Dibenzocycloheptadien- oder Dibenzocyclohepta- trien-Derivate, die in 5-Stellung einen offenkettigen basischen Rest, wie beispielsweise den Dimethyl- aminopropylidenr oder einen N-Alkyl-piperidyl'iden- (4)-Rest enthalten, sind bekannt.
Ihre Herstellung erfolgt in analoger Weise durch Grignardierung von Dibenzo[a,e]cycloheptatrien-5-on oder Dibenzo[a,d]- cycloheptal[1,4]dien-5-on. mit der entsprechenden ba sischen Halogenverbindung, anschliessende Hydrolyse und Wasserabspaltung.
Die verfahrensgemäss hergestellten neuen Verbin dungen unterscheiden sich von den bekannten un gesättigten Dibenzocycloheptan-Derivaten dadurch, dass sie in 5-Stellung des ungesättigten Dibenzocyclo- heptan-Gerüstes einen 1-Alkyl-piperidyl-alkylid'enL oder einen 1-Alkyl-pyrrolidyl-alkyliden-Rest besitzen.
Das erfindungsgemässe Verfahren gestaltet sich beispielsweise wie folgt: Eine mit wasserfreiem Tetra- hydrofuran überschichtete Kupfer-Magnesium-Legie- rung (nach Gilman) wird mit der Lösung von 1- Methyl-piperidyl-(2)-äthyl-chlorid in Tetrahydrofuran versetzt.
Die Bildung der Grignardverbindung wird vorzugsweise durch Zusatz von wenig Äthylenbromid gefördert. Anschliessend wird das Reaktionsgemisch noch 30 bis 60 Minuten zum Sieden erhitzt. Anstelle von Kupfer-Magnesium-Legierung können auch Ma gnesiumspäne, die vorzugsweise mit Joddämpfen an geätzt worden sind, verwendet werden.
Die so bereitete Grignardlösung wird darauf mit einer Verbindung der Formel II, z. B. Dibenzo[a,d]- cyclohepta[1,4]dien - 5 - an oder Dibenzo[a,e]cyclo- heptatrien-5-on, versetzt und das Gemisch zur Ver vollständigung der Reaktion noch einige Zeit erhitzt. Anschliessend hydrolysiert man das Reaktionsgemisch in der Kälte mit wässeriger Ammoniumchloridlösung und extrahiett mit einem mit Wasser nicht misch baren organischen Lösungsmittel, vorzugsweise Chlo roform.
Das als Zwischenprodukt erhaltene 5-Hy- droxy-Derivat der Formel IV kann gewünschtenfalls durch Kristallisation gereinigt und mit anorganischen oder organischen Säuren in geeignete Salze über geführt oder direkt weiterverarbeitet werden.
Die Wasserabspaltung erfolgt beispielsweise durch Erhitzen der in Eisessig gelösten Substanz mit konz. Salzsäure. Sie gelingt indessen auch mit ande ren Agenzien, z. B. mit Phosphoroxychlorid, Thionyl- chlorid oder Zinkchlorid. Das Endprodukt wird nach bekannten Methoden isoliert und gereinigt und ge- wünschtenfalls in Säureadditionssalze und in quar- täre Verbindungen übergeführt.
Die verfahrensgemäss hergestellten, neuen un gesättigten Dibenzocycloheptan-Derivate sind bei Raurritemperatur ölig oder kristallin. Es sind basische Verbindungen, die mit anorganischen: oder organi schen Säuren bei Raumtemperatur beständige, kri stallisierte Salze bilden.
Die erfindungsgemäss hergestellten neuen un gesättigten Dibenzocycloheptan-Derivate sowie die Hydroxyderivate der Formel IV, sind auf Grund ihrer ausgezeichneten pharmakodynamischen Eigen schaften zur Verwendung als Heilmittel geeignet, in dem sie sich durch vielfältige Wirkungen auf das Nervensystem auszeichnen.
