Neue Wuchsstoffe und ihre Herstellung
Aus Materialien biologischen Ursprungs, insbe- sondere Pflanzenmaterial, tierischen Organen und Mikroorganismen, sind bereits eine grosse Zahl biologisch und physiologisch aktiver Stoffe isoliert worden. Über das weitverbreitete Vorhandensein einer bestimmten ; Gruppe von Wuchsstoffen war jedoch nichts bekannt.
Es wurde nun gefunden, Idass man aus Mikroor- ganismen und deren Extrakten Wuchs, stoffe in reiner bzw. angereicherter Form erhalten kann,, die im fol- genlden Antisideromycinle genanX werden.
Die Antisideromycine sinld stickstoff-unld eisenhaltige organische Verbindungenl, deren Säurehydro- lysate ninhydrinpositive Substanzen enthalben. Sie besitzen eine rote Farbe uinid sind leicht löslich in Säuren und stark polaren Lösungsmitteln, wie Was ; ser, Dimethylformamid, Glykol, Äthylenglykolmonomethyläther, sowie in nielderen aliphatisohen Alkoholen, wie Methanol. Auch in höheren aliphatischen Alkoholen sowie in armomlatischen Alkoholen und Phenolen besitzen sie eine beschränkte Löslichkeit, z. B. in Butanol, Ben ylalkoh, ol, Phenol.
Ein charakteristischer Vertreter dieser Gruppe ist das Antisideromycin Al (auch Fenioxamin B ge nannt), das im Patent Nr. 413 853 beschriehen ist. Es wird als Hauptprodukt z. B. von Streptomyceten pro duziert. Danebe, n findet man drei weitere Stoffe, die als Antisideromycin A2 und A3 umd B bezeichnet werden.

Bei der Hochspannungs-Elektrophorese auf Papier in 1/3-n. Essigsäure wandern Antisideromycin A2 und A3 innert 4 Stunden Laufzeit (1000 Volt) 8, 4 cm bzw. 12 cm weit.
Bei der Hydrolyse mit Säuren erhält man Produkte, die zum Teil mit Ninhydrin eine positive Farbreaktion geben.
Dia freien Basen der Antisideromycine sind leicht in iiblicher Weise aus ihren Salzen zugänglich, aus dem Sulfat z. B. durch Umsetzung in wässrigem ausgearbeitete Test von Bonifas [V. Bonifas, Experientia 8, 234 (1952)] sinngemäss angewendet werden kans.
Die bereits genannten Substanzen Ferrichrom, Terregens Factor und Coprogen gleichen in ihrem Vitamincharaktar fiir Arthrobacter terregems und Athrobacter flavescens den Antisideromycinen.
Hingegen ist die oben geschilderte, für die Antiside romycine typische, wachsbumsfördernde Wirkung auf Baoillus subtils, Micrococcus pyogenes, Saccharomyces cerevisiae, Ustilago sphaerogena und Chlamy domonias eugametos für die drei Substanzen Ferrichrom, Terregens Factor und Coprogen nicht be kannt. Zud ! em fehlt diesen Substanzen, die starke und typisch, e Antisi, deromycinlwirkung.
Wie in Tabe31e 1 gezeigt worden ist, unterscheidet sich das Ferrichrom weitssr Volz den Antisideromyci- nen durch sefin papierchromatographisches Verhal- ten. Anderseits unterschenden sich Coprogen unld der Terregens Factor durch ihren Eisengehalt. Ersteres enthält 6, 61 /o Journ [Amer. Chem. Soc. 74, 1362 (1952)] und letzterer nur Spuren Eisen [Can. Jours.
Biochem. an, d Physiol. 32, 400 (1954)]. Im Gegensatz dazu wurde fiir Antisideromycin Al 4, 5 bis 5, 5 /o Eisen gefunden.
Die Herstellung des Gemisches der Anísidero- mycine A2, A3 und B kann nach an sich bekannten Methoden unter Beriieksichtigung der oben gegebenen physikalischen Daten und insbesondere mit Hilfe des zur Verfolgung der Isolierung geeigneten biologi eschez Testes erfolgen. Das erfindun, gsgemässe Verfahren ist dadurch gekennzeichnet, sdass man einen Antisideromycin erzeugenden Mikroorganismen- stamm züchtet, bis das Kulturmedium eine wesentliche Antisideromycinwirkung aufweist, unld aus dem Kulturfiltrat das Gemisch der Antisideromycine A2, A3 und B isoliert.
Als Ausgangsstoffe für die Gewinnung der Anti side, romycinid werden Kulturen von Mikroarganis- mfen, vor allem von Vertretern der Gattung Streptomyces, von Bakterien, z. B. von B. subtilis, oder von Hefen, z. B. von Saccharomyces cerevisiae, verwen- dot.
Eine besonders bdvorzugte Quelle sind, Kulturen vonz Streptomyceben, die anhand der von Ettlinger et al. [Arch. Mikrobiol. 3, 326 (1958)] vorgeschlagenen Merkmale der folgenden Arten zugeordnet werden : Streptomyces griseoflavus (Krainsky) Waksman et Henrici, Streptomyces laven, dulae (Waksman et Curtis) Waksman et Henfici, Streptomyces galilaeus Ett luger et al., Streptomyces pilosus Ettlinger et al., Streptomyces polychromogenus Hagemann, Pinasse et Teillon, Streptomyces viridochromogenes (Krainsky) Waksman et Henrici, Streptomyces aureofaciens Duggar,
Streptomyces divaceus (Waksman) Waksman et Henrici, Streptomyces griseus (Krainsky) Waksman et Henrici, Streptomyces glau cescens Gause et al.
