cycloalipha- tischen, cycloaliphatisch-aliphatischen, araliphatischen oder heterocyclisch-aliphatischen Rest steht, R3 ein Wasserstoffatom oder einen niederen Alkylrest und R1 ein Wasserstoffatom, einen Alkylrest, einen Hy- droxyalkylrest, einen Halogenalkylrest, einen Oxa- alkylrest,
einen Cycloalkyl- oder Cycloalkylalkylrest, einen Aralkylrest, einen Heterocyclyl-alkylrest oder einen höchstens zweikernigen Arylrest oder hetero- cyclischen Rest bedeutet, oder ihrer Salze.
In den neuen Verbindungen kommen als Alkyl- reste insbesondere niedere Alkyh-este, wie Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, Pentyl- (1)-, Pentyl-(2)-, Pentyl-(3)-, 2-Methyl-butyl-(3)- oder Hexylreste,
als Halogenalkylreste oder Hydroxyalkyl- reste beispielsweise Chloräthyl- oder Hydroxyäthyl- reste und als Oxa-alkylreste z. B. 3-Oxapentyl-(5)- oder 3-Oxa-heptyl-(6)-reste in Betracht.
Cycloalkyl- oder Cycloalkylalkylreste sind bei spielsweise Cyclopentyl- oder Cyclohexylreste, oder Cyclopentyl- oder Cyclohexyl-methyl-, -äthyl- oder -propylreste. Als Aralkyl- und Arylreste kommen insbesondere Phenylalkylreste, wie 1- oder 2-Phenyläthyl- oder Phenylmethylreste, oder Phenylreste in Frage,
in de nen die aromatischen Kerne Substituenten tragen kön nen, wie niedere Alkylreste oder freie oder substi tuierte Hydroxy-, Amino- oder Mercaptogruppen, Halogenatome, Trifluormethyl- oder Nitrogruppen.
In den genannten substituierten Hydroxy-, Mercapto- oder Aminogruppen sind die Substituenten insbeson dere solche der oben genannten Art, vor allem nie dere Alkylreste, so dass es sich z.
B. um Methoxy-, Äthoxy-, Propoxy- oder Butoxygruppen, entspre chende Alkylmercaptogruppen, Alkylendioxygruppen, wie Methylendioxygruppen, Mono- oder Dialkyl- aminogruppen, wie Mono- oder Dimethyl-, -äthyl-, -propyl-, -butyl- oder -pentylaminogruppen handelt. Als Halogenatome sind vor allem Fluor-, Chlor- oder Bromatome zu nennen.
Die Aralkylreste können meh rere Arylreste enthalten, wie z. B. im Diphenylmethyl- rest.
Als heterocyclische Reste bzw. Heterocyclylalkyl- reste seien beispielsweise Pyridyl-, Thienyl-, Furyl-, Thenyl- oder Furfurylreste, die im heterocyclischen Rest wie oben für die Arylreste angegeben, substi tuiert sein können, genannt.
Besonders in Frage kommt als Rest R, ein niede rer verzweigter oder gerade, an beliebiger Stelle ver bundener Alkylrest, wie ein Methyl-, Äthyl-, Propyl-, Isopropyl-, Butyl-, Isobutyl-, Pentyl-(1)-, Pentyl-(2)-, Pentyl-(3)-, 2-Methyl-butyl-(3)- oder Hexylrest; ein Cyclo-alkylrest, wie ein Cyclopentyl- oder Cyclohexyl- rest;
ein Cycloalkyl-alkylrest, wie ein Cyclohexyl-me- thyl-, -äthyl- oder -propylrest; ein Oxaalkylrest, wie der 3-Oxa-pentyl- oder 5-Oxaheptyl-(2)-rest; ein Hy- droxy- oder Halogen-alkylrest, wie ein Hydroxy- oder Chloräthyl- oder -propyl-rest; oder besonders ein Amino- oder Ammonium-alkylrest, z.
B. ein solcher, dessen Amino- bzw. Ammoniumgruppe durch Koh- lenwasserstoffreste, insbesondere niedere Alkyl- oder Alkylenreste, mono- oder vorzugsweise di- bzw. tri- substituiert ist, wobei die Kohlenwasserstoffreste in der Kohlenstoffkette auch durch Sauerstoff-, Schwe fel- oder Stickstoffatome unterbrochen sein können.
