dans laquelle - R représente un atome d'hydrogène ou un radical alcoyle ou alcoyloxyle contenant de 1 à 4 atomes de carbone, et - A et A', identiques ou différents, représentent cha cun une simple liaison ou un radical alcoylène droit ou ramifié contenant de 1 à 6 atomes de carbone et sont situés en méta ou para l'un par rapport à l'autre.
Les composés d e formule (I) peuvent exister sous des formes stéréoisomères et le procédé de l'invention permet d'obtenir aussi bien ces différentes formes que leurs mélanges.
Le procédé selon l'invention est caractérisé en ce que l'on condense du thiophosgène CSC12 sur une di amine de formule générale

en présence d'un excès de thiophosgène et d'un ac cepteur d'acide. On opère généralement à une tempé rature voisine de la température ambiante, de préfé rence entre 0 et 300 C. Il est avantageux d'opérer dans l'eau, en présence d'un agent basique susceptible de neutraliser l'acide chlorhydrique formé au cours de la réaction.
Les formes stéréonsomères des composés de for mule (I) peuvent être obtenues soit directement en partant de matières premières de formes stéréoiso- mères correspondantes, soit par séparation selon les méthodes connues des formes stéréoisomères à partir du mélange les contenant.
Les nouveaux diisothiocyanates de formule (I) pré sentent d'intéressantes propriétés chimiothérapeuti- ques : ils se sont révélés comme des anthelminthiques très actifs, en particulier sur cestodes, et comme des fongicides.
Les exemples suivants montrent comment le pro cédé de l'invention peut être mis en pratique <I>Exemple 1</I> On ajoute lentement en agitant 52,0 g de thio- phosgène à une suspension de 60;0 g d'adipate neutre de diamino-1,4 cyclohexane, forme cis, et de 112,0 g de carbonate de calcium dans 325 cm3 d'eau, en main tenant la température entre 0 et 100 C. On laisse la réaction se poursuivre en agitant pendant 16 heures à 251, C. On ajoute alors 400 cm3 de chloroforme, filtre l'insoluble, le lave avec 200 cm3 d'eau, puis 100 cm3 de chloroforme et joint les lavages au filtrat.
La masse liquide est décantée et la solution chloroformique est séchée sur sulfate de sodium. L'extrait chloroformique est évaporé sous vide (25 mm Hg). Le résidu huileux est dissous dans 500 cm3 de chlorure de méthylène et la solution ainsi obtenue est chromatographiée sur une colonne de 4 cm de diamètre contenant 360 g d'alu mine spéciale pour chromatographie. Le produit fixé sur la colonne est élué avec 780 cm3 de chlorure de méthylène et l'éluat est ensuite éva poré à sec sous vide (25 mm Hg).
Après recristallisa- tion dans 50 cm3 d'oxyde d'isopropyle, on obtient 13,3 g de diisothiocyanato-1,4 cyclohexane, forme cis, fondant à 60-62 C. Le diamino-1,4 cyclohexane, forme cis, utilisé comme produit de départ, a été préparé selon A.T. Nielsen, J. Org. Chem. 27 1998 (1962). _ <I>Exemple 2</I> En opérant comme à l'exemple 1, mais à partir de 16,7g de diamino-1,4 cyclohexane, forme trans, 44,2 g de carbonate de calcium et 35,4 g de thiophos- gène, on obtient, après recristallisation dans le cyclo- hexane, 15,5 g de diisothiocyanato-1,4 cyclohexane, forme trans, fondant à 1460 C.
Le diamino-1,4 cyclohexane, forme trans, utilisé comme produit de départ, a été préparé selon A.T. Nielsen, J. Org. Chem. _27 1998 (1962).
<I>Exemple 3</I> En opérant comme à l'exemple 1, mais à partir de 115,0 g de bis(amino-2 éthyl)-1,4 cyclohexane, forme trans, 148,6 g de carbonate de calcium et 176,6 g de thiophosgène, on obtient, :après chromatographie et recristallisation ,dans le cyclohexane, 92 g de bis(iso- thiocyanato-2 éthyl)-1,4 cyclohexane, forme trans, fon dant à 52-53() C.