So sind antihistamin- artige, narkosepotenzierende, adrenolytische, sedative, antipyretische, antiemetische, serotoninhemmende und hypotherme Wirkungen festgestellt worden, wo bei diese Wirkungen je nach der Struktur der basi schen Seitenkette variieren können. Sie sollen thera- peutisch verwendet werden. Ausserdem dienen die neuen Verbindungen aber auch als Zwischenprodukte zur Herstellung von Medikamenten.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden und sind un- korrigiert.
<I>Beispiel 1</I> 5-(2'-[1"-Methyl-piperidyl-(2")]-äthylidenl- dibenzo[a,d]eyclohepta[1,4]dien 2,8 g aktivierte Kupfer-Magnesium-Legierung (nach Gilman) werden mit 15 cm3 Tetrahydrofuran überschichtet und mit 0,4 cm3 Äthylenbromid ver setzt.
Sobald die Reaktion eingesetzt hat, lässt man eine Lösung von 16,2 g 1-Methyl-piperidyl-(2)-äthyl- chlorid in 20 cm3 Tetrahydrofuran innert 20 Min. zutropfen. Nach zweistündigem Erhitzen zum Sieden am Rückfluss wird bei 50 eine Lösung von 10,4 g Dibenzo[a;d]cyclohepta[1,4]dien-5-on (Smp.34-35 ) in 10 cm3 Tetrahydrofuran zugesetzt. Man erhitzt an schliessend nochmals 1 Std. zum Sieden.
Die erkal tete Lösung wird darauf in 300 cm?, einer 10%igen Ammoniumchloridlösung gegossen. Man filtriert von Metallrückständen ab, schüttelt das Filtrat dreimal mit Chloroform aus, trocknet die vereinigten Chloro- formextrakte über Kaliumcarbonat und dampft die Lösung ein. Der ölige Rückstand wird in 200 cm3 Petroläther aufgenommen, von harzartigen Flocken filtriert, das Filtrat eingeengt und auf - 20 abgekühlt.
Bei längerem Stehen kristallisiert aus der konzen trierten Lösung das 5-Hydroxy-5-@2'-[1"-methyl- piperidyl- (2")] - äthyl} -dibenzo[a,d]cyclohepta[1,4]- dien vom Smp. 116-118 aus.
Die erhaltene Verbindung wird in 100 cm-3 Eis essig und 40 cm3 konz. Salzsäure gelöst und 1 Std. unter Rückfluss zum Sieden erhitzt. Die Lösung wird anschliessend im Vakuum eingedampft, der Rück stand in Wasser gelöst, die wässerige Lösung mit Kalilauge alkalisch gemacht und mit Äther extrahiert.
Der über Kaliumcarbonat getrocknete Ätherextrakt wird durch Destillation vom Lösungsmittel befreit und das zurückbleibende Öl bei einem Druck von 0,01 mm Hg destilliert, wobei das 5-@2'-[1"-Methyl- piperidyl - (2")] - äthyliden@ -dibenzo [a,d] cyclohepta- [1,4]dien bei einer Luftbadtemperatur von 180 bis 200 übergeht.
Hydrobromid: Man versetzt die Lösung der Base in Methanol mit so viel wässeriger Bromwasserstoff säure, bis die Lösung gerade kongosauer reagiert. Nach Eindampfen der Lösung wird der Rückstand in Aceton aufgenommen und die Lösung langsam mit Äther versetzt, wobei das 5-f2'-[1"-Methyl-piperidyl- (2")] - äthyliden @-dibenzo[a,d]cyclohepta[1,4]dien-hy- drobromid kristallisiert. Nach Umkristallisieren aus Äthanol/Äther schmilzt das Hydröbromid bei 189 bis 190 .
Das Jodmethylat wird hergestellt, indem man die Lösung der Base in abs. Äthanol mit der berechneten Menge Methyljodid versetzt. Smp. 228-230 (Zers.) aus Methanol. <I>Beispiel 2</I> 5-@[1'-Methyl-piperidyl (3')]-methylen@- dibenzo[a,d]cycloheptadien[1,4] 2,8 g aktivierte Kupfer-Magnesium-Legierung (nach Gilman)
werden mit 15 cm3 Tetrahydrofuran überschichtet und mit 0,5 cm3 Äthylenbromid ver setzt. Sobald die Reaktion kräftig in Gang gekommen ist, lässt man eine Lösung von 14,8 g 1-Methyl-piperi- d'yl-3-methylchlorid in 20 cm3 Tetrahydrofuran zu tropfen.
