Procédé de préparation de nouveaux dérivés de l'acide 5-thiazole carboxylique
La présente invention a pour objet un procédé de préparation de nouveaux dérivés de l'acide 5-thiazole carboxylique, répondant à la formule générale:

dans laquelle R représente un reste alcoyle dérivé d'un alcool aliphatique éventuellement substitué.
Le substituant R est de préférence le reste alcoyle d'un alcool aliphatique comportant de 1 à 10 atomes de carbone, le reste alcoyle d'un alcool aryloxyaliphatique dont le reste aliphatique comporte de 1 à 6 atomes de carbone, le reste alcoyle d'un alcool alcoylhétérocyclique, dont le reste aliphatique comporte de 1 à 6 atomes de carbone et dont le reste hétérocyclique est le groupe furyle, le groupe thiényle ou le groupe pyridyle.
Ces composés se distinguent par des propriétés phar macologiques intéressantes. Ils possèdent en particulier une activité hypolipémiante remarquable qui se manifeste par une baisse du taux des triglycérides très significative. Ils sont dénués d'effets secondaires gênants, n'ayant qu'une action vasodilatatrice très faible ou nulle.
Parmi ces composés, présentent un intérêt particulier:
- le 5-thiazole carboxylate de (3'-pyridyl) méthyle,
- le 5-thiazole carboxylate de 2'-p-chlorophénoxy
2'-méthylpropyle,
- le 5-thiazole carboxylate d'isopropylidène pyri
doxine,
- le 5-thiazole carboxylate de N-diéthylamino éthyle,
et ses sels d'acides minéraux ou organiques,
- le 5-thiazole carboxylate de 2',2'-diméthyl l'-pro
pyle.
Le procédé, objet de l'invention, est caractérisé en ce que l'on fait agir un alcool sur l'acide 5-thiazol carboxylique ou encore un dérivé fonctionnel de cet acide en présence d'une amine tertiaire et obtient l'ester de l'acide 5-thiazol carboxylique cherché. Dans le cas où l'acide 5-thiazol carboxylique est estérifié par un aminoalcool, disubstitué sur la fonction amine, on peut transformer cet ester en sel par action d'un acide minéral ou organique.
Le procédé peut être exécuté avantageusement dans les conditions suivantes:
1. L'estérification de l'acide 5-thiazol carboxylique est effectuée par action d'un halogénure de l'acide 5thiazol carboxylique sur un alcool, en présence d'un accepteur d'hydracide.
2. L'accepteur d'hydracide est une amine tertiaire et, de préférence, la triéthylamine.
Le produit de départ, L'acide 5-thiazol carboxylique, est obtenu selon le procédé décrit par H. Erlenmeyer et
H. Von Meyenburg Helv. Chem. Acta 20, 204, 1937 ou
H. Erlenmeyer et R. Marbet Helv. Chem. Acta 29, 19461949, (1946).
Exemple 1
5-thiazol carboxylate de (3'-pyridyl) méthyle

On verse une solution de 28,5 g de chlorure de l'acide 5-thiazole carboxylique dans 50cm3 d'éther, dans une solution de 21 g de 3-pyridyl méthanol et 23,4 g de tri- éthylamine dans 400 cm3 d'éther, en maintenant la température à 200 C; on agite le mélange réactionnel pendant une heure trente minutes à température ambiante, filtre, lave la phase éthérée avec une solution aqueuse de carbonate de potassium, puis à l'eau et évapore le solvant sous pression réduite; on obtient le 5-thiazol carboxylate de (3'-pyridyl) méthyle sous forme de cristaux incolores, qui, après recristallisation dans l'éther isopropylique, fondent à 640 C (Rendement: 47 5S).

Le chlorure de l'acide 5-thiazole carboxylique peut être obtenu à partir de l'acide 5-thiazol carboxylique selon les méthodes de chloruration usuelles.
Exemple 2
5-thiazol carboxylate de 2'-p-chlorophénoxy
2'-méthylpropyle

On verse une solution de 14,7 g de chlorure de l'acide 5-thiazol carboxylique dans 90 cm3 d'acétone, dans une solution de 18,15g de 2-p-chlorophénoxy 2méthyl 1-propanol et 12,21 g de triéthylamine dans 50 cm3 d'acétone, en maintenant la température entre 200 et 250 C; on agite pendant une heure à température ambiante, filtre et distille le solvant sous pression réduite: on reprend le résidu à l'éther, lave la solution éthérée avec une solution aqueuse de carbonate de sodium, puis à l'eau; on distille l'éther sous pression réduite et obtient le 5-thiazole carboxylate de 2'-p-chlorophénoxy 2'-méthylpropyle sous forme de cristaux incolores qui, après recristallisation dans l'isopropanol, fondent à 770 C (Rendement: 76 %).

