Verfahren zur Gewinnung von Kryptoäscin aus Rosskastanienextrakt
Die Erfindung bezieht sich auf ein Verfahren zur Gewinnung von Kryptoäscin aus Rosskastanienextrakt, welches dadurch gekennzeichnet ist, dass aus Rosskastanienextrakt durch Einwirkung von Chole sterin die hämolytisch wirkenden Saponine gefällt werden und dass aus dem Filtrat durch Ansäuern auf einen pH-Wert von 2 oder darunter das Kryptoäscin ausgefällt wird. Kryptoäscin ist ein neues Saponin aus dem Extrakt von Teilen, insbesondere Samen, der Rosskastanie - und zwar ein Saponin, das in einer seiner beiden Formen von einer hämolytischen Wirkung völlig frei und in der anderen seiner Formen von bei der Injektion seiner Lösung in die Blutbahn schädlichen hämolytischen Wirkungen frei ist.
Die Erfinder haben diesem neuentdeckten wasserlöslichen Saponin die Bezeichnung Kryptoäscin > gegeben, wobei Kryptoäscin A die nichthämolytische und Kryptoäscin B, in das das Kryptoäscin A übergeht, eine hämolytische Form darstellt.
Während lange Zeit die Meinung herrschte, dass die therapeutische Wirkung des per os eingenommenen Rosskastanienextraktes zurückzuführen sei auf den Gehalt dieser Extrakte an Saponin, und in neuerer Zeit festgestellt wurde, dass beim Entfernen des beim Einspritzen in die Blutbahn hämolytisch wirkenden, die Injizierung des Extraktes also verhindernden Saponins, z.
B. durch Behandeln mit Cholesterin und Abtrennen des Feststoffes, ein Restextrakt erhalten wird, der nicht nur die wertvollen therapeutischen Eigenschaften des ursprünglichen - saponinhaltigen Extraktes aufweist, sondern bei dem Injizieren zusätzliche wertvolle Wirkungen hervorruft, haben die Erfinder festgestellt, dass in dem wässerig-alkoholischen oder wässerigen Extrakt von Teilen, insbesondere Samen, der Rosskastanie nach Ausfällung des hämolytisch wirkenden Saponin-Anteils, insbesondere durch Behandeln des Extraktes mit Cholesterin und Abfiltrieren des Niederschlags, noch ein Saponinkörper vorliegt, der durch das Behandeln mit Cholesterin nicht entfernt wurde und der sich isolieren lässt durch Behandeln mit Säure und gegebenenfalls mit organischein, mit Wasser nicht bzw.
nur teilweise misch- baren aliphatischen Alkoholen oder Ketonen oder halogenierten Kohlenwasserstoffen, insbesondere in Mischung mit solchen Alkoholen oder Ketonen.
Die anderen Inhaltsstoffe des Rosskastanienextraktes, z. B. Flavone, bleiben nach Abscheiden dieses Saponins, z. B. in Lösung in dem organischen Lösungsmittel, nun frei von allen saponinhaltigen Körpern zurück, während das Kryptoäscin für sich frei von anderen Saponinen gewonnen wird.
Bei Injizieren einer Lösung dieses Kryptoäscins treten keine schädlichen hämolytischen Wirkungen auf; das Kryptoäscin wirkt in starkem Masse auf das Gefäss-System und weist Diuresewirkung und Antihistaminwirkung auf.
Dieses einheitliche wasserlösliche Saponin liegt in einer unhämolytischen A-Form vor und in einer eine leichte, jedoch unschädliche Hämolyse bewirkenden B-Form. Der Übergang von der A-Form in die - isomere - B-Form geht in wässriger Lösung vor sich; er wird mit steigender Temperatur und sinkendem pH Wert beschleunigt; bei Raumtemperatur und pH 6-7 bleibt die A-Form auch in wässriger Lösung über einen längeren Zeitraum stabil.
Das Kryptoäscin A - dem wahrscheinlich die im Aglukon des Äscin-Moleküls vorhandene Doppelbindung zwischen C12 und C13 fehlt - unterscheidet sich von dem bisher in dem Rosskastanienextrakt aufgefundenen Saponin Äscin durch einen Mehrgehalt an einem Mol Arabinose.