Nach 11stündigem Erhitzen am Rückfluss lässt man bei 60 eine Lösung von 10,4 g Dibenzo- [a,d] cyclohepta [1,4] dien - 5 - an (Smp. 34-35 ) in 10 cm3 Tetrahydrofuran zutropfen und erhitzt an schliessend noch 2 Std.
am Rückfluss. Die erkaltete Lösung wird darauf in 300 cm3 einer 10 % igen Am moniumehloridlösung gegossen, mit Chloroform ver rührt, von Metallrückständen abfiltriert und das Fil trat mit Chloroform ausgeschüttelt. Nach Trocknen des Chloroformextraktes über Magnesiumsulfat und Abdampfen:
des Lösungsmittels wird das rohe 5- Hydroxy - 5 - {[1' - methyl-piperidyl-(3')]-methyll}-di- benzo[a,d]cycloheptadien[1,4] aus Äthanol umkristal lisiert. Smp. 160-161'.
Die erhaltene Verbindung wird in 125 cm3 Eis essig und 50 cm' konz. Salzsäure 90 Min. am Rück fluss erhitzt. Die Lösung wird anschliessend im Va kuum eingedampft und der Rückstand in Wasser ge löst. Beim Abkühlen kristallisiert das 5-f,[1'-Methyl piperidyl-(31]-methylen}-dibenzo[a,d]cycloheptadien- [1,4]hydrochlorid-hydrat aus. Es wird aus Wasser und Aceton umkristallisiert. Smp.140-145 unter Abgabe von Kristallwasser.
<I>Beispiel 3</I> 5-{2'-[ 1 "-Methyl'-piperidyl-(2")]-äthyliden}- dibenzo[a,e]cycloheptatrien 2,8 g aktivierte Kupfer-Magnesium-Legierung (nach Gilman) werden mit 15 cm3 Tetrahydrofuran überschichtet und mit 0,4 cm3 Äthylen'bromid ver setzt.
Sobald die Reaktion eingesetzt hat, lässt man eine Lösung von 16,2 g 1-Methyl-piperidyl-(2)-äthyl- chlorid in 20 cm3 Tetrahydrofuran innert 20, Min. zu tropfen.
Nach zweistündigem Erhitzen am Rückfluss wird bei 30 portionenweise 10,3 g Dibe:nzo[a,e]- cycloheptatrien-5-on (Smp. 89-90 ) zugegeben und noch 1 Std.
am Rückfluss erhitzt. Die erkaltete Lö sung wird darauf in 300 cm3 einer 10%igen Am moniumchloridlösung gegossen, mit Chloroform ver rührt, von Metallrückständen abfiltriert und das Fil trat dreimal mit Chloroform extrahiert. Nach Trock nen der vereinigten Chloroformextrakte über Kalium- carbonat und Abdampfen des Chloroforms wird der Rückstand mit Hexan verrieben. Ungelöste braune Flocken werden über Tierkohle filtriert und das Fil trat eingedampft.
Das zurückbleibende zähe Öl wird mit 125 cma Eisessig und 50 cm3 konz. Salzsäure 1 Std, am Rückfluss erhitzt. Die Lösung wird an schliessend im Vakuum eingedampft, der Rückstand in Wasser gelöst, die wässerige Lösung mit Natron- lauge alkalisch gemacht und die ausgeschiedene Sub stanz in Äther aufgenommen.
Der über Kaliumcar- bonat getrocknete Ätherextrakt wird durch Destilla tion vom Lösungsmittel befreit, das zurückbleibende Öl in Äthanol gelöst und die äthanolische Lösung mit der berechneten Menge Fumarsäure versetzt. Nach Zusatz von Äther kristallisiert das saure Fumarat des 5 - f 2'-[1"-Methyl-piperidyl-(2")]-äthyliden@-dibenzo- [a,e]cycloheptatriens aus.