Zur Gewinnung grösserer Mengen von Antisideromycin werden vorteilhaft Kulturen der genannten Me, clium mit Bariumhydroxyd, Neutralisierung des überschüssigen Baryts mit Kohlendioxyd sowie Abtrennung des Bariumcarbonat- und -sulfatnieder schlages und Isolierung der freiexl Base mitbels Ge- friertrocknung. Eivfacher erfolgt die Herstellung aus den Salzen unter Verwendung eines basischen Anionenaustauschers.
Die Salze der Antisideromycine können sich von den bekannben anorganischen und organischen Salzen ableiten, beispielsweise von der Salzsäure, den Schwefelsäuren unld Phosphorsäuren, der Essig-, Propion-, Valerian-, Palmitin- oder Ölsäure, der Bernsteinsäure, Zitronensäure, Weinsäure, Mandel- säure, Glutaminsäure oder Pantothensäure. Sie stellen neutrale oder saure Salze, diar. Ihre Herstellung kann durch Einwirkung der entsprechenden Säuren auf die fraie Base oder durch, doppelbe Umsetzung vong Salzen, beispielsweise von Antisideromycin-Sulfat mit pantothensaurem Calcium erfolgen.
Die Antisideromycine besitzen wachstumsfördernde Eigenschaften für eine grosse Zahl von Organismen. So besitzen sie einen solchen Effekt auf Bacillus subtils, Micrococcus pyogenes var. aureus, Saccharomyces cerevisiae, Ustilago sphaerogena und Chlamydomonas eugametos.
Andere Organismen, z. B. Vertreter Ider Gattung Arthrobacter, wie Arthrobacter terregens und Arth robacter flavescens, entwickeln sich überhaupt nur in Anwesenheit von Antisideromycin. In diesen Fällen besitzen die Antisideromycine also einen lihnlichen Vitamincharaktor, wie es far die Wirksubstanzen Ferrichrom, Terregens Factor und Coprogen gezeigt worden zist. [Bact. Rev. 21, 101 (1957)].
Eine weitere biologische Eigenschaft der Antisideromycine besteht darin, dass sie im Stande sind, die Wirkung von Antibiotika, die der Gruppelder Sidero- mycine angehören, gagenüber grampositiven Erre- gern kompetitiv aufzuheben. Zu den Sideromycinten gehören u. a. die eisenhaltigen Antibiotika Grisein, Albomycin, Ferrimycin, das Antibiotikum 1787 H. Thrum, Naturwiss. 44, 561 (1957) und die Substanzen L. A. 5352 und L. A. 5937 [P. Sensi und M. T. Timbal, Antibiotics & Chemotherapy 9, 160 (1959)]. Anscheinend besteht ein Antagonismus zwischen den Antisideromycinen einerseits und den.
Sideromycinen anderseits, der sowohl in vitro als auch in vivo nachgewiesen werden kann. Von dieser Wirkung ist im übrigen auch der Name Antisi- deromycine abgeleitet.
Die geschilderte antagonsistische Wirkung beschränkt sich ausschliesslich auf die Sideromycin Antibiotika und konnte bei anderen Antibiotika, wie Neomycin, Viomycin, Streptomycin, Streptothricin, Penicillin, Brythromycin oder Novobiocin, nicht beobachtet werden.
Die Aufhebung der antibiotischen, Wirkung der Sideromycine in vitro eignet sich gut für eine qualitative Erkennung un, d quantitative Bestimmung der Antisideromycine, indem der an und für sich zur Bestimmung von synergistisch wirkender Substanzen Mikroorganismen verwendet. Besonders bewährt haben sich in dieser Beziehung die oben erwähnten Streptomyces-Stämme"die sich leicht in grösserem Massstab züchten lassen.
Die vorliegende Erfindung ist daher nichet auf die Verwendung der Vertreter der genannten Arten beschränkt, sondern betrifft auch die Verwendung vonl Antisiderornycin-bildenden Stämmen anderer Arten und insbeson, dere von Vari- anten aller dietser Organismen, wie sie z. B. durch Selektionierung oder Mutatlon, inisbesondere von Ultraviolett-oder Röntgenstrahlen oder von Stickstoffsenfölen gewonnen werden.
Zur Gewinaungider Antisideromycine in grosse- rem MAssstab wird z. B. ein die Eigenschaften der oben angeführten streptomyceten aufweisencder Stamm, z. B. in wässriger, KohlenNhydrate, stickstoilE- haltige Verbindungen sowie anorganische Salze enthaltender Nährlösung aerob gezüchtet, bis diese eine wesentliche Antisideromycinwirkung zeigt, und die Antisideromycine hierauf isoliert. Man kann atber auch Bakte. rien, wie B. subtils, züchten und aus den betreffenden Kulturen die Antisideromycine in reine-r bzw. angereicherter Form isolieren.
