Verfahren zur Herstellung von d 5-19-Hydroxy-steroiden Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von d5-19-Hydroxystero- iden aus 5a-Halogen-6ss,19-oxido-steroiden. Die ge nannten 19-oxigenierten Steroide sind wichtige Zwi schenprodukte zur Gewinnung pharmakologisch wich tiger 19-Nor-steroide, z.
B. von Derivaten des 19- Nor-testosterons und 19-Nor-progesterons. Insbeson dere das 19-Nor-17a-methyl-testosteron, das 19-Nor- 17a-äthenyl-testosteron und gewisse Ester des 19- Nor-testosterons haben in letzter Zeit therapeutische Verwendung gefunden.
Alle diese Verbindungen waren bisher nur durch Reduktion von Steroidverbindungen mit einem aro matischen Ring A zugänglich, welche ihrerseits aus ungesättigten 3-Keto-steroiden durch thermische Eli minierung der angulären C-19-Methylgruppe und gleichzeitige Aromatisierung gewonnen werden muss ten.
Die gemäss der vorliegenden Erfindung nunmehr leicht zugänglichen 19-oxigenierten d5-Steroide er möglichen nun die Herstellung von 19-Nor-steroiden auf äusserst einfache Weise, ohne dass dabei der Ring A zuerst aromatisiert werden muss.
Das erfindungsgemässe Verfahren besteht darin, dass man 5a-Halogen-6P,19-oxido-steroide reduktiv spaltet.
Die Ausgangsstoffe können z. B. der Spirostan-, Androstan-, Pregnan-, Cholan-, Cholestan-, Stigma- stan-, Spirostan- und Cardanolidreihe angehören, welche im Ringsystem, insbesondere in einer oder mehreren der Stellungen 1, 2, 3, 4, 7, 8, 9, 11, 12, 14, 15, 16, 17, 20 und 21 weitere Substituenten auf weisen können, wie freie oder funktionell abgewan delte Oxogruppen,
veresterte oder verätherte Hy- droxylgruppen, Alkyl- (z. B. Methyl-) Gruppen und/ oder Halogenatome. Unter funktionell abgewandelten Oxogruppen kommen ketalisierte oder in Enolderi- vate, z. B. Enoläther oder Enolester, übergeführte Oxogruppen in Frage.
Ausserdem können die Aus gangsstoffe auch Doppelbindungen oder Oxidogrup- pen aufweisen, z. B. 9,11- oder 16,17-Stellung.
Besonders wichtige Ausgangsstoffe sind solche 5a-Halogen-6ss,19-oxyde, die in 3-Stellung eine freie, veresterte oder verätherte Hydroxygruppe oder eine ketalisierte Oxogruppe aufweisen, da dann anschlie ssend an die verfahrensgemässe Reduktion in bekann ter Weise leicht eine d4-3-Oxogruppierung eingeführt werden kann, welche die Abspaltung des angulären C-19-Substituenten erleichtert.
Die Ausgangsstoffe können durch Einwirkung oxydierend wirkender Schwermetallacylate, insbeson dere Bleitetraacylate, gegebenenfalls in Gegenwart von Jod, auf 19-unsubstituierte 5a-Halogen-6ss-hy- droxy-steroide hergestellt werden.