Es schmilzt nach Kristalli sation aus Methanol bei 188190 (Zers.).
Das Hydrobromid schmilzt nach Kristallisation aus Methanol bei 187-190 (Zers.). <I>Beispiel 4</I> 5-1[1'-Methyl-piperidyl-(3')]-methylen@- d'ibenzo[a,e]cycloheptatrien In der gleichen Weise wie in Beispiel 2 beschrie ben wird die Grignardverbindung aus 2,8g Kupfer- Magnesium-Legierung und 14,8 g 1-Methyl-piperid'yl- (3)-methyl-chlorid in 15 cm3 Tetrahydrofuran her gestellt.
Die Grignardverbindung versetzt man darauf bei 30 portionenweise mit 10,3g Dibenzo[a,e]cyclo- heptatrien (Smp. 89-90 ) und erhitzt 1 Std. am Rück fluss. Das Reaktionsgemisch wird anschliessend in 300 cm3 einer 10 % igen Ammoniumchloridlösung gegossen, mit Chloroform verrührt, von Metallrück ständen filtriert und dreimal mit Chloroform ausge schüttelt.
Nach Trocknen der vereinigten Chloro- formextrakte über Kaliumcarbonat und Abdampfen des Chloroforms wird der Rückstand aus Aceton um kristallisiert. 5-Hydroxy-5-1 [ 1'-Methyl-piperidyl-(3,#]- methyl}-dibenzo[a,e]cycloheptatrien schmilzt bei 144 bis 145 .
13 g dieser Verbindung werden in 125 cm3 Eis essig und 50 cms konz. Salzsäure 1 Std. am Rück fluss erhitzt. Die Lösung wird darauf im Vakuum ein gedampft und der Rückstand aus Aceton ,/Äther kri stallisiert.
Nach nochmaligem Umkristallisieren aus Aceton oder Äthanol/Äther schmilzt das 5-{[1'-Me- thyl - piperidyl - (3')] - me:thylen@ - dibenzo [a,e] cyclo- heptatrien-hydrochlorid bei 237-239 (Zers.). Aus Wasser kristallisiert das Hydrochlorid mit Kristall wasser.
Die aus dem Hydrochlorid durch Schütteln mit Chloroform und Natronlauge hergestellte Base kri stallisiert aus Hexan oder Aceton in Prismen vom Smp. 117-119<B>0</B>.
<I>Beispiel 5</I> 5-{2'-[1"-Methyl-pyrrolidyl-(2")]-äthyliden.}- dibenzo[a,d]cyclohepta[1,4]dien a) 1,3g mit Jod aktivierte Magnesiumsp'äne wer den mit 5 cm3 abs. Tetrahydrofuran überschichtet und mit 0,2 cm3 Äthylenbromid versetzt. Sobald die Reaktion kräftig in Gang gekommen ist, lässt man die Lösung von 8,9 g 1-Methyl-pyrrolidyl-(2)
-äthylchlorid in 10 cm3 Tetrahydrofuran zutropfen. Nach 11/2stün- digem Erhitzen unter Rückfluss lässt man bei 50-60 die Lösung von 10,4g Dibenzo[a,d]cyclohepta[1,4]- dien-5-on (Smp. 34-35 ) in 10 cm3 Tetrahydrofuran zutropfen. Man erhitzt nochmals 11/2 Std.
unter Rückfluss. Die erkaltete Lösung wird darauf in 150 cm3 einer 10 % igen Ammoniumchloridlösung ge gossen und mit Chloroform ausgeschüttelt. Nach Trocknen des Chloroformextraktes über Magnesium sulfat und Abdampfen des Lösungsmittels wird der Rückstand aus Aceton umkristallisiert, wobei das 5 -Hydroxy-5-{2'-[1"-@methyl-pyrrolidyl-(2")]-äthyl@- dibenzo[a,d]cyclohepta[1,4]dien in feinen Nadeln vom Smp. l15-117 erhalten wird.