Für die Züchtung der genaneten Pilze oder B, akberien kommlen als assi milierbare Kohlenhydrate z. B. Glukose, Saccharose, Laktose, Mannit, Stärke sowie Glycerin in Frage. Als stickstoffhaltige Nährstoffe und gegebenenfalls wachs tumsfördernde Stoffe seien genannt : Aminosäuren, Peptide und Proteine sowie, defen Abbauprodukte, wie Pepton o, dcr Trypton, ferner Fleischextrakte, wasserlösliche Anteile von Getreidekörnern, wie Mais und Weizen, von Destillationsrückständen der Alkoholherstellung, von Hefe, Samen, insbesondere der Raps- und Soyapflanze, der Baumwollpflanze usw., aber auch Ammoniumsalze und Nitrate.
Von anderen anorganischen Salzen kann die Nährlösung beispielsweise Chloride, Carbonate, Sulfate von Alkalien, Erdalkalien, Magnesium, Eisen, Zink und Mangan enthalten.
Die Züchtun, g erfolgt im allgemeinen aerob, also heispielsweise in ruherder Oberflächenkultur oder vorzugsweise submers unter Schütteln oder Rühren mit Luft oder Sauerstoff in Schüttelflaschen oder den b-kannten Fermentern,. Als Tem-peratur eignet sich eine solche zwischen 18 und 40 . Eine wesentliche Antisideromycinwirkung zeigt die Nährlösung dabei im allgemeinen nach 1¸ bis 5 Tagen.
Man trennt vorzugsweise das Mycel vom Kulturfiltrat ab, wonach die Hauptmenge, der Antisideromy- cime im Kulturfiltrat gefunden wird. Es bleiben aber trotzdem namhafte Mengen davon am Mycel adsorbiert. Es ist daher vorteilhaft, letzteres gut auszuwaschen. Dazu eignen sich Wasser und wässrige organ, i- sche Lösungsmittel, wie Alkohole, z. B. wässriges Methanol.
In ah. nlicher Weise kans man auch Bakterien, z. B. B. subtils, züchten und das Kulturfiltr. at als Quelle fiir die Isolierung von Antisideromycin verwenden.
Für die Isolierung der Antisideromycine aus den genannten Materialien, insbesondere den Kulturfiltraten von Pilz- oder Bakterienzüchtungen, kann man nach an sich bekannten Methoden vorgehen und z. B. eine der nachfolgend angegebenen Arbeitsweisen oder Kombinationen davon verwenden.
1) Man kann Adsorptionsmittel verwendet, z. B.
Aktivkohlen, wie aNorita, aktivierte Erden, wie Frankonit , Fullereie oder Floridin , oder Haradsorber, wie Asmit . Die Elution der Adsorbate erfolgt zweckmässig mit Gemischen von mit Wasser mischbaren orgnaischen Lösungsmitteln mit Wassex, z. B. mit Wasser-Meth, anol-, Wasser-Pyridin-, ver dünnte Essigsäure-Methanol-oder Wasser-Methanol Eisessig-Butanol-Gemischen. Als besonders orteil- haft fiir die Elution eines Frankonit - oder Norit adsorbates hat sich das Gemisch von Wasser (4 Volumenteile) und Pyridin (1 Volumteil) erwiesen.
2) Eine zweite Methode zur Abtrennun, g der An tisideromycine besteht IdLarin, dass man, six an Katio nenaustauschern aldisorbiert, wobei besonders Saure- gruppen enthaltende Harze, wie Amberlite IRC-50 (vgl. US-Patent 2 340 111) geeignet sind.. Letzteres kann sowohl in der Säureform als auch in der Natriumform verwendet werden, doch hat sich auch ein Gemisch dieselr beideID Formen bewährt. Die Elution geschieht zweckmässig mit verdünnten Säuren, z. B. mit methanolischer Salzsäure.
3) Weiter könnenf die Antisi, dieromycine einer wässrigen Lösung mittels organischen Lösungsmitteln entzogen werden. Für diese Extraktionsverfahren haben sich höhere organische Alkohole, z. B. Benzylalkohol oder Isopropylalkohol, besonders bewährt.
Zweckmässigerweise wird dabei dier wässrigen Phase ein anorganlsches Salz, z. B. Ammoniumlsulfat oder Natriumchlorid, zugesetzt. Aus den erhaltenen orga nischlen ExtrakteD können die Antisideromycine entweder durch Abdampfen des Lösungsmittels oder durch Ausfällung mitbels eines geeigneten organi- schen Lösungsmittels, z. B. Sither, Petroläther oder Athylacetat, in angereichlerter Form erhalten werden.
4) Eine Anreicherung der Antisilderomycine wird auch dadurch erreicht, dass man konzentrierte, wäss ; riga oder alkoholisch-wässrige Lösungen des Salzes mit einem Oberschuss an organischen, mit Wasser mischbaren Lösungsmitteln, wie Aceton, Dioxan, etc., versetzt, wobei die Salze in fester Form ausgefällt werden.
5) Eine weitere Methode der AnreicherungSder Antisideromycine besteht darin, dass man wässrige Lösungen desselben mit Lösungen von Phenol in Chloroform extrahiert, wobei sowohl das pH der wässrigen Lösuntg als auch der Phenolgehalt der Chloroformlösung variiert werden. So befinden sich die Antisideromycine z.
B. bei cirer Verteilung zwi schen, einer Lösung, die auf 100 cm3 chloroform 100 g Phenol enthält, und einer wässrigen Phase von pH 1 bis 6 fia, st ausschliesslich in derorganischen Phase, während sie bei der Verwendung einer Lösung, die auf 100 cm3 Chloroform nur 33 g Phenol enthält, der wdssrigen Phase lediglich bei einem pH zwischen 4 unl 6 annähernd vollständig entzogen werden.
