Verfahren zur Herstellung von neuen heterocyclischen Verbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von neuen 4-Aza-thioxanthen-Deri- vaten der Formel I,

Das Verfahren gemäss Erfindung ist dadurch gekennzeichnet, dass man ein 4-Aza-thioxanthon der Formel II

Das Verfahren wird beispielsweise wie folgt ausgeführt : Mit Tetrahydrofuran oder Diäthyläther über schichtete Magnesiumspäne werden mit der Lösung eines basisch substituierten Halogenalkyl-Derivates in Tetrahydrofuran oder Diäthyläther versetzt.
Erfindungsgemäss können als basisch substituierte Halogenalkyl-Derivate verwendet werden : Dialkyl aminopropyl-halogenide, z. B. Dimethylaminopropyl chlorid, Diäthylaminopropylchlorid oder Diäthyl- aminopropylbromid; 1-Alkyl-4-halogen-piperidine, z. B. l-Methyl-4-chlor-piperidin, 1-Athyl-4-chlor- piperidin, 1-Isopropyl-4-chlor-piperidin oder 1-Me- thyl-4-brom-piperidin.
Die Bildung der Grignardveibrndunfg wird vorzugsweise durch Zusatz einer kleinen Menge Athylenbromid, Athylbromid oder Methyljodid und einer Spur Jod gefördert. Das Reaktionsgemisch wird gegebenenfalls zur Vervollständigung der, Raaktion am Riickfluss erhitzt.
Anstelle vonMagnesiumspänen kann auchKupfer- Magnesium-Legiorung (nach Gilman) verwendet werden.
Diese Grignardlösung wind darauf bei Q-30 mit einem 4-Aza-thioxanthon der Formel II, beispielsweise 7-Chlor-4-aza-thioxanthon, 7-Methyl-4-aza thioxanthonoder3-Methyl-7-chlor-4-aza-thioxanthon, versetzt, und das Gemisch zweckmässig noch einige Zeit bei Raumtemperatur gerührt.
Anschliessend hydrolysiert man das Reaktions- gemisch in derKälte mit wässrigerAmmoniumchlorid- lösung und extrahiert mit einem mit Wasser nicht mischbaren organischen Lösungsmittel, vorzugsweise mit Methylenchlorid, Diäthyläther oder Benzol. Das als Zwischenprodukt erhaltene 4-Aza-thioxanthydrol- Derivat wird durch Kristallisation gereinigt und ge wünschten, falls mit anorganischen oder organischen Säuren in geeignete Salze übergeführt oder direkt weiter verarbeitet.
Die Wasserabspaltung erfolgt beispielsweise durch Erhitzen mit hochkonzentrierten Säuren, z. B. mit 80-proz. Schwefelsäure oder mit einem Gemisch aus Eisessig und konz. Salzsäure. Sie gelingt indessen aber auch mit andern Agenzien. z. B. mit Phosphoroxychlorid, Thionylchlorid oder mit Essigsäureanhydrid.
Das Endprodukt wird nach bekannten Methoden iso liert und gereinigt, gewünschtenfalls in Säureadditions- salze übergeführt, und diese gegebenenfalls in ihre stereoisomeren Formen getrennt.
Die verfahrensgemäss hergestellten neuen 4-Aza thioxanthen Derivate sind bei Raumtemperabur gröss- tenteils kristallin. Es sind basische Verbindungen, die mit anorganischen oder organischen Säuren bestän dige, bei Raumtemperatur kristallisierte Salze bilden, die gewiinschtenfalls in ihre stereoisomeren Formen getrennt werden können. Solche Salze sind z. B. die mit anorganischen Säuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure oder Schwefelsäure, oder mit organischen Säuren, wie Essigsäure, Apfelsäure, Fumarsäure, Maleinsäure, Weinsäure, Naphthalin-1, 5disulfonsäure.
Die erfindungsgemäss hergestellten neuen 4-Aza thioxanthen ; Derivate sind auf Grund ihrer ausgezeichneten pharmakodynamischen Eigenschaften zur Verwendung als Heilmittel geeignet. So entfalten die neuen Verbindungen eine bemerkenswerte histaminund serotoninhemmende Wirkung. Ausserdem besitzen diese Verbindungen auch eine starke neurolepti- sche Wirkung, die sich z. B. durch Adrenalinantago nismus und durch Narkosepotenzierung äussert. Die neuen Verbindungen können dementsprechend als Antihistamin, ika, Serotoninantagonisten, aber auch als Neuroleptika oder Antidepressiva therapeutisch verwendet werden.