Die Verbindung enthält neben Aglukon, a-Methyl ss-hydroxy-buttersäure, Essigsäure, Arabinose, Xylose,
Glukose, Glukoronsäure und Wasser (Co9H94027. H20).
Der Schmelzpunkt von Kryptoäscin A liegt bei 218-220 . Das Molekulargewicht beträgt 1253. Das ohne Änderung des Molekulargewichtes aus dem Kryptoäscin A durch Isomerisierung in wässriger Lösung beim Erwärmen entstehende Kryptoäscin B zeigt einen Schmelzpunkt von 220-2210.
Die Kryptoäscinlösung kann als solche für therapeutische Zwecke verwendet werden, oder aber sie kann zur Anreicherung eines Rosskastanienextraktes, insbesondere eines Rosskastanienextraktes, dessen mit Cholesterin fällbare Saponinanteile entfernt sind, verwendet werden; die nach Abtrennen des zu isolierenden Kryptoäscins von allen Saponinanteilen befreite oder praktisch freie Rosskastanienextraktlösung kann für therapeutische Zwecke mit Vorteil dann verwendet werden, wenn die therapeutische Wirkung des Kryptoäscins nicht erwünscht ist, dagegen die therapeutische Wirkung der übrigen Inhaltsstoffe des Extraktes, z. B. der Flavonstoffe.
Nach Abscheiden des mit Cholesterin fällbaren Anteils des Extraktes der Rosskastanie fällt beim Ansäuern auf einen pH-Wert von 2 oder darunter das Kryptoäscin aus, das von der Flüssigkeit mechanisch, z. B. durch Zentrifugieren, abgetrennt und durch Umlösen gereinigt werden kann; nach einer bevorzugten Ausführungsform der Erfindung geschieht die Abtrennung durch Behandeln des Gemisches von Flüssigkeit und ausgefälltem Stoff bzw. der Lösung bei Ansäuern auf einen weniger sauren pH-Wert, der nicht zu einem Ausfällen in sichtbarer Form führt, mit mit Wasser nicht oder teilweise mischbaren or ganischen Lösungsmitteln und die Isolierung durch Abdampfen der von dem übrigen Extrakt getrennten organischen Lösung.
Gegebenenfalls wird von Extrakten ausgegangen, deren Gehalt an Begleitstoffen wie z. B. Flavonolglycoside durch Behandeln mit Aluminiumoxyd und vollständiges Entmineralisieren durch Kationenaustauschersäulen abgetrennt ist.
Die Erfindung schafft nicht nur die Möglichkeit, einen Rosskastanienextrakt zu erzeugen, der von Saponin-Stoffen völlig oder im gewünschten Masse frei ist, sondern schafft weiter die Möglichkeit, das therapeutisch wirksame, keine schädliche Hämolyse bei dem Einspritzen in die Blutbahn bewirkende Krypto äscin als solches zu verwenden, was die Möglichkeit genauer Dosierung mit sich bringt, und die Möglichkeit, den Gehalt eines Rosskastanienextraktes an diesem neu entdeckten therapeutisch wirkungsvollen Kryptoäscin zu erhöhen und/oder zu dosieren.
Als organische Lösungsmittel eignen sich mit Wasser nicht mischbare organische Lösungsmittel, insbesondere bei üblichen Bedingungen flüssige, einwertige, aliphatische Alkohole wie n-Butanol, sowie iso-Butanol, n-Amylalkohol, iso-Amylalkohol sowie Hexylalkohol und Heptylalkohol und aliphatische, mit Wasser nicht mischbare Ketone, wie Methyl äthylketon, Methylpropylketon, Diäthylketon, sowie mit Wasser nicht mischbare halogenierte Kohlenwasserstoffe mit bis zu zwei Kohlenstoffatomen, z. B.
Methylenchlorid, Chloroform oder Trichloräthylen, insbesondere im Gemisch mit aliphatischen Alkoholen oder Ketonen mit bis zu drei Kohlenstoffatomen, z. B. mit Methylalkohol, Äthylalkohol oder Propylalkohol, bzw. Aceton; diese Alkohole sind für sich nicht geeignet, da sie mit Wasser mischbar sind, eignen sich aber im Gemisch mit den halogenierten Kohlenwasserstoffen ausgezeichnet als Extraktionsmittel gemäss der Erfindung.
