Das Verfahren wird beispielsweise so ausgeführt, dass man die Lösung von 2-(1'-Methyl-piperidyl-2')- 1-chloräthan in einem wasserfreien offenkettigen oder cyclischen Äther, z. B. Diäthyläther oder Tetrahydrofuran, mit Magnesiumspänen, die bei hoher Temperatur, vorzugsweise zwischen 105-110 , im Vakuum getrocknet und mit Jod angeätzt wur den, versetzt, und das Reaktionsgemisch bei Rück flusstemperatur des angewandten Äthers behandelt.
Anstelle von mit Jod aktiviertem Magnesium kann auch eine Magnesium-Kupfer-Legierung (nach Gilman) verwendet werden. Die so bereitete Gri- gnardlösung wird bei Siedetemperatur portionen- weise mit dem im entsprechenden Äther gelösten oder suspendierten Thioxanthon-Derivat der Formel 1I versetzt, und das Reaktionsgemisch während meh rerer Stunden erhitzt.
Anschliessend entfernt man das Lösungsmittel im Vakuum, behandelt das Reak tionsgemisch in der Kälte mit wässeriger Ammonium chloridlösung und extrahiert anschliessend mit einem mit Wasser nicht mischbaren organischen Lösungs mittel, vorzugsweise Essigsäureäthylester. Nach Trocknen der Lösung wird das Lösungsmittel ver dampft, und das als Zwischenprodukt entstandene Thioxanthenal-9-Derivatgegebenenfalls durch Destil lation unter vermindertem Druck gereinigt.
Die Wasserabspaltung erfolgt durch Behandlung mit wasserentziehenden Mitteln, wie beispielsweise Phosphoroxychlorid, bei erhöhter Temperatur. Zur Isolierung des Endproduktes der Formel I wird die auf Zimmertemperatur abgekühlte Reaktionslösung auf Eis gegossen, mit einer Alkalihydroxydlösung alkalisch gemacht und mit einem mit Wasser nicht mischbaren organischen Lösungsmittel, vorzugsweise Chloroform, extrahiert. Nach Entfernen des Lösungs mittels wird das Endprodukt der Formel 1 durch Destillation unter vermindertem Druck gereinigt und gegebenenfalls in die sieroisomeren Formen getrennt und/oder gewünschtenfalls in Säureadditionssalze übergeführt.
Die erfindungsgemäss hergestellten Thioxanthen- Derivate sind bei Zimmertemperatur ölig oder fest. Es sind basische Verbindungen, die mit Säuren be ständige kristallisierte Salze bilden. Gewünschten falls können sie in ihre steroisomeren Formen ge trennt werden.
Die nach dem vorliegenden Verfahren hergestellten, bisher unbekannten Thioxanthen-Derivate sind auf Grund ihrer ausgezeichneten pharmakodynamischen Eigenschaften zur Verwendung als Heilmittel in hohem Masse geeignet. So wirken sie narkosepoten zierend, adrenolytisch und sedativ. Ausserdem zeich nen sie sich durch antihistaminartige, antipyretische und hypotherme Wirkungen aus.
Die als Ausgangsmaterialien zur Verwendung gelangenden Thioxanthon-Derivate der Formel 1I werden hergestellt, indem man Thiosalicylsäureester mit S-Alkyl-p-bromthiophenol kondensiert und das Reaktionsprodukt mit Phosphoroxychlorid cyclisiert.
In den nachfolgenden Beispielen erfolgen alle Temperaturangaben in Celsiusgraden und sind nicht korrigiert.
<I>Beispiel 1</I> 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl-2")- äthyliden-1']-thioxanthen a) 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyl-1']-thioxanthenol-(9). Zur Herstellung des Ausgangsmaterials, 2-Me- thylmercapto-thioxanthon-(9), wird Thiosalicylsäure mit p-Bromthioanisol in Gegenwart von Kupfer und einem Alkalicarbonat kondensiert, und das Reak tionsprodukt mit Thionylchlorid/Aluminiumchlorid cyclisiert.