Zu nennen sind vor allem Dimethylamino-, Diäthyl- amino-, Pyrrolidino-, Piperidino-, Morpholino-, Pi- perazino-, N-Methyl-piperazino-äthyl-,
-propyl- oder -butyl-reste. Als araliphatische oder heterocyclyl-ali- phatische Reste sind besonders die oben erwähnten Aralkyl- und Heterocyclylalkylreste zu nennen.
Die neuen Verbindungen und ihre Salze besitzen wertvolle pharmakologische Eigenschaften. Insbeson dere sind sie coronarerweiternd wirksam. Die neuen Verbindungen können somit als Heilmittel, vor allem bei Durchblutungsstörungen des Herzmuskels, aber auch als Zwischenprodukte zur Herstellung solcher Heilmittel dienen.
Besonders hervorzuheben als coronarerweiternde Mittel sind die Verbindungen der Formel

Das erfindungsgemässe Verfahren zur Herstel lung der neuen Verbindungen ist dadurch gekenn- zeichnet, dass man ein 1-R1-3-R3-4-Oxo-4,5-dihydro- 6-R,-pyrazolo[3,4-d]pyrimidin mit einem den Rest R.- abgebenden Mittel umsetzt, wobei R1, R3, R, und R, die oben gegebene Bedeutung haben.
Dies kann vor allem durch Behandlung mit reaktionsfähigen Estern von Alkoholen der Formel R.OH geschehen. Als reaktionsfähige Ester sind dabei solche starker an organischer oder organischer Säuren, z. B. Halogen- wasserstoffsäuren, Schwefelsäure oder organischer Sulfonsäuren, z. B. Arylsulfonsäuren, wie Toluolsul- fonsäuren, zu nennen, während als Alkohole beson ders niedere Alkanole, Alkoxyalkanole oder z.
B. wie oben angegeben substituierte Aminoulkanole aufzu führen sind. Die Reaktion kann in üblicher Weise, zweckmässig in Anwesenheit basischer Kondensa- tionsmittel, wie Alkalihydroxyden, -alkoholaten, -hy- driden oder -amiden durchgeführt werden. Anstelle der reaktionsfähigen Ester kann man gegebenenfalls auch den Rest R, einführende Verbindungen mit an lagerungsfähiger Doppelbindung oder kondensations fähige Epoxyde verwenden.
Erhaltene tertiäre Amine können in üblicher Weise, z. B. mit reaktionsfähigen Estern von Alkoho len, wie Alkanolen oder Phenylalkanolen quaternisiert werden.
Erhaltene Verbindungen von basischem Charak ter können Salze mit anorganischen oder organischen Säuren bilden. Als salzbildende Säuren kommen bei- spielsweise therapeutisch verwendbare in Frage:
Ha- logenwasserstoffsäuren, Schwefelsäuren, Phosphor säuren, Salpetersäure, Perchlorsäure; aliphatische, ali- cyclische, aromatische oder heterocyclische Carbon- oder Sulfonsäuren, wie Ameisen-, Essig-, Propion-, Oxal-, Bernstein-, Glykol-, Milch-, Äpfel-.
Wein-, Zitronen-, Ascorbin-, Hydroxymalein-, Dihydroxy- malein- oder Brenztraubensäure; Phenylessig , Ben- zoe-, p-Aminobenzoe-, Anthranil-, p-Hydroxyben- zoe-, Salicyl- oder p-Aminosalicylsäure;
Methansul- fon-, Äthansulfon-, Oxyäthansulfon-, Äthylensulfoii- säure; Toluolsulfo#n-, Naphthalinsulfonsäuren oder Sulfanilsäure; Methionin, Tryptophan, Lysin, Arginin, Cystein oder Glutaminsäure. Erhaltene Salze lassen sich in die freien Basen, freie Basen in ihre Salze überführen.
Die neuen, pharmakologisch wertvollen Verbin dungen, ihre Salze oder entsprechende Gemische kön nen z. B. in Form pharmazeutischer Präparate Ver wendung finden. Diese enthalten die genannten Ver bindungen in Mischung mit einem für die enterale oder parenterale Applikation geeigneten pharmazeu tischen organischen oder anorganischen Trägermate rial.
Sofern die beim erfindungsgemässen Verfahren verwendeten Ausgangsstoffe neu sind, lassen sie sich nach an sich bekannten oder hier beschriebenen Me thoden herstellen.