Die folgenden Ausführungen und Beispiele veranschaulichen die Erfindung.
Wird aus einem 100/eigen, wässrigen Rosskastanienextrakt durch Versetzen mit Cholesterin (100 g auf 101 Extrakt) in ätherischer Lösung, nach Abdampfen des Ethers und Zentrifugieren das beim Injizieren in die Blutbahn hämolytisch wirkende Saponin abgetrennt, so verbleibt ein Restextrakt ohne hämolytische Wirkung von bitterem Geschmack, der eine gewisse Schaumkraft besitzt. Beim Ansäuren dieses Restextraktes, z. B. auf pH 2, entsteht ein Nie- derschlag von Kryptoäscin A, der abzentrifugiert werden kann. Noch in Lösung verbleibendes Krypto äscin kann mittels eines mit Wasser nicht oder teilweise mischbaren organischen Lösungsmittels, z. B. n-Butanol, ausgeschüttelt werden.
Aus diesem Niederschlag und den Extrakten können begleitende Flavonolglycoside durch Behandeln mit Aluminiumoxyd abgetrennt werden. Eine weitere Reinigung ist möglich durch sorgfältiges Waschen des organischen Lösungsmittels mit Wasser. Die noch in sehr geringer Menge verbleibenden gefärbten Verunreinigungen lassen sich mit Tierkohle in methanolischer Lösung entfernen. Bei Reinigung über Aluminiumoxyd muss die wässrige Saponinlösung über einen Kationenaustauscher entmineralisiert werden.
Aus der sauer abfliessenden wässrigen Lösung kann das reine Saponin mit mit Wasser nicht oder teilweise mischbaren organischen Lösungen wiederum extrahiert werden.
Der Hämolyse-Index (HI) des Kryptoäscins liegt sehr niedrig; der von Kryptoäscin A ist Null; die Toxizität ist geringer als die des cholesterinfällbaren Saponin-Anteils des Rosskastanienextraktes, jedoch liegen die Werte DL 50 weit oberhalb von störenden Werten.

Pharmakologische Untersuchungen von Krypto äscin A und Kryptoäscin B brachten sehr befriedigende Ergebnisse, die Stoffe zeichneten sich durch starke Gefässwirksamkeit, diuretische Wirkung sowie Antihistaminwirkung aus.
Die folgenden Beispiele veranschaulichen bevorzugte Ausführungsformen der Erfindung:
Beispiel 1
Mit Cholesterin vorbehandelter Extrakt von Rosskastaniensamen (1 Liter, 100/oig) wurde auf pH 2 angesäuert, der entstehende Niederschlag wurde abzentrifugiert. Der Überstand wurde mehrmals mit n-Butanol (4 X200 ml) extrahiert. In den vereinigten Extrakten wurde der Niederschlag gelöst. Die Lösung wurde mit Wasser (4X100 ml) einige Male zur Befreiung von Mineralsäuren behandelt.
Zur Abtrennung von Flavonolglycosiden wurde mit Aluminiumoxyd (Woelm, neutral) behandelt.
Nach Abdestillieren des Butanols im Vakuum wurde der Rückstand in 400 ml Wasser gelöst und über einem Kationenaustauscher (40 ml, Dowex 50, x-4) entmineralisiert.
Aus der abfliessenden sauren Lösung wurde das Kryptoäscin mit n-Butanol extrahiert; es wurde nach Abtreiben des Extraktionsmittels im Vakuum in 200 ml Methanol gelöst und mit Tierkohle entfärbt; dann im Vakuum zur Trockne eingeengt.
Die an Aluminiumoxyd adsorbierten Flavonolglycoside wurden mit verdünnten Mineralsäuren abgelöst und über ihre Bleisalze gereinigt. Die von Blei befreiten Flavonolglycoside stellten ein gut wasserlösliches Produkt dar.
Aus Kunstharzaustauscher liessen sich keine Rosskastanieninhaltsstoffe isolieren, da dieser nur Kationen festhält.

Weisses, amorphes und hygroskopisches Pulver von stark bitterem Geschmack mit Reizwirkung auf die Schleimhaut.
Leicht löslich in Methanol, mässig in Wasser und Äthanol, schlecht löslich in Aceton und unlöslich in Äther und Kohienwasserstoffen.
