Verfahren zur Herstellung von neuen Sulfonamiden und von deren Salzen
Es sind Sulfonamidderivate bekannt, die imstande sind, nach oraler Verabreichung den Blutzuckerspiegel zusenken. So wurde diese Wirkung am 2- (p-Aminobenzolsulfonamido)-5-isopropylthiodiazol von M. Janbon und Mitarb. (Montpellier Med. 21/22 [1942], S. 441) aufgefunden. In neuerer Zeit sind weitere Sulfonamidderivate des Thiodiazols (brit. Patentschrift Nr. 822, 947) sowie des Oxadiazols (brit. Patent-

Die nach dem erfindungsgemässen Verfahren hergestellten Sulfonamide sind bei oraler Applikation blutzuckersenkende Mittel, deren starke und lang anhaltende Wirkung den bekannten Mitteln zum Teil überlegen ist.
Eine Reihe der erfindungsgemäss hergestellten Verbindungen zeichnet sich dazu noch durch eine ganz besonders niedrige Toxizität aus, wie sie für ein Dauertherapeutikum, das jahrelang täglich eingenommen werden muss, besonders wünschenswert ist.
In der nachfolgenden Tabelle 1 ist die mittlere tödliche Dosis (LDso) getestet an Ratten nach intravenöser Applikation, sowie die in Prozenten des Ausgangswertes angegebene maximale Blutzuckersenkung an nüchternen Kaninchen nach einer oral applizierten Dosis von 500 mg/kg angeführt, wobei zum Vergleich der in der medizinischen Praxis am meisten verwendete N- (4-Methylbenzolsulfonyl)-N2-n-butyl- - harnstoff (bekannt unter dem Handelsnamen"Rastinon"bzw."Tolbutamide") unter Nr. 1 an die Spitze gestellt wurde.

Zum therapeutischen Gebrauch können die erfindungsgemäss hergestellten Substanzen als freie Sulfonamide, als Salze mit physiologisch vertretbaren anorganischen und bzw.
oder organischen Basen, wie z. B.
Natrium-, Lithium-, Kalzium-, Ammoniumhydroxy, Aminen, wie Methylglucamin, Morpholin, Piperazin, Äthanolamine u. a., oder auch in Form von Mischungen der freien Sulfonamide mit einem geeigneten Alkalibicarbonat bzw. Alkalicarbonat verabreicht werden.
Die Konfektionierung der Substanzen kann ohne oder mit den in der galenischen Pharmazie üblichen Zusätzen, Trägersubstanzen, Geschmackskorrigentien u. ähnl. erfolgen, beispielsweise in Pulverform, als Tabletten, Dragees, Kapseln, Pillen, in Form von Suspensionen oder Lösungen.
Beispiel l : Zu einer Lösung von 12, 5 g 2-Amino-S-methoxypyrimidin in 100 ml Wasser werden 15 g Kalziumcarbonat und 20 g Toluolsulfochlorid gegeben. Das Gemisch wird innerhalb 1/2 h auf 800C erwärmt und 2 h bei dieser Temperatur gerührt, bis alles Sulfochlorid verschwunden ist. Dann werden

20 ml 30% igue Natronlauge zugegeben und die Lösung mit Kohle geklärt. Durch Fällung mit konzentrierter Salzsäure werden 21, 5 g 2- (4-Methylbenzolsulfonamido) -5-methoxypyrimidin vom F = 196 - 1970C erhalten, Beispiel 2 : 15, 3 g 2-Amino-5-n-propoxypyrimidin werden in 60ml Pyridin gelöst und mit 19 g i Toluolsulfochlorid 2 h bei 60 C gerührt.
Die Lösung wird dann in 300 ml Wasser gegossen, der Niederschlag abgesaugt und mit Wasser gewaschen, Zur Reinigung wird das Filtergut in 300 mln-Natronlauge gelöst, mit Kohle geklärt und filtriert. Durch Fällung mit Salzsäure werden 29 g 2- (4-Methylbenzolsulfon- amido)-5-n-propoxypyrimidin vom F = 198 - 2000C erhalten.
Beispiel 3 : 25 g 2-Amino-5-methoxypyrimidin werden in 250 ml trockenem Benzol gelöst. Dann ) wird unter Kühlung eine Lösung von 16 g p-Toluolsulfenylchlorid in 150 ml Benzol zugetropft. Das Gemisch wird anschliessend noch 1/2 h bei 300C gerührt. Nach dem Abkühlen werden die abgeschiedenen

