglichster Ungiftigkeit bei diesen Präparaten mit Dauermedika tion wurde die subakute Toxizität in der Weise be stimmt, dass an je 10 Ratten 4 Wochen hindurch täg lich 100 mg/kg Substanz per os verabfolgt wurden, worauf die Leber der getöteten Tiere histologisch un tersucht wurde.

als Salze mit physiologisch vertretbaren anor ganischen und bzw. oder organischen Basen, wie z. B. gestellt und zeigen bei den untersuchten Substanzen der erfindungsgemäss hergestellten Gruppe ebenfalls eine - teilweise sogar starke - Überlegenheit gegen über N1 - (4 - Methylbenzolsulfonyl) - N2-n-butylharn- stoff, wobei diese Verbindung von den oben erwähn ten drei in der Praxis eingeführten Sulfonylharnstoffen zweifellos die verträglichste ist. Natrium-, Lithium-, Kalzium-, Ammoniumhydroxy, Aminen, wie Methylglukamin, Morpholin, Piperazin, Äthanolamine u. a., oder auch in Form von Mischun gen der freien Sulfonamide mit einem geeigneten Alkalibicarbonat bzw. Alkalicarbonat verabreicht werden.
Die Konfektionierung der Substanzen kann ohne oder mit den in der galenischen Pharmazie üblichen Zusätzen, Trägersubstanzen, Geschmackskorrigentien u. ä. erfolgen, beispielsweise in Pulverform, als Ta bletten, Dragées, Kapseln, Pillen, in Form von Sus pensionen oder Lösungen. <I>Beispiel 1</I> Zu einer Lösung von 12,5 g 2-Amino-5-methoxy- pyrimidin in 100 ml Wasser werden 15g Kalzium- carbonat und 20g Toluolsulfochlorid gegeben. Das Gemisch wird innerhalb 1/2 Stunde auf 80 C er- wärmt und 2 Stunden bei dieser Temperatur gerührt, bis alles Sulfochlorid verschwunden ist.
Dann werden 20 ml 30 % ige Natronlauge zugegeben und die Lö sung mit Kohle geklärt. Durch Fällung mit konzen trierter Salzsäure werden 21,5 g 2-(4-Methylbenzol- sulfonamido) - 5 - methoxypyrimidin vom F. 196 bis 197 C erhalten. <I>Beispiel 2</I> 15,3 g 2-Amino-5-n-propoxypyrimidin werden in 60 ml Pyridin gelöst und mit 19 g Toluolsulfochlorid 2 Stunden bei 60 C gerührt. Die Lösung wird dann in 300 ml Wasser gegossen, der Niederschlag abge saugt und mit Wasser gewaschen. Zur Reinigung wird das Filtergut in 300 ml n-Natronlauge gelöst, mit Kohle geklärt und filtriert.
Durch Fällung mit Salzsäure werden 29 g 2-(4-Methylbenzolsulfon- amido)-5-n-propoxypyrimidin vom F. 198-200 C er halten.
<I>Beispiel 3</I> 25 g 2-Amino-5-methoxypyrimidin werden in 250 ml trockenem Benzol gelöst. Dann wird unter Kühlung eine Lösung von 16 g p-Toluolsulfenylchlo- rid in 150 ml Benzol zugetropft. Das Gemisch wird anschliessend noch 1/2 Stunde bei 30 C gerührt. Nach dem Abkühlen werden die abgeschiedenen Kristalle abgesaugt und an der Luft getrocknet. Zur Entfer nung des mitausgefallenen 2-Amino-5-methoxypyri- midin-hydrochlorids wird das Filtergut mit 100 ml Wasser verrührt und abgenutscht. Es wird dann noch feucht in 300 ml Eisessig gelöst und durch 12stündiges Stehenlassen nach Zugabe von 60 ml Perhydrol oxydiert.
Anschliessend wird die Lösung im Vakuum auf 100 ml eingeengt und mit 300 ml Wasser verdünnt. Die abgeschiedenen Kristalle wer den abgesaugt und aus 50 % igem Aceton umkristalli siert. Ausbeute: 21,5 g 2-(4-Methylbenzolsulfon- amido)-5-methoxypyrimidin vom F. 196-197 C.
