Verfahren zur Herstellung von p-Alkyl-benzyl-tropiniumderivateai In der Literatur wurden quaternäre Tropeine be schrieben, in welchen die quaternisierende; Gruppe ein Aralkylrest, vorzugsweise ein durch Halogen atome p-subsituierter Benzylrest, ist. Diese Verbin dungen zeigen. eine sehr starke ganglienblockierende. Wirkung, ihre parasympatholytische Wirkung ist da gegen - besonders im Vergleich mit den entsprechen den tertiären Verbindungen - sehr schwach.
In der Literatur ist ferner die Herstellung eines quaternären Tropeins beschrieben, in welchem der quaternisie- rende Benzyl'rest in der p-Stellung durch eine Phenylgruppe substituiert ist.
Dieser Typ der quater- nären: Tropeine ist in therapeutischer Hinsicht vor- teilhafter, da bei diesen Verbindungen die ganglien- blockierende Wirkung von einer milden parasympa- tholytischen Wirkung begleitet wird.
Neben dieser vorteilhaften Eigenschaft zeigt aber der typische Vertreter dieser Verbindungen, das p-Phenylbenzyl-. methyl-( )-tropeyl-tropiniumbromid den Nachteil, dass es im Wasser wenig lösbar ist, und deshalb müssen zur Bereitung von Injektionslösungen Lö sungsvermittler angewendet werden. Auch die Mög lichkeiten der peroralen Anwendung dieser Verbin dung werden durch die schlechte Löslichkeit be schränkt.
Bei der Untersuchung von analogen Verbindun gen haben wir überraschenderweise gefunden, dass die Verbindungen, bei welchen die N-Benzyl- gruppe in der p-Stellung durch einen 2-8 Kohlen stoffatome enthaltenden Alkylrest substituiert ist, wesentlich günstigere therapeutische Eigenschaften zeigen. So hat z.
B. das p-Äthyl-benzyl-atropinium- bromid bei oraler oder intraperitonealer Verabrei- chung eine etwa dreifach stärkere Wirkung, als die analoge p-Phenyl-benzyl-Verbindung, seine Löslich keit ist ebenfalls wesentlich besser und wird auch vom Magen besser resorbiert.
Die neuen p-Alkyl- benzyltropiniumderivate zeigen den weiteren, auch aus therapeutischen Gesichtspunkten wichtigen Vor teil, dass sie neben der guten ganglienblockierenden Wirkung auch eine erheblich: - parasympatholytische Wirksamkeit (etwa ein Drittel jener des Atropins) aufweisen;
ihre Toxizität ist dabei wesentlich gerin ger, etwa 1/,1 jener der analogen p-Phenylbenzinderi- vate. Diese parasympatholytische Wirkung ist beson ders.
bei jenen Derivaten hervortretend, in welchen der in p-Stellung befindliche Substituent der Benzyl- gruppe 2-4. Kohlenstoffatome enthält. Bei der wei teren Verlängerung der Kohlenstoffkette dieser p- Substituenen nimmt die parasympatholytische Wir kung rasch ab,
und parallel tritt eine papaverinartige spasmolytische Wirkung gegenüber Spasmen der glatten Muskulatur hervor, welche mit der Verlän gerung der p-Substituenten der Benzylgruppe immer stärker wird. So zeigt z.
B. das p-Oktylbenzylatropin- niumbromid eine gegenüber jener des Papaverins etwa siebenfach stärkere und das p-n-Butylbenzyl-3tt- (phenyl-cyclohexyl)-acetyl-tropiniumbromid eine etwa zwölffach stärkere spasmolytische Wirkung. Derartige Wirkungen haben die obigen in der Literatur be schriebenen analogen Verbindungen überhaupt nicht gezeigt.
Die vorliegende Erfindung betrifft die Herstel lung von neuen Alkyl-N-benzyltropiniumderivaten der Formel:

worin R für einen geraden oder verzweigten Alkyl- rzst von 2-8 Kohlenstoffatomen, Acyl für einen Carbonsäurerest und X für das Anion einer einen reaktionsfähigen Ester bildenden Säure stehen; die Acyloxygruppe kann dabei zum Stickstoffatom in syn-(a)- oder in anti-(f)-Stellung stehen.