Beispiel 5 : 17, 7 g 2-Amino-5-cyclohexyloxypyrimidin werdenin 60 ml Pyridin gelöst. Nach Zu- gabe von 17 g Benzolsulfochlorid wird das Gemisch 2 h auf 600C erwärmt. Eswirq dann in 300 ml Wasser gegossen, der gebildete Niederschlag abfiltriert und in verdünntem Ammoniak gelöst. Nach Klären der Lö- sung mit Kohle werden 30 g 2-Benzolsulfonamido-5-cyclohexyloxypyrimidin durch Fällen mit Essigsäure erhalten. F = 225-226 C.
Beispiel 6 : 12, 5 g 2-Amino-5-methoxypyrimidin werden in 60 ml Pyridin gelöst und mitl7g
Isoamylsulfochlorid versetzt. Das Gemisch wird 3 h bei Zimmertemperatur gerührt und dann in 300 ml
Wasser gegossen. Der Niederschlag wird abfiltriert, mit Wasser gewaschen und zur Reinigung in verdünn- tem Ammoniak gelöst. Nach Klären der Lösung mit Kohle werden durch Fällen mit Essigsäure 20 g 2-Iso- amylsulfonamido-5-methoxypyrimidin vom F=163 - 1650C erhalten.
Beispiel 7 : 12, 5g2-Amino-5-methoxypyrimidin werden in Pyridin mit 18 g Äthoxyäthyl- sulfochlorid 3 h bei Zimmertemperatur gerührt. Dann wird das Pyridin im Vakuum abuestilliert und der
Rückstand mit 200 ml Wasser 1 h gerührt. Der Niederschlag wird abfiltriert und zur Reinigung aus ver- dünntem Ammoniak umgefällt. Man erhält so 20 g 2-Äthoxyäthylsulfonamido-5-methoxypyrimidin vom
F = 124-1250C.
Beispiel 8 : Zu einer Lösung von 25 g 2-Amino-5-methoxypyrimidin in 300 ml absolutem Äther werden unter Kühlung 14 g Isoamylsulfensäurechlorid, gelöst in 100 ml absolutem Äther, zugetropft. Das Reaktionsgemisch wird anschliessend noch 1/2 h bei leichtem Sieden gerührt. Das dabei ausfallende Gemisch von 2-Isoamylsulfenamido-5-methoxypyrimidin und 2-Amino-5-methoxypyrimidin-hydrochlorid wird abgesaugt, mit 100 ml Wasser verrührt und erneut filtriert.
Der Filterrückstand wird in 200 ml Eisessig gelöst und nach Zugabe von 60 ml Perhydrol 24 h bei Zimmertemperatur stehengelassen. Anschlié- ssend wird die Lösung im Vakuum auf etwa 50 ml eingeengt und die Restlösung mit 200 ml Wasser verdünnt, wobei sich kristallines 2-Isoamylsulfonamido-5-methoxypyrimidin abscheidet, das nach Abtrennung und Umkristallisieren aus 50o/oigem Alkohol bei 163 - 1650C schmilzt. Die Ausbeute beträgt 12 g.
B e i s p i e l 9: 25 g 2-Amino-5-äthoxypyrimidin werden in 200 ml absolutem Äther gelöst und bei 50C unter Rühren mit einer Lösung von 15 g Isoamylsulfinsäurechlorid in 100 ml Äther versetzt. Es wird noch 2 h bei Zimmertemperatur gerührt. Der Niederschlag wird abgesaugt und analog Beispiel 8 mit Perhydrol oxydiert. Nach dem Umkristallisieren aus 50% igemAceton werden 14 g 2-Isoamylsulfonamido- - 5-äthoxypyrimidin vom F = 170-172 C. erhalten.

Beispiel 80 : 62 g 2-Bcnzolsulfonamido-5-methoxyäthoxypyrimidin werden mit 8 gNaOHin 250 ml Äthanol gelöst. Die Lösung wird zur Trockne eingedampft und der Rückstand in 300 ml Aceton suspendiert. Das Salz wird abgesaugt, mit Aceton gewaschen und getrocknet.
Ausbeute etwa 60 g, F = 220-2230C.
In analoger Weise wurde 2- (4-Methylbenzolsulfonamido)-5-methoxypyrimidin-natrium dargestellt das bei 273 - 2750C schmilzt.
Beispiel 81 : 61 g 2-Benzolsulfonamido-5-methoxyäthoxypyrimidin werden in 250 ml HO mit 7, 4 g Ca (OH) heiss gelöst. Die Lösung wird filtriert und im Vakuum bis zur Trockne eingeengt. Der Rückstand wird mit 300 ml Aceton verrührt und abfiltriert. Nach dem Trocknen erhält man 60 g Kalziumsalz mit einem F von etwa 150 C.
Beispiel 82 : 25 g 2-Benzolsulfonamido-5-methylpyrimidin werden mit 15 g Äthanolamin in 300 ml Äthanol heiss gelöst. Nach dem Abkühlen wird der dabei entstandene Niederschlag abgesaugt und mit Äthanol gewaschen. Nach dem Trocknen bei etwa 500C erhält man 26 g Äthanolaminsalz vom F = 138-1400C.