Versteht man unter dem Verteilungskoeffizienten der Antisideromycine das Verhältnis von, Konzentration in der organischen Phase zur Konzen°ration in dFer wässrigen, so geht aus, den obigen Angaben hervor, dass der Verteilungskoeffizient mit steigendem Phenolgehalt der organischen Phase zunimmt und mit sinken, deml pH Ider wässrigen Phase abnimmt. Da es somit möglich ist, jeden beliebigen Verteilungskoeffi zientenl der Antisideromycine in diesam. System einzustellen, kann man, durch Kombinatiorn von wenigen Verteilungsoperationen einen Grossteil inaktiver Verunreinigurllgen abtrennen.
6) Eine andere Anreicherungsmethode für die Antisideromycine stellt die Chromatographie dar, wie Adsorptionschromatographie an verschiedenen, Materialien, z. B. an Norit , Aluminiumoxyd, Magnesiumsilikaten, Silicagel , Calciumsulfat, so- wie Verteilungschromatographie mit Cellulose, Stärke, Silicagel , Celite und dergl. als Rager- substanzen, oder aber Chromatographie an Ionenaustauscherharzen, z. B. an Dowex 50 , Amberlite IRC-50 undSdergl.
7) Weiter können die Antisideromycine durch Gegenstromverteilung nach Craig zwischen, zwei n ! icht mischbaren Lbsungsmittelphasen angereich-ert werden,. Folgende Lösungsmittelsysteme haben sich dabei besonders bewährt : a) Benzylalkohol - 20 %ige wässrige Lösung von Ammoniumsulfat. b) n-Butanol (100 Volumteile)-Benzylalkohol (200 Volumteile)-1-n. Salzsaure (6 Volumteile)- Wasser (300 Volumteile) - wässrige, bei 19 gesättigte Natriumchloridlösung (60 Volumteile).
8) Schliesslich evgnet sich die praparative Elektrophorese an einler Säule mit Trägermaterial gut zur Reinigung, Anreicherung und Auftennung von AntiF sideromycin-'raparaten.Duesewirvorzugsvvei.se als Hochspannungselektrophorese bei 500-4000 Volt durchgefuhrt. Eine weitere Verbesserung besteht darin, dass man dieses Verfahren nach dem sogenannten Gegenstromprinzip durchführt.
Dabei werden die als Kationen vorliegenden Antisilderomycine auf der Trägersäule örtlich festgehalten, indem man die du. rch das elektrische Feld hervorgerufene Bewegung mittels einer in entgegengesetzter Richtung verlau fenden Strömung, des Elektrolyts genau kompensiert.
Dadurch wird erreicht, dass Stoffe mit anderer elektrischer Beweglichkeit die Trägersäule an den beiden Eiektrodenenden verlassen°.
Das so erhaltene Gemisch der Antisidleromycine kan, nl zur Herstellung Ider einzelnen Komponenten, Antisideromycin A2, A3 und B1 verwendet werden, indem man das Gemisch in die Komponenten zerlegt ; dies kann vor allem durch Papierchromatographie erfolgen.
Die Antisideromtycirlie können als Wuchsstoffe fur verschiedene Organismen verwen, det werden, und zwar entweder aUein oder inr Form von speziellen Präparaten.
In dlea nachfolgenden Beispielen sind bdie Temperaturen in Celsiusgradieni angegeben
Beispiel 1
Die Züchtung, des Strelptomyces pailosus, Stamm A 21748, (Institut für spezielle Botanik, Eidgenöfssische Technische Hochschule, Zürich) wird nach dem Submersverfahren durchgeführt. Man verwendet eine Nahrlosumg, die pro Liter Leitungswaslser 20 g Soyamehl und 20 g Maunit enthält. Die Nährlösung wird in den Impfkolben oder in den Fermentern 20-30 Minuten bei 1 atii sterilisiert. Die sterilisierte Nährlö- sung zeigt ein, pH von 7, 2-7, 6.
Die Beimpfung erfolgt mit bis zu 20% einer teilweise sporulierenden vegetativeni Kulturldes, genannten Organismus. Man inkubiert unter gutem Schütteln oder Rühren bei 24-30 , wobei die Kulturen in Fermentern mit ca. 2 volumen Luft je Volumen Lösung pro Minute belüf- tet werden. Nach 72-144 Stuniden Bebrdtung hat die Kultwrlosun : g den hochsten, Gehalt an Antisideromy- cin erreicht.
Man unterbricht die Kultur, trennt dias Mycel sowie andere feste Bestandteile von der die Hauptmenge der Antisideromycin enthaltenden Lösung mittels Filtration, oder Zentrifugation ab, wobei gegebenenfalls der Kulturlösung vor der Filtration etwa 1 /o, eines Filterhilfsmittels, z. B. Hyflo Supercel , zugesletzt wird. Die Filterrückstände wäscht man mit Wasser oder wässrigem Methanol und vereinigt die Waschflüssigkeiten mit dem Kulturfiltrat. Das gewonnene Kulturfiltrat wird unter stetem Rühren mit 2 % Tonerde, z. B. Frankonit , versetzt.