b) 9,4 g dieser Verbindung werden in<B>100</B> cm3 Eisessig und 40 cms konz. Salzsäure 1 Std. unter Rückfluss erhitzt. Die Lösung wird anschliessend im Vakuum eingedampft, der Rückstand in Wasser ge löst, die Lösung alkalisch gestellt und das ausgeschie dene Produkt in Äther aufgenommen. Nach Trock nen der ätherischen. Lösung über Kaliumcarbonat und Abdampfen des Äthers wird das zurückbleibende Öl in Methanol gelöst,
die Lösung mit der berechneten Menge Bromwasserstoffsäure versetzt und im Va kuum eingedampft. Der Rückstand, das 5-{2'-[1'- Methyl=pyrrolidyl (2")]-äthyliden}-dibenzo[a,d]cyclo- hepta[1,4]dien-hydrobromid, wird aus Aceton umkri stallisiert. Smp. 15l-153 (Zers.).
<I>Beispiel 6</I> 5-{2'-[1"-Methyl-pyrrolidyl-(2")]-äthyliden}- dibenzo[a,e]cycloheptatrien Auf die gleiche Weise wie in Beispiel 5 beschrie ben erhält man aus 8,9 g 1-Methyl-pyrrolidyl-(2)- äthylchlorid und 10,3g Dibenzo[a,e]cycloheptatrien- 5-on (Smp.89-90 ) das 5-Hydroxy-5-{2'-[1"-Me- thyl - pyrrolidyl - (2'@] -äthyl}-dibenzo[a,
e]cyclohepta- trien. Die Verbindung schmilzt nach dem Umkristal- lisieren aus Aceton bei 129-131'.
Die Wasserabspaltung aus dem 5-Hydroxy-Deri- vat erfolgt ebenfalls in der gleichen Weise wie in Bei spiel 5 beschrieben. Das 5-{2'-[1"-Methyl-pyrrolidyl- (2'@] - äthyliden@ - dibenzo[a,e]cycloheptatrien hydro- bromid schmilzt nach Kristallisation aus Isopropanol/ Aceton bei 164-166 (Zers.).
<I>Beispiel 7</I> 5-{[I'-Methyl-pyrrolidyl-(3@]-methylen}- dibenzo[a,d]eyclohepta[ 1,4]d'ien a) 1,7 g mit Jod aktivierte Magnesiumspäne wer den mit 5 cm3 abs. Tetrahydrofuran überschichtet und mit 0,2 cm3 Äthylenbromid versetzt.
Sobald die Reaktion eingesetzt hat, lässt man die Lösung von 10,7 g 1- Methyl - pyrrolidyl - (3) - methylchlorid in 15 ein?- Tetrahydrofuran zutropfen. Nach einstündi gem Erhitzen am Rückfluss wird bei 40 die Lösung von<B>13,5</B> g Dibenzo[a,d]cyclohepta[1,4]dien-5-on in 15 cm3 Tetrahydrofuran zugetropft. Man erhitzt an schliessend nochmals 2 Std. zum Sieden.
Das erkal tete Reaktionsgemisch wird darauf in 150 cm3 einer 10%igen Ammoniumehloridlösung gegossen. Man schüttelt mehrmals mit Äther aus, trocknet die ver- einigten Ätherextrakte über Natriumsulfat und dampft die Lösung ein.
Der Rückstand wird aus Aceton um kristallisiert, wobei 5-Hyd@roxy-5-1[1'-methyl-pyrroli- dyl- (3')] - methyl} - d'ibenzo [a,d] cyclohepta[1,4]dien vom Smp. 134-135 erhalten wird.
b) 12 g der unter a erhaltenen Verbindung wer den mit 100 cm3 Eisessig und 40 em3 konz. Salz säure 1 Std. am Rückfluss zum Sieden erhitzt. Die Lö sung wird anschliessend im Vakuum eingedampft, der Rückstand in Wasser gelöst, die wässerige Lö sung alkalisch gestellt und die abgeschiedene Substanz in Äther aufgenommen. Das nach Trocknen über Ka- liumcarbonat und Abdampfen des Lösungsmittels zu rückbleibende Öl wird in 30 em3 Äthanol gelöst und die Lösung mit 2,2 g Fumarsäure versetzt.