Ausserdem dienen sie aber auch als Zwischenprodukte zur Herstellung von Medikamenten.
Die als Ausgangssubstanzen verwendeten 4-Azathioxanthone der Formel II sind neue Verbindungen.
Ihre Herstellung erfolgt, falls Ri Wasserstoff bedeutet, in der Weise, dass man 2-Chlor-nicotinsäure mit einem in p-Stellung durch Halogen oder eine niedere Alkylgruppe substituierten Thiophenol zum 3-Carb oxy-pyridyl-(2)-phenylsulfid-Derivat kondensiert und dieses anschliessend durch Erhitzen mit Polyphosphorsäure zum entsprechenden 4-Aza-thioxanthon- Derivat cyclisiert.
Ausgangsprodukte der Formel II, worin Ri eine niedere Alkylgruppe bedeutet, können zweckmässigerweise erhalten werden, indem man ein in 6-Stellung durch niedere Alkylgruppen substituiertes 2-Halogen-3-cyanpyridin, beispielsweise 2-Chlor-3-cyan6-methyl-pyridin, mit einem in p-Stellung durch Halogen oder durch niedere Alkylgruppen substituierten AIizalithiophenolat blei erhöhter Temperatur kondien- siert, das erhaltene 3-Cyan-pyridyl- (2)-phenylsulfid- Derivat zum 3-Carboxy-Derivat hydrolysiert und anschliessend, wie bereits oben beschrieben, mit Polyphosphorsäure cyclisiert.
In den nachfolgendem Beispielen, die die Ausfüh- rung des Verfahrens erläutern, die Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind nicht korrigiert.
Beispiel 1 a) 7-Chlor-9- [I'-methyl-piperidyl- (4')]-4-aza- thioxanthydrol
2, 43 g mit Jod aktivierte Magnesiumspäne werden mit 10 ccm Tetrahydrofuran überschichtet und mit 0, 3 ccm Athylenbromid versetzt, wodurch die Reaktion in Gang gebracht wird. Sodann lässt man die Lösung von 14, 7 g (0, 11 Mol) l-Methyl-4-chlor- piperidin in 25 ccm abs. Tetrahydrofuran in solcher Geschwindigkeit zutropfen, dass die Reaktions- mischung ständig siedet. Anschliessend erhitzt man noch 1-2 Std., bis das Magnesium zum grössten Teil gelöst ist, kühlt ab und gibt bei 20-25 portionen- weise 12, 4 g (0, 05 Mol) 7-Chlor-4-aza-thioxanthon zu. Nach 20 Min.
Rühren bei Raumtemperatur giesst man das Reaktionsgemisch auf 300 ccm 10-proz.
Ammoniumchloridlösung und schüttelt mit Methylenchlorid aus. Nach Trocknen über Magnesiumsulfat und Verdampfen des Lösungsmittels wird der Rück- stand aus Aceton umkristallisiert. Smp. 225-226 . b) 7-Clzlor-9- [I'-methyl-piperidyliden- (4')]-4- aza-thioxanthen
10 g 7-Chlor-9- [1'-methyl-piperidyl- (4')]-4-aza- thioxanthydrol werden mit einer Mischung von 25 ccm Wasser und 75 ccm konz. Schwefelsäure 20 Min. auf 140 erhitzt. Dann giesst man auf 500 ccm Eiswasser, stellt die Lösung mit 50-proz. Natronlauge alkalisch und schüttelt mit Methylenchlorid aus. Nach Trocknen über Kaliumcarbonat und Abdampfen des Lö sungsmittels wird der Rückstand aus Aceton umkristallisiert. Smp. 150-152 .
