Dans cette formule et dans les formules suivan tes R1 est un atome de fluor ou d'iode ou un groupe éthyle, méthoxy, méthylmercapto, formyle, -acétyle, cyano ou nitroso et R2 est un atome d'hydrogène, de fluor, de chlore ou de brome ou un groupe méthyle.
L'anion associé au cation de formule (I) peut être un équivalent d'anion de n'importe quel acide phar- maceutiquement acceptable, par exemple un équiva lent d'anion chlorure, bromure, iodure, méthylsulfate ou sulfate. Par rapport aux composés divulgués dans le bre vet principal, les composés contenant un cation de formule (I) présentent une activité sélective contre le Nematospiroïdes dubius, et une activité inférieure contre le Nippostrongylus mugis.
Le cation préféré de formule (I) est le cation N- benzyl-N-[2-(o-fluoro-phénoxy)-éthyl] -N,N-diméthyl- ammonium.
Les sels préférés du cation de formule (I) sont ceux qui sont peu solubles dans l'eau, par exemple de solubilité inférieure à 2 % en poids/volume dans l'eau.
Par exemple, l'équivalent d'anion associé à un cation de formule (I) à 20 C peut être un équiva lent d'anion borofluorure, perchlorate, laurylsulfate, dodécylbenzènesulfonate, p-toluènesulfonate, p-chlo- robenzènesulfonate, p-bromobenzènesulfonate, p-acyl- amidobenzènesulfonate, 4,4'-diamino-stilbène-2,3'-di- sulfonate, diphényl-4-sulfonate, naphtalène- 1-sulfo- nate, naphtalène-2-sulfonate, naphtalène- 1,5-disulfo- nate, naphtalène-2,7-disulfonate, 1-naphtol-3,

Le procédé selon l'invention est caractérisé en ce que l'on fait réagir une amine tertiaire contenant tous les groupes désirés dans le composé d'ammo nium quaternaire sauf un, avec un dérivé réactif du groupe que l'on désire introduire.
Par exemple, les composés peuvent être préparés par quaternisation d'un 1-(N-benzyl-N-méthyl-ami- no)-2-phénoxy-éthane avec une proportion molécu laire d'un agent de méthylation, par exemple d'iodure de méthyle, de diméthylsulfate ou d'un sulfonate de méthyle de formule CH3 - O - S02 - Y dans la quelle Y est un hydrocarbure substitué ou non substi tué tel qu'un groupe p-toluyle. Cette réaction peut être effectuée dans un solvant, par exemple l'acé tone ou le méthanol. Pratiquement, il est en général préférable d'utiliser un peu plus que la quantité théorique d'agent de méthylation pour avoir un bon rendement en produit désiré.
Les composés peuvent encore être préparés par quaternisation d'un 1-diméthylamino-2-phénoxy- éthane avec un agent de benzylation, ou par quater- nisation d'une N-benzyl-N,N-diméthyl-amine avec un dérivé réactif d'un groupe phénoxy-éthyle. Dans ces réactions, l'agent de benzylation et le dérivé réactif de phénoxyéthyle contiennent un groupe réactif X, qui peut par exemple être un groupe chlorure, bro mure, ou iodure, un groupe d'ester sulfonique -0-SO2-Y, Y étant tel que défini ci-dessus. Les deux réactions peuvent être effectuées dans un sol vant tel que l'isopropanol ou l'acétone.
La première réaction se produit facilement, et peut être assistée par la présence d'iodure de potassium, le composé d'ammonium quaternaire étant alors isolé en général sous forme de son iodure. Par contre, la seconde opé ration nécessite un chauffage prolongé, et peut prati quement être mise en @uvre en chauffant les réactifs ensemble en l'absence d'un solvant.
Le sel produit par les réactions décrites ci-dessus peut être converti en sel d'un autre anion par double décomposition, soit pendant soit après les réactions, par exemple en solution ou sur une colonne échan- geuse d'anions. Ceci peut être spécialement avanta geux lorsqu'on désire un sel peu soluble dans l'eau.
