Diese Sulfonamide, insbesondere das bekannte 4- Sulfanilamido-2,6-dimethoxy-pyrimidin, zeichnen sich durch hohe Wirksamkeit gegen pathogene Keime aus.
Das erfindungsgemässe Verfahren ist dadurch ge kennzeichnet, dass man eine Polyhalogenverbindung der Formel

in welcher R ein Halogenatom, eine Alkoxygruppe oder eine Alkylthiogruppe darstellt, mit einem Alkali salz von N4 Aeylsulfanilamid oder von Sulfanilamid umsetzt und das Umsetzungsprodukt mittels eines Al kohols der Formel Rl-OH der Alkoholyse unter wirft, wobei eine im Umsetzungsprodukt gegebenen falls vorhandene N4 Acylgruppe vor oder nach der Alkoholyse abgespalten wird.
Dihalogenverbindungen der obigen allgemeinen Formel 11 kann man z. B. wie folgt gewinnen: Ein 2-Alkoxy-4,6-dihydroxy-pyrimidin bzw. 2 Alkylthio-4,6-dihydroxy-pyrimidin wird mit einem Phosphorhalogenid in Gegenwart eines basischen Mittels behandelt, wodurch das entsprechende 2-Alk- oxy-4,6-dihalogen-pyrimidin bzw. 2-Alkylthio-4,6-di- halogen-pyrimidin entsteht.
2,4,6-Trihalogen-pyrimidine sind bekannte Ver bindungen.
Die Umsetzung der genannten Polyhalogenver- bindungen mit dem Alkalisalz von Sulfanilamid oder dessen Acylderivat erfolgt mit Vorteil in einem Lö sungsmittel, welches das Alkalisalz des Acylsulfanil- amids bzw. Sulfanilamids wenigstens teilweise löst, selbst jedoch an der Reaktion nicht teilnimmt. Zweck mässig haben sich für diesen Zweck Dimethylform- amid, Acetamid oder Dioxan erwiesen.
Zwecks Erzielung einer günstigen Ausbeute und zur Gewinnung reiner Endprodukte ist es vorteilhaft, die Ausgangsmaterialien im Verhältnis von 1 Mol der Polyhalogenverbindung zu 2 Mol des Alkalisalzes von N4 Acylsulfanilamid bzw. Sulfanilamid für die erfindungsgemässe Reaktion einzusetzen. Bei Anwen dung anderer Mengenverhältnisse sinken sowohl Aus beute als auch Reinheit der Reaktionsprodukte.
Ge mäss einer besonders günstigen Ausführungsform der Erfindung lässt man die Umsetzung, ausgehend von 2-Alkoxy-4,6-dihalogen-pyrimidin bzw. 2-Alkylthio- 4,6-dihalogen-pyrimidin während einiger Stunden bei 80-120 in Dimethylformamid oder Acetamid vor sich gehen, digeriert die Reaktionsmischung an schliessend mit Wasser, trennt das dabei anfallende überschüssige Sulfanilamid bzw.
Acylsulfanilamid ab und gewinnt aus dem Filtrat 2-Alkoxy-4-(N4 acyl- sulfanilamido)-6-halogen-pyrimidin bzw. 2-Alkoxy-4- sulfanilamido-6-halogen-pyrimidin bzw. 2-Alkylthio- 4-sulfanilamido-6-halogen-pyrimidin durch Ansäuern. Wenn man das gewonnene Reaktionsprodukt, z. B.
das durch Umsetzung von 2-Methoxy-4,6-chlor-pyri- midin mit Acetylsulfanilamidnatrium erhältliche 2- Methoxy - 4-(N4 acetyl-sulfanilamido)-6-chlor-pyrinü- din, mit Alkalimethylat in Methanol unter Druck er- wärmt und anschliessend mit wässerigem Alkali be handelt, bildet sich das bekannte 2,6-Dimethoxy-4- sulfanilamido-pyrimidin. In gleicher Weise,
jedoch bei entfallender Behandlung mit wässerigem Alkali kann aus dem durch Umsetzung von 2-Methoxy-4,6- dichlor-pyrimidin mit Sulfanilamidnatrium herstell- baren 2 - Methoxy - 4-sulfanilamido-6-chlor-pyrimidin das 2,6-Dimethoxy-4-sulfanilamido-pyrimidin gewon nen werden.