Nach gründlicher Durchmischung filtriert man ab und widerholt mit dem erhaltenen Filtrat den Ab sorptionsvongang l-2mal. Die Filterrückstände werden vereinigt und mehrmals mit Wasser und wässrigem Methanol gewaschen Anschliessend werden die Filternückstände 2-3mal mit einem Pyridin-Wassergemisch (1 : 4) eluiert. Das Idurch Filtration geklärte Eluat wird am Vakuum eingeengt. Die erhaltenen Konzentrate können entweder direkt weiter verarbeitet werden (siehe Beispiel 3) odeur man kann daraus mittels Gefr, ielrtrocknung ein Gemisch von Antiside- romycinen, in roher Form isolieren ;.
Vrwendet man anstelle der oben angegebenen Nährlösung solche, die pro Liter Leitungswasser die folgenden Nährstoffe enthalten, so erhält man nach analoger Züchtung und Aufarbeitung KulturfiltDate von ähnlich hohen Gehalt an Antisideromycin. a) Saccharose 20 g

Anstelle des angegebemen Stammes dler Art Streptomyces pilosus können die folgenden Stämme verwentdet werdell (unter den angegebenen Stamm- Nummern werden die Stämme im Institut fur spezielle Botanik, Eidgenössische Technische Hochschule, Zürich, aufbewahrt.) Nach analoger Züch tung und Aufanbeitunlg eIhält man Kulturfiltrate von ähnlich hohem Ferrioxamingehalt.

Der Stamm A 23 978 der Art Stremptomyces aureofaciens (Institut für spezielle Botanik, Eidgenössische Technische Hochschule, Zürich) wird submers gezüchtet auf einer Nährlösung, die pro Liter Leitungswasser 20 g Malzextrakt, 20 g Distillers solubles enthält. Die Züchtung und Aufarbeitung erfolgt gleich wie in Beispiel 1, wobei Kulturfiltrate erhalten werden mit ähnlich hohem Antisideromycingehalt.
Beispiel 3
60 Liter Kultur werden in der in Beispiel 1 targe- stellteru Weise hergesbellt und aufgearbeitet. Das ge- wonnene Pyridin-Wasser-Eluat (ca. 6 Liter) wird im Vakuum auf 3 Liter eingeengt. In diesem Konzentrat werden 870 g Amlmonsulfat golöst. Die Lösung wird durch Filtration oder Zentrifugation, ggegebenenfalls unber Zugabe von 1 % Hyflo Supercel , geklärt.
Durch 3-4maliges Ausschutteln mit Benzylalkohol odier Isopropylalkohol werdlen die Antisideromycine in das organische L6sungsmittel iibergefiihrt. Die orgnischen Phasen werd-en vereiniigt und, mit Hilfe von Natriumsulfat getrocknet. Man gibt, einen, Ober- schu, ss von Ather oder Athylacetat zu un^S filtriert die ausgefällten Antisiderlomycine ab. Der. Zusatz von Filterhilfsmittel, z. B. Hyflo Supercel , vor der Ausfällung erleichtert die Gewinnung des Niederschlages.
Aus diesem können die Antisideromycine mit Methanol oder Wasser ausgewaschen werden. Diese Eluate werden eingedampft oder lyophilisiert und man erhält so ein, Prdparat von Antisideromycin in angereicher- ter Forum.
Beispiel 4
Einem gemäss Beispiel 1 oder 2 erhaltenen Kul turfiltrat werden pro Liter 20 g Kochsalz zugefügt.
Die klare Lösung wird 3mal mit 1/10 Volumen Chloroform-Phenol-Gemisch (1 Volumteil Chlorofarm, 1 Gewichtsteil Phenol) ausgezogen. Die organischen Phasen werden vereinigt, unter Zusatz von Hyflo Supercel filtriert und mit eanem tJberschuss an Ather versetzt. Durch mehrmaliges Ausschütteln mit wenig Wasser werden die Antisideromycine in d-ie wässrigen Anteile übergeführt. Die erhaltenen wäss- rigen Extrakte werden vereinligt und zur vollständi- gen Entfernung des Phenols 2mal mit Äther ausgeschüttelt.
Durch Gefriertrocknung, gewinnt m ! an ein orange bis braunrotes Präparat von Antisideromycinen, das durch Papierchromatographie zerlegt werden kann.
Beispiel 5
Ein gemäss Beispiel 3 erhaltenles Rohprodukt von Antisideromycin wird einer Gegenstromverteilung user 83 Stufen unterworfen, wobei das Lösungsmit- telsystem Benzylalkohol-20 /0ige wässrige Ammoni- umsulfatlösung verwendet wird. Der Inhalt der einW zelnen Vertealumgsgefasse wird hierauf kolorimetrisch (Absorption bei 425 mou,) und biologisch (Bonifas Test) untersucht. Dabei können zwei Maxima festgestellt werden, und zwar bei Stufe 32 und. bei Stufe 76.
Das erstere enthält Antisideromycin A1 und das zweite die Antisideromycine A2, A3 und-, B. Die Stu- fen 32-42 werden verein, igt und bis zur Sättigung mit Ammoniumsulfat versetzt. Dam wird de Benzylal- koholphase, die nun die genannte Antisideromycin Aktivität enthält, abgetrennt und mit dem gleichen Volumen Ather versetzt. Diesem Gemisch wird die Antisideromycin-Aktivität mit werig Wasser entzogen. Durch Gefriertrocknung der konzentrierX wässrigen Lösung erhält man Antisideromycin A1 in Form eines braunroten Pulvers, das in bezu, g auf das Ausgangsmaterial der Gegenstromverteilung ca. 10mal angereichert ist.