Spezifische Ausgangsstoffe für das vorliegende Verfahren sind z. B. die folgenden Verbindungen: 3 - Hydroxy - 5 a - halogen -6ss,19-oxido-spirostane und ihre Ester, z. B. das 3ss-Acetoxy-5a-chlor-6ss,19-oxido-spirostan, 3ss,17ss-Dihydroxy-5a-chlor- und 5a-brom-6ss,19-oxido-androstan und seine Ester, das 3ss-Hydroxy-5a-chlor- und -5a-brom-6ss,19-oxido-17-oxo-androstan und seine Ester, 3ss,17ss-Dihydroxy-5a-chlor- und -5a-brom-6ss,19-oxido-17a-alkyl ,
insbesondere -17a-methyl- und -17a-äthyl-androstan und seine Ester, 3ss,17ss-Dihydroxy-Sa-chlor- und 5a-brom-6ss,19-oxido-17a-allyl-androstan und seine Ester, 3ss,20-Dihydroxy-5a-chlor- und -5a-brom-6ss,19-oxido-pregnan und seine Ester, 3ss-Hydroxy-5a-chlor- und -5a-brom-6ss,19-oxido-20-oxo-pregnan und seine Ester, 3ss-Hydroxy-5a-chlor- und -5 a-brom-6ss,19-oxido-16,17a-oxido-20-oxo- pregnan und seine Ester, 3ss,
17a-Dihydroxy-5a-chlor- und -5a-brom-6ss,19-oxido-20-oxo-pregnan. Die 5a-Halogen-6ss,19-oxyde sind gegenüber hydrolytischen Mitteln sehr stabil. überraschender weise gelingt aber die Öffnung der 6ss,19-Sauerstoff- brücke leicht unter reduktiven Bedingungen, insbeson dere mit solchen Reduktionsmitteln, welche für die Reduktion von Halogenhydrinen zu Olefinen ver wendet werden können.
Besonders geeignet sind me tallische Reduktionsmittel, wie Alkali- oder Erdalkali- metalle, wie Natrium, Kalium, Lithium oder beson ders Calcium, insbesondere gelöst in flüssigem Am moniak oder einem Amin, wie Äthylamin, Isopropyl- amin, Äthylendiamin usw., gegebenenfalls unter Zu satz eines Verdünnungsmittels wie Äther, Tetrahydro- furan oder Dioxan.
Unter den angegebenen Reaktionsbedingungen bilden sich d5-19-Hydroxy-steroide. Gleichzeitig mit der reduktiven Öffnung der 6ss,19-Oxyde können an dere funktionelle Gruppen, wie andere Oxyde, z. B. 16,17-Oxyde oder Oxogruppen, falls sie nicht durch Ketalisierung oder Enolätherbildung geschützt sind, ebenfalls reduziert werden.
In den Estern der verfahrensgemäss erhaltenen Produkte können die Säurereste solche von gesättig ten oder ungesättigten aliphatischen oder cycloalipha- tischen, von aromatischen oder heterocyclischen Car- bonsäuren sein, beispielsweise solche der Ameisen säure, Essigsäure, Propionsäure, der Buttersäuren, Valeriansäuren, wie n-Valeriansäure oder Trimethyl- essigsäure, der Capronsäuren,
wie ss-Trimethylpro- pionsäure, der önanth-, Capryl-, Pelargon-, Caprin-, Undecylsäuren, z. B. der Undecylensäure, der Lau rin-, Myristin-, Palminn- oder Stearinsäuren, z. B.
der Ölsäure, der Cyclopentyl-, Cyclohexyl- oder Phe- nyl-essigsäuren oder -propionsäuren, der Benzoe- säure, Phenoxy-alkansäuren, wie Phenoxy-essigsäure, p - Chlor - phenoxy-essigsäure, 2,4-Dichlor-phenoxy- essigsäure, 4-tert.-Butyl-phenoxy-essigsäure, 3-Phen- oxy-propionsäure, 4-Phenoxy-buttersäure,
der Furan- 2 - carbonsäure, 5 - tert. - Butyl-furan-2-carbonsäure, 5 - Brom - furan - 2 - carbonsäure, der Nicotinsäu- ren, ferner von Dicarbonsäuren, wie Oxal'-, Bernstein- oder Glutarsäuren, von substituierten Carbonsäuren, wie ss-Ketocarbonsäuren, z.