Als Ausgangsstoffe werden gemäss der vorliegen den Erfindung vorzugsweise diejenigen verwendet, die zu den eingangs als besonders wertvoll geschilderten Endstoffen führen. Die Ausgangsstoffe können auch gegebenenfalls in Form ihrer quaternären Ammo- niumverbindungen oder Salze verwendet werden. Die als Ausgangsstoffe verwendeten 1-R1-3-R3-4-Hydro- xy-6-RB pyrazolo[3,4-d]pyrimidine können z.
B. ge- mäss dem Verfahren des Schweizer Patentes. Nr. 398 626 durch Umsetzung von 2-R1-3-Amino-4-carb- äthoxy-5-R3-pyrazolen mit Nitrilen der Formel R,CN unter Verwendung von Natrium als Kondensations mittel erhalten werden.

In den nachfoulenden Beispielen wurde bei den ass Ausgangsstoffe verwendeten Pyrazolo,[3,4-d]pyrimidi- nen die Schreibweise mit den meisten Kerndoppel bindungen gewählt. Die Temperaturen sind in Cel siusgraden angegeben.
<I>Beispiel 1</I> Eine Lösung von 5,4 g 1-Iso#propyl-4-hydroxy-6- benzyl-pyrazolo,[3,4-d]pyrimidin in 30 cm3 1-n. Na tronlauge wird mit 1,95 cm3 Dimethylsulfat versetzt, und es wird während 1 Stunde bei Zimmertemperatur gerührt.
Man zieht hierauf die alkalische Lösung mit Äther aus und kristallisiert den Ätherrückstand aus Petroläther. 1-Isopropyl-5-methyl-6-b@enzyl-4-oxo-4,5- dihydro-pyrazolo[3,4-d]pyrimidin wird so, in farb losen Kristallen vom F. 96-97 erhalten.
Das Ausgangsmaterial wird z. B. wie folgt er halten: In 50 cm3 Benzylcyanid werden 2,3 g Natrium fein zerkleinert und 9,9 g 2-Isopropyl-3-amino-4- carbäthoxy-pyrazo,1 eingetragen. Man erhitzt dann während 4 Stunden unter Rühren auf 110-120 , ver setzt nach dein. Erkalten mit 100 cm, Alkohol und dampft im Vakuum zur Trockne ein. Der Rückstand wird in 150 cm3 2-n.
Natronlauge aufgenommen, die alkalische Lösung wird zur Abtrennung von Unge löstem mit Chloroform ausgeschüttelt und dann mit 6-n. Salzsäure auf pH 5-6 gestellt, wobei ein festes Produkt ausfällt. Letzteres wird aus wenig Alkohol umkristallisiert. Man erhält so 1-Isopropyl-4-hydroxy- 6-benzyl-pyrazolo,[3,4-d]pyrimidin der Formel

in farblosen Kristallen vom F. l65-166 .
<I>Beispiel 2</I> Zu 14,1 g 1-sek.-Butyl-4-hydroxy-6-benzyl-pyra- zolo[3,4-d]pyrimidin in 60 cm3 abs. Alkohol gibt man eine Lösung von 1,15 g Natrium in 40 cm3 abs. Al kohol und kocht nach Zusatz von 7,5g Diäthylamino- äthylchlorid während 4 Stunden am Rückfluss. Nach dem Abkühlen nutscht man die ausgefallenen Kri stalle ab, dampft das Filtrat ein,
versetzt den Rück stand mit 50 cm3 1-n. Salzsäure und filtriert von we nig Ungelöstem ab. Das Filtrat stellt man mit 2-n. Na tronlauge auf pH 10 und extrahiert mit Äther. Das aus dem Ätherrückstand erhaltene Öl wird in 30 cm3 abs. Alkohol gelöst und mit 28,4 cm3 1,49-n. alkoho- lischer Salzsäure versetzt.
Aus der eingedampften Reaktionslösung erhält man als Rückstand das 1-sek. Butyl-5-(ss-diäthylaminoäthyl)-6-benzyl-4-oxo-4,5-di- hydro-pyrazolo[3,4-d]pyrimidin in Form des Hydro chlorids vom F. 147-148 .