Eine andere günstige Ausführungsform der Erfin dung besteht darin, dass man die Umsetzung, aus gehend von 2,4,6-Trihalogen-pyrimidin, bei Raum temperatur in Dimethylformamid vor sich gehen lässt, das Reaktionsgemisch anschliessend mit Wasser be handelt, das überschüssige Acylsulfanilamid mittels Kohlendioxyd abscheidet und abtrennt, worauf man aus dem Filtrat durch Ansäuern 2,6-Dihalogen-4-(N4 acyl-sulfanilamido)-pyrimidin gewinnt.
Wenn man nun das 2,6-Dihalogen-4-(N4 acyl-sulfanilamido)-py- rimidin, insbesondere das durch Umsetzung von 2,4, 6-Trichlor-pyrimidin mit Acetylsulfanilamidnatrium erhältliche 2,6 - Dichlor - 4-(N4 acetyl-sulfanilamido)- pyrimidin mit Alkalimethylat in Methanol bei Raum temperatur ohne Druck reagieren lässt, entsteht 2- Methoxy - 4-(N4 acetyl-sulfanilamido)
-6-chlor-pyrimi- din, welches bereits gemäss der weiter oben angegebe nen Arbeitsweise erhältlich war.
Von diesem 2 - Methoxy - 4 - (N4 acetyl-sulfanil- amido)-6-chlor-pyrimidin kann man die Acetylgruppe durch Behandlung mit etwa 0,5n Natronlauge bei etwa 95-100 während etwa 1-4 Stunden abspalten, wodurch das neue 2 - Methoxy - 4 - sulfanilamido-6- chlor-pyrimidin erhalten wird.
Das durch Umsetzung von 2,4,6-Trichlor-pyri- midin mit Acetylsulfanilamidnatrium nach der oben angeführten Arbeitsweise gewonnene 2,6-Dichlor-4- (N4 acetyl-sulfanilamido)-pyrimidin kann man in ab solut alkoholischer Lösung der Einwirkung von Säure aussetzen, wodurch die Acetylgruppe abgespalten wird und 2,6-Dichlor-4-sulfanilamido-pyrimidin ent steht.
Am besten geht man dabei so vor, dass das 2,6 Dichlor-4-(N4 acetyl-sulfanilamido)-pyrimidin mit ge sättigter methanolischer Salzsäure während einiger Stunden zum Rückfluss erhitzt und das gebildete 2,6- Dichlor - 4 - sulfanilamido-pyrimidin-hydrochlorid er wünschtenfalls in die freie Base übergeführt wird. Letztere kann durch Erwärmen mit Alkalimethylat in Methanol unter Druck in das bekannte 2,6-Dimeth- oxy-4-sulfanilamido-pyrimidin übergeführt werden.
Behandelt man das 2,6-Dichlor-4-sulfanilamido-pyri- midin dagegen mit Alkalimethylat in Methanol bei Raumtemperatur ohne Anwendung von Druck, wird nur ein Chloratom durch die Methoxygruppe ersetzt, und es entsteht das obenerwähnte 2-Methoxy-4-sul- fanilamido-6-chlor-pyrimidin.
Das durch Umsetzung von 2,4,6-Trichlor-pyri- midin mit Acetylsulfanilamidnatrium erhaltene 2,6- Dichlor - 4 - (N4 acetyl-sulfanilamido)-pyrimidin kann man direkt durch Erwärmen mit Alkalimethylat in Methanol unter Druck und anschliessender Behand- lung mit wässrigem Alkali in das bekannte 2,6-Di- methoxy-4-sulfanilamido-pyrimidin überführen.
Zu diesem Zwecke ist es empfehlenswert, das 2,6-Di- chlor-4-(N4 acetyl-sulfanilamido)-pyrimidin mit über schüssigem Alkalimethylat in Methanol auf über 100 und unter Druck während einiger Stunden zu er wärmen, den Alkohol abzudestillieren, den Rück stand mit wässerigem Alkali in der Wärme zu be handeln, worauf beim Ansäuern der Reaktions mischung das 2,6-Dimethoxy-4-sulfanilamido-pyri- midin abgeschieden wird.
Verwendet man als Polyhalogenverbindung ein 2-Alkylthio-4,6-dihalogen-pyrimidin, insbesondere 2- Methylthio-4,6-dichlor-pyrimidin, und behandelt das durch Umsatz mit einem Alkalisalz von Sulfanilamid bzw. von N4 Acyl-sulfanilamid, insbesondere N4 Ace- tyl-sulfanilamid, erhältliche 2-Alkylthio-6-halogen-4- sulfanilamido- bzw.