B. der Acetessig , Propionylessig-, Butyrylessig- oder Ca- prinoylessigsäure, von Aminosäuren usw.
Zur Herstellung der therapeutisch wichtigen 19- Nor-steroide können die verfahrensgemäss erhaltenen 19-oxigenierten d5-3-Hydroxy-steroide durch Oxy dation, z. B. nach der Methode von Oppenauer, in 44-3-Oxo-steroide verwandelt werden. Aus den er haltenen 44-3-Oxo-19-hydroxy Steroiden lässt sich die anguläre Hydroxymethylgruppe, gegebenenfalls nach Oxydation zur Aldehyd- oder Säuregruppe durch Ein wirkung alkalischer Mittel, z.
B. durch Erwärmen mit einem Alkalimetallhydroxyd in wasserfreier oder wässriger Lösung, unter Bildung von 19-Nor-Verbin- dungen abspalten. Man erhält so z. B. aus den ver fahrensgemäss erhaltenen 44-3,-Oxo-19-hydroxy-Ver- bindungen z. B. das bekannte 19-Nor-testosteron, das 19-Nor-progesteron usw.
Die Temperaturen sind in den nachstehenden Bei spielen in Celsiusgraden angegeben.
<I>Beispiel 1</I> In einem Vierhalskolben mit Rührer, Gasablei- tungsrohr, Tropftrichter und Rückflusskühler, der mit einem Feuchtigkeitsabschluss und einem Gasablei- tungsrohr versehen ist, werden 100 ml Ammoniak kondensiert (Badtemperatur - 60 ).
Portionenweise wird nun 1,00g Lithium-Metall zugegeben, das sich im Ammoniak mit blauer Farbe auflöst, und an schliessend unter Rühren in schneller Folge eine Lö sung von 3,00 g 3ss-Acetoxy-5a-chloro-(6ss---> 19) oxido-cholestan in 50 ml abs. Äther zugetropft. Nach der Entfernung des Kühlbades wird unter Rühren und Überleiten eines trockenen Stickstoffstromes innert 2 Stunden der Ammoniak abgedampft, wobei man in Abständen von 20 Minuten jeweils 25 ml abs. Äther zufügt.
Das überflüssige Lithium wird mit Methanol zerstört und das Reaktionsgemisch nach Ansäuern mit verdünnter Schwefelsäure und Verdünnen mit Äther aufgearbeitet. Die nacheinander mit Wasser, Natriumhydrogencarbonatlösung und Wasser ge waschene und mit Natriumsulfat getrocknete äthe rische Lösung liefert nach Eindampfen im Vakuum 2,52 g eines öligen Produktes, das beim Bespritzen mit Methanol kristallisiert. Nach einmaligem Umkri- stallisieren aus Methylenchlorid-Methanol erhält man 1,130 g d5-3ss,
19-Dihydroxy-cholesten (19-Hydroxy- cholesterin) mit F. 153-155 C. Das IR-Spektrum weist u. a charakteristische Absorptionsbanden bei 2,8,0, 6,86, 7,30, 7,36, 7,5,0, 9,25, 9,70, 9,85, 10,30, 10,48, 10,87, 11,30 und 11,90,u auf.
Vom Kernresonanzspektrum fehlt das Maximum der C-19-Methylgruppe, dagegen treten bei 153 bis 184 Hz und 45-48,5 Hz Maxima auf, die den Pro tonen der -CH2-OH bzw. der -C=CH-Gruppierung zugeordnet werden können.
In analoger Weise erhält man aus 5,0 g 3ss-Acet- oxy-5a-chlor-6ss,19-oxido-spirostan 3,6 g 19-Hy- droxy-diosgenin.
<I>Beispiel 2</I> In 100 ml flüssigem Ammoniak werden unter Rühren bei - 60 750 mg Lithiummetall in Portio nen aufgelöst. Die blaue Lösung versetzt man mit 1,00 g 3ss - Acetoxy - 5a-chloro-(6ss -->19)-oxido-17- oxo-androstan und splült mit 50 ml abs. Äther nach. Unter Rühren, ohne externe Kühlung wird anschlie ssend während 1,5 Stunden ein trockener Stickstoff strom über die Reaktionslösung geleitet und der ab- gedampfte Ammoniak durch abs. Äther ersetzt.