<I>Beispiel 4</I> 25 g 2-Amino-5-methoxypyrimidin werden in 300 ml Benzol gelöst und bei 5 C einer Lösung von 22 g p-Toluolsulfinsäurechlorid in 100 ml Benzol versetzt. Es wird noch 2 Stunden bei Zimmertempe ratur gerührt. Der Niederschlag, der ein Gemisch von 2 - (4 -Methylbenzolsulfinamido)-5-methoxypyrimidin und 2-Aminomethoxypyrimidin-hydrochlorid dar- stellt, wird abgesaugt und nach Extraktion des Hy drochlorids mit Wasser, wie unter Beispiel 3 angege ben, oxydiert. Ausbeute: 23 g 2-(4-Methylbenzolsul- fonamido)-5-methoxypyrimidin vom F. 196-197 C. <I>Beispiel 5</I> 17,7 g 2-Amino-5-cyclohexyloxypyrimidin wer den in 60 ml Pyridin gelöst. Nach Zugabe von 17 g Benzolsulfochlorid wird das Gemisch 2 Stunden auf 60 C erwärmt.
Es wird dann in 300 ml Wasser ge gossen, der gebildete Niederschlag abfiltriert und in verdünntem Ammoniak gelöst. Nach Klären der Lö sung mit Kohle werden 30 g 2-Benzolsulfonamido- 5-cyclohexyloxypyrimidin durch Fällen mit Essig säure erhalten. F. 225-226 C.
<I>Beispiel 6</I> 12,5 g 2-Amino-5-methoxypyrimidin werden in 60 ml Pyridin gelöst und mit 17 g Isoamylsulfochlorid versetzt. Das Gemisch wird 3 Stunden bei Zimmer temperatur gerührt und dann in 300 ml Wasser ge gossen. Der Niederschlag wird abfiltriert, mit Wasser gewaschen und zur Reinigung in verdünntem Ammo niak gelöst. Nach Klären der Lösung mit Kohle wer den durch Fällen mit Essigsäure 20 g 2-Isoamyl- sulfonamido-5-methoxypyrimidin vom F. 163-165 C erhalten.
<I>Beispiel 7</I> 12,5 g 2-Amino-5-methoxypyrimidin werden in Pyridin mit 18g Äthoxyäthylsulfochlorid 3 Stunden bei Zimmertemperatur gerührt. Dann wird das Pyri- din im Vakuum abdestilliert und der Rückstand mit 200 ml Wasser 1 Stunde gerührt. Der Niederschlag wird abfiltriert und zur Reinigung aus verdünntem Ammoniak umgefällt. Man erhält so 20 g 2-Äthoxy- äthylsulfonamido-5-methoxypyrimidin vom F. 124 bis <B>1250</B> C.
<I>Beispiel 8</I> Zu einer Lösung von 25 g 2-Amino-5-methoxy- pyrimidin in 300 ml absolutem Äther werden unter Kühlung 14 g Isoamylsulfensäurechlorid, gelöst in 100 ml absolutem Äther, zugetropft. Das Reaktions gemisch wird anschliessend noch 1/2 Stunde bei leich tem Sieden gerührt. Das dabei ausfallende Gemisch von 2-Isoamylsulfenamido-5-methoxypyrimidin und 2-Amino-5-methoxypyrimidin-hydrochlorid wird ab gesaugt, mit 100 ml Wasser verrührt und erneut fil triert. Der Filterrückstand wird in 200 ml Eisessig gelöst und nach Zugabe von 60 ml Perhydrol 24 Stunden bei Zimmertemperatur stehengelassen.
An schliessend wird die Lösung im Vakuum auf etwa 50 ml eingeengt und die Restlösung mit 200 ml Was ser verdünnt, wobei sich kristallines 2-Isoamylsulfon- amido-5-methoxypyrimidin abscheidet, das nach Ab trennung und Umkristallisieren aus 50 % igem Alko hol bei 163-165 C schmilzt. Die Ausbeute beträgt 12 g.
<I>Beispiel 9</I> 25 g 2-Amino-5-äthoxypyrimidin werden in 200 ml absolutem Äther gelöst und bei 5 C unter Rühren mit einer Lösung von 15 g Isoamylsulfin- säurechlorid in 100 ml Äther versetzt. Es wird noch 2 Stunden bei Zimmertemperatur gerührt. Der Nie derschlag wird abgesaugt und analog Beispiel 8 mit Perhydrol oxydiert. Nach dem Umkristallisieren aus 50 % igem Aceton werden 14 g 2-Isoamylsulfonamido- 5-äthoxypyrimidin vom F. 170-172 C erhalten.