Le produit de départ, le 2-p-chlorophénoxy 2-méthyl l-propanol, peut être obtenu de la manière suivante:
On ajoute à une solution de 56,4 g de p-chlorophénoxy isobutyrate d'éthyle dans 250 cm3 d'éther, une suspension de 8,8 g d'hydrure de lithium-aluminium dans 250 cm3 d'éther, en maintenant la température entre 00 et + 50 C; on agite le mélange réactionnel pendant une heure trente minutes, à température ambiante, ajoute 20cm3 d'eau et filtre; le filtrat est lavé à Peau et distillé sous pression réduite; on obtient 39,60g de 2-p-chlorophénoxy 2-méthyl 1-propanol, sous forme d'un liquide bouillant à 990-1020 C sous 0,3 mm de mercure (Rendement: 85%), /n/2D2 = 1,5281.

On ajoute à une solution de 5,55 g de chlorure de l'acide 5-thiazole carboxylique dans 40 cm3 d'acétone.
une solution de 8,11g d'isopropylidène pyridoxine et 4,7 g de triéthylamine dans 160 cm3 d'acétone, en maintenant la température entre 200 et 250 C; on agite pendant une heure trente minutes à température ambiante, filtre et évapore le solvant sous pression réduite; on reprend le résidu à l'éther, lave la phase éthérée avec une solution aqueuse à 10 % de carbonate de potassium, puis à l'eau et distille l'éther; on obtient le 5thiazole carboxylate d'isopropylidène pyridoxine sous forme de cristaux incolores qui, après recristallisation dans l'éthanol, fondent à 1320 C (rendement: 40 %).

Exemple 4
Dichlorhydrate de 5-thiazole carboxylate de
N-diéthylamino éthyl

On ajoute à 2,77 g de chlorure de l'acide 5-thiazol carboxylique, 40 cm3 de diéthylamino éthanol en refroidissant; puis on chauffe le mélange à 1000 C pendant une demi-heure et distille l'excès de diéthylaminoéthanol sous pression réduite; on reprend le résidu par50cm3 d'éthanol et ajoute 2,87 g detriéthanolamine; on filtre,distille l'huile résiduelle et recueille la fraction bouillant entre 101o et 1540 C sous 10mm de mercure; on obtient le 5-thiazol carboxylate de diéthylamino éthyle avec un rendement de 49 %.
En traitant cette base par la quantité stoechiomé- trique d'acide chlorhydrique dans l'éthanol, on obtient le dichlorhydrate de 5-thiazole carboxylate de diéthylamino éthyle sous forme de cristaux incolores qui, après recristallisation dans l'éthanol, fondent à 1160 C (Rendement: 45,5 %).

Exemple 5 5-thiazol carboxylate de 2',2'-diméthyl l'-propyle

On ajoute lentement une solution de 5,5 g de chlorure de l'acide 5-thiazole carboxylique dans 20 cm3 d'acétone à une solution de 3,10g d'alcool néopentylique et de 4,73 g de triéthylamine dans 20cm3 d'acétone; on filtre, évapore le solvant sous pression réduite et reprend le résidu à l'éther; on lave la solution éthérée avec une solution aqueuse de carbonate de potassium à 10%, puis à 'leau; on chasse l'éther et distille l'huile résiduelle; on recueille la fraction passant de 1300 à 1320 C sous une pression de 15 mm de mercure et obtient le 5-thiazole carboxylate de 2',2'-diméthyl 1'propyle, avec un rendement de 35,5 %, sous forme d'un liquide, //2S = 1,4929-1,4928.

Ainsi qu'il a été indiqué plus haut, les produits obtenus par le procédé, objet de l'invention, sont dotés de propriétés pharmacologiques intéressantes. Ils possèdent notamment une action hypolipémiante remarquable qui se manifeste par une baisse du taux des triglycérides très significative.
Ils peuvent être utilisés pour le traitement de l'hyperlipémie aiguë ou chronique, de rathéromatose, des stéatoses hépatiques ou toxiques et des néphroses lipoidiques.
Ils sont utilisés par voie buccale ou rectale.
Ils peuvent se présenter sous forme de oomprimés, de comprimés enrobés, de cachets, de capsules, de granulés, d'émulsions, de sirops et de suppositoires.
La posologie utile est d'environ 0,2 g à 1 g par prise et 1 g à 5 g par jour chez l'adulte en fonction de la voie d'administration et, de préférence 1,5 g.
Les formes pharmaceutiques telles que: comprimés, comprimés enrobés, cachets, capsules, granulés, émulsions, sirops et suppositoires, sont préparées selon les procédés usuels.
Process for the preparation of novel 5-thiazole carboxylic acid derivatives
The present invention relates to a process for the preparation of novel derivatives of 5-thiazole carboxylic acid, corresponding to the general formula:

in which R represents an alkyl residue derived from an optionally substituted aliphatic alcohol.
The substituent R is preferably the alkyl residue of an aliphatic alcohol containing from 1 to 10 carbon atoms, the alkyl residue of an aryloxyaliphatic alcohol in which the aliphatic residue contains from 1 to 6 carbon atoms, the alkyl residue of a alkylheterocyclic alcohol, the aliphatic residue of which contains from 1 to 6 carbon atoms and the heterocyclic residue of which is the furyl group, the thienyl group or the pyridyl group.
These compounds are distinguished by interesting pharmacological properties. They have in particular a remarkable lipid-lowering activity which is manifested by a very significant drop in the level of triglycerides. They are devoid of bothersome side effects, having only very little or no vasodilator action.
Among these compounds, are of particular interest:
- (3'-pyridyl) methyl 5-thiazole carboxylate,
- 2'-p-chlorophenoxy 5-thiazole carboxylate
2'-methylpropyl,
- isopropylidene pyri 5-thiazole carboxylate
doxin,
- N-diethylamino ethyl 5-thiazole carboxylate,
and its salts of mineral or organic acids,
- 2 ', 2'-dimethyl l'-pro 5-thiazole carboxylate
pyle.
The process which is the subject of the invention is characterized in that an alcohol is made to act on 5-thiazol carboxylic acid or else a functional derivative of this acid in the presence of a tertiary amine and the ester of the sought 5-thiazol carboxylic acid. In the case where the 5-thiazol carboxylic acid is esterified with an aminoalcohol, disubstituted on the amine function, this ester can be converted into a salt by the action of an inorganic or organic acid.
The process can be carried out advantageously under the following conditions:
1. The esterification of 5-thiazol carboxylic acid is carried out by the action of a halide of 5-thiazol carboxylic acid on an alcohol, in the presence of a hydracid acceptor.
2. The hydracid acceptor is a tertiary amine and preferably triethylamine.
The starting product, 5-thiazol carboxylic acid, is obtained according to the process described by H. Erlenmeyer and
H. Von Meyenburg Helv. Chem. Acta 20, 204, 1937 or

Example 1
(3'-pyridyl) methyl 5-thiazol carboxylate

5-thiazole carboxylate dihydrochloride

Is added to 2.77 g of 5-thiazol carboxylic acid chloride, 40 cm3 of diethylamino ethanol while cooling; then the mixture is heated at 1000 ° C. for half an hour and the excess of diethylaminoethanol is distilled off under reduced pressure; the residue is taken up in 50 cm3 of ethanol and 2.87 g of triethanolamine are added; the residual oil is filtered and distilled and the fraction boiling between 101o and 1540 C under 10mm of mercury is collected; diethylamino ethyl 5-thiazol carboxylate is obtained in a yield of 49%.
By treating this base with the stoichiometric quantity of hydrochloric acid in ethanol, diethylamino ethyl 5-thiazole carboxylate dihydrochloride is obtained in the form of colorless crystals which, after recrystallization in ethanol, melt at 1160 C ( Yield: 45.5%).

Example 5 2 ', 2'-dimethyl-propyl 5-thiazol carboxylate

We slowly add a solution of 5.5 g of 5-thiazole carboxylic acid chloride in 20 cm3 of acetone to a solution of 3.10 g of neopentyl alcohol and 4.73 g of triethylamine in 20 cm3 of acetone ; filtered, the solvent evaporated off under reduced pressure and the residue is taken up in ether; the ethereal solution is washed with an aqueous solution of potassium carbonate at 10%, then with water; the ether is removed and the residual oil distilled; the fraction passing from 1300 to 1320 C under a pressure of 15 mm of mercury is collected and the 2 ', 2'-dimethyl 1'propyl 5-thiazole carboxylate is obtained, with a yield of 35.5%, in the form of a liquid, // 2S = 1.4929-1.4928.

As far as we know, this compound is not described in the literature.
As indicated above, the products obtained by the process, which is the subject of the invention, are endowed with advantageous pharmacological properties. In particular, they have a remarkable lipid-lowering action which manifests itself in a very significant drop in the level of triglycerides.
They can be used for the treatment of acute or chronic hyperlipemia, atheromatosis, hepatic or toxic steatosis and lipoid nephrosis.
They are used orally or rectally.
They can be in the form of tablets, coated tablets, cachets, capsules, granules, emulsions, syrups and suppositories.
The useful dosage is approximately 0.2 g to 1 g per dose and 1 g to 5 g per day in adults depending on the route of administration and preferably 1.5 g.
The pharmaceutical forms such as: tablets, coated tablets, cachets, capsules, granules, emulsions, syrups and suppositories are prepared according to the usual methods.