Der Überschuss an Lithium zersetzt man mittels Methanol und lässt das Gemisch während 18 Stunden bei Raum temperatur stehen. Nach Ansäuern des Reaktions gemisches mit verdünnter Schwefelsäure und Extrak tion mittels Chloroform werden 857 mg eines Öls iso liert. Dieses kristallisiert beim Bespritzen mit Essig ester/Äther. Man erhält so das d5-3ss,19,17-Tri- hydroxy-androsten vom F. 231-233 .
<I>Beispiel 3</I> Aus einer Lösung von 350 mg 45-3ss,19-Di- hydroxy-cholesten in 15 ml abs. Toluol und 5 ml Cyclohexanon werden etwa 5 ml Lösungsmittel ab destilliert und das Reaktionsgemisch nach Zugabe einer Lösung von 470 mg Aluminiumisopropylat in 10 ml Toluol unter Rühren 15 Stunden am Rückfluss gekocht. Das abgekühlte und mit verdünnter Schwe felsäure angesäuerte Gemisch wird in Äther auf genommen, neutralgewaschen, getrocknet und im Vakuum eingeengt.
Den Rückstand unterwirft man zwecks Abtrennung des Cyclohexanons und Cyclo- hexanols einer Wasserdampfdestillation, nimmt die nichtflüchtigen Anteile in Äther auf und isoliert auf übliche Weise das Reaktionsprodukt. Dieses fällt in Form eines nicht kristallisierenden Öls an und wird durch Chromatographie an Aluminiumoxyd gereinigt. In einer Ausbeute von 15-20 % wird auf diese Weise das amorphe 19-Nor-d4-3-oxo-cholesten erhalten. Die reine Verbindung weist im IR-Absorptionsspektrum u. a.
Banden bei 6,00, 6,85, 7,25, 7,35, 7,55, 8,30, 9,00, 10,36 und 11,30,u auf.
<I>Beispiel 4</I> In einem 500-ml-Rührkolben kondensiert man 200 ml Ammoniak und gibt anschliessend 1,5 g Lithiummetall in kleinen Portionen zu. Zur entstan denen blauen Lösung lässt man innert 15 Minuten 1,95 g 3ss,20ss - Diacetoxy - 5a -chlor - 6ss,19 - oxido- pregnan in 20 ml Tetrahydrofuran zutropfen und rührt schliesslich noch 1,5 Stunden weiter.
Dann gibt man langsam ein Gemisch von 20 ml Methanol und 100 ml Äther zu und lässt schliesslich den Ammoniak verdampfen. Schliesslich gibt man nochmals 100 ml Tetrahydrofuran zu, leitet während 1 Stunde Stick stoff durch die Suspension, verdünnt mit 200 ml Was ser und extrahiert mehrmals mit Methylenchlorid. Die Extrakte werden mit verdünnter Schwefelsäure und mit Wasser gewaschen, getrocknet und ein gedampft.
Man erhält 1,82 g rohes 45-3ss,19,20ss- Trihydroxy-pregnen. Dieses wird in 200 ml Aceton gelöst und bei 0 mit 5,0 ml einer auf 5,0,0 ml mit Wasser aufgefüllten Lösung von 13,3 g Chromtrioxyd in 11,5 ml konz. Schwefelsäure 35 Minuten gerührt. Dann setzt man 50 g krist. Natriumacetat in 80 ml Wasser zu, verdünnt mit 500 ml Benzol,
wäscht die Lösung mehrmals mit Wasser und extrahiert die wäss- rige Lösung mehrmals mit Benzol. Der Rückstand der getrockneten und im Wasserstrahlvakuum ein- gedampften Lösung wird in 1-00 ml Chloroform auf genommen und bei -I- 10 mit Chlorwasserstoffgas gesättigt. Man lässt 16 Stunden bei 0 stehen und dampft anschliessend im Wasserstrahlvakuum ein.