Dans les exemples suivants, les températures sont données en degrés centigrades. <I>Exemple 1</I> On chauffe à reflux un mélange de 60 g d'o- éthyl-phénol, 123,6 g de dibromure d'éthylène et 100 ml d'eau, en agitant et en ajoutant progressive ment une solution de 18,9 g d'hydroxyde de sodium dans 50 ml d'eau. On termine l'addition en 1 1/2 h et on continue à chauffer le mélange pendant encore 6 h. On extrait la couche organique à l'éther et sé pare l'o-éthyl-phénol non transformé au moyen d'hy droxyde de sodium aqueux 4 N. On sèche la solu tion éthérée sur du chlorure de calcium, on la filtre et on l'évapore. On distille le résidu sous vide et on obtient le 1-bromo-2-(o-éthyl-phénoxy)-éthane, p. éb. 134-1370/21 mm.
On fait réagir 13 g de ce bromure avec 17,2 g de benzylméthylamine dans 17 ml de benzène. Après avoir chauffé pendant 3 h au bain-marie, on secoue le mélange avec un excès d'hydroxyde de sodium 10 N et on enlève la couche aqueuse. On sèche la couche résiduelle sur de l'hydroxyde de potassium solide, on filtre et on évapore. On distille le résidu sous vide et on obtient le 1-(N-benzyl-N-méthyl- amino)-2-(o-éthyl-phénoxy)-éthane; p. éb. 120-124o/ 0,01 mm.
On fait réagir 11,5 g de cette base avec 6,0 g d'iodure de méthyle dans 35 ml d'acétone. Une ré action se produit spontanément. Après 3 h on sépare par filtration l'iodure de N-benzyl-N-[2-(o-éthyl-phé- noxy)-éthyl]-N,N-diméthyl-ammonium et on le re- cristallise dans de l'éthanol, p.f. 138-139 .
<I>Exemple 2</I> On ajoute 7 g de chlorure d'o-chloro-benzyle à une solution de 7 g de 1-diméthylamino-2-(o-mé- thoxy-phénoxy)-éthane dans 7 ml d'acétone, et on chauffe la solution à reflux pendant 45 min. Par addi tion d'éther au mélange réactionnel refroidi, on obtient le chlorure de N-(o-chloro-benzyl)-N-[2-(o-méthoxy- phénoxy)-éthyl]-N,N-diméthyl-ammonium sous forme d'une gomme incristallisable. On décante la phase liquide et on dissout la gomme résiduelle dans 20 ml d'eau. On ajoute 7 g d'iodure de potassium à la solu tion, et il se sépare une gomme qui cristallise par la suite.
On recueille ce produit solide, qui est l'iodure de N-(o-chloro-benzyl)-N-[2-(o-méthoxy-phénoxy)- éthyl]-N,N-diméthyl-ammonium, on le lave à l'eau et on le recristallise dans du méthanol, p.f. 145-146o.
Dans le tableau I, on énumère d'autres composés d'ammonium quaternaire pouvant être préparés par des méthodes analogues à celles décrites dans l'exem ple 1.

The anion associated with the cation of formula (I) can be an anion equivalent of any pharmaceutically acceptable acid, for example an equivalent of chloride, bromide, iodide, methylsulfate or sulfate anion. Compared to the compounds disclosed in the main patent, the compounds containing a cation of formula (I) exhibit selective activity against Nematospiroides dubius, and lower activity against Nippostrongylus mugis.
The preferred cation of formula (I) is the N-benzyl-N- [2- (o-fluoro-phenoxy) -ethyl] -N, N-dimethylammonium cation.

The process according to the invention is characterized in that a tertiary amine containing all the desired groups is reacted in the quaternary ammonium compound except one, with a reactive derivative of the group which is desired to be introduced.