In einer gut getrockneten Apparatur werden 2,43g Magnesiumspäne, die zuvor mit Joddämpfen aktiviert wurden, mit 20 cm?, abs. Tetrahydrofuran überschichtet, einige Tropfen Äthylenbromid zuge setzt und mit 29,5 g einer 50 z'r)igen Lösung von 2-(1'-Methylpyrrolidyl-2')-äthylchlorid im gleichen Lösungsmittel zur Reaktion gebracht. Das Reaktions gemisch wird 5 Stunden unter Rückfluss erhitzt, wo bei das Magnesium grösstenteils in Lösung geht.
Nun gibt man mittels Extraktionsapparatur 5,5 g 2-Me- thylmercapto-thioxanthon-(9) zu und erhitzt weitere 12 Stunden zum Sieden. Das Lösungsmittel wird nun unter vermindertem Druck teilweise abdestilliert, der Rückstand in eine etwa 20 ?; ige wässerige Lösung von Ammoniumchlorid eingetragen und dieses Ge misch mit Chloroform extrahiert. Aus dem Chloro- formextrakt wird nach dem Entfernen des Lösungs mittels das rohe 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyl-1']-thioxanthenol-(9) erhalten.
Zur Trennung der racemischen Isomeren werden 20 g der rohen Thioxanthenolverbindung in 50 cm3 abs. Äthanol gelöst und mit 3,13 g Fumarsäure auf gekocht. Durch fraktionierte Kristallisation wird das rohe Halbfumarat vom Smp. 187-189<B>'</B> (rac. a-Form) und das rohe Halbfumarat vom Smp. etwa 150 (rac. ss-Form) erhalten.
Nach viermaligem Um kristallisieren des Halbfumarates vom Smp. <B>187</B> bis l89 (rac. a-Form) aus 50 Ö igem Äthanol erhält man das analysenreine a-2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyl-1']-thioxanthenol-(9)-halbfumarat. Smp. 200,5\ (Zers.).
Das aus obigem Salz freigesetzte a-2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyl-1']-thioxanthenol-(9) schmilzt nach Kristallisation aus 95 'ö igem Äthanol bei 105,5-106,6''.
Das Halbfumarat vom Smp. etwa 150'F (rac. @- Form) löst man in heissem Isopropanol, filtriert von wenig schwerlöslichem r,-Isomerem ab und dampft die Lösung zur Trockne ein. Nach zweimaligem Um kristallisieren des Rückstandes aus abs. Äthanol schmilzt das analysenreine /)'-2 -Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyl-1']-thioxanthenol-(9)-halbfumarat bei 150-1531.
Das aus obigem Halbfumarat der rac. f-Form freigesetzte j3-2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyl-1']-thioxanthenol-(9) schmilzt, aus 95 O' igem Athanol umkristallisiert, bei 120,5-121-.
b) 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyliden-1']-thioxanthen. Zur Wasserabspaltung erhitzt man 10 g des oben erwähnten rohen Isomerengemisches der Thio- xanthenolbase mit 20 cm-- Acetanhydrid, dem 1,0 g Natriumacetat beigefügt wird, während 5 Stunden zum Sieden. Nach Entfernen des überschüssigen Acetanhydrids unter vermindertem Druck versetzt man mit Natriumcarbonatlösung und extrahiert mit Chloroform.
Nach Entfernen des Lösungsmittels aus dem Chloroformextrakt wird das 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyliden-1']-thioxanthen im Kugelrohr bei 180-185 /0,1 mm Hg destilliert.
Maleinat: 6,73 g der obigen im Kugelrohr destil lierten Thioxanthenbase und 2,32 g Maleinsäure werden in 34 cm3, siedendem Äthanol gelöst, filtriert und bei 0 kristallisiert. Das 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyliden-1']-thioxanthen-maleinat schmilzt nach zweimaligem Umkristallisieren aus abs. Äthanol bei 142-145 (Zerr.).
Fumarat: Smp. 153-156 nach Umkristallisieren aus abs. Athanol.
Sowohl aus der - als auch aus der P-Form von 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyl-1']-thioxanthenol-(9) wird durch Wasserabspaltung mit Acetanhydrid 2-Methylmercapto-9-[2'-(N-methylpyrrolidyl- 2")-äthyliden-1']-thioxanthen erhalten, das im Kugelrohr bei 0,1 mm Hg bei 180 bis 185 destilliert. Fumarat: Smp. 153-156 aus abs. Äthanol.