Aus dem Rückstand erhält man durch Chromato- graphie an 30 g Aluminiumoxyd 81,0 mg 19-Nor- progesteron vom F. 141-144 .
Process for the preparation of d5-19-hydroxy steroids The present invention relates to a process for the preparation of d5-19-hydroxysteroids from 5a-halogen-6ss, 19-oxido-steroids. B. of derivatives of 19-nor-testosterone and 19-nor-progesterone. In particular 19-nor-17a-methyl-testosterone, 19-nor-17a-ethenyl-testosterone and certain esters of 19-nor-testosterone have recently found therapeutic use.
All these compounds were previously only accessible by reducing steroid compounds with an aromatic ring A, which in turn had to be obtained from unsaturated 3-keto steroids by thermal elimination of the angular C-19 methyl group and simultaneous aromatization.
The 19-oxygenated d5-steroids, which are now easily accessible according to the present invention, now enable the production of 19-nor-steroids in an extremely simple manner, without ring A first having to be aromatized.
The process according to the invention consists in cleaving 5a-halo-6P, 19-oxido-steroids reductively.
The starting materials can, for. B. belonging to the spirostan, androstan, pregnan, cholan, cholestan, stigma stan, spirostan and cardanolide series, which in the ring system, in particular in one or more of the positions 1, 2, 3, 4, 7, 8, 9, 11, 12, 14, 15, 16, 17, 20 and 21 can have further substituents, such as free or functionally modified oxo groups,

in particular -17a-methyl- and -17a-ethyl-androstane and its esters, 3ss, 17ss-dihydroxy-Sa-chloro- and 5a-bromo-6ss, 19-oxido-17a-allyl-androstane and its esters, 3ss, 20 -Dihydroxy-5a-chloro- and -5a-bromo-6ss, 19-oxido-pregnan and its esters, 3ss-hydroxy-5a-chloro- and -5a-bromo-6ss, 19-oxido-20-oxo-pregnan and its esters, 3ss-hydroxy-5a-chloro- and -5 a-bromo-6ss, 19-oxido-16,17a-oxido-20-oxo-pregnane and its esters, 3ss,
17a-dihydroxy-5a-chloro- and -5a-bromo-6ss, 19-oxido-20-oxo-pregnane. The 5a-halogen-6ss, 19-oxides are very stable towards hydrolytic agents. Surprisingly, however, the opening of the 6ss, 19-oxygen bridge easily succeeds under reductive conditions, in particular with reducing agents which can be used for the reduction of halohydrins to olefins.
Particularly suitable are metallic reducing agents, such as alkali or alkaline earth metals, such as sodium, potassium, lithium or especially calcium, in particular dissolved in liquid ammonia or an amine such as ethylamine, isopropylamine, ethylenediamine, etc., optionally below set of a diluent such as ether, tetrahydrofuran or dioxane.

To prepare the therapeutically important 19 nor-steroids, the 19-oxygenated d5-3-hydroxy steroids obtained according to the process can be dated by oxidation, eg. B. by the method of Oppenauer, are converted into 44-3-oxo-steroids. <I> Example 1 </I> In a four-necked flask with stirrer, gas discharge pipe, dropping funnel and reflux condenser, which is provided with a moisture seal and a gas discharge pipe, 100 ml of ammonia are condensed (bath temperature - 60).
In portions, 1.00 g of lithium metal is now added, which dissolves in ammonia with a blue color, and then a solution of 3.00 g of 3ss-acetoxy-5a-chloro- (6ss ---> 19) oxido-cholestane in 50 ml of abs. Ether added dropwise. 19-dihydroxy-cholesterol (19-hydroxy-cholesterol) with F. 153-155 C. The IR spectrum shows u. a characteristic absorption bands at 2.8.0, 6.86, 7.30, 7.36, 7.5.0, 9.25, 9.70, 9.85, 10.30, 10.48, 10, 87, 11.30 and 11.90, u.