Entwickelt in wässriger Lösung, noch 1:20.000 verdünnt, beim Schütteln kräftig Schaum. Die Verbindung fällt bei pH 2 aus mässig verdünnten Lösungen aus, nicht aber in Anwesenheit von cholesterinfällbarem Saponin.
Durch Lösen von Kryptoäscin A in Wasser und Erhitzen kann, wie das folgende Bei spiel 2 zeigt, Kryptoäscin B gewonnen werden.
Beispiel 2
10 g eines nach Beispiel 1 erhaltenen Krypto äscin A wurden in 250 ml Wasser gelöst und unter Rückfluss 1 Stunde in kochendem Wasser gehalten.
Nach Abkühlung wurde die Lösung im Vakuum eingeengt und die Substanz zur Trockne gebracht.
Ausbeute: lOg Eigenschaften: Schmelzpunkt (Säure) = 220-221"C;
Schmelzpunkt (Natriumsalz) = 255-257 C.
HI = 1:6.000
Die Substanz gleicht in allen sonstigen Eigenschaften dem Kryptoäscin A.
Der Drehwert von Kryptoäscin A beträgt minus 7,9O, der von Kryptoäscin B minus 10,7O.
Beispiel 3
Aus einem Liter eines cholesterinbehandelten Rosskastanienextraktes, in dem die Flavonolglycoside und Kryptoäscin in einem Mischungsverhältnis von etwa 1:4 vorliegen, wurden nach dem Verfahren des Beispiels 1 etwa 10 g Kryptoäscin isoliert. Der verbleibende Extrakt wurde mit von Aluminiumoxyd abgelösten Flavonoiglycosiden (Beispiel 1) versetzt.
Erhalten wurde ein therapeutisch wirksamer, nicht hämolytischer Rosskastanienextrakt mit einem Gehalt von 2,5 g Flavonolglycosiden pro Liter.
Beispiel 4
Ein durch Behandeln von Rosskastanienextrakt mit Cholesterin und Abtrennen der Ausfällung gewonnener Extrakt, der pro 50 ml 150 mg Kryptoäscin bei 3 o/o Trockenrückstand enthielt und der als Infusion ohne weiteres vom Menschen vertragen wurde, wurde in 5 ml Ampullen-Portionen, die 15 mg Krypto äscin A bzw. B enthielten, unterteilt; jeder dieser Portionen wurden je 15 mg des nach der Erfindung isolierten Kryptoäscin A zugesetzt.
Eine schädliche hämolytische oder toxische Wirkung war nicht zu beobachten.

The colored impurities still remaining in very small quantities can be removed with animal charcoal in a methanolic solution. When cleaning with aluminum oxide, the aqueous saponin solution must be demineralized using a cation exchanger.
The pure saponin can again be extracted from the acidic draining aqueous solution with organic solutions which are immiscible or partially miscible with water.
The hemolysis index (HI) of cryptoascin is very low; that of cryptoascin A is zero; the toxicity is lower than that of the cholesterol-precipitable saponin content of the horse chestnut extract, but the DL 50 values are far above disturbing values.

No horse chestnut ingredients could be isolated from synthetic resin exchanger, since it only holds cations.

By dissolving Kryptoäscin A in water and heating, as the following Example 2 shows, Kryptoäscin B can be obtained.
Example 2
10 g of a cryptoascin A obtained according to Example 1 were dissolved in 250 ml of water and kept under reflux for 1 hour in boiling water.

Example 3
From one liter of a cholesterol-treated horse chestnut extract in which the flavonol glycosides and cryptoescin are present in a mixing ratio of about 1: 4, about 10 g of cryptoascin were isolated by the method of Example 1. The remaining extract was mixed with flavonoiglycosides detached from aluminum oxide (Example 1).
A therapeutically effective, non-hemolytic horse chestnut extract with a content of 2.5 g of flavonol glycosides per liter was obtained.
Example 4
An extract obtained by treating horse chestnut extract with cholesterol and separating the precipitate, which contained 150 mg cryptoescin per 50 ml at 3 o / o dry residue and which was easily tolerated by humans as an infusion, was in 5 ml ampoule portions containing 15 mg Contained cryptoascin A and B, divided; 15 mg of the cryptoescin A isolated according to the invention were added to each of these portions.