4-(N4 Acyl-sulfanilamido)-pyrimi- din mit Alkalimethylat in der Wärme und unter Druck, so erhält man, wie gefunden wurde, in über wiegender Menge 2,6 - Dimethoxy - 4 - sulfanilamido- bzw. 4-(N4- Acyl-sulfanilamido)-pyrimidin. Das N4 Acylderivat kann sodann mittels wässerigem Alkali verseift werden.
Bei der Methanolyse der Alkylthioverbindung wird somit nicht nur das 6-Halogenatom durch die Methoxygruppe ersetzt, sondern, mindestens teil weise, auch die 2-Alkylthiogruppe. <I>Beispiel 1</I> Eine Lösung von 33g O-Methyl-harnstoffchlorid in 100 ml abs. Methanol wird auf -8 (Badtemp.) abgekühlt.
Unter Rühren lässt man langsam eine Lö sung von 13,8g Natrium in 140 ml abs. Methanol zutropfen. Sofort darauf werden 39,6 g Malonsäure- dimethylester rasch zum Reaktionsgemisch zugeführt. Man lässt noch 4 Stunden im Eisbad stehen und be wahrt den Ansatz 3 Tage unter Feuchtigkeitsaus schluss bei Zimmertemperatur auf. Die Kristallmasse wird abgenutscht. Das Filtrat wird mit verdünnter Salzsäure neutralisiert und im Vakuum zur Trockne eingedampft.
Die beiden vereinigten Kristallrück- stände werden mit 100 ml Wasser versetzt und, falls nötig, erneut mit verdünnter Salzsäure eine Reaktion von pH 4-5 eingestellt. Die durch Kühlung vervoll ständigte Abscheidung wird abgenutscht und mit 35 ml Eiswasser nachgewaschen. Der Kristallbrei von 2-Methoxy-4,6-dihydroxy-pyrimidin wird ab gesaugt, im Vakuum auf dem Wasserbad vorgetrock net und im Trockenschrank bei 105 vollständig was serfrei gemacht.
Ausbeute über 80 %; Zersetzungs punkt 190 (unter Paraffinbläschenbildung). 7,2 g des staubfein getrockneten 2-Methoxy-4,6- dihydroxy-pyrimidins werden vorsichtig in eine Mischung von 25 ml Phosphoroxychlorid und 12,5 ml Dimethylanilin eingetragen. Man kühlt den Kolben von aussen, damit die Reaktion nicht plötzlich erfolgt.
Lässt die Wärmeentwicklung nach, so erwärmt man mit aufgesetztem Kühler zunächst eine Stunde bei l05 und dann 11/2 Stunden bei 125 auf dem Ölbad unter Rückfluss (Siedesteinchen). Nach Abkühlen wird der Kolben mit absteigendem Kühler versehen und überschüssiges Phosphoroxychlorid im Vakuum von 12 mm bei etwa 50 abdestilliert. Der dickflüs sige Kolbeninhalt wird auf Eis gegossen. Es scheiden sich Kristalle ab. Nach 1/9 Stunde wird mit Äther überschichtet und im Schütteltrichter dreimal mit Äther ausgeschüttelt.
Die vereinigten Ätherauszüge werden zweimal mit Wasser neutral gewaschen und mit Natriumsulfat getrocknet. Nach Abdestillieren des Äthers wird im Vakuum destilliert. Dabei geht das 2- Methoxy - 4,6 - dichlor - pyrimidin bei<B>1051111</B> mm über. In der Vorlage erstarrt es zu einer farblosen Masse vom Schmelzpunkt 59 . Ausbeute 6,4 g (70%).
16,1 g (2 Mol) N4 Acetyl-sulfanilamidnatrium werden in 70 ml trockenem Dimethylformamid ein getragen und unter gelegentlichem Schütteln im Öl bad auf 130' erwärmt. Es löst sich unvollständig, nach Abkühlen auf 80 werden unter Rühren 6,1 g 2-Methoxy-4,6-dichlor-pyrimidin (1 Mol) eingetra gen. Dabei entsteht eine feine Suspension. Es wird auf 105 erwärmt und 4 Stunden bei dieser Temperatur belassen.
Dann wird das Dimethylformamid im Va kuum auf dem Wasserbad abdestilliert. Der sirup artige Rückstand wird mit Wasser digeriert (etwa 45 ml) wobei sich das überschüssige N4 Acetyl- sulfanilamid kristallin abscheidet. Die Lösung zeigt alkalische Reaktion. Man lässt 1/2 Stunde im Eis schrank stehen und saugt ab. Das Filtrat wird mit Eisessig angesäuert, wobei sich das 2- Methoxy-4- (N4 - Acetylsulfanilamido)-6-chlor-pyrimidin abschei det.