Man löst durch kurzes Erwärmen und kühlt ab, wobei das neutrale Fumarat des 5-f[1'-Methyl-pyrrolidyl-(3')]- methylen@-dibenzo[a,d]cyclohepta[1,4]diens kristalli siert. Es wird zweimal aus Methanol/Äthanol umkri stallisiert. Smp. 194-l96 (Zers.).
<I>Beispiel 8</I> 5-{[1'-Methyl-pyrrolidyl-(3')]-methylen}- dibenzo[a,e]cycloheptatrien Die wie im vorangehenden Beispiel hergestellte Grignardlösung aus 1,7 g Magnesiumspänen, <B>10,7</B> g 1-Methyl-pyrrolidyl-(3)-methylchlorid und 20 cm3 Tetrahydrofuran versetzt man portionenweise bei 30 mit 13,4 g Dibenzo[a,e]cycloheptatrien-5-on. An schliessend gibt man noch 10 cm3 Tetrahydrofuran zu und erhitzt 2 Std.
am Rückfluss. Das erkaltete Reak tionsgemisch wird darauf in<B>150</B> em3 einer 10 % igen Ammoniumchloridlösung gegossen. Man schüttelt mehrmals mit Äther aus, trocknet die vereinigten Atherextrakte über Natriumsulfat und dampft die Lösung ein. Der verbleibende ölige Rückstand wird mit 200 cm3 Eisessig und 80 em3 konz. Salzsäure 1 Std. am Rückfluss zum Sieden erhitzt.
Die Lösung wird darauf im Vakuum eingedampft, der zähe Rück stand in Wasser gelöst und mit Äther ausgeschüttelt. Die wässerige Phase wird alkalisch gestellt und das abgeschiedene Öl in Äther aufgenommen. Man trock net den Atherextrakt über Kaliumcarbonat, dampft das Lösungsmittel ab und destilliert das viskose Öl im Hochvakuum. Das bei einer Badtemperatur von 150 bis 180 bei einem Druck von 0,05 mm Hg destil lierende gelbe, viskose Öl wird in 40 cm3 Äthanol gelöst und die Lösung mit 2,36g Fumarsäure ver setzt.
Man löst durch kurzes Erwärmen und kühlt ab, wobei das neutrale Fumarat des 5-{[1'-Methyl- pyrrolidyl- (3')] -methylen@ -dibenzo [a,e]cyclohepta- triens kristallisiert. Es wird zweimal aus Methanol Athanol umkristallisiert. Smp. 202-204 (Zers.).

The new compounds produced according to the process differ from the known unsaturated dibenzocycloheptane derivatives in that they have a 1-alkyl-piperidyl-alkylid'enL or a 1-alkyl-pyrrolidyl-alkylidene in the 5-position of the unsaturated dibenzocycloheptane structure - Own the rest.
The process according to the invention is structured as follows, for example: A copper-magnesium alloy (according to Gilman) coated with anhydrous tetrahydrofuran is mixed with a solution of 1-methyl-piperidyl- (2) -ethyl chloride in tetrahydrofuran.
The formation of the Grignard compound is preferably promoted by adding a little ethylene bromide. The reaction mixture is then heated to boiling for a further 30 to 60 minutes. Instead of copper-magnesium alloy, magnesium shavings, which have preferably been etched with iodine vapors, can also be used.
The Grignard solution prepared in this way is then treated with a compound of formula II, e.g. B. dibenzo [a, d] - cyclohepta [1,4] diene - 5 - one or dibenzo [a, e] cycloheptatrien-5-one, added and the mixture heated for some time to complete the reaction. The reaction mixture is then hydrolyzed in the cold with aqueous ammonium chloride solution and extraheatly with a water-immiscible organic solvent, preferably chloroform.