Das als Ausgangsstoff verwendete 1-sek.-Butyl-4- hydroxy-6-benzyl-pyrazolo[3,4-d]pyrimidin kann wie folgt erhalten werden: Zu 200 cm3 Benzylcyanid gibt man 13,8 g Na trium in kleinen Stückchen und anschliessend 63,2 g 2-sek: Butyl-3-amino-4-carbäthoxy-pyrazol. Man er hitzt in ca. 30 Minuten auf 110-120 und rührt dann bei dieser Temperatur noch während 5 Stunden wei ter. Nach dem Abkühlen versetzt man mit abs. Al kohol und engt im Vakuum ein.
Zum Rückstand gibt man verdünnte Natronlauge und extrahiert mit Chlo roform. Die wässrige alkalische Lösung wird mit Aktivkohle behandelt und filtriert. Das klare Filtrat stellt man mit 5-n.
Salzsäure auf pH 6, worauf sich das 1-sek.-Butyl-4-hydroxy-6-benzyl-pyrazolo[3,4-d]- pyrimidin der Formel

ausscheidet, das nach Umkristallisieren aus Alkohol bei 154-155 schmilzt.
<I>Beispiel 3</I> In eine Natriumäthylat-Lösung, hergestellt aus 1,2 g Natrium und 300 cm3 wasserfreiem Alkohol, werden 13,4 g 1-Isopropyl-4-hydroxy-6-benzyl-pyra- zolo[3,4-d]pyrimidin eingetragen. Zur Bildung des Natriumsalzes wird während 1 Stunde bei Zimmer- temperatur gerührt. Man versetzt mit 5,5 g ss-Dime- thylamino-äthylchlorid, erhitzt während 4 Stunden zum Sieden, dampft dann im Vakuum zur Trockne ein, löst den Rückstand in 100 cm3 1-n.
Salzsäure, stellt mit Natronlauge auf pH 10 und nimmt das aus gefallene Öl in Äther auf. Der Ätherrückstand wird aus Petroläther umkristallisiert. 1-Isopropyl-5-(ss-di methyla,mnoäthyl)-6-benzyl-4-oxo-4,5-dihydro-pyra- zolo[3,4-d]pYrimidin wird so in farblosen Kristallen vom F.115-117 erhalten. Das Hydrochlorid schmilzt von 229-231 .
<I>Beispiel 4</I> In eine Natriumäthylat-Lösung, hergestellt aus 1,2 g Natrium und 300 cm3 wasserfreiem Alkohol, werden 13,4 g 1-Iso@propyl-4-hydroxy-6-benzyl-pyra- zolo[3,4-d]pyrimidin eingetragen.
Zur Bildung des Natriumsalzes wird während 1 Stunde bei Zimmer temperatur gerührt. Man versetzt mit 7 g ss-Diäthyl- aminoäthylchlorid, erhitzt während 4 Stunden zum Sieden, dampft dann im Vakuum zur Trockne ein, löst den Rückstand in 100 cm3 1-n. Salzsäure, stellt mit Natronlauge auf pH 10 und nimmmt das aus gefallene Öl in Äther auf. Der Ätherrückstand wird mit alkoholischer Salzsäure versetzt.
Letzteres wird eingedampft und der Rückstand wird aus Essigester umkristallisiert. Man erhält so das Hydrochlorid des 1-Isopropyl-5-(ss-diäthylaminoäthyl)-6-benzyl-4-oxo- 4,5-dihydro-pyrazolo[3,4-d]pyrin-idin in farblosen Kristallen vom F. 202-203 .
<I>Beispiel 5</I> In eine Natriumäthylat-Lösung, hergestellt aus 1,2 g Natrium und 300 cm' wasserfreiem Alkohol, werden 13,4 g 1-Isopropyl-4-hydroxy-6-benzyl-pyra- zolo[3,4-d]pyrimidin eingetragen. Zur Bildung des Natriumsalzes wird während einer Stunde bei Zim mertemperatur gerührt. Man versetzt dann mit 8 g y-Diäthylamino-propylchlorid, erhitzt während 4 Stun den zum Sieden, dampft dann im Vakuum zur Trockne ein,
löst den Rückstand in 100 cm3 1-n. Salzsäure, stellt mit Natronlauge auf pH 10 und nimmt das ausgefallene Öl in Äther auf. Der Äther rückstand wird aus Petroläther umkristallisiert. 1-Iso propyl-5-(y-diäthylarninopropyl)-6-benzyl-4-oxo-4,5- dihydro-pyrazolo[3,4-d]pyrimidin wird so in farb losen Kristallen vom F.<B>70-71'</B> erhalten. Das Hydro- chlorid schmilzt von 173-175 .