Die neuen, in der Benzylgruppe p-substituierten Tropiniumderivate der Formel (I) zeigen also - wie oben erwähnt - neben der für diese Verbindungen charakteristischen ganglienblockierenden Wirkung eine erhebliche parasympatholytische bzw. spasmo- lytische Wirksamkeit, bessere Löslichkeit und Resor- bierbarkeit sowie eine geringere Toxizität als die bekannten ähnlichen Verbindungen.
Die erwähnten sehr vorteilhaften pharmakologi schen Eigenschaften der neuen Verbindungen der Formel I sind der eigenartigen Wirkung der p-Sub- stituenten der Benzylgruppe zuzuschreiben und sind von der Natur der zur Esterbildung verwendeten Acylgruppe bzw. des salzbildenden Anions weit gehend unabhängig. Die beiden letztgenannten Reste können daher zwischen weiten Grenzen variiert wer den, ohne dass dabei die wertvollen pharmakologi schen Eigenschaften dieser Verbindungen nachteilig beeinflusst würden.
Es versteht sich aber von selbst, dass aus praktischen Gründen am ehesten solche Acylgruppen bzw. solche salzbildenden Säurereste in Betracht kommen, welche die gute Löslichkeit bzw. Resorbierbarkeit der Verbindungen nicht herab setzen. Aus diesen Gründen werden diejenigen Pro dukte vorgezogen, in welchen als Acylgruppe niedere aliphatische Säurereste, z. B. die Acetyl- oder Pro- pionylgruppe, aromatische Säurereste, z.
B. Benzoyl- gruppe, oder aromatisch substituierte Oxysäurereste, z. B. die Tropeylgruppe, und als salzbildendes Anion Halogenide, z. B. Bromid oder Chlorid, ferner Sulfonsäurereste, z. B. die Methansulfonyl- oder p-Toluolsulfonylgruppe, stehen.
Die neuen p-Alkylbenzyl-tropiniumderivate der Formel (I) werden erfindungsgemäss derart herge stellt, dass Tropeine (Carbonsäureester von Tropan- 3a-ol bzw.
Tropan-3ss-ol) der Formel (II) CH2 CH CH2 N-C H3 C H-O-Acyl (II) C112 CH CH2 mit einem reaktionsfähigen Ester eines p-alkyl- substituierten Aralkylalkohols der Formel (III) R / \ -CH2X (III) in -Reaktion gebracht werden.
Die, neuen p-Alkylbenzyl-tropiniumderivate der Formel (I) können mit den bekannten Methoden zu pharmazeutischen Präparaten, wie Tabletten, In jektionen usw., verarbeitet werden; zur Bereitung die ser Präparate können die üblichen Hilfsstoffe, wie Trägerstoffe, Verdünnungs- bzw. Lösungsmittel usw., verwendet werden. <I>Beispiel 1</I> 7,35 g (0,03 Mol) Benzoyl-cz-tropin werden in 50 ml Aceton gelöst, dann mit einer Lösung von 6,2 g p-Äthylbenzylbromid in 25 ml Aceton ver setzt und 2 Stunden am Wasserbad unter Rückfluss erwärmt.
Die Ausscheidung der Quaternärverbindung beginnt schon am Anfang des Erwärmens; nach Be endigung der Reaktion wird das Reaktionsgemisch. abgekühlt und das ausgeschiedene Produkt filtriert, mit Aceton gewaschen und getrocknet. Es werden 12,6 g p-Äthylbenzyl-3a-benzoyl-tropiniumbromid (94,7% der theoretischen Menge) erhalten, Schmp. 226 (Zers.). Nach Umkristallisieren aus einem Ge misch von Äthanol und Methanol steigt der Schmelz punkt auf 229-233 .
<I>Beispiel 2</I> Es wird gemäss Beispiel 1 vorgegangen, mit dem Unterschied, dass an Stelle von p-Äthylbenzylbromid 7,0 g p-tert.-Butylbenzylbromid angewandt werden. Es werden derart 14,0 g p-tert.-Butylbenzyl-3a- benzoyl-tropiniumbromid (98,5 % der theoretischen Menge) erhalten; Schmp. 223 . Nach Umkristallisie-. ren aus einem Gemisch von Isopropanol und Äther schmilzt das gereinigte Produkt bei 228'.
<I>Beispiel 3</I> 8,7 g (0,03 Mol) Atropinbase werden in 50 ml Aceton unter gelindem Erwärmen gelöst, dann mit einer Lösung von 6 g p-Äthylbenzylbromid in 25 ruf Aceton versetzt und 2 Stunden am Wasserbad unter Rückfluss erwärmt.
