Process for the preparation of α-dialkylthio- (dithio) -phosphoryl-α-phenylacetic acid esters of cyanohydrin and use of these esters as a component of pesticides. The present invention relates to the preparation of a new class of thio- and di-thiophosphoric acid esters of the general formula :
EMI0001.0004
where R 'and R "are lower alkyl groups, R' is identical to or different from R", R1, R2 are hydrogen, alkyl or aryl radicals and X is oxygen or sulfur.
In laboratory tests it has been observed that these compounds are more or less toxic to plants or house parasites.
Some of them show a particularly high toxic effect against mites and their eggs.
It is therefore also the use of the compounds of the formula given above as a component of pesticides claims.
The preparation of the substances of the formula (1) is characterized in that a metal or ammonium compound of a 0,0-dialkylthio (or dithio) phosphoric acid is used
EMI0001.0011
wherein Me is an alkali metal or ammonium with an α-halo-α-phenylacetic acid ester of cyanohydrin
EMI0001.0014
wherein Hal is chlorine, bromine or iodine, and the metal or ammonium halide formed separates from.
The reaction can be carried out in the presence of solvents for one or both reaction components; Particularly suitable solvents for this are ketones and, in particular, acetones.
It may be useful to use an excess of metal or ammonium dialkyl phosphate. The reaction, which also takes place at room temperature, can be accelerated by heating to 50 to 600 C.
The metal or ammonium halide formed as a by-product can, for. B. either filtered off or by adding water, where the inorganic salt dissolves and at the same time the ge desired product precipitates, be separated.
The phosphoric esters of the general formula (1) are oily, colorless or pale yellow liquids that are almost insoluble in water, but are soluble in alcohol, benzene, acetone, dioxane and other commonly used organic solvents with the exception of aliphatic hydrocarbons .
The α-halo-α-phenylacetic acid esters of the cyanohydrins (which are used as starting materials in the preparation of compounds of the first-mentioned general formula) can be prepared by adding the halide of the α-halophenylacetic acid to a mixture of cyanohydrin, anhydrous Pyridine and petroleum ether are prepared to the mixture cooled to 011 C. The temperature of the reaction mass must always be kept at 00 C.
The pyridine halide thus formed is separated off while the liquid is concentrated on a water bath under reduced pressure.
The residue is diluted ver with methylene chloride, shaken with ice water and then neutralized with sodium bicarbonate.
After drying the organic layer with sodium sulfate, the substance is concentrated on a water bath. The residue consists of the α-halophenylacetic acid ester of a cyanohydrin. The product can be purified by vacuum distillation.
Among the products of general formula (1), the α- (0,0-diethyldithiophosphoryl) -α-phenylacetic acid esters of acetone cyanohydrin showed:
EMI0002.0010
particular effectiveness. Tests have shown that this substance has an acaricidal activity which is approximately 18 times higher than that of parathion (p-nitrophenyl diethyl thiophosphate).
It is also surprising that this new compound shows a higher parasiticidal activity than the other compounds with a similar constitution and than the phosphoric acid esters usually used as parasiticides, not only against mites in their various growth stages, but also against their eggs.
The compounds produced according to the invention can be used in various ways. In general, they can be diluted with solid, liquid or semi-solid carriers; wetting agents, adhesives, dispersing agents and emulsifying agents can be added. The agent can be made into a paste; the active substances can be dispersed in the air by smoking or as aerosols. Other substances with insecticidal activity can be added (phosphoric acid ester, dichlorodiphenyltrichloroethane, γ-isomer of hexachlorocyclohexane, octachloro-4,7-methane-tetrahydroindane, etc.).
Example 1 a) Preparation of the starting product α-bromo-α-phenylacetic acid ester of acetone cyanohydrin, which was previously unknown in the chemical literature. 278 g of α-phenylacetic acid bromide are slowly added to a cooled mixture of 85 g of acetone cyanohydrin, 79 g of anhydrous pyridine and 180 ml of petroleum ether.
The pyridine hydrobromide thus formed is separated off, the liquid is concentrated on a water bath under reduced pressure.
The residue is diluted ver with methylene chloride, shaken with ice water and neutralized with sodium bicarbonate. After drying the organic layer with sodium sulfate, it is evaporated on a water bath and 219 g of α-bromo-α-phenylacetic acid ester of acetone cyanohydrin are obtained as residue.
The product can be purified by vacuum distillation, collecting the fraction that passes over at 12411 C and 0.6 mm Hg.
b) 28.2 g of α-bromo-α-phenylacetic acid ester of acetoney anhydrin, dissolved in 30 ml of acetone, are added to a solution of 26 g of sodium 0,0-dimethyl-di-thiophosphate dihydrate in 60 ml of acetone.
The mixture heats up and NaBr separates.
The reaction mixture is heated under reflux for 30 min and, after cooling, the NaBr formed (9 g) is filtered off. The liquid is now concentrated to half the volume under reduced pressure and then diluted ver with 200 ml of water. The oil which separates out is extracted with methylene chloride and neutralized with a little aqueous saturated sodium bicarbonate solution.
The organic layer is dried over sodium sulfate and evaporated on a water bath. The residue is washed 5 times with approximately 10 ml of n-hexane each time in a separating funnel; the fraction dissolved in n-hexane is discarded.
The oily liquid obtained consists essentially of α- (0,0-dimethyldithiophosphoryl) -α-phenylacetic acid ester of acetone cyanohydrin