Le bis(amino-2 éthyl)-1,4 cyolohexane, forme trans, utilisé comme produit de départ, a été préparé selon P.P. Garcia et J.H. Wood, J: Org. Chem. 26 4167 (1961).
<I>Exemple 4</I> En opérant comme à l'exemple 1, mais à partir de 32,8 g de bis(amino-2 éthyl)-1,4 cyelohexane, forme cis, 38,6 g de carbonate de calcium et 44,5 g de thio- phosgène, on obtient, après chromatographie et re- cristallisation dans l'éther de pétrole, 20;0 g de bis (isothiocyanato-2 éthyl)-1,4 cyclohexane, forme cis, fondant à 38-39 C.
Le bis(amino-2 éthyl)-1,4 cyclohexane, forme cis, utilisé comme produit de départ, a été préparé selon P.P. Garcia et J.H. Wood, J. Org. Chem. 26 4167 (l961).
<I>Exemple 5</I> En opérant comme à l'exemple 1, mais à partir de 18,7- de dichlorhydrate de bis(aminométhyl)-1,4 cyclo- hexane, forme cis, 26,1 g de carbonate de calcium et 20,9 g de thiophosgène, on obtient, après chromato graphie et recristallisation dans le cyclohexane, 12,7 g de bis(isothiocyanatométhyl)-1,4 cyclohexane, forme cis, fondant à 84-86o C.
Le bis(aminométhyl)-1,4 cyclohexane, forme cis, utilisé comme produit de départ, a été préparé selon R. Malachowski et coll., Ber. 71759 (1938).
<I>Exemple 6</I> En opérant comme à l'exemple 1, mais à partir de 32,3 g de dichlorhydrate de bis(aminométhyl)-1,4 cy- elohexane, forme trans, 45,0 g de carbonate de calcium et 47,2 g de thiophosgène, on obtient, après chromato graphie et recristallisation dans le cyclohexane, 9,3 g de bis(isothiocyanatométhyl)-1,4 cyclohexane, forme trans, fondant à 1070 C. Le bis(aminométhyl)-1,4 cyclohexane, forme trans, utilisé comme produit de départ, a été préparé selon R. Malachowski et coll., Ber. 71 759 (1938).
<I>Exemple 7</I> En opérant comme à l'exemple 1, mais à partir de 22,8g de diamino-1,3 cyclohexane, mélange des formes cils et trans, 44,0 g de carbonate de calcium et 52,2 g de thiophosgène, on obtient, après chromato graphie et recristallisation dans l'oxyde d'isopropyle, 14,2 g de diisothiocyanato-1,3 cyclohexane, mélange des formes cis et trams, fondant à 48-500 C.
Le diamino-1,3 cyclohexane, utilisé comme produit de départ, a été préparé selon F.R. Hewgill et P.R. Jefferies, J. Chem. Soc., 805 (1956).
<I>Exemple 8</I> On ajoute lentement 48,7g de thiophosgène à une suspension de 20 g de diamino-1,4 cyclohexane (mé- langedes formes cis et trans) et de 55,5 g de carbonate de calcium dans 70 cm3 -d'eau, en maintenant la tem pérature entre 0 et 100 C. On laisse la réaction se pour suivre pendant 16 heures à 250 C. On acidifie alors le mélange réactionnel par 25 cm3 d'acide chlorhydrique (d = 1,.l9), puis on l'extrait avec 150 cm3 de chloro forme. L'extrait chloroformique est évaporé sous vide et le résidu ainsi obtenu est dissous dans 250 cm3 de chlorure de méthylène.
