EMI0001.0015
Process for the preparation of phenoxazine derivatives In French patent N 1242849, the holder described a process for the preparation of 3-mono alkyl-amino-7-dialkoylamino-N-alkyl-phenoxazim-2, of formula VI
EMI0001.0006
in which R, R ', R "and R' are simple or branched aliphatic radicals comprising from 2 to 4 carbon atoms, R and R 'possibly being united in the form of a piperidic or pyrrolidic ring optionally bearing branches , these compounds being of interest because of the anti-tuberculosis activity with which they are endowed.
However, it has now been found, and this forms the subject of the present invention, that these compounds can be accessed by a shorter route.
The preparation process according to the present invention consists in converting the 5-nitro-benzimidazol of formula I
EMI0001.0015
as a quaternary ammonium salt of 5-nitro N, N'-di-alkyl-benzimidazol, of formula II,
preferably under the action of an alkyl halide on the potassium derivative
EMI0001.0020
One then carries out the opening of the penta gonal ring with elimination of a molecule of formal acid in general by means of a strong alkali -and obtains a 3-nitro-o-phenylene-diamine of formula III
EMI0001.0024
By reduction of the phenylene diamine of formula III, the latter is converted into the corresponding triamins of formula IV
EMI0001.0027
including condensation in general in a homogeneous phase and a buffered medium, in pyridine,
with a 2-nitroso-5-dialkoyl-amino-phenol of formula V
EMI0001.0029
provides the expected 3-monoalkylamino-7, dialkoylamino-N-alkyl-phenoxazimes-2 expected by the above-mentioned formula VI.
Example 1 7-Diethyla zino-3-butylatnirao-N-butylphenoxazim-2 (formula VI: R and R '= ethyl, R "= butyl, R"' = butyl) 5-nitrobenzimidazol is butylated by heating for 3 hours in water bath in methyl ethyl ketone with butyl bromide, in the presence of potassium carbonate, giving a pale yellow body, with a melting point of 78o C.
Nitrobutylbenzidazol, heated for 3 hours at 130 ° C. of butyl iodide, gives the crystallized quaternary ammonium iodide of formula
EMI0002.0018
By boiling for half an hour with 3N alcoholic soda, a molecule of formic acid is removed and 3-nitro-N, N'-dibutyl-o-phenylene-di-amine precipitates on cooling in the form of crystals. brick red with a melting point of 76o C.
By catalytic reduction (platinum -1- hydrogen) in a hydrochloric alcoholic medium, the corresponding triaminobenzene dihydrochloride is obtained, in the form of white crystals, by adding dioxane to the highly concentrated alcoholic solution.
308 g (1 mol.) Of this dihydrochloride are dissolved in 750 cm 3 of pyridine. Into this hand held solution below 25 ° C., with good stirring, is introduced dropwise a solution of 232 g of 2-nstroso-5-diethyl-aminophenol in 750 cm3 of pyridine. A blue coloration occurs immediately and intensifies over the following hours.
After standing overnight, 1200 cm3 of methanol, 2500 cm3 of petroleum ether, 200 g of diatomaceous earth and, with vigorous mechanical stirring, 1000 cm 2 of 10N sodium hydroxide are added, then, gradually, 4000 cm3 of water. After 30 minutes of stirring, poured onto a filter, the two layers are allowed to settle and the brick red petroleum ether is separated. The diatomaceous earth is again extracted with petroleum ether, the ethers together which are washed with water, then with soda, then with water.
Distilled petroleum ether leaves a residue weighing 104 g which partially crystallizes. The crystals collected are recrystallized from aqueous acetone at 80 9 / o. (Red needles of melting point 1120 C.) The product is not described in the literature. In this particular case, one can advantageously isolate the dye in the phosphate state by stirring the ethereal solution with a solution of monosodium phosphate or phosphoric acid, the acid phosphate separates into brownish green crystals, almost insoluble in water and acetone, poorly soluble in alcohol.
Example 2 7-Piperidiiio-3-birtylattzino-N-propylphenoxaziitze-2 Formula VI:
EMI0002.0052
(3-Hydroxyphenyl) -piperidine is nitrosated in the usual manner and the hydrochloride of the 4-nitrose derivative is condensed with 1-butylamino-2-propylamino-4-aminobenzene dihydrochloride, itself obtained by reduction with zinc in hydrochloric alcohol of the corresponding nitro derivative of melting point 74 C.
EMI0002.0060
This is prepared as it was said in Example 1, by replacing butyl iodide, by propyl iodide, to obtain the quaternary iodide.
The condensation is carried out in pyridine as above, the final extraction with petroleum ether results in an incrystallizable red oil which is subjected to chromatography in benzene on an alumina column.
The sought dye goes overhead and is obtained crystallized by distillation of benzene. It is recrystallized from petroleum ether, it melts at 760 C.
It is not described in the literature. Example 3 7-Dibutylatnino-3-propylatnitio-N-ethylphéiioxazirne-2 (formula VI: R and R '= butyl, R "= propyl, R"' = ethyl) The starting materials for the condensation.
here are 2-nitroso-5-dibutyl-ni-amino-phenol hydrochloride (pale yellow, well crystallized) and 1-propylam.ino-2-ethylamino-4-aminobenzene dichlorhydrate, obtained by reduction catalytic converter of the nitro derivative with a melting point of 106o C, itself obtained by the same series of operations as in Example 1.
The treatment of the condensation product and the chromatography are carried out as before, but the low melting point (around 300 C) and the extreme solubility of bhenoxazim make its isolation difficult. base state, the dye is therefore isolated in the phthalate state within the dioxane. We obtain green crystals, golden brown, hardly soluble in water.
This product is not described in the literature.