Es fällt zuerst harzig aus, kristallisiert dann aber in harten Brocken, die in der Mutterlauge zerrieben werden. Der weisse Niederschlag wird abgesaugt und gewaschen. 11,4 g Rohausbeute, Schmelzpunkt 230 . Zur Analyse löst man in Sodalösung und trägt in überschüssige verdünnte Salzsäure ein; der ge waschene und getrocknete Niederschlag wird unter Vermeidung unnötig langer Erhitzungszeit aus 80%igem Alkohol umgelöst; Schmelzpunkt 233 .
6 g des erhaltenen 2-Methoxy-4-(N4 acetyl-sulfa- nilamido)-6-chlor-pyrimidins werden mit einer Lö sung von 1,95g Natrium (5 Mol) in 35 ml absolutem Methanol 5 Stunden im Glasautoklaven bei 120 im Ölbad erhitzt. Beim Abkühlen setzt sich Kochsalz am Boden des Gefässes ab. Das Methanol wird im Vakuum abdestilliert und der Rückstand mit 17 ml In Natronlauge aufgenommen und bei 90 auf dem Wasserbad 1 Stunde verseift.
Man versetzt mit etwas Kohle, filtriert und säuert mit Eisessig an; durch Fil tration des Niederschlages erhält man 4,95 g 2,6- Dimethoxy-4-sulfanilamido-pyrimidin vom Schmelz- punkt 194-196 . Dieses Produkt wird mit einer zur Lösung unzureichenden Menge 96 % igen Alkohols einige Zeit aufgekocht und darauf mit Wasser bis auf etwa 80 Ö igen Alkohol verdünnt; nach 12 Stunden Stehen im Eis resultieren 4,6 g reines Produkt vom Schmelzpunkt 202 .
<I>Beispiel 2</I> 3 g 2 - Methoxy - 4 - (N4 acetyl-sulfanilamido)-6- chlor-pyrimidin (gemäss Beispiel 1 hergestellt) wer den in 67,2 ml 0,5n Natronlauge (4 Mol) gelöst und im Wasserbad 3 Stunden bei 98 Badtemperatur er hitzt. Nach dieser Zeit wird die klare Lösung von aussen mit Eiswasser gekühlt und unter Rühren in Salzsäure (12 ml konz. Salzsäure und 21 ml Wasser) eingetropft. Es scheiden sich weisse Flocken ab. Man lässt 1 Stunde im Eisschrank stehen und saugt ab.
Unter Eiskühlung und Rühren wird das Filtrat trop fenweise mit konz. Ammoniak auf pl=I 4-5 neutrali siert. Dabei fällt 2-Methoxy-4-sulfanilamido-6-chlor- pyrimidin flockig aus. Man lässt über Nacht im Eis schrank stehen, wobei der Niederschlag kristallisiert. 2,1g Rohausbeute (Schmelzpunkt 160-166 ). Nach Umkristallisieren aus Methanol-Wasser resultieren 1,85 g vom Schmelzpunkt 174 .
Diese Verbindung kann, wie im Beispiel 6 beschrieben, durch Methano- lyse in 2,6 - Dimethoxy - 4 - sulfanilamido - pyrimidin übergeführt werden. <I>Beispiel 3</I> 48 g trockenes, fein gepulvertes N4 Acetyl-sulfa- nilamidnatrium werden in 160 ml Dimethylformamid suspendiert und die Mischung auf -15 gekühlt. Unter Schütteln werden sodann 18,2 g 2,4,6-Trichlor- pyrimidin innert 10 Minuten zugetropft. Dabei steigt trotz Aussenkühlung die Temperatur auf - 5 an.
Unter gelegentlichem Schütteln lässt man sodann 1 Stunde bei 0 und 30 Minuten bei 20 stehen, wobei fast alles in Lösung geht. Der Ansatz wird sodann auf 1 Liter Wasser gegossen (fast neutrale Reaktion) und mit Kohlendioxyd völlig neutralisiert. Nach 2 Stunden Stehen bei 0 wird das ausgeschiedene N4 Acetyl-sulfanilamid isoliert. Das Filtrat- wird mit ver dünnter Salzsäure (10 ml konz. Salzsäure und 40 ml Wasser) angesäuert und 1-2 Stunden bei 0 auf bewahrt.