La solution obtenue est chro- matographiée sur une colonne de 3 cm de diamètre contenant 200 g d'alumine spéciale pour chromatogra phie. Le produit fixé sur la colonne est élué avec 565 cm3 de chlorure de méthylène et l'éluat est ensuite évaporé à sec sous vide. Après recristallisation dans 50 cm3 de cyclohexane, on obtient 17 g de diisothio- cyanato-1,4 cyclohexane (mélange des formes cis et trans) fondant à 721, C.

The compounds of formula (I) can exist in stereoisomeric forms and the process of the invention makes it possible to obtain both these different forms and their mixtures.
The process according to the invention is characterized in that the thiophosgene CSC12 is condensed on a di amine of general formula

in the presence of an excess of thiophosgene and an acid acceptor. The operation is generally carried out at a temperature close to room temperature, preferably between 0 and 300 C. It is advantageous to operate in water, in the presence of a basic agent capable of neutralizing the hydrochloric acid formed during of the reaction.
The stereonsomeric forms of the compounds of formula (I) can be obtained either directly by starting from starting materials of corresponding stereoisomeric forms, or by separation according to known methods of the stereoisomeric forms from the mixture containing them.
The new diisothiocyanates of formula (I) exhibit interesting chemotherapeutic properties: they have been shown to be very active anthelmintics, in particular on cestodes, and as fungicides.
The following examples show how the process of the invention can be put into practice <I> Example 1 </I> 52.0 g of thiophosgene are slowly added with stirring to a suspension of 60.0 g of adipate. Neutral of 1,4-diamino-cyclohexane, cis form, and 112.0 g of calcium carbonate in 325 cm3 of water, keeping the temperature between 0 and 100 C. The reaction is allowed to continue with stirring for 16 hours at 251 ° C. 400 cm3 of chloroform are then added, the insoluble matter is filtered off, washed with 200 cm3 of water, then 100 cm3 of chloroform and the washings are joined to the filtrate.
The liquid mass is decanted and the chloroform solution is dried over sodium sulfate. The chloroform extract is evaporated under vacuum (25 mm Hg). The oily residue is dissolved in 500 cm3 of methylene chloride and the solution thus obtained is chromatographed on a column 4 cm in diameter containing 360 g of special aluminum for chromatography. The product fixed on the column is eluted with 780 cm3 of methylene chloride and the eluate is then evaporated to dryness under vacuum (25 mm Hg).
After recrystallization from 50 cm3 of isopropyl ether, 13.3 g of 1,4-diisothiocyanato cyclohexane, cis form, melting point 60-62 C. The 1,4-diamino cyclohexane, cis form, is used. as starting material, was prepared according to AT Nielsen, J. Org. Chem. 27 1998 (1962). _ <I> Example 2 </I> By operating as in Example 1, but starting from 16.7 g of 1,4-diamino cyclohexane, trans form, 44.2 g of calcium carbonate and 35.4 g of thiophos- gene, after recrystallization from cyclohexane, 15.5 g of 1,4-diisothiocyanato-cyclohexane, trans form, melting at 1460 C.

<I> Example 8 </I> 48.7 g of thiophosgene are slowly added to a suspension of 20 g of 1,4-diamino cyclohexane (mixture of cis and trans forms) and 55.5 g of calcium carbonate in 70 cm3 of water, maintaining the temperature between 0 and 100 C. The reaction is left to follow for 16 hours at 250 C. The reaction mixture is then acidified with 25 cm3 of hydrochloric acid (d = 1, .19), then extracted with 150 cm3 of chloroform. The chloroform extract is evaporated off under vacuum and the residue thus obtained is dissolved in 250 cm3 of methylene chloride.
The solution obtained is chromatographed on a column 3 cm in diameter containing 200 g of special alumina for chromatography. The product fixed on the column is eluted with 565 cm3 of methylene chloride and the eluate is then evaporated to dryness under vacuum. After recrystallization from 50 cm3 of cyclohexane, 17 g of 1,4-diisothiocyanato-cyclohexane (mixture of the cis and trans forms) are obtained, melting at 721 C.