Das als Ausgangssubstanz verwendete 7-Chlor-4 aza-thioxanthon wird wie folgt hergestellt :
78 g (0, 5 Mol) 2-Chlor-nicotinsäure und 145 g (1 Mol) p-Chlorthiophenol werden 3 Std. auf 170 er- wärmt. Dann kühlt man ab und verrührt das kristal line Reaktionsprodukt mit 500 ccm Äther und 1000 ccm gesättigter Natriumhydrogencarbonatlösung, bis alles gelöst ist. Die ¯therschicht wird dann abgetrennt und ; nochmals mit Natriumhydrogencarbonatlösung ausgeschüttelt. Die vereinigten wässerigen Extrakte werden mit Essigsäure auf ein pH von 5 gestellt. Die ausgefallene Substanz, das 3-Carboxy-pyridyl-(2)-p- chlorphenylsulfid, wird abfiltriert, getrocknet und aus Methanol umkristallisiert.
Smp. 216-217 .
35 g 3-Carboxy-pyridyl-(2)-p-chlorphenylsulfid werden mit 350 g Polyphosphorsäure 1 Std. auf 150 , dann 2 Std. auf 180 erhitzt. Nach Abkühlen auf ca.
100¯ lÏsst man das Reaktionsgemisch unter gutem Rühren in 1500 ccm Wasser einfliessen. Die ausgefallene Substanz wird abfiltriert, dann mit 200 ccm 10proz. Natronlauge 10 Min^. gerührt, wieder abfiltriert, gut mit Wasser gewaschen, getrocknet und aus Eisessig umkristallisiert. Das 7-Chlor-4-aza-thioxanthon schmilzt bei 194-195 .
Beispiel 2 a) 7-Brom-9- [I'-methyl-piperidyl- (4')]-4-a. za- thioxanthydrol
Diese Verbindung wird in analoger Weise zu Beispiel 1, a) durch Grignardienmg von l-Methyl-4- chlor-piperidin, Umsetzung mit 7-Brom-4-aza-thiox- anthon und anschliessende Hydrolyse hergestellt. Smp.
227-237 (Zers.) aus ¯thanol. b) 7-Brom-9- [I'-methyl-piperidyliden- (4')]-4-aza- thioxanthen
Die Herstellung erfolgt durch Wasserabspaltung aus dem Thioxanthydrol analog Beispiel l, b). Smp.
166-168 aus Aceton.
Die verwendete Ausgangssubstanz, das 7-Brom 4-aza-thioxanthon vom Smp. 190-193 aus Eisessig, wird analog der Ausgangssubstanz von Beispiel 1 hergestellt, indem man 2-Chlor-nicotinsäure mit p Bromthiophenol kondensiert und das erhaltene 3 Carboxy-pyridyl-(2)-p-bromphenylsulfid (Smp. 220221¯ aus Methanol) mit Polyphosphorsäure cycli- siert.
Beispiel 3 a) 7-Methyl-9- [I'-methyl-piperidyl- (4')]-4-aza- thioxanthydrol
Diese Verbindung wird in analoger Weise zu Bei spiel 1, a) durch Grignardierung von 1-Methyl-4 chlor-piperidin, Umsetzung mit 7-Methyl-4-aza-thioxanthon und anschliessende Hydrolyse hergestellt.
Smp. 185-186 aus ¯thanol. b) 7-Methyl-9- [I'-methyl-piperidyliden- (4')]-4 aza-thioxanthen
Die Herstellung erfolgt durch Wasserabspaltung aus dem ThioxanthyOol analog Beispiel 1, b3. Smp.
130-132 aus Hexan.
Die verwendete Ausgangssubstanz, das 7-Methyl 4-aza-thioxanthon vom Smp. 153-154¯ aus Aceton, wird in analoger Weise wie das in Beispiel 1 beschrie- bene 7-Chlor-4-aza-thioxanthon hergestellt, indem man 2-Chlor-nicotinsäure mit p-Methyl-thiophenol kondensiert und das erhaltene 3-Carboxy-pyridyl- (2)- p-methyl-phenylsulfid (Smp. 217-218 aus Sithanol) mit Polyphosphorsäure cyclisiert.
Beispiel 4 a) 3-Methyl-7-chlor-9- [I'-methyl-piperidyl- (4')]-
4-aza-thioxanthydrol
Diese Verbindung wird in analoger Weise zu Beispiel 1, a) durch Grigaardierung von l-Methyl-4-chlorpiperidin, Umsetzung mit 3-Methyl-7-chlor-4-azathioxanthon und anschliessende Hydrolyse hergestellt.