<I>Beispiel 10</I> 20g wasserfreie p-Toluolsulfosäure und 17 g 2-Acetylamino-5-methoxypyrimidin werden in 200 ml absolutem Äthanol gelöst. Dann werden innerhalb von 2 Stunden 150 ml Äthanol abdestilliert. Der

<I>Beispiel 80</I> 62 g 2-Benzolsufonamido-5-methoxyäthoxypyri- midin werden mit 8 g NaOH in 250 ml Äthanol ge löst. Die Lösung wird zur Trockne eingedampft und der Rückstand in 300 ml Aceton suspendiert. Das Salz wird abgesaugt, mit Aceton gewaschen und ge trocknet. Ausbeute etwa 60 g, F. 220-223 C.
In analoger Weise wurde 2-(4-Methylbenzolsul- fonamido) - 5 - methoxypyrimidin-natrium dargestellt, das bei 273-275 C schmilzt.
<I>Beispiel 81</I> 61 g 2-Benzolsulfonamido-5-methoxyäthoxypyri- midin werden in 250 ml H20 mit 7,4 g Ca(OH)2 heiss gelöst. Die Lösung wird filtriert und im Vakuum bis zur Trockne eingeengt. Der Rückstand wird mit 300 ml Aceton verrührt und abfiltriert. Nach dem Trocknen erhält man 60g Kalziumsalz mit einem F. von etwa 150 C. <I>Beispiel 82</I> 25 g 2 - Benzolsulfonamido - 5 - methylpyrimidin werden mit 15g Äthanolamin in 300 ml Äthanol heiss gelöst. Nach dem Abkühlen wird der dabei ent standene Niederschlag abgesaugt und mit Äthanol gewaschen. Nach dem Trocknen bei etwa 50 C erhält man 26g Äthanolaminsalz vom F.138-140 C.
<I>Beispiel 83</I> Zur eventuellen äusseren Anwendung löst man 21,46 g 2-Benzolsulfonamido-5-methoxyäthoxypyri- midin-natrium, entsprechend 20 g freiem Sulfonamid, zu 100 ml mit destilliertem Wasser. 1 ml dieser Lö sung enthält 200 mg Sulfonamidwirkstoff. Gegebe nenfalls kann eine solche Lösung in üblicher Weise sterilisiert werden.

To remove the co-precipitated 2-amino-5-methoxypyrimidine hydrochloride, the filter material is stirred with 100 ml of water and suction filtered. It is then dissolved in 300 ml of glacial acetic acid while still moist and oxidized by leaving it to stand for 12 hours after adding 60 ml of Perhydrol.

<I> Example 4 </I> 25 g of 2-amino-5-methoxypyrimidine are dissolved in 300 ml of benzene and a solution of 22 g of p-toluenesulfinic acid chloride in 100 ml of benzene is added at 5 ° C. <I> Example 6 </I> 12.5 g of 2-amino-5-methoxypyrimidine are dissolved in 60 ml of pyridine, and 17 g of isoamyl sulfochloride are added. The mixture is stirred for 3 hours at room temperature and then poured into 300 ml of water. The precipitate is filtered off, washed with water and dissolved in dilute ammonia for purification. <I> Example 8 </I> To a solution of 25 g of 2-amino-5-methoxypyrimidine in 300 ml of absolute ether, 14 g of isoamylsulfenic acid chloride, dissolved in 100 ml of absolute ether, are added dropwise with cooling. The reaction mixture is then stirred for a further 1/2 hour at gentle boiling. The resulting mixture of 2-isoamylsulfenamido-5-methoxypyrimidine and 2-amino-5-methoxypyrimidine hydrochloride is sucked off, stirred with 100 ml of water and filtered again. The filter residue is dissolved in 200 ml of glacial acetic acid and, after adding 60 ml of Perhydrol, left to stand for 24 hours at room temperature.
The solution is then concentrated in vacuo to about 50 ml and the remaining solution is diluted with 200 ml of water, crystalline 2-isoamylsulfonamido-5-methoxypyrimidine separating out, which after separation and recrystallization from 50% alcohol at 163 -165 C melts. The yield is 12 g.
<I> Example 9 </I> 25 g of 2-amino-5-ethoxypyrimidine are dissolved in 200 ml of absolute ether and a solution of 15 g of isoamylsulfinic acid chloride in 100 ml of ether is added at 5 ° C. while stirring. It is stirred for a further 2 hours at room temperature. The precipitate is suctioned off and oxidized analogously to Example 8 with Perhydrol.