<I>Beispiel 6</I> Zu 12 g 1-Methyl-4-hydroxy-6-benzyl-pyrazolo- [3,4-d]pyrimidin in 75 cm3 Alkohol werden 1,2 g Na trium in 25 cm' Äthanol gegeben. Man kocht eine Stunde unter Rückfluss und gibt hierauf 6 g ss-Di- äthylamino-äthylchlorid zu, worauf weitere 3 Stun den zum Sieden erwärmt wird. Das ausgeschiedene Natriumchlorid wird abfiltriert, das Filtrat dampft man ein und kristallisiert den Rückstand aus Cyclo- hexan um.
Man erhält das 1-Methyl-5-(ss-Diäthyl amin6äthyl)-6-benzyl-4-oxo-4,5-dihydro-pyrazolo[3,4- d]pyrimidin in Kristallen vom F. 83-85 . Das Hydro chlorid schmilzt bei 219 .
Das als Ausgangsstoff verwendete 1-Methyl-4-hy- droxy-6-benzyl-pyrazolo[3,4-d]pyrimidin wird z. B. wie folgt erhalten: Zu einer Lösung von 8,5 g 2-Methyl-3-amino-4.- carbäthoxy-pyrazol in 50 cms Benzylcyanid gibt man 2,
3 g Natrium in kleinen Stücken und erwärmt an- schliessend unter Rühren auf 110-120 . Nach 4 Stunden wird das Reaktionsgemisch abgekühlt und niit 100 cm3 Äthanol versetzt. Die Lösung wird im Vakuum zur Trockene eingedampft. Zum Rückstand gibt man 150 cm3 2-n.
Natronlauge und extrahiert das überschüssige Benzylcyanid mit Chloroform. Die wäsr serige Phase wird durch Zugabe von 5-n.
Salzsäure auf pH 5-6 gestellt, wobei ein fester Niederschlag aus fällt, den man abfiltriert und mehrmals aus Äthanol umkristallisiert. Man erhält das 1-Methyl-4-hydroxy- 6-benzyl-pyrazo@lo[3,4-d]pyrimidin der Formel

Das ausgeschiedene Natriumchlo- rid wird abfiltriert und das Filtrat wird im Vakuum zur Trockne eingedampft. Den festen Rückstand kri stallisiert man aus Cyclohexan-Petroläther um und erhält so das 1-Phenyl-5-(ss-diäthylaminoäthyl)-6-ben- zyl-4-oxo-4,5-dihydro-pyrazolo[3,4-d]pyrimidin in Kristallen vom F. 103-105 . Das Hydrochlorid schmilzt bei 225 .
Das als Ausgangsstoff verwendete 1-Phenyl-6- benzyl-4-hydroxy-pyrazolo[3,4-d]pyrimidin wird z. B. wie folgt erhalten: In eine Mischung von 12 g 2-Phenyl-3-amino-4- carbäthoxy-pyrazol und 50 g Benzylcyanid gibt man 2,3 g Natrium in kleinen Stücken und erwärmt unter Rühren 4 Stunden auf 140 .
Nach dem Abkühlen versetzt man das Reaktionsgemisch mit 100 cm' Al kohol und gibt 2-n. Salzsäure bis pH 5-6 zu. Es fällt ein Niederschlag aus, den man aus Chloroform-Petrol- äther umkristallisiert. Man erhält so das 1-Phenyl-4- hydroxy-6-benzyl-pyrazolo[3,4-d]pyrimidin der For mel

Das als Ausgangsstoff verwendete 1,6 Di-isopro- pyl-4-hydroxy-pyrazolo,[3,4-d]pyrimidin wird z. B. wie folgt erhalten: In 160 cm3 Isobutyromtril werden 39,6 g 2-Iso- propyl-3-amino.4-carbäthoxy-pyrazol und 9,2 g Na trium fein zerkleinert eingetragen. Man erhitzt inner halb einer Stunde auf 110 und hält während 4 Stun den unter Rühren diese Temperatur.
Nach dem Er- kalten versetzt man mit 15 ctns Äthanol, dampft im Vakuum zur Trockne ein, nimmt den Rückstand in 100 cms 2-n. Natronlauge auf und schüttelt die al- kalische Lösung mit Chloroform aus. Die wässrige Phase wird durch Zugabe von 5-n.