The maximum of the C-19 methyl group is absent from the nuclear magnetic resonance spectrum, whereas at 153 to 184 Hz and 45-48.5 Hz maxima occur which can be assigned to the protons of the -CH2-OH or the -C = CH group .
In an analogous manner, 3.6 g of 19-hydroxy-diosgenin are obtained from 5.0 g of 3ss-acetoxy-5a-chloro-6ss, 19-oxido-spirostane.
<I> Example 2 </I> In 100 ml of liquid ammonia, 750 mg of lithium metal are dissolved in portions at -60 while stirring. The blue solution is mixed with 1.00 g of 3ss - acetoxy - 5a-chloro- (6ss -> 19) -oxido-17-oxo-androstane and rinsed with 50 ml abs. Ether after. With stirring, without external cooling, a stream of dry nitrogen is then passed over the reaction solution for 1.5 hours and the ammonia evaporated through abs. Ether replaced.
The excess of lithium is decomposed using methanol and the mixture is left to stand at room temperature for 18 hours. This gives the d5-3ss, 19,17-trihydroxy androstene from F. 231-233.

Toluene and 5 ml of cyclohexanone, about 5 ml of solvent are distilled off and, after adding a solution of 470 mg of aluminum isopropoxide in 10 ml of toluene, the reaction mixture is refluxed for 15 hours with stirring. The cooled mixture acidified with dilute sulfuric acid is taken up in ether, washed neutral, dried and concentrated in vacuo.
In order to separate off the cyclohexanone and cyclohexanol, the residue is subjected to steam distillation, the non-volatile components are taken up in ether and the reaction product is isolated in the usual way. This is obtained in the form of a non-crystallizing oil and is purified by chromatography on aluminum oxide. The amorphous 19-nor-d4-3-oxo-cholesten is obtained in this way in a yield of 15-20%. The pure compound shows in the IR absorption spectrum u. a.
Bands at 6.00, 6.85, 7.25, 7.35, 7.55, 8.30, 9.00, 10.36 and 11.30, u.
<I> Example 4 </I> 200 ml of ammonia are condensed in a 500 ml stirred flask and 1.5 g of lithium metal are then added in small portions. 1.95 g of 3ss, 20ss -diacetoxy-5α-chlorine-6ss, 19-oxido-pregnane in 20 ml of tetrahydrofuran are added dropwise to the resulting blue solution within 15 minutes, and the mixture is finally stirred for a further 1.5 hours.
A mixture of 20 ml of methanol and 100 ml of ether is then slowly added and the ammonia is finally allowed to evaporate. Finally, 100 ml of tetrahydrofuran are added again, nitrogen is passed through the suspension for 1 hour, diluted with 200 ml of water and extracted several times with methylene chloride. The extracts are washed with dilute sulfuric acid and with water, dried and evaporated.
1.82 g of crude 45-3ss, 19.20ss-trihydroxy-pregnen are obtained. This is dissolved in 200 ml of acetone and concentrated at 0 with 5.0 ml of a solution made up to 5.0.0 ml with water of 13.3 g of chromium trioxide in 11.5 ml. Sulfuric acid stirred for 35 minutes. Then 50 g of crystalline are used. Sodium acetate in 80 ml of water, diluted with 500 ml of benzene,
the solution was washed several times with water and the aqueous solution extracted several times with benzene. The residue of the dried and evaporated solution in a water jet vacuum is taken up in 1-00 ml of chloroform and saturated at -I- 10 with hydrogen chloride gas. The mixture is left to stand at 0 for 16 hours and then evaporated in a water-jet vacuum.
By chromatography on 30 g of aluminum oxide, 81.0 mg of 19-norgestesterone with a melting point of 141-144 are obtained from the residue.
