in which R1, R4 and R5 have the meaning given above and X is halogen or an alkoxy or alkylthio group containing at most 3 carbon atoms, with ammonia or
a primary or secondary amine of the formula HNR.R3, for example diethylamine, piperidine, morpholine, N-methyl piperazine, alkylated alkylenediamines and the like. To form quaternary ammonium derivatives, the reaction product can subsequently be treated with a quaternizing agent.
The reaction of the imide halide, imido ether or imido thioether with the amine is conveniently carried out by adding the components, depending on their physical properties, possibly using an inert diluent such as dioxane, xylene, mesitylene, decalin and the like , heated for 1 to 3 days to a temperature preferably above 150 C,
be it by reflux or using the containment tube. The amine is preferably used in at least a three-fold molar excess. The reaction is often catalyzed by acid, for which purpose a few drops of glacial acetic acid (e.g. 5 drops per 10 g of the diazepine compound) can be added to the reaction mixture.
After concentrating the reaction mixture ver, the residue is conveniently divided between ether and water and the base formed is removed e.g. B. by extraction with dilute hydrochloric acid or acetic acid.
The base can be precipitated with ammonia from the hydrochloride or acetate solution clarified with charcoal if necessary and, if it crystallizes well and is sparingly soluble in ether, it can be isolated directly by filtration, otherwise. take up in ether and work up the ethereal solution in the usual way by washing with water and drying with sodium sulfate.
Further purification can be carried out by recrystallization or high vacuum distillation.
It goes without saying that the basic side chain, if R2 and R3 are not simultaneously hydrogen, can also be introduced gradually by first reacting the diazepine compound of the formula II with ammonia or a primary amine and subsequently introducing R2 and / or R3,
possibly with simultaneous quaternization. If necessary, further N atoms of the basic side chain can also be alkylated subsequently.
If a starting material with R1 = hydrogen was assumed, an alkyl group can optionally also be introduced subsequently in the 5-position.
Any quaternization of the reaction product takes place, for example, by treatment with one or two molar equivalents of a dialkyl sulfate, alkyl halide or sulfonic acid alkyl ester.
The bases obtained are yellow, in many cases crystallizable, otherwise they can be distilled undecomposed in a high vacuum, and due to the amidine grouping, apart from any other basic nitrogen atoms, they have sufficient base strength to react with inorganic or organic acids, for example hydrochloric acid, Hydrobromic acid, sulfuric acid, nitric acid, phosphoric acid, acetic acid, oxalic acid, malonic acid,
Succinic acid, maleic acid, malic acid, tartaric acid, toluenesulfonic acid and the like to form stable salts in water.
The imido-thioethers are preferred as starting materials of the formula II because they are the easiest to access. They are obtained, for example, by thermal cyclization of appropriately substituted o-nitro-.diphenylamino-o'-carboxylic acids to the lactam, the tautomeric form of which corresponds to the formula 11, in which X is a hydroxyl group.
By treating the lactam with phosphorus pentasulfide in boiling pyridine, the yellow thiolactam (formula II; X = SH; tautomeric form), which is soluble in alkali, is obtained, and from it the imido-thiolactam (formula I; X =) by alkylation with alkali and dialkyl sulfate S-alkyl), both stages in good yield.
The bases obtained in the manner according to the invention, their quaternary ammonium derivatives and acid addition salts are new compounds which can be used as active ingredients in medicaments, in particular as analgesics, chemotherapeutic agents, antihistamines, antiallergics, sedatives,
Adrenolytics and neuroplegics. Some of them are suitable for the treatment of psychotic states.
For example, the 8-chloro-1 1- (N-methyl) piperazino-SH-dibenzo: [b, e], [1,4]: diazepine obtained according to Example 27 shows in animal experiments the properties of a neuroplegic with strong analgesic properties , parasympathicolytic and sympathicolytic effects.
The analgesic effect is determined by measuring the pain threshold during electrical stimulation of the tooth pulp in rabbits. It is compared to the effect of known analgesics in Table I below.

Example 1 3.4 g of 11-methylthio-5H-dibenzo [b, e] [l, 4] diazepine are dissolved in 40 ml of absolute xylene, 4.3 g of N-methylpiperazine are added and afterwards Add 2 to 3 drops of glacial acetic acid and reflux for 5 hours. The reaction mixture is mixed with water and made alkaline with concentrated sodium hydroxide solution. The xylene layer is separated off, washed neutral with water and extracted with hydrochloric acid.
The acidic extracts are made alkaline with concentrated ammonia solution and the precipitated oil is etherified. The ethereal solution is washed with water and evaporated. The residue is crystallized from acetone / petroleum ether. 3.6 g of 11- (N-methyl) piperazino-5H dibenzo- [b, e] [1,4] diazepine with a melting point of 184-185 ° C. are obtained in a yield of 87% of theory.
Example 2 A solution of 2.1 g of 5-methyl-11- (N-methyl) -piperazino-5H-dibenzo [b, e] [1,4] diazepine, prepared according to the later Example 15, in 10 ml of benzene, is mixed with a solution of 0.87 g (equimolar amount) of neutral dimethyl sulfate in 30 ml of benzene, a quaternary ammonium salt immediately precipitating in crystalline form with heating. After leaving it to stand overnight, it is sucked off and washed with ether.
The yield is almost quantitative, so it is the monoquaternary salt. It is hygroscopic and cannot be recrystallized. The fact that the UV-absorption spectrum. is identical to that of the starting material, speaks for the fact that the basic nitrogen atom of the N-methylpiperazine residue, which already carries a methyl group, has been quaternized.
Example 3 1.5 g of 11- (N - methyl) piperazino - 5H - dibenzo- [b, e] [1,4] -diazepine, prepared according to Example 1, who with 10 ml of methanol and 4.0 ml (6.9 g) methyl bromide in the containment tube heated to 100 ° C. for 15 hours.
The reaction product taken up in more methanol is, after filtering off a small amount of insoluble precipitate, evaporated to dryness. Recrystallization from isopropanol / ethyl acetate gives 1.93 g of a very strongly hygroscopic yellow salt with a decomposition point of 198-200 C.
It is a bisquaternary salt with the formula:
EMI0003.0044
In a manner analogous to that in Example 1 described above, the products mentioned in Table II below are obtained from the corresponding starting materials. In the last column, Ae means ether, Pe means petroleum ether and Ae means acetone.