Maleinat: Smp. 142-143 (Zers.) nach zwei maligem Umkristallisieren aus abs. Äthanol. Der Misch-Smp. mit dem sub b erhaltenen Maleinat zeigt keine Depression.
<I>Beispiel 2</I> 2-Äthylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyliden-1']-thioxanthen Zur Herstellung des Ausgangsmaterials, 2-Athyl- mercaptothioxanthon-(9), wird Thiosalicylsäure mit S-Äthyl-p-brom-thiophenol in Gegenwart von Kupfer und einem Alkalicarbonat kondensiert und das Re aktionsprodukt mit Thionylchlorid/Aluminiumchlorid cyclisiert.
In einer gut getrockneten Apparatur werden 6,07g Magnesiumspäne, die zuvor mit Joddämpfen aktiviert wurden, mit 75 cm3 abs. Tetrahydrofuran überschichtet, einige Tropfen Äthylenbromid zuge setzt und mit 40,4 g 2-(1'-Methylpiperidyl-2')-äthyl- chlorid zur Reaktion gebracht. Das Reaktionsgemisch wird 11/., Stunden unter Rückfluss erhitzt, wobei das Magnesium grösstenteils in Lösung geht.
Nun gibt man mittels Extraktionsapparatur 27,2 g 2-Äthyl- mercapto-thioxanthon-(9) zu und erhitzt weitere 12 Stunden zum Sieden. Das Lösungsmittel wird nun unter vermindertem Druck teilweise abdestilliert, der Rückstand in eine etwa 20%ige wässerige Lösung von Ammoniumchlorid eingetragen und dieses Ge misch mit Chloroform extrahiert. Dieser Auszug liefert nach dem Entfernen des Lösungsmittels 42 g des rohen 2-Äthylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyl-1']-thioxanthenols-(9).
Halbfumarat: Smp. 180,5-181'.
Zur Wasserabspaltung erhitzt man 47,5 g der rohen Thioxanthenolbase mit 32,5 cm3 Acetanhy- drid, dem 1,6 g Natriumacetat beigefügt werden, während 5- Stunden zum Sieden. Nach Entfernen des überschüssigen Acetanhydrids unter vermindertem Druck versetzt man mit Natronlauge und extrahiert mit Chloroform. Aus dem Chloroformextrakt wird das Lösungsmittel abdestilliert, und der Rückstand im Kugelrohr bei 190-200 /0,03 mm Hg destilliert.
Zur weiteren Reinigung wird das Destillat an Alu miniumoxyd chromatographiert und mit einem Ge misch von vier Teilen Chloroform und einem Teil Cyclohexan eluiert. Maleinat: 4,5 g Base aus der chromatographi- schen Hauptfraktion werden mit 1,44 g Maleinsäure und 25 cms abs. Äthanol heiss gelöst und bei 0 kristallisieren gelassen.
Nach zweimaligem Um kristallisieren aus Äthanol/Äther schmilzt das analy senreine 2-Athylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyliden-1']-thioxanthen-maleinat bei 116-l18 (Zers.). <I>Beispiel 3</I> 2-Methyhnercapto-9-[2'-(N-methylpiperidyl- 2")-äthyliden-1']-thioxanthen a) 2-Methylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyl-1']-thioxanthenol-9. Zur Herstellung des Ausgangsmaterials, 2-Methyl- mercapto-thioxanthon-9,
wird Thiosalicylsäuremethyl- ester mit p-Bromthioanisol kondensiert und das Re aktionsprodukt mit Phosphoroxychlorid oder Alumi niumchlorid cyclisiert. In einer gut getrockneten Apparatur werden 0,79g Magnesiumspäne, die bei 110 getrocknet und mit Joddämpfen angeätzt wurden, mit 10 cm3 abs. Tetrahydrofuran über schichtet und innerhalb einer Stunde mit einem Ge misch von 5,24 g 2-(1'-Methylpiperidyl2')
-1-chlor- äthan und etwa 30 Tropfen Äthylbromid in 10 cm 3 abs. Tetrahydrofuran bei Rückflusstemperatur behan delt. Nachdem alles Magnesium in Lösung gegan gen ist, lässt man die Lösung von 1,8 g 2-Methyl- mercapto-thioxanthon-9 in 10 cms abs. Tetrahydro- furan zutropfen und erhitzt das Reaktionsgemisch zum Sieden am Rückfluss während 5 Stunden.