For example, the compounds can be prepared by quaternization of a 1- (N-benzyl-N-methyl-amino) -2-phenoxy-ethane with a molecular proportion of a methylating agent, for example of. Methyl iodide, dimethyl sulphate or a methyl sulphonate of the formula CH3 - O - SO2 - Y in which Y is a substituted or unsubstituted hydrocarbon such as a p-toluyl group. This reaction can be carried out in a solvent, for example acetone or methanol. In practice, it is generally preferable to use a little more than the theoretical amount of methylating agent in order to have a good yield of the desired product.
The compounds can further be prepared by quaternization of a 1-dimethylamino-2-phenoxyethane with a benzylating agent, or by quaternization of an N-benzyl-N, N-dimethyl-amine with a reactive derivative of 'a phenoxy-ethyl group. In these reactions, the benzylating agent and the reactive phenoxyethyl derivative contain a reactive group X, which may for example be a chloride, bromide, or iodide group, a sulfonic ester group -O-SO2-Y, Y being as defined above. Both reactions can be carried out in a solvent such as isopropanol or acetone.
The first reaction takes place easily, and can be assisted by the presence of potassium iodide, the quaternary ammonium compound then being generally isolated in the form of its iodide. In contrast, the second operation requires prolonged heating, and can practically be carried out by heating the reactants together in the absence of a solvent.
The salt produced by the reactions described above can be converted to the salt of another anion by double decomposition, either during or after the reactions, for example in solution or on an anion exchange column. This can be especially advantageous when a salt which is poorly soluble in water is desired.
In the following examples, temperatures are given in degrees centigrade. <I> Example 1 </I> A mixture of 60 g of o-ethylphenol, 123.6 g of ethylene dibromide and 100 ml of water is heated to reflux, with stirring and the gradual addition of a solution of 18.9 g of sodium hydroxide in 50 ml of water. The addition is completed in 11/2 hours and heating the mixture is continued for a further 6 hours. The organic layer is extracted with ether and the unconverted o-ethyl-phenol separated with 4N aqueous sodium hydroxide. The ethereal solution is dried over calcium chloride, filtered and dissolved. evaporates it. The residue is distilled in vacuo and 1-bromo-2- (o-ethyl-phenoxy) -ethane is obtained, p. eb. 134-1370 / 21 mm.
13 g of this bromide are reacted with 17.2 g of benzylmethylamine in 17 ml of benzene. After heating for 3 h in a water bath, the mixture is shaken with excess 10 N sodium hydroxide and the aqueous layer is removed. The residual layer is dried over solid potassium hydroxide, filtered and evaporated. The residue is distilled off in vacuo to give 1- (N-benzyl-N-methyl-amino) -2- (o-ethyl-phenoxy) -ethane; p. eb. 120-124o / 0.01mm.
11.5 g of this base are reacted with 6.0 g of methyl iodide in 35 ml of acetone. A reaction occurs spontaneously. After 3 h, the N-benzyl-N- [2- (o-ethyl-phenoxy) -ethyl] -N, N-dimethyl-ammonium iodide is filtered off and recrystallized from sodium chloride. ethanol, pf 138-139.
<I> Example 2 </I> 7 g of o-chloro-benzyl chloride are added to a solution of 7 g of 1-dimethylamino-2- (o-metoxy-phenoxy) -ethane in 7 ml of acetone, and the solution is heated under reflux for 45 min. By adding ether to the cooled reaction mixture, N- (o-chloro-benzyl) -N- [2- (o-methoxy-phenoxy) -ethyl] -N, N-dimethyl-ammonium chloride is obtained under form of an incrystallizable eraser. The liquid phase is decanted and the residual gum is dissolved in 20 ml of water. 7 g of potassium iodide are added to the solution, and a gum separates which subsequently crystallizes.
This solid product, which is N- (o-chloro-benzyl) -N- [2- (o-methoxy-phenoxy) - ethyl] -N, N-dimethyl-ammonium iodide, is collected, washed with water and recrystallized from methanol, mp 145-146o.
In Table I, other quaternary ammonium compounds are listed which can be prepared by methods analogous to those described in Example 1.