The 5-hydroxy derivative of the formula IV obtained as an intermediate product can, if desired, be purified by crystallization and converted into suitable salts with inorganic or organic acids or further processed directly.
The water is split off, for example, by heating the substance dissolved in glacial acetic acid with conc. Hydrochloric acid. However, you can also use other agents such. B. with phosphorus oxychloride, thionyl chloride or zinc chloride. The end product is isolated and purified by known methods and, if desired, converted into acid addition salts and into quaternary compounds.
The new unsaturated dibenzocycloheptane derivatives prepared according to the process are oily or crystalline at Raurrite temperature. They are basic compounds that form stable, crystallized salts with inorganic or organic acids at room temperature.
The novel unsaturated dibenzocycloheptane derivatives prepared according to the invention and the hydroxy derivatives of the formula IV are suitable for use as medicinal products due to their excellent pharmacodynamic properties, in which they are characterized by diverse effects on the nervous system.
So antihistamine-like, narcosis-potentiating, adrenolytic, sedative, antipyretic, antiemetic, serotonin-inhibiting and hypothermic effects have been found, where these effects can vary depending on the structure of the basic side chain. They should be used therapeutically. In addition, the new compounds also serve as intermediates in the manufacture of drugs.
In the following examples, all temperatures are given in degrees Celsius and are uncorrected.
<I> Example 1 </I> 5- (2 '- [1 "-Methyl-piperidyl- (2")] -äthylidenl-dibenzo [a, d] cyclohepta [1,4] diene 2.8 g of activated copper -Magnesium alloy (according to Gilman) are covered with 15 cm3 of tetrahydrofuran and 0.4 cm3 of ethylene bromide is added.
As soon as the reaction has started, a solution of 16.2 g of 1-methyl-piperidyl- (2) -ethyl chloride in 20 cm3 of tetrahydrofuran is added dropwise within 20 minutes. After two hours of refluxing, a solution of 10.4 g of dibenzo [a; d] cyclohepta [1,4] dien-5-one (melting point 34-35) in 10 cm3 of tetrahydrofuran is added at 50. The mixture is then heated to the boil for a further 1 hour.
The cooled solution is then poured into 300 cm ?, a 10% ammonium chloride solution. Metal residues are filtered off, the filtrate is extracted three times with chloroform, the combined chloroform extracts are dried over potassium carbonate and the solution is evaporated. The oily residue is taken up in 200 cm3 of petroleum ether, the resinous flakes are filtered off, the filtrate is concentrated and cooled to -20.
On prolonged standing, the 5-hydroxy-5- @ 2 '- [1 "-methyl piperidyl- (2")] - ethyl} -dibenzo [a, d] cyclohepta [1,4] - crystallizes from the concentrated solution serve from m.p. 116-118.
The compound obtained is concentrated in 100 cm-3 of ice cream and 40 cm3. Dissolved hydrochloric acid and heated to boiling under reflux for 1 hour. The solution is then evaporated in vacuo, the residue was dissolved in water, the aqueous solution made alkaline with potassium hydroxide and extracted with ether.

Hydrobromide: The solution of the base in methanol is mixed with enough aqueous hydrobromic acid until the solution just reacts acidic to the Congo. After evaporation of the solution, the residue is taken up in acetone and the solution is slowly treated with ether, the 5-f2 '- [1 "-methyl-piperidyl- (2")] - ethylidene @ -dibenzo [a, d] cyclohepta [ 1,4] diene hydrobromide crystallized. After recrystallization from ethanol / ether, the hydrobromide melts at 189 to 190.
The iodine methylate is prepared by the solution of the base in abs. Ethanol mixed with the calculated amount of methyl iodide. M.p. 228-230 (decomp.) From methanol. <I> Example 2 </I> 5 - @ [1'-methyl-piperidyl (3 ')] - methylene @ - dibenzo [a, d] cycloheptadiene [1.4] 2.8 g of activated copper-magnesium alloy (after Gilman)

The ether extract, dried over potassium carbonate, is freed from the solvent by distillation, the remaining oil is dissolved in ethanol and the calculated amount of fumaric acid is added to the ethanolic solution. After the addition of ether, the acidic fumarate of 5 - f 2 '- [1 "-Methyl-piperidyl- (2")] - ethylidene @ -dibenzo- [a, e] cycloheptatriene crystallizes out.