Smp. 208-210 aus Aceton. b) 3-Methyl-7-chlor-9- [l'-methyl-piperidyliden- (4')]-4-aza-thioxanthen
Die Herstellung erfolgt durch Wasserabspaltung aus 3-Methyl-7-chlor-9- [l'-methyl-piperidyl- (4')]-4- aza-thioxanthydrol analog Beispiel 1, b). Smp. 155- 156 aus Aceton.
Die verwendete Ausgangssubstanz, das 3-Methyl 7-chlor-4-aza-thioxanthen, wird wie folgt hergestellt :
19 g 2-Chlor-3-cyan-6-methyl-pyridin, 23 g p Chlor-thiophenol-natrium und 160 ccm abs. Dioxan werden 20 Std. unter Rückfluss erhitzt. Das ausgefallene Salz wird sodann abfiltriert und die Dioxanlösung bei einem Druck von 15 mm Hg eingedampft. Die Lösung des Rückstands in Methylenchlorid wird darauf mit Wasser ausgeschüttelt, über Magnesiumsulfat getrocknet und eingedampft. Anschliessend wird der Rückstand, das 3-Cyan-6-methyl-pyridyl- (2)-p-chlor- phenylsulfid, aus Aceton umkristallisiert. Smp. 135 136 .
28 g des erhaltenen Sulfids werden mit einer Mi schung von 125 ccm Wasser und 125 ccm konz.
Schwefelsäure 16 Std. auf 140 erhitzt. Dann giesst man die Reaktionslösung in 1500 ccm Wasser ein, filtriert die ausgefallene Substanz ab und löst sie darauf unter Erwärmen in 5-proz. Natronlauge. Die Lösung wird darauf filtriert und mit Essigsäure auf ein pH von 5 gestellt. Die ausgefallene Substanz wird wieder abfiltriert und getrocknet. Das 3-Carboxy-6-me thyl-pyridyl- (2)-p-chlor-phenylsulfid schmilzt bei 212 bis 213 .
Die anschliessende Cyclisierung zum 3-Methyl-7 chlor-4-aza-thioxanthon erfolgt mit Polyphosphor- säure wie in Beispiel 1 beschrieben. Smp. 235-236 aus Eisessig.
Beispiel 5 a) 7-Chlor-9- (3'-dimethylamino-propyl)-4-aza- thioxanthydrol
Diese Verbindung wird in analoger Weise zu Bei spiel 1, a) durch Grignardierung von 3-Dimethyl- amnno-propylchlorid, Umsetzung mit 7-Chlor-4-azathioxanthon und anschliessende Hydrolyse hergestellt.
Smp. 175-177¯ aus EssigsÏureÏthylester. b) 7-Chlor-9- (3'-dimethylamino-propyliden)- 4-aza-thioxanthen
12 g 7-Chlor-9- (3'-dimethylamino-propyl)-4-aza- thioxanthydrol werden mit 120 ccm Eisessig und 48 ccm konz. Salzsäure 1 Std. unter R ckfluss erhitzt.
Anschliessend dampft man das Reaktionsgemisch unter einem Druck von 15 mm Hg ein, schüttelt den Rückstand mit verd. Natronlauge und Methylenchlo- rid durch, trocknet die organische Phase über Kaliumcarbonat und dampft sie ein. Die zurückbleibende ölige Base (9, 1 g) wird in 20 ccm Athanol gelöst und mit der zur Herstellung des neutralen Fumarats berechneten Menge Fumarsäure versetzt. Die durch gelindes Erwärmen erhaltene Lösung wird abgekühlt und mit Äther versetzt, wobei das Gemisch der cistrans-isomeren Fumarate kristallisiert. Das in grös- serer Menge vorhandene neutrale Fumarat des Isomeren A wird nach zweimaligem Umkristallisieren aus Methanol rein erhalten,. Das neutrale Salz schmilzt bei 179-181¯ (Zers.).
Beispiel 6 a) 7-Methyl-9- (3'-dimethylamino-propyl)-4-aza- thioxanthydrol
Diese Verbindung wird in analoger Weise zu Beispiel l, a) durch Grignardierung von 3-Dimethylamino propylchlorid, Umsetzung mit 7-Methyl-4-aza-thio- xanthon und anschliessende Hydrolyse hergestellt.
