Process for the preparation of substituted triphenylethylenes
The subject of the invention is a process for the preparation of substituted triphenylethylenes. These substances are generally characterized by their anti-estrogenic activity in the endocrine system, by their anti-inflammatory activity, their gonadotrophic activity and / or their activity which allows them to lower the levels of cholesterol in the blood.
The substituted triphenylethylenes obtained according to the invention have the following formula:
EMI1.1
wherein the group -OCnH2nA is attached in the meta or para position to the benzene ring, n is an integer between 2 and 4, A is a group. eg amino, alkyllamino, dialkoylamino or a di (haloalkyl) amino group, the alkyl part of these last 3 groups containing from 1 to 4 carbon atoms, or an N-piperidyl, N-morpholinyl or N-pyrrolidinyl group or a group 4 -methyl-1-piperazinyl, where R is hydrogen or halogen or methyl or methoxy, and where X is halogen.
As estrogen antagonists, these new compounds are useful in the treatment of hyperestrogenism and disorders resulting from this condition, for example endometriosis, haemophilia or blood loss, Kleinfelter's syndrome, dysmenrhea. , menopausal disorders and the like. They are active both buccally and parenterally and can therefore be administered by either method, although the buccal route is preferably employed in most cases.
Some of these compounds possess high activity as estrogen antagonists and are preferably non-estrogenic. Some of these compounds, endowed with antiestrogenic activity, are also uterotrophic.
As anti-inflammatory agents, the new products are interesting in that they improve the symptoms of collagenous diseases, such as arthritis and rheumatism, and also in all inflammation treatments. Certain of these products simultaneously possess the anti-inflammatory activity and the antiestrogenic activity, which constitutes a new association, and are more particularly advantageous for this reason.
As agents for lowering blood cholesterol levels, the new products are of value in the treatment of arteriosclerosis.
As gonadotropic inhibiting agents, the new products are of value in the treatment of disorders related to fertility and infertility problems, and can for this purpose be administered orally or parenterally.
The new compounds can be administered orally or parenterally at doses of between 25 mg and 4 grams per day depending on the conditions of the treatment. For various uses, the products in question can be incorporated into creams, ointments or lotions in concentrations of up to 10%. In aerosol form, the concentration can be in the range of about 0.05 to 1%.
The new compounds can be isolated and are generally usable in the form of salts of such mineral acids. as hydrochloric or hydrobromic acid or organic acids such as citric or oxalic acid. They can however be used in the basic form.
These novel substituted triphenylethylenes are prepared, according to the invention, by halogenation of triphenylethylene or of the corresponding triphenylethanol.
The haloethylenes can be prepared by halogenation of the corresponding ethylenes using bromine, chlorine or N-chloro or N-bromo succinimide. They can also be prepared by halogenation of the hydrochloride or hydrobromide of the corresponding ethylenes as when operating, respectively, with N-chloro- or N-bromosuccinimide. They can also be prepared by halogenation of the corresponding ethanols by means of bromine.
The preparation of the substituted triphenylethanols from which the corresponding novel triphenylethylenes are prepared is described in detail in Swiss Patent No. 358435. When A is an amino group, the corresponding substituted triphenylethanols can be prepared by catalytic hydrogenolysis of the corresponding substituted triphenylethanols. - dants in which A is a dibenzylamino group.
These latter compounds can be prepared by reacting a suitably substituted benzylmagnesium halide with a substituted benzophenone having a -OC ,, H ,,, [N (PhCH.,) *,] Group, and this benzophenone can be obtained. by reaction of a hydroxybenzophenone with a dibenzylaminoalkyl halide.
Substituted triphenylethylenes in which A is a (di-haloalkyl) amino group can be prepared by reacting an alkali metal salt of a hydroxybenzophenone with an alkyl halide such as ethylene chlorobromide to obtain a haloalkoxybenzophenone, then reacting reacting the latter derivative with a benzylmagnesium halide, then, after dehydration, reaction with a suitable dialkanolamine followed by reaction with a halogenating agent such as a thionyl halide.