Nach Entfernen des Lösungsmittels im Vakuum wird der Rückstand mit einer eisgekühlten Ammoniumchlorid- lösung behandelt. Man extrahiert das Reaktionspro dukt mit Essigsäureäthylester, trocknet die Lösung über MgS04 und destilliert das Lösungsmittel ab.
Das 2-Methylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyl-1']-thioxanthenol-9 schmilzt nach Umkristallisieren aus Isopropanol bei 123-124 . Halb-Fumarat, Smp. 209 (Zers.). b) 2-Methylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyliden-1']-thioxanthen. Zu 4,11 g 2-Methylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyl-1']-thioxanthenol-9 werden 9,9 cm3 POCl,
beigefügt und 5 Stunden zum Sieden am Rückfluss erhitzt. Nach Abkühlen tropft man das Gemisch auf 70 g Eis, stellt mit NaOH- Lösung alkalisch, extrahiert mehrmals mit Chloro form, trocknet die Chloroformextrakte über Magne- siumsulfat und destilliert das 2-Methylmercapto-9-[2'-(N methylpiperidyl- 2")-äthyliden-1']-thioxanthen nach Entfernen des Lösungsmittels im Kugelrohr bei einem Druck von 0,
04 Torr und einer Badtempe- ratur von 180-190 . Sdp. 219 /0,05 Torr, gelbes Öl, das glasig erstarrt.
10 g des erhaltenen Isomerengemisches von cis- und trans- 2-Methylmercapto-9-[2'-(N methylpiperidyl- 2")-äthyliden-1']-thioxanthen werden in 30 cm3 Aceton gelöst und bei 0 stehen gelassen, bis keine weiteren Kristalle mehr ausfallen. Nach Umkristallisieren aus Aceton erhält man die eine Isomerenform analysenrein vom Smp.105-107 .
Zur Herstellung des Maleinates vereinigt man die heissen Lösungen von 3,0 g 2-Methylmercapto-9-[2'-(N methylpiperidyl- 2")-äthyliden-1']-thioxanthen (Smp. 105-107 ) in 8 cm3 Zsopropanol und 1,
0 g Maleinsäure in 10 cm3 Isopropanol. Das im Eis schrank auskristallisierte Maleinat zeigt nach Um kristallisieren aus 40 cm3 Isopropanol den kon°tan- ten Smp. 157-159 . Fumarat: Smp. l41-143 aus Äthanol. Isomeres B: Die nach Abtrennung des Isomeren A erhaltene Aceton-Mutterlauge wird zur Isolierung des Isomeren B eingeengt.
Der Rückstand (6,5 g) wird in 16 em3 Benzol/Petroläther (1 : 1) gelöst und an einer Säule von 185g Aluminiumoxyd adsorbiert. Nachdem noch vorhandenes Isomeres A mit 585 cm3 Benzol/Petroläther (1:
l) abgetrennt worden ist, wird mit 455 cm3 Benzol das Isomere B eluiert. Die Benzolfraktion wird eingeengt und der Rückstand mit 0,40 g Fumarsäure zusammen in 7,8 cms sie dendem abs. Äthanol gelöst, filtriert und zum Aus kristallisieren auf 0 abgekühlt.
Die ausgeschiedenen Kristalle liefern nach zweimaligem Umkristallisieren aus abs. Äthanol das analysenreine Isomere B des 2 Methylmercapto-9-[2'-(N-methylpiperidyl- 2")-äthyliden-1']-thioxanthen-fumarats (C22H25NS2 ' 1/2 C4H404) vom konstanten Smp. 169 bis 171 .