Zur Isolierung der Antisideromycine A2, A3 und B vereinigt man die StuMen 65-82 usnd behandolt sie in gleicher Weise. Das Gemisch von Antisideromycin A2, A3 unld B stellt ebenfalls ein braunrotes Pulver dar. Es kann durch Papierchromatographie zerlegt werden.
Beispiel 6
100 g frische Bäckerhefe wird in einem Liter Methanol aufgeschgwemmt undl 30 Minuten kräftig gerührt. Die Aufschwemmung filtriert man und engt die methanolische Lösun'g am Vakuum auf 200 ml eiIr. Zu diesem Konzentrat werden 200 ml Wasser zugesetzt und die Lösung durch Filtration oder Zen- trifugation geklärt. Die geklärte Lösung wird 3mal mit einem Chloroform-Phenol-Gemisch (1 Volumteil Chloroform, 1 Gewichtsteil Phlenol) ausgezogen. Die vereinigten organischen Phasen versetzt man mit wenig Filterhilfsmittel ( Hyflo Supercel ) und gibt zu dieser Aufschwemmung unter stetem Rühren einen Überschuss an Äther zu.
Die ausgefallenen Antisideromycine werden zusammen mit dem Filterhilfsmittel abgetrenst, mit Äther gewaschen und getrocknet. Aus dem Filterhilfsmittel werden die Antisideromycine mit Methano1 eluiert. Die methanolische Lösung wird am Vakuum schonend eingedampft, wobei die Anti sideromycine m Form eines aktiven rotbraunen Har zes erhalten werden, aus dern sich mittels Papierchromatographie die einzelnen Faktoren abtrennen lassen.
Zu Präparaten mit ähnlich hohem Gehalt an Antisideromycin gelangt man, wenn man anstelle der Bäckerhefe einen käuflichen Hefeextrakt, abgepresste Brauereihefe oder frische Hefekulturen, wie beschrieben aufarbeitet.
Beispiel 7
Die Züchtung von Bacillus subtilis wird nach dem Submersverfahren durchgeführt. Man verwendet eme Nährlösung, Idie pro Liter Leitungswasser 10 g Roh- glukose, 5 g Pepton, 3 g Fleischextrakt, 5 g Kochsalz und 10 g Kalk enthält. Man inkubiert die Kulturen bei 33-37 unter Belüftung und unter stetem Schütteln oder Riihren. Nach 48-96 Stunden Bebrütung werden díe Kulturen unfiltriert mit 2 % Tonerde ( Frankonit ) versetzt, kräftig gerührt und anschliessend durch Filtration, oder Zentifugation geklärt.
Das Sedomlent wind mit Methlanol gewaschen und an- schliesseud mit einem Pyrid, in-Wassergemisch (1 : 4) eluiert. Das Eluat enthält die Hauptmenge der Antisideromycine, die gemäss den Angaben in Beispiel 3 isoliert wer, den.

They are red in color and are easily soluble in acids and strongly polar solvents, such as what; ser, dimethylformamide, glycol, ethylene glycol monomethyl ether, and in lower aliphatic alcohols such as methanol. They also have limited solubility in higher aliphatic alcohols and in aromatic alcohols and phenols, e.g. B. in butanol, Ben ylalkoh, ol, phenol.
A characteristic representative of this group is the antisideromycin Al (also called fenioxamine B), which is described in Patent No. 413,853. It is used as the main product e.g. B. produced by Streptomycetes. In addition, there are three other substances that are known as antisideromycin A2 and A3 and B.

In contrast, the above-described, typical for the antiside romycine, the growth-promoting effect on Baoillus subtils, Micrococcus pyogenes, Saccharomyces cerevisiae, Ustilago sphaerogena and Chlamy domonias eugametos is not known for the three substances ferrichrome, terregens factor and coprogen. As has been shown in Table 1, the ferric chromium differs from the antisideromycins by its own paper chromatographic behavior. Jours.

The preparation of the mixture of the anisideromycins A2, A3 and B can be carried out by methods known per se, taking into account the physical data given above and in particular with the aid of the biological eschez test suitable for tracking the isolation. The method according to the invention is characterized in that an antisideromycin-producing strain of microorganisms is cultivated until the culture medium has a substantial antisideromycin effect and the mixture of antisideromycins A2, A3 and B is isolated from the culture filtrate.
Cultures of micro-organisms, especially of representatives of the genus Streptomyces, of bacteria such as bacteria, are used as starting materials for obtaining the anti side, romycinid. B. from B. subtilis, or from yeasts, e.g. B. from Saccharomyces cerevisiae, used dot.

They represent neutral or acidic salts. Other organisms, e.g. B. Representatives of the Arthrobacter genus, such as Arthrobacter terregens and Arth robacter flavescens, only develop at all in the presence of antisideromycin. In these cases the antisideromycins have a similar vitamin character, as has been shown for the active substances ferrichrome, terregens factor and coprogen. [Bact. Rev. 21, 101 (1957)].