Smp. 172-174 aus Aceton. b) 7-Methyl-9- (3'-dimethylamino-propylen)-4-aza- thioxanthen
Die Wasserabspaltung aus dem Thioxanhydrol erfolgt analog wie in Beispiel 5, a) beschrieben. Die erhaltene ölige Base wird in das saure Fumarat übergeführt, wobei aus Athanol/Ather das Gemisch der cis-trans-isomeren Fumarate kristallisiert. Das saure Fumarat des Isomeren A wird nach dreimaligem Umkristallisieren aus Methanol rein erhalten. Das saure Salz schmilzt bei 195-196 (Zers.).

The process is carried out, for example, as follows: Magnesium turnings coated with tetrahydrofuran or diethyl ether are mixed with a solution of a basic substituted haloalkyl derivative in tetrahydrofuran or diethyl ether.
According to the invention, the basic substituted haloalkyl derivatives can be used: Dialkyl aminopropyl halides, eg. B. dimethylaminopropyl chloride, diethylaminopropyl chloride or diethyl aminopropyl bromide; 1-alkyl-4-halopiperidines, e.g. B. 1-methyl-4-chloro-piperidine, 1-ethyl-4-chloro-piperidine, 1-isopropyl-4-chloro-piperidine or 1-methyl-4-bromo-piperidine.
The formation of the Grignard vein is preferably promoted by adding a small amount of ethylene bromide, ethyl bromide or methyl iodide and a trace of iodine. If necessary, the reaction mixture is refluxed to complete the action.

The reaction mixture is then hydrolyzed in the cold with aqueous ammonium chloride solution and extracted with a water-immiscible organic solvent, preferably with methylene chloride, diethyl ether or benzene. The 4-aza-thioxanthydrol derivative obtained as an intermediate is purified by crystallization and, if desired, converted into suitable salts with inorganic or organic acids or further processed directly.
The elimination of water occurs, for example, by heating with highly concentrated acids, e.g. B. with 80 percent. Sulfuric acid or with a mixture of glacial acetic acid and conc. Hydrochloric acid. However, it can also be achieved with other agents. z. B. with phosphorus oxychloride, thionyl chloride or with acetic anhydride.
The end product is isolated and purified by known methods, if desired converted into acid addition salts, and these are optionally separated into their stereoisomeric forms.
The new 4-aza thioxanthene derivatives produced according to the process are for the most part crystalline at room temperature. They are basic compounds which, with inorganic or organic acids, form stable salts which crystallize at room temperature and which, if desired, can be separated into their stereoisomeric forms. Such salts are e.g. B. those with inorganic acids such as hydrochloric acid, hydrobromic acid or sulfuric acid, or with organic acids such as acetic acid, malic acid, fumaric acid, maleic acid, tartaric acid, naphthalene-1,5-disulfonic acid.
The new 4-aza thioxanthene prepared according to the invention; Derivatives are suitable for use as medicaments because of their excellent pharmacodynamic properties. The new compounds develop a remarkable histamine and serotonin-inhibiting effect. In addition, these compounds also have a strong neuroleptic effect. B. expressed by adrenaline antagonism and by narcosis potentiation. The new compounds can accordingly be used therapeutically as antihistamines, ika, serotonin antagonists, but also as neuroleptics or antidepressants.
They also serve as intermediate products in the manufacture of drugs.
The 4-azathioxanthones of the formula II used as starting substances are new compounds.
If Ri is hydrogen, they are prepared in such a way that 2-chloro-nicotinic acid is condensed with a thiophenol substituted in the p-position by halogen or a lower alkyl group to give the 3-carb oxy-pyridyl- (2) -phenyl sulfide derivative and this is then cyclized by heating with polyphosphoric acid to the corresponding 4-aza-thioxanthone derivative.
Starting products of the formula II, in which Ri is a lower alkyl group, can conveniently be obtained by adding a 2-halo-3-cyanopyridine substituted in the 6-position by lower alkyl groups, for example 2-chloro-3-cyan6-methyl-pyridine an alizalithiophenolate substituted in the p-position by halogen or by lower alkyl groups is condensed at elevated temperature, the 3-cyano-pyridyl- (2) -phenylsulfide derivative obtained is hydrolyzed to the 3-carboxy derivative and then, as already described above, cyclized with polyphosphoric acid.