Salzsäure auf pH 5-6 gestellt, wobei ein festes Produkt ausfällt. Letz teres wird mehrmals aus Äthanol umkristallisiert. 1- Isopropyl-4-hydroxy-6 isopropyl-pyrazolo[3, 4-d]pyri- miäin der Formel

Der Ätherrückstand wird destilliert. 1,6-Di-isopropyl-5- (ss-diäthylamin6äthyl)-4-oxo-4,5 -dihydro-pyrazolo- [3,4-d]pyrimidin geht bei 138-140 bei einem Druck von 0,05 mm Hg über.
<I>Beispiel 10</I> In eine Natriumäthylat-Lösung, hergestellt aus 0,35 g Natrium und<B>150</B> cm3 Äthanol, werden 5,2 g 1-Isopropyl-4-hydroxy-6-diphenylmethyl-pyrazolo- [3,4-d]pyrimidin eingetragen.
Zur Bildung des Na- triumsalzes wird während einer Stunde bei Zimmer- temperatur gerührt. Man versetzt mit 2,1g ss-Diäthyl- amino-äthylchlorid, erhitzt während 4 Stunden zum Sieden, dampft im Vakuum zur Trockne ein und kri- stallisiert den Rückstand aus Petroläther. 1-Isopropyl- 6-diphenylmethyl-5-(ss-diäthylaminoäthyl)-4-oxo-4,
5- dihydro-pyrazolo[3,4-d]pyrimidin wird so in farblosen Kristallen vom F. 124-125 erhalten.
Das als Ausgangsstoff verwendete 1-Isopro@pyl-4- hydroxy-6-diphenylmethyl-pyrazolo[3, 4-d]pyrimidin wird z. B. wie folgt erhalten: 20 g Diphenylacetonitril und 19,7 g 2-Isopropyl- 3-amino-4-carbäthoxy-pyrazol werden auf 70 er- wärmt und mit 2,3g Natrium in kleinen Stücken ver setzt.
Man erhitzt dann, während 4 Stunden auf 110- 120 , lässt erkalten, zerstört das überschüssige Na trium mit Äthanol und dampft im Vakuum zur Trockne ein. Der Rückstand wird mit 300 cm3 Was ser versetzt und es wird mit 2-n. Salzsäure auf pH 3 gestellt, worauf sich ein fester Niederschlag ausschei det.
Von letzterem putscht man ab, kocht diesen mit viel Petroläther zur Abtrennung von noch vorhande nem Ausgangsmaterial aus und kristallisiert den in Petroläther nicht gelösten Anteil aus Äthanol um, 1-Isopropyl-4-hydroxy-6-diphenylmethyl-pyrazolo- [3,4-d]pyriniidin der Formel

This can be done primarily by treatment with reactive esters of alcohols of the formula R.OH. As reactive esters are those strong in organic or organic acids, eg. B. hydrohalic acids, sulfuric acid or organic sulfonic acids, z. B. arylsulfonic acids, such as toluenesulfonic acids, while the alcohols FITS lower alkanols, alkoxyalkanols or z.

To form the sodium salt, the mixture is stirred at room temperature for 1 hour. 5.5 g of β-dimethylamino-ethyl chloride are added, the mixture is heated to boiling for 4 hours, then evaporated to dryness in vacuo, the residue is dissolved in 100 cm3 of 1N.

The ether residue is recrystallized from petroleum ether. 1-Isopropyl-5- (ss-di methyla, mnoethyl) -6-benzyl-4-oxo-4,5-dihydro-pyrazolo [3,4-d] pYrimidine is thus converted into colorless crystals of F.115- 117 received. The hydrochloride melts from 229-231.
<I> Example 4 </I> In a sodium ethylate solution, prepared from 1.2 g of sodium and 300 cm3 of anhydrous alcohol, 13.4 g of 1-iso @ propyl-4-hydroxy-6-benzyl-pyrazolo are added [3,4-d] pyrimidine added.