Another biological property of the antisideromycins is that they are able to competitively cancel the effect of antibiotics belonging to the group of sideromycins in return for gram-positive pathogens. The sideromycins include u. a. the iron-containing antibiotics grisein, albomycin, ferrimycin, the antibiotic 1787 H. Thrum, Naturwiss. 44, 561 (1957) and the substances L.A. 5352 and L.A. 5937 [P. Sensi and M. T. Timbal, Antibiotics & Chemotherapy 9, 160 (1959)]. Apparently there is an antagonism between the antisideromycins on the one hand and the.
Sideromycins on the other hand, which can be detected both in vitro and in vivo. The name antisideromycine is also derived from this effect.
The described antagonistic effect is limited exclusively to the sideromycin antibiotics and could not be observed with other antibiotics such as neomycin, viomycin, streptomycin, streptothricin, penicillin, brythromycin or novobiocin.
The abolition of the antibiotic effect of the sideromycins in vitro is well suited for a qualitative recognition and quantitative determination of the antisideromycins by using microorganisms in and of themselves for the determination of synergistic substances. The above-mentioned Streptomyces strains "which can be easily grown on a larger scale" have proven particularly useful in this regard.
The present invention is therefore not restricted to the use of the representatives of the species mentioned, but also relates to the use of antisiderornycin-producing strains of other species and in particular of variants of all diets organisms, such as those found, for. B. by selection or mutation, in particular from ultraviolet or X-rays or from nitrogen mustard oils.
To win antisideromycins on a larger scale, z. B. a strain having the properties of the streptomycetes listed above, e.g. B. cultured aerobically in aqueous, carbohydrates, nitrogen-containing compounds as well as inorganic salts containing nutrient solution until it shows a substantial antisideromycin effect, and the antisideromycins are then isolated. Nitrogen-containing nutrients and possibly growth-promoting substances may be mentioned: amino acids, peptides and proteins as well as defen degradation products such as peptone or tryptone, also meat extracts, water-soluble parts of grains such as corn and wheat, of distillation residues from alcohol production, of yeast, seeds , especially the rapeseed and soya plants, the cotton plant, etc., but also ammonium salts and nitrates.
Of other inorganic salts, the nutrient solution can contain, for example, chlorides, carbonates, sulfates of alkalis, alkaline earths, magnesium, iron, zinc and manganese.
The cultivation is generally carried out aerobically, for example in resting surface culture or preferably submerged with shaking or stirring with air or oxygen in shake flasks or the fermenters known from b. A suitable temperature is between 18 and 40. The nutrient solution generally shows a substantial antisideromycin effect after 1¸ to 5 days.
The mycelium is preferably separated from the culture filtrate, after which the main amount, the antisideromycime, is found in the culture filtrate. Nevertheless, significant amounts of it remain adsorbed on the mycelium. It is therefore advantageous to wash out the latter well. Water and aqueous organic solvents, such as alcohols, e.g. B. aqueous methanol.

For the isolation of the antisideromycins from the materials mentioned, in particular the culture filtrates from cultures of fungi or bacteria, one can proceed according to methods known per se and, for. B. use one of the following working methods or combinations thereof.
1) Adsorbents can be used, e.g. B.
Activated carbons such as aNorita, activated earths such as Frankonit, Fullereie or Floridin, or Haradsorbers such as Asmit. The elution of the adsorbates is expediently carried out with mixtures of water-miscible organic solvents with Wassex, e.g. B. with water-meth, anol, water-pyridine, ver dilute acetic acid-methanol or water-methanol glacial acetic acid-butanol mixtures. The mixture of water (4 parts by volume) and pyridine (1 part by volume) has proven to be particularly advantageous for the elution of a Frankonite or Norit adsorbate.
2) A second method of separating the antisideromycins consists in that one aldisorbates on cation exchangers, resins containing acid groups such as Amberlite IRC-50 (cf. US Pat. No. 2,340,111) being particularly suitable. The latter can be used both in the acid form and in the sodium form, but a mixture of the two forms has also proven useful. The elution is conveniently done with dilute acids, e.g. B. with methanolic hydrochloric acid.

The aqueous phase is expediently an inorganic salt, e.g. B. ammonium sulfate or sodium chloride added. The antisideromycins can be extracted from the organic extracts obtained either by evaporation of the solvent or by precipitation with a suitable organic solvent, e.g. B. Sither, petroleum ether or ethyl acetate, can be obtained in enriched form.
4) Enrichment of the antisilderomycins is also achieved by using concentrated, aqueous; riga or alcoholic-aqueous solutions of the salt with an excess of organic, water-miscible solvents such as acetone, dioxane, etc., added, the salts being precipitated in solid form.
5) Another method of enriching the antisideromycins consists in extracting aqueous solutions of the same with solutions of phenol in chloroform, varying both the pH of the aqueous solution and the phenol content of the chloroform solution. So are the antisideromycins z.

The cultivation of Strelptomyces pailosus, strain A 21748, (Institute for Special Botany, Swiss Federal Institute of Technology, Zurich) is carried out using the submerged method. A nutrient solution is used which contains 20 g of soya flour and 20 g of Maunit per liter of tap water. The nutrient solution is sterilized in the inoculation flask or in the fermenters for 20-30 minutes at 1 atii. The sterilized nutrient solution shows a pH of 7.2-7.6.
The inoculation takes place with up to 20% of a partially sporulating vegetative culture of the said organism. Incubation is carried out with vigorous shaking or stirring at 24-30, the cultures being aerated in fermenters with approx. 2 volumes of air per volume of solution per minute. After 72-144 hours of incubation, the cult phrase has reached the highest level of antisideromycin.