In the following examples, which explain the execution of the method but are not intended to restrict the invention in any way, all temperature data are given in degrees Celsius and are not corrected.
Example 1 a) 7-Chloro-9- [I'-methyl-piperidyl- (4 ')] -4-aza-thioxanthydrol
2.43 g of magnesium shavings activated with iodine are covered with a layer of 10 cc of tetrahydrofuran and 0.3 cc of ethylene bromide is added, which starts the reaction. The solution of 14.7 g (0.11 mol) of 1-methyl-4-chloropiperidine in 25 ccm of abs. Add tetrahydrofuran dropwise at such a rate that the reaction mixture boils continuously. The mixture is then heated for a further 1-2 hours until most of the magnesium has dissolved, cooled, and 12.4 g (0.05 mol) of 7-chloro-4-aza-thioxanthone are added in portions at 20-25 . After 20 min.
Stirring at room temperature, the reaction mixture is poured to 300 ccm 10 percent.
Ammonium chloride solution and extracted with methylene chloride. After drying over magnesium sulfate and evaporation of the solvent, the residue is recrystallized from acetone. M.p. 225-226. b) 7-chloro-9- [I'-methyl-piperidylidene- (4 ')] -4-aza-thioxanthene
10 g of 7-chloro-9- [1'-methyl-piperidyl- (4 ')] - 4-aza- thioxanthydrol are concentrated with a mixture of 25 cc of water and 75 cc. Sulfuric acid heated to 140 for 20 min. Then it is poured onto 500 ccm of ice water, the solution is made with 50 percent. Sodium hydroxide solution is alkaline and extracted with methylene chloride. After drying over potassium carbonate and evaporation of the solvent, the residue is recrystallized from acetone. M.p. 150-152.
The 7-chloro-4 aza-thioxanthone used as the starting substance is produced as follows:
78 g (0.5 mol) of 2-chloro-nicotinic acid and 145 g (1 mol) of p-chlorothiophenol are heated to 170 for 3 hours. It is then cooled and the crystalline reaction product is stirred with 500 cc of ether and 1000 cc of saturated sodium hydrogen carbonate solution until everything has dissolved. The ether layer is then separated and; Shaken out again with sodium hydrogen carbonate solution. The combined aqueous extracts are adjusted to pH 5 with acetic acid. The precipitated substance, the 3-carboxy-pyridyl- (2) -p-chlorophenyl sulfide, is filtered off, dried and recrystallized from methanol.

35 g of 3-carboxy-pyridyl- (2) -p-chlorophenyl sulfide are heated to 150 for 1 hour and then to 180 for 2 hours with 350 g of polyphosphoric acid. After cooling down to approx.
100¯ the reaction mixture is allowed to flow into 1500 cc of water with thorough stirring. The precipitated substance is filtered off, then with 200 ccm 10proz. Caustic soda 10 min ^. stirred, filtered off again, washed well with water, dried and recrystallized from glacial acetic acid. The 7-chloro-4-aza-thioxanthone melts at 194-195.
Example 2 a) 7-Bromo-9- [I'-methyl-piperidyl- (4 ')] - 4-a. zathioxanthydrol
This compound is prepared in a manner analogous to Example 1, a) by Grignardization of 1-methyl-4-chloro-piperidine, reaction with 7-bromo-4-aza-thioxanthone and subsequent hydrolysis. M.p.

The starting substance used, the 7-bromo-4-aza-thioxanthone with a melting point of 190-193 from glacial acetic acid, is prepared analogously to the starting substance of Example 1 by condensing 2-chloro-nicotinic acid with p-bromothiophenol and the 3-carboxy-pyridyl- (2) -p-bromophenyl sulfide (melting point 220221¯ from methanol) cyclized with polyphosphoric acid.
Example 3 a) 7-Methyl-9- [I'-methyl-piperidyl- (4 ')] -4-aza-thioxanthydrol
This compound is prepared in a manner analogous to Example 1, a) by Grignardation of 1-methyl-4-chloro-piperidine, reaction with 7-methyl-4-aza-thioxanthone and subsequent hydrolysis.