To form the sodium salt, the mixture is stirred at room temperature for 1 hour. 7 g of SS-diethylaminoethyl chloride are added, the mixture is heated to boiling for 4 hours, then evaporated to dryness in vacuo, the residue is dissolved in 100 cm3 of 1N. The latter is evaporated and the residue is recrystallized from ethyl acetate. The hydrochloride of 1-isopropyl-5- (ss-diethylaminoethyl) -6-benzyl-4-oxo-4,5-dihydro-pyrazolo [3,4-d] pyrynidine is obtained in colorless crystals with a melting point of 202 -203.
<I> Example 5 </I> In a sodium ethylate solution, prepared from 1.2 g of sodium and 300 cm 'of anhydrous alcohol, 13.4 g of 1-isopropyl-4-hydroxy-6-benzyl-pyrazolo [ 3,4-d] pyrimidine entered. To form the sodium salt, the mixture is stirred at room temperature for one hour. 8 g of y-diethylamino-propyl chloride are then added, the mixture is heated to boiling for 4 hours, then evaporated to dryness in vacuo,
dissolves the residue in 100 cm3 1-n. Hydrochloric acid, adjusts to pH 10 with sodium hydroxide solution and absorbs the precipitated oil in ether. The ether residue is recrystallized from petroleum ether. 1-Iso propyl-5- (y-diethylarninopropyl) -6-benzyl-4-oxo-4,5-dihydro-pyrazolo [3,4-d] pyrimidine is thus formed in colorless crystals of F. <B> 70- 71 'received. The hydrochloride melts from 173-175.
<I> Example 6 </I> 1.2 g of sodium in 25 cm of ethanol are added to 12 g of 1-methyl-4-hydroxy-6-benzyl-pyrazolo- [3,4-d] pyrimidine in 75 cm 3 of alcohol given. The mixture is refluxed for one hour and then 6 g of β-diethylamino-ethyl chloride are added, whereupon the mixture is heated to boiling for a further 3 hours. The precipitated sodium chloride is filtered off, the filtrate is evaporated and the residue is recrystallized from cyclohexane.
The 1-methyl-5- (ß-diethylamine-6ethyl) -6-benzyl-4-oxo-4,5-dihydro-pyrazolo [3,4-d] pyrimidine is obtained in crystals with a melting point of 83-85. The hydrochloride melts at 219.
The 1-methyl-4-hydroxy-6-benzyl-pyrazolo [3,4-d] pyrimidine used as starting material is z. B. obtained as follows: To a solution of 8.5 g of 2-methyl-3-amino-4.- carbethoxy-pyrazole in 50 cms of benzyl cyanide is added 2,
3 g sodium in small pieces and then heated to 110-120 while stirring. After 4 hours, the reaction mixture is cooled and 100 cm3 of ethanol are added. The solution is evaporated to dryness in vacuo. 150 cm3 of 2-n are added to the residue.
Sodium hydroxide solution and extracted the excess benzyl cyanide with chloroform. The aqueous phase is by adding 5-n.
Hydrochloric acid adjusted to pH 5-6, a solid precipitate falling out, which is filtered off and recrystallized several times from ethanol. The 1-methyl-4-hydroxy-6-benzyl-pyrazo @ lo [3,4-d] pyrimidine of the formula is obtained

<I> Example 7 </I> A solution of 800 mg of sodium in 10 cm3 of ethanol is added to a suspension of 10 g of 1-phenyl-6-benzyl-4-hydroxypyrazolo [3,4-d] pyrimidine in Entered 100 cm3 of ethanol and refluxed for 30 minutes. Then 5 g of diethylaminoethyl chloride in 10 cm3 of alcohol are added and the mixture is boiled for a further 3 hours.
The precipitated sodium chloride is filtered off and the filtrate is evaporated to dryness in vacuo. The solid residue is crystallized from cyclohexane-petroleum ether and 1-phenyl-5- (s-diethylaminoethyl) -6-benzyl-4-oxo-4,5-dihydro-pyrazolo [3,4-d ] pyrimidine in crystals of F. 103-105. The hydrochloride melts at 225.
The 1-phenyl-6-benzyl-4-hydroxy-pyrazolo [3,4-d] pyrimidine used as starting material is z. B. obtained as follows: 2.3 g of sodium are added in small pieces to a mixture of 12 g of 2-phenyl-3-amino-4-carbethoxy-pyrazole and 50 g of benzyl cyanide and heated to 140 for 4 hours while stirring.
