Procédé de préparation de composés pharmacologiquement actifs
La présente invention a pour objet un procédé de préparation de composés pharmacologiquement actifs, et plus particulièrment de nouvrelles pénicillines semi-synthétiques de formule:

dans laquelle X représente un groupe alcoylène, et de leurs sels non toxiques, par exemple de sodium, de potassium, d'aluminium, d'ammonium et d'ammonium substitué obtenus par réaction avec les amines employées couramment dans le domaine des pénicillines, telles que la prochaine, la dibenzylamine et
Ia N,N'-dibenzyl-éthylènediamine.
Conformément au procédé selon l'invention, on fait réagir de l'acide 6-amino-pénicillanique, de formule:

avec un acide alpha-phényl-#-carbobenzoyaminé en présenoe d'une amine tertiaire, par exemple la triéthylamine, et on scinde le groupe protecteur par hydrogénation catalytique. Les composés possèdent un atome de carbone asymétrique et les diastéréoiso- mères correspondants peuvent être préparés de la manière usuelle.
Par exenîple, les formes optique ment actives de l'a beta-amino-alpha-phényl-éthyl- pénicilline sont préparées à partir des acides alphaphényl-bêta-carbobenzoxyamino-propioniques opti quement actifs, eux-mêmes obtenus à partir du mé lange racémique par l'intermédiaire de leurs ses avec des bases optiquement actives, comme la quinine, la cinchonine, la sïtrychnine et la morphine.
On donne dans le tableau ci-dessous les concentrations inhibitrices minimum in vitro, en v/ml, de quelques-unes des nouvelles pénicillines contre un certain nombre de micro-organismes pathogènes.

Les nouvelles pénicillines résistent très bien aux acides et à la pénicillinase. Le tableau ci-dessous montre le comportement de quelques-unes de ces nouvelles pénicillines dans l'acide chlorhydrique (PH 1), en comparaison avec la phénoxyéthyl pénicilline, qui est réputée être l'une des pénicillines les moins sensibles aux milieux acides. Les pourcen tages de pénicilline non dégradée sont indiqués pour différents temps.

Exemple 1
A 82, 5 g d'acide bêta-am1no-alpha-phényl-pro- pionique dans 200 mi de NaOH à 10 /o, on ajoute goutte à goutte 98 g de chlorocarbonate de benzyle à 150 C, en agitant. On continue à agiter à la température ordinaire pendant une heure et demie.

On extrait à l'éther le précipité huileux, on lave les extraits à l'eau, on les sèche et on les évapore, obtenant ainsi 140 g d'acide N-carbobenzoxybêta-amino-alpha-phényl-propionique; p.f. 124-7 C.
A 155 g de quinine dans 700 ml d'alcool éthylique à 900/o, on ajoute une solution chaude de 140 g d'acide N-carbobenzoxy-bêta-amino-alphaphényl-propionique dans 500 mi d'alcool éthylique.
On refroidit le mélange et on filtre le précipité. Quantité: 13 g; p.f. 176-7 C; [alpha]D=-125 (0,2% dans l'alcool éthylique). On suspend le sel de quinine dans Il d'eau, on acidule le mélange avec du HCl 10% et on l'extrait à l'éther. On réunit les extraits, on les lave à l'eau, on les sèche sur du Na2SO4 et on les évapore. On obtient 55 g d'acide (-)-N-carbo- benzoy-bêta-amino-alpha-phényl-propionique ; p.f.
99-100 C; [alpha]D25=-93 (1% dans l'alcool éthylique).
A un mélange refroidi à 0 C de 52g d'acide (-)-N-carbobenzoxy-bêta-amino-alpha-phényl-propionique, 1300 mi d'acétone anhydre et 27 mi de tri- éthylamine anhydre, on ajoute goutte à goutte, en agitant, 18,5 ml de chlorocarbonate d'éthyle, après quoi on continue à agiter pendant 10 min puis on refroidit le mélange à -50 C et on lui ajoute progressivement 37,5 g d'acide 6-amino-péniclllanique dans 1000 ml de solution de bicarbonate de sodium à 4%, en maintenant la température en dessous de
Os C. On continue à agiter pendant 30 min à la température ordinaire.
On extrait le mélange à l'éther, puis on l'amène au pH 2 avec du HOl dilué et on l'extrait à nouveau à l'éther. On lave les extraits éthérés à l'eau et on les amène au pH 6,2 avec du NaHCO3 40/o. On ajoute la solution du sel de sodium de la (-)-N-oarbo- benzoxy-bêta-amino-alpha-phényl-éthylpénicilline à une suspension aqueuse de Pd/CaCO8 préalablement saturée d'hydrogène, et on secoue le mélange pendant une heure. On filtre le mélange, on l'amène au pH 2, on l'extrait à l'éther, on amène la couche aqueuse au pH 4,6 et on la concentre à un petit volume, sous vide et sous congélation.
Quantité: 40 g de (-)-bêta-amino-alpha-phényl-éthylpénicilline ; p. f. 225-300 C (déc.), [alpha]D20 = -2030.

In accordance with the process according to the invention, 6-amino-penicillanic acid, of formula:

with alpha-phenyl - # - carbobenzoyamine acid in the presence of a tertiary amine, for example triethylamine, and the protecting group is cleaved by catalytic hydrogenation. The compounds have an asymmetric carbon atom and the corresponding diastereoisomers can be prepared in the usual manner.
For example, the optically active forms of α-beta-amino-alpha-phenyl-ethyl-penicillin are prepared from the optically active alpha-phenyl-beta-carbobenzoxyamino-propionic acids, themselves obtained from the racemic mixture. through their ses with optically active bases, such as quinine, cinchonine, strychnine and morphine.
The table below gives the minimum inhibitory concentrations in vitro, in v / ml, of some of the new penicillins against a number of pathogenic microorganisms.

The new penicillins are very resistant to acids and penicillinase.