Die Ausscheidung des Quaternär- salzes beginnt schon in den ersten Minuten des Er- wärmens. Das mit einer Ausbeute von 96,5% er haltene rohe p-Äthylbenzyl-atropiniumbomid wird aus Acetonitril oder aus einem Gemisch von Iso- propanol und Äther umkristallisiert; Schmp. 177'. <I>Beispiel 4</I> Es wird gemäss Beispiel 3 vorgegangen, an Stelle von Aceton wird aber Dimethylsulfoxyd als Lösungs mittel verwendet.
Das rohe p-Athylbenzyl-atropi- niumbromid wird mit nahezu theoretischer Aus beute erhalten. <I>Beispiel S</I> Es wird gemäss Beispiel 3 vorgegangen, an Stelle von p-Äthylbenzylbromid wird aber die äquivalente Menge von p-Isopropylbenzylbromid verwendet. Das derart hergestellte und aus Äthanol umkristallisierte p - Isopropylbenzyl - atropiniumbromid schmilzt bei 205 .
<I>Beispiel 6</I> 8,7 g Atropinbase werden in 50 ml Aceton gelöst, mit einer Lösung von 8,6 g p-Octylbenzyl- bromid versetzt und 2 Stunden unter Rückfluss er-. wärmt. Da das Quaternärsalz zum Teil in der Lö sung bleibt, wird das Reaktionsgemisch unter Va kuum zur Trockne verdampft und der Rückstand zuerst aus Aceton/Äther, dann aus Acetonitril/Ather umkristallisiert. Das derart erhaltene p-Octylbenzyl- atropiniumbromid schmilzt bei<B>1611.</B>
<I>Beispiel 7</I> 10,7 g Phenylcyclohexyl-acetyltropin werden in 50 ml Aceton gelöst, mit einer Lösung von 7 g p-n-Butylbenzylbromi'd in 25 ml Aceton versetzt und bei 60 2 Stunden lang erwärmt. Das mit einer Ausbeute von 82 % erhaltene rohe p-n-Butylbenzyl- 3 a. - (phenyl - cyclohexyl)-acetyltropiniumbromid wird aus Athanoli umkristallisiert Schmp. 245-252 (Zers.).
<I>Beispiel 8</I> 4,34 g (0,015 Mol) 3ss-( )-Tropeyl-tropin (y- Atropin, Schmp. 91-93 ) werden in 30 ml Aceton unter gelindem Erwärmen gelöst, mit einer Lösung von 3 g p-Äthylbenzylbromid in 10 mi Aceton ver setzt und 5 Stunden am Wasserbad unter Rückfluss erwärmt. Das anfangs in öligem Zustand ausschei dend; Produkt kristallisiert über Nacht. Es wird filtriert und mit Aceton gewaschen.
Das erhaltene p-Athylbenzyl - 3ss - ( )-tropeyl tropini'umbromid ist sehr hygroskopisch, sein Schmelzpunkt kann daher mit annähernder Genauigkeit bestimmt werden; es schmilzt bei 95-100 unter Zersetzung.

In addition to this advantageous property, however, the typical representative of these compounds, p-phenylbenzyl-. methyl- () -tropeyl-tropinium bromide has the disadvantage that it is not very soluble in water, and therefore solubilizers must be used for the preparation of injection solutions. The possibilities of oral use of this compound are also limited by its poor solubility.
When investigating analogous compounds, we surprisingly found that the compounds in which the N-benzyl group in the p-position is substituted by an alkyl radical containing 2-8 carbon atoms show significantly more favorable therapeutic properties. So has z.
B. p-ethyl-benzyl-atropinium bromide, when administered orally or intraperitoneally, has an effect about three times stronger than the analogous p-phenyl-benzyl compound, its solubility is also much better and is also better absorbed by the stomach .
The new p-alkylbenzyltropinium derivatives show the further advantage, which is also important from a therapeutic point of view, that, in addition to the good ganglion-blocking effect, they also have a considerable: - parasympatholytic activity (about a third of that of atropine);
Their toxicity is much lower, about 1/1 of that of the analogous p-phenylbenzene derivatives. This parasympatholytic effect is special.
in the case of those derivatives in which the p-position substituent of the benzyl group 2-4. Contains carbon atoms. As the carbon chain of these p-substituents is further lengthened, the parasympatholytic effect decreases rapidly,
and at the same time there is a papaverine-like spasmolytic effect on spasms of the smooth muscles, which becomes stronger as the p-substituents of the benzyl group lengthen. So shows z.