After cooling, the reaction mixture is treated with 100 cm of alcohol and 2-n is added. Hydrochloric acid to pH 5-6. A precipitate separates out and is recrystallized from chloroform-petroleum ether. This gives 1-phenyl-4-hydroxy-6-benzyl-pyrazolo [3,4-d] pyrimidine of the formula

<I> Example 8 </I> A solution of 11 g 1,6-diisopropyl-4-hydroxy-pyrazolo [3,4-d] pyrimi.din in 75 cms 2-n. 6.3 g of dimethyl sulfate are added to sodium hydroxide solution, and the mixture is stirred at room temperature for 2 hours and left to stand overnight.
The precipitate is then wiped off and crystallized from petroleum ether. 1,6-Di-isopropyl-5-methyl-4-oxo-4,5-dihydro-pyrazolo [3,4-d] pyrimidine is obtained in this way in colorless crystals with a melting point of 75-77.
The 1,6 di-isopropyl-4-hydroxypyrazolo, [3,4-d] pyrimidine used as starting material is z. B. obtained as follows: 39.6 g of 2-isopropyl-3-amino.4-carbethoxy-pyrazole and 9.2 g of sodium are finely comminuted in 160 cm3 of isobutyromtrile. The mixture is heated to 110 within an hour and maintained at this temperature for 4 hours with stirring.
After cooling, 15 ctns of ethanol are added, the mixture is evaporated to dryness in vacuo, and the residue is taken up in 100 cms of 2N. Sodium hydroxide solution and shakes out the alkaline solution with chloroform. The aqueous phase is by adding 5-n.
Hydrochloric acid adjusted to pH 5-6, whereby a solid product precipitates. The latter is recrystallized several times from ethanol. 1- isopropyl-4-hydroxy-6 isopropyl-pyrazolo [3, 4-d] pyramid of the formula

<I> Example 9 </I> 10 g of 1,6-diisopropyl-4-hydroxypyrazolo [3,4-d] pyrimidine are added to a sodium ethylate solution prepared from 1.05 g of sodium and 150 cm3 of ethanol registered. To form the sodium salt, the mixture is stirred for 1 hour at room temperature. It is mixed with 6.5 g of SS-diethylaminoethyl chloride, heated to boiling for 4 hours,
then evaporates to dryness in vacuo, dissolves the residue in 100 cm3 of 1-n. Hydrochloric acid, adjusts to pH 10 with sodium hydroxide solution and absorbs the precipitated oil in ether. The ether residue is distilled. 1,6-Di-isopropyl-5- (ss-diethylamin6ethyl) -4-oxo-4,5-dihydro-pyrazolo- [3,4-d] pyrimidine goes at 138-140 at a pressure of 0.05 mm Hg about.
<I> Example 10 </I> 5.2 g of 1-isopropyl-4-hydroxy-6-diphenylmethyl are added to a sodium ethylate solution prepared from 0.35 g of sodium and <B> 150 </B> cm3 of ethanol -pyrazolo- [3,4-d] pyrimidine added.
To form the sodium salt, the mixture is stirred for one hour at room temperature. 2.1 g of SS-diethylamino-ethyl chloride are added, the mixture is heated to boiling for 4 hours, evaporated to dryness in vacuo and the residue is crystallized from petroleum ether. 1-isopropyl-6-diphenylmethyl-5- (ss-diethylaminoethyl) -4-oxo-4,
5-dihydro-pyrazolo [3,4-d] pyrimidine is obtained in this way in colorless crystals with a melting point of 124-125.
The 1-Isopro @ pyl-4-hydroxy-6-diphenylmethyl-pyrazolo [3, 4-d] pyrimidine used as starting material is z. B. obtained as follows: 20 g of diphenylacetonitrile and 19.7 g of 2-isopropyl-3-amino-4-carbethoxy-pyrazole are heated to 70 and small pieces of 2.3 g of sodium are added.
The mixture is then heated to 110-120 for 4 hours, allowed to cool, the excess sodium is destroyed with ethanol and evaporated to dryness in vacuo. 300 cm3 of water are added to the residue and 2-n. Hydrochloric acid adjusted to pH 3, whereupon a solid precipitate ausschei det.
The latter is popped off, boiled with a lot of petroleum ether to separate any starting material that is still present and the portion not dissolved in petroleum ether is recrystallized from ethanol, 1-isopropyl-4-hydroxy-6-diphenylmethyl-pyrazolo- [3,4- d] pyriniidin of the formula