After crystallization from methanol, it melts in 188190 (decomp.).

After the combined chloroform extracts have been dried over potassium carbonate and the chloroform has been evaporated off, the residue is recrystallized from acetone. 5-Hydroxy-5-1 [1'-methyl-piperidyl- (3, #] - methyl} -dibenzo [a, e] cycloheptatriene melts at 144-145.
13 g of this compound are concentrated in 125 cm3 of glacial vinegar and 50 cms. Hydrochloric acid heated to reflux for 1 hour. The solution is then evaporated in vacuo and the residue is crystallized from acetone / ether.
After repeated recrystallization from acetone or ethanol / ether, the 5 - {[1'-methyl - piperidyl - (3 ')] - methylene @ - dibenzo [a, e] cycloheptatriene hydrochloride melts at 237-239 (Dec.). The hydrochloride crystallizes from water with crystal water.
The base produced from the hydrochloride by shaking with chloroform and sodium hydroxide solution crystallizes from hexane or acetone in prisms with a melting point of 117-119.
<I> Example 5 </I> 5- {2 '- [1 "-Methyl-pyrrolidyl- (2")] - ethylidene.} - dibenzo [a, d] cyclohepta [1,4] diene a) 1, 3g magnesium shavings activated with iodine are the 5 cm3 abs. <I> Example 6 </I> 5- {2 '- [1 "-Methyl-pyrrolidyl- (2")] - ethylidene} - dibenzo [a, e] cycloheptatriene In the same way as described in Example 5 is obtained from 8.9 g of 1-methyl-pyrrolidyl- (2) -ethyl chloride and 10.3 g of dibenzo [a, e] cycloheptatrien-5-one (mp 89-90) the 5-hydroxy-5- {2'- [1 "-Methyl-pyrrolidyl- (2 '@] -ethyl} -dibenzo [a,
e] cycloheptatriene. The compound melts at 129-131 'after recrystallization from acetone.
The elimination of water from the 5-hydroxy derivative also takes place in the same way as described in Example 5. The 5- {2 '- [1 "-Methylpyrrolidyl- (2' @] - ethylidene @ - dibenzo [a, e] cycloheptatriene hydrobromide melts after crystallization from isopropanol / acetone at 164-166 (decomp.).
<I> Example 7 </I> 5 - {[I'-methyl-pyrrolidyl- (3 @] - methylene} - dibenzo [a, d] eyclohepta [1,4] d'ien a) 1.7 g with Iodine activated magnesium shavings with 5 cm3 abs. Tetrahydrofuran covered with a layer of 0.2 cm3 of ethylene bromide.
As soon as the reaction has started, the solution of 10.7 g of 1-methyl-pyrrolidyl- (3) -methyl chloride in 15% -tetrahydrofuran is added dropwise. After refluxing for one hour, the solution of 13.5 g of dibenzo [a, d] cyclohepta [1,4] dien-5-one in 15 cm3 of tetrahydrofuran is added dropwise at 40. The mixture is then heated to the boil for a further 2 hours.
The cooled reaction mixture is then poured into 150 cm3 of a 10% ammonium chloride solution. It is extracted several times with ether, the combined ether extracts are dried over sodium sulfate and the solution is evaporated.
The residue is recrystallized from acetone, 5-Hydroxy-5-1 [1'-methyl-pyrrolidyl- (3 ')] -methyl} -d'ibenzo [a, d] cyclohepta [1,4 ] diene of m.p. 134-135 is obtained.
b) 12 g of the compound obtained under a are treated with 100 cm3 of glacial acetic acid and 40 cm3 of conc. Hydrochloric acid heated to boiling under reflux for 1 hour. The solution is then evaporated in vacuo, the residue is dissolved in water, the aqueous solution is made alkaline and the separated substance is taken up in ether.