The thioxanthene derivatives prepared according to the invention are oily or solid at room temperature. They are basic compounds that form constant crystallized salts with acids. If desired, they can be separated into their steroisomeric forms.
The hitherto unknown thioxanthene derivatives produced by the present process are highly suitable for use as medicaments because of their excellent pharmacodynamic properties. They have a decorative, adrenolytic and sedative effect. They are also characterized by antihistamine-like, antipyretic and hypothermic effects.
The thioxanthone derivatives of the formula 1I which are used as starting materials are prepared by condensing thiosalicylic acid esters with S-alkyl-p-bromothiophenol and cyclizing the reaction product with phosphorus oxychloride.
In the following examples, all temperatures are given in degrees Celsius and have not been corrected.
<I> Example 1 </I> 2-Methylmercapto-9- [2 '- (N-methylpyrrolidyl-2 ") - ethylidene-1'] - thioxanthene a) 2-Methylmercapto-9- [2 '- (N- methylpyrrolidyl- 2 ") - ethyl-1 '] - thioxanthenol- (9). Maleate: 6.73 g of the above thioxanthene base distilled in a bulb tube and 2.32 g of maleic acid are dissolved in 34 cm3 of boiling ethanol, filtered and crystallized at 0. The 2-methylmercapto-9- [2 '- (N-methylpyrrolidyl- 2 ") - ethylidene-1'] - thioxanthene maleate melts after two recrystallization from absolute ethanol at 142-145 (Zerr.).
Fumarate: m.p. 153-156 after recrystallization from abs. Ethanol.
Both the - and the P-form of 2-methylmercapto-9- [2 '- (N-methylpyrrolidyl- 2 ") - ethyl-1'] -thioxanthenol- (9) becomes 2-methylmercapto by elimination of water with acetic anhydride -9- [2 '- (N-methylpyrrolidyl- 2 ") - ethylidene-1'] - thioxanthene, which distills at 180 to 185 in a bulb tube at 0.1 mm Hg. Fumarate: m.p. 153-156 from abs. Ethanol.
Maleate: m.p. 142-143 (decomp.) After recrystallizing twice from abs. Ethanol. The mixed smp. with the maleate obtained with sub b shows no depression.
<I> Example 2 </I> 2-Ethylmercapto-9- [2 '- (N-methylpiperidyl- 2 ") - ethylidene-1'] - thioxanthene For the preparation of the starting material, 2-ethyl mercaptothioxanthone- (9), thiosalicylic acid is condensed with S-ethyl-p-bromothiophenol in the presence of copper and an alkali metal carbonate and the reaction product is cyclized with thionyl chloride / aluminum chloride.
In a well-dried apparatus, 6.07 g of magnesium shavings, which were previously activated with iodine vapors, are mixed with 75 cm3 abs. Tetrahydrofuran is covered, a few drops of ethylene bromide are added and made to react with 40.4 g of 2- (1'-methylpiperidyl-2 ') - ethyl chloride. The reaction mixture is heated under reflux for 11/1 hours, the magnesium for the most part going into solution.
27.2 g of 2-ethyl-mercapto-thioxanthone- (9) are now added using an extraction apparatus and the mixture is heated to the boil for a further 12 hours. The solvent is now partially distilled off under reduced pressure, the residue is introduced into an approximately 20% strength aqueous solution of ammonium chloride and this mixture is extracted with chloroform. After removing the solvent, this extract gives 42 g of the crude 2-ethylmercapto-9- [2 '- (N-methylpiperidyl- 2 ") - ethyl-1'] - thioxanthenols- (9).
Semi-fumarate: m.p. 180.5-181 '.
To split off water, 47.5 g of the crude thioxanthenol base are heated to the boil for 5 hours with 32.5 cm 3 of acetanhydride, to which 1.6 g of sodium acetate are added. After removing the excess acetic anhydride under reduced pressure, sodium hydroxide solution is added and the mixture is extracted with chloroform. The solvent is distilled off from the chloroform extract and the residue is distilled in a bulb tube at 190-200 / 0.03 mm Hg.
For further purification, the distillate is chromatographed on aluminum oxide and eluted with a mixture of four parts of chloroform and one part of cyclohexane.