B. p-octylbenzylatropinium bromide has a spasmolytic effect about seven times stronger than that of papaverine and p-n-butylbenzyl-3tt- (phenyl-cyclohexyl) -acetyl-tropinium bromide about twelve times stronger. The present invention relates to the production of new alkyl-N-benzyltropinium derivatives of the formula:

where R is a straight or branched alkyl ring of 2-8 carbon atoms, acyl is a carboxylic acid residue and X is the anion of an acid which forms a reactive ester; the acyloxy group can be in the syn (a) or anti (f) position to the nitrogen atom.
The new tropinium derivatives of the formula (I) substituted in the benzyl group thus show - as mentioned above - in addition to the ganglion-blocking effect characteristic of these compounds, considerable parasympatholytic or spasmolytic activity, better solubility and resorbability and lower toxicity than the known similar compounds.
The mentioned very advantageous pharmacological properties of the new compounds of the formula I are attributable to the peculiar effect of the p-substituents of the benzyl group and are largely independent of the nature of the acyl group or the salt-forming anion used for ester formation. The last two radicals mentioned can therefore be varied between wide limits without the valuable pharmacological properties of these compounds being adversely affected.
It goes without saying, however, that for practical reasons those acyl groups or those salt-forming acid residues are most likely to come into consideration which do not reduce the good solubility or resorbability of the compounds. For these reasons, those pro products are preferred in which the acyl group is lower aliphatic acid residues, eg. B. the acetyl or propionyl group, aromatic acid residues, z.
B. benzoyl group, or aromatically substituted oxy acid residues, z. B. the tropeyl group, and as the salt-forming anion halides, z. B. bromide or chloride, also sulfonic acid residues, z. B. the methanesulfonyl or p-toluenesulfonyl group.
The new p-alkylbenzyl-tropinium derivatives of the formula (I) are manufactured according to the invention in such a way that tropeins (carboxylic acid esters of tropan-3a-ol or
Tropan-3ss-ol) of the formula (II) CH2 CH CH2 NC H3 C HO-acyl (II) C112 CH CH2 with a reactive ester of a p-alkyl-substituted aralkyl alcohol of the formula (III) R / \ -CH2X (III) be brought into reaction.

<I> Example 2 </I> The procedure is as in Example 1, with the difference that instead of p-ethylbenzyl bromide, 7.0 g of p-tert-butylbenzyl bromide are used. 14.0 g of p-tert-butylbenzyl-3-benzoyl-tropinium bromide (98.5% of the theoretical amount) are obtained in this way; M.p. 223. The crude p-ethylbenzyl-atropinium bromide is obtained with almost theoretical yield. <I> Example S </I> The procedure is as in Example 3, but the equivalent amount of p-isopropylbenzyl bromide is used instead of p-ethylbenzyl bromide. The p-isopropylbenzyl-atropinium bromide which has been prepared in this way and recrystallized from ethanol melts at 205.
<I> Example 6 </I> 8.7 g of atropine base are dissolved in 50 ml of acetone, a solution of 8.6 g of p-octylbenzyl bromide is added and the mixture is refluxed for 2 hours. warms. Since some of the quaternary salt remains in the solution, the reaction mixture is evaporated to dryness under vacuum and the residue is recrystallized first from acetone / ether and then from acetonitrile / ether. <I> Example 7 </I> 10.7 g of phenylcyclohexyl-acetyltropine are dissolved in 50 ml of acetone, treated with a solution of 7 g of p-n-butylbenzylbromide in 25 ml of acetone and heated at 60 for 2 hours. The crude p-n-butylbenzyl-3 a obtained with a yield of 82%. - (Phenyl - cyclohexyl) acetyltropinium bromide is recrystallized from Ethanoli, melting point 245-252 (decomp.).
<I> Example 8 </I> 4.34 g (0.015 mol) 3ss- () -tropyl-tropine (y-atropine, melting point 91-93) are dissolved in 30 ml acetone with gentle warming, with a solution of 3 g of p-ethylbenzyl bromide in 10 ml of acetone are set and refluxed for 5 hours on a water bath.