The starting substance used, the 7-methyl 4-aza-thioxanthone with a melting point of 153-154¯ from acetone, is prepared in a manner analogous to the 7-chloro-4-aza-thioxanthone described in Example 1 by adding 2- Chloronicotinic acid is condensed with p-methyl-thiophenol and the 3-carboxy-pyridyl- (2) - p-methyl-phenylsulfide obtained (melting point 217-218 from sithanol) is cyclized with polyphosphoric acid.
Example 4 a) 3-Methyl-7-chloro-9- [I'-methyl-piperidyl- (4 ')] -
4-aza-thioxanthydrol
This compound is prepared in a manner analogous to Example 1, a) by Grigaardation of 1-methyl-4-chloropiperidine, reaction with 3-methyl-7-chloro-4-azathioxanthone and subsequent hydrolysis.

It is prepared by splitting off water from 3-methyl-7-chloro-9- [l'-methyl-piperidyl- (4 ')] -4-aza-thioxanthydrol analogously to Example 1, b). 155-156 from acetone.
The starting substance used, 3-methyl 7-chloro-4-aza-thioxanthene, is produced as follows:
19 g of 2-chloro-3-cyano-6-methyl-pyridine, 23 g of sodium chloro-thiophenol and 160 ccm of abs. Dioxane is refluxed for 20 hours. The precipitated salt is then filtered off and the dioxane solution is evaporated at a pressure of 15 mm Hg. The solution of the residue in methylene chloride is then extracted by shaking with water, dried over magnesium sulphate and evaporated. The residue, 3-cyano-6-methyl-pyridyl- (2) -p-chlorophenyl sulfide, is then recrystallized from acetone. M.p. 135 136.
28 g of the sulfide obtained are concentrated with a mixture of 125 cc of water and 125 cc.
Sulfuric acid heated to 140 for 16 hours. The reaction solution is then poured into 1500 ccm of water, the precipitated substance is filtered off and then dissolved in 5 percent strength while warming. Caustic soda. The solution is then filtered and adjusted to pH 5 with acetic acid. The precipitated substance is filtered off again and dried. The 3-carboxy-6-methyl-pyridyl- (2) -p-chlorophenyl sulfide melts at 212 to 213.
The subsequent cyclization to 3-methyl-7-chloro-4-aza-thioxanthone takes place with polyphosphoric acid as described in Example 1. M.p. 235-236 from glacial acetic acid.
Example 5 a) 7-Chloro-9- (3'-dimethylamino-propyl) -4-aza-thioxanthydrol
This compound is prepared in a manner analogous to Example 1, a) by Grignardation of 3-dimethyl-amnno-propyl chloride, reaction with 7-chloro-4-azathioxanthone and subsequent hydrolysis.
Melting point 175-177¯ from ethyl acetate. b) 7-chloro-9- (3'-dimethylamino-propylidene) -4-aza-thioxanthene
12 g of 7-chloro-9- (3'-dimethylamino-propyl) -4-aza-thioxanthydrol are concentrated with 120 cc of glacial acetic acid and 48 cc. Hydrochloric acid heated under reflux for 1 hour.
The reaction mixture is then evaporated under a pressure of 15 mm Hg, the residue is shaken through with dilute sodium hydroxide solution and methylene chloride, the organic phase is dried over potassium carbonate and evaporated. The remaining oily base (9.1 g) is dissolved in 20 cc of ethanol and the amount of fumaric acid calculated for the preparation of the neutral fumarate is added. The solution obtained by gentle heating is cooled and ether is added, the mixture of the cistrans-isomeric fumarates crystallizing. The neutral fumarate of isomer A, which is present in large quantities, is obtained in pure form after two recrystallizations from methanol. The neutral salt melts at 179-181¯ (dec.).
Example 6 a) 7-Methyl-9- (3'-dimethylamino-propyl) -4-aza-thioxanthydrol
This compound is prepared in a manner analogous to Example 1, a) by Grignardation of 3-dimethylamino propyl chloride, reaction with 7-methyl-4-aza-thioxanthone and subsequent hydrolysis.

The elimination of water from the thioxanhydrol takes place analogously to that described in Example 5, a). The oily base obtained is converted into the acidic fumarate, the mixture of the cis-trans isomeric fumarates crystallizing from ethanol / ether. The acidic fumarate of isomer A is obtained in pure form after three recrystallization from methanol. The acid salt melts at 195-196 (dec.).