The culture is interrupted, the mycelium and other solid constituents are separated from the solution containing the majority of the antisideromycin by means of filtration or centrifugation, optionally adding about 1 / o of a filter aid, e.g. B. Hyflo Supercel is added. The filter residues are washed with water or aqueous methanol and the washing liquids are combined with the culture filtrate. Example 2
The strain A 23 978 of the species Stremptomyces aureofaciens (Institute for Special Botany, Eidgenössische Technische Hochschule, Zurich) is grown submerged on a nutrient solution containing 20 g of malt extract and 20 g of distillers solubles per liter of tap water. The cultivation and work-up are the same as in Example 1, culture filtrates being obtained with a similarly high antisideromycin content.
Example 3
60 liters of culture are produced and worked up in the manner described in Example 1. The pyridine-water eluate obtained (approx. 6 liters) is concentrated to 3 liters in vacuo. 870 g of ammonium sulfate are dissolved in this concentrate. The solution is clarified by filtration or centrifugation, if necessary without the addition of 1% Hyflo Supercel.
By shaking 3-4 times with benzyl alcohol or isopropyl alcohol, the antisideromycins are converted into the organic solvent. The organic phases are combined and dried with the aid of sodium sulfate. An excess of ether or ethyl acetate is added and the precipitated antisiderlomycins are filtered off. Of the. Addition of filter aids, e.g. B. Hyflo Supercel, before precipitation facilitates the extraction of the precipitate.
From this the antisideromycins can be washed out with methanol or water. These eluates are evaporated or lyophilized and a preparation of antisideromycin in an enriched forum is obtained.
Example 4
A culture filtrate obtained according to Example 1 or 2 is added per liter of 20 g of common salt.
The clear solution is extracted 3 times with 1/10 volume of a chloroform-phenol mixture (1 part by volume of Chlorofarm, 1 part by weight of phenol). The organic phases are combined, filtered with the addition of Hyflo Supercel and treated with an excess of ether. By shaking out several times with a little water, the antisideromycins are converted into the aqueous components. The aqueous extracts obtained are combined and extracted twice with ether to completely remove the phenol.
By freeze drying, m wins! an orange to brown-red preparation of antisideromycins that can be broken down by paper chromatography.
Example 5
A crude product of antisideromycin obtained in accordance with Example 3 is subjected to countercurrent distribution in 83 stages, the solvent system being benzyl alcohol-20/0 aqueous ammonium sulfate solution. The contents of the individual vertical vessels are then examined colorimetrically (absorption at 425 mou) and biologically (Bonifa's test). Two maxima can be determined, namely at level 32 and. at level 76.
The former contains antisideromycin A1 and the second the antisideromycins A2, A3 and -, B. The stages 32-42 are combined and ammonium sulphate is added to saturation. The benzyl alcohol phase, which now contains the aforementioned antisideromycin activity, is then separated off and the same volume of ether is added. The antisideromycin activity is removed from this mixture with a little water. By freeze-drying the concentrated aqueous solution, antisideromycin A1 is obtained in the form of a brown-red powder which is enriched approximately 10 times in relation to the starting material of the countercurrent distribution.
To isolate the antisideromycins A2, A3 and B, the StuMen 65-82 are combined and treated in the same way. The mixture of antisideromycin A2, A3 and B is also a brown-red powder. It can be broken down by paper chromatography.
Example 6
100 g of fresh baker's yeast are suspended in one liter of methanol and vigorously stirred for 30 minutes. The suspension is filtered and the methanolic solution is concentrated in vacuo to 200 ml of egg. 200 ml of water are added to this concentrate and the solution is clarified by filtration or centrifugation. The clarified solution is extracted 3 times with a chloroform-phenol mixture (1 part by volume of chloroform, 1 part by weight of phlenol). A little filter aid (Hyflo Supercel) is added to the combined organic phases and an excess of ether is added to this suspension with constant stirring.
The precipitated antisideromycins are separated off together with the filter aid, washed with ether and dried. The antisideromycins are eluted from the filter aid with Methano1. The methanolic solution is gently evaporated in vacuo, the anti-sideromycins being obtained in the form of an active red-brown resin, from which the individual factors can be separated using paper chromatography.
Preparations with a similarly high content of antisideromycin are obtained if, instead of baker's yeast, a commercially available yeast extract, pressed brewer's yeast or fresh yeast cultures are processed as described.
Example 7
The cultivation of Bacillus subtilis is carried out according to the submerged method. A nutrient solution is used which contains 10 g of raw glucose, 5 g of peptone, 3 g of meat extract, 5 g of table salt and 10 g of lime per liter of tap water. The cultures are incubated at 33-37 with aeration and constant shaking or stirring. After 48-96 hours of incubation, the unfiltered cultures are mixed with 2% clay (Frankonit), stirred vigorously and then clarified by filtration or centrifugation.
The Sedomlent is washed with methanol and then eluted with a pyrid in water mixture (1: 4). The eluate contains the majority of the antisideromycins which are isolated according to the information in Example 3, the.
PATENT CLAIMI
Process for the production of new growth substances, the antisideromycins, characterized in that an antisideromycin-producing microorganism strain is cultivated until the culture medium has a substantial antisideromycin effect, and the mixture of the antisideromycin A2, A3 and B is isolated from the culture filtrate.