Die Filtration ergibt 36 g rohes 2,6-Dichlor- 4 - (N4 - acetyl-sulfanilamido)-pyrimidin vom Zerset zungspunkt 238-245 (Tröpfchen ab 230 ). Das Rohprodukt wird in einem Gemisch von 360 ml Al kohol und 50 ml Wasser heiss gelöst. Nach Abkühlen auf 20 und Filtration gewinnt man 30 g des Pro duktes vom Zersetzungspunkt 238-240 (Reste bis 248 ). Aus der Mutterlauge erhält man beim Kühlen auf 0 noch 1 g des gleichen Produktes; Gesamtaus beute 31 g.
30 g des erhaltenen 2,6-Dichlor-4-(N4 acetyl- sulfanilamido)-pyrimidins werden mit 150 ml abs. Methanol und 65 ml 8 % iger absolut methanolischer Salzsäure zum Rückfluss erhitzt. Nach 10 Minuten ist vollständige Lösung eingetreten, nach 15-20 Minuten beginnt neue Kristallisation unter heftigem Stossen des Ansatzes. Nach einer Kochdauer von 110 Minuten lässt man auf 20 auskühlen und filtriert.
Man erhält so 20 g 2,6-Dichlor-4-sulfanilamido-pyrinüdin-hydro- chlorid vom Zersetzungspunkt 180-200 (67 % der Theorie). 20g Hydrochlorid werden mit Natriumbicarbo- natlösung (200 ml Wasser) und 10g Natriumbicar- bonat übergossen, wobei unter Kohlendioxyd-Ent- wicklung bis auf geringe Reste alles in Lösung geht. Nach der Filtration wird mit Essigsäure (10 ml Eis essig und 10 ml Wasser) angesäuert. Nach einigem Stehen auf Eis wird filtriert.
Man erhält 17,8 g der rohen Base vom Schmelzpunkt 204-208 . Das Roh produkt wird in einem Gemisch von 200 ml Alkohol und 15 ml Wasser heiss gelöst und die klare Lösung mit 220 ml heissem Wasser versetzt. Nach Abkühlen auf 0 und Filtrieren erhält man 14,7 g reines 2,6- Dichlor - 4 - sulfanilamido - pyrimidin vom Schmelz punkt 208 . Aus der Mutterlauge lassen sich durch teilweises Abdestillieren des Alkohols noch 1,2 g vom Schmelzpunkt 199-206 gewinnen.
1,6 g des erhaltenen 2,6-Dichlor-4-sulfanilamido- pyrimidins vom Schmelzpunkt 208 werden zu 25 ml abs. Methanol gegeben, welches 0,46 gNatrium enthält. Sodann wird im Glasautoklaven 7 Stunden auf 125 (Radtemperatur) erhitzt. Hierauf wird der Methyl alkohol im Vakuum abdestilliert und der Rückstand in 20 ml Wasser gelöst. Nach Filtration von einer geringen Trübung wird mit verdünnter Salzsäure kongosauer gemacht, wobei nach vorübergehender Fällung alles wieder in Lösung geht. Schliesslich wird die saure Lösung mit Ammoniak neutralisiert.
Man erhält so nach dem Trocknen 1,45 g 2,6-Dimethoxy- 4-sulfanilamido-pyrimidin vom Schmelzpunkt 190 bis 196 . Durch Umlösen aus verdünntem Alkohol steigt der Schmelzpunkt auf 198-201'.
<I>Beispiel 4</I> 1 g 2,6-Dichlor-4-sulfanilamido-pyrimidin (her gestellt nach Beispiel 3) werden zu einer eiskalten Lösung von 0,33g Natrium in 10 ml abs. Methanol gegeben. Die klare Lösung wird 1 Stunde bei 0 be lassen und dann 14 Stunden bei 20 aufbewahrt (Ab scheidung von Natriumchlorid). Hierauf destilliert man ungefähr die Hälfte des Lösungsmittels im Va kuum bei 30 ab und versetzt mit verdünnter Essig säure (2 ml Eisessig und 20 ml Wasser).
Nach 1 Stunde Stehen bei 20 werden die ausgeschiedenen Kristalle isoliert. Man erhält so 0,89 g 2-Methoxy-4- sulfanilamido - 6 - chlor-pyrimidin vom Schmelzpunkt 155-170 . Durch Umlösen aus Methanol steigt der Schmelzpunkt auf 171-174 . Die erhaltene Verbin dung kann, wie im Beispiel 6 beschrieben, durch Methanolyse in 2,6-Dimethoxy-4-sulfanilamido-pyri- midin übergeführt werden.
<I>Beispiel 5</I> 4,8 g trockenes, fein gepulvertes N4 Acetyl-sulfa- nilamidnatrium werden in 20 ml absolutem Dioxan suspendiert und mit 1,12 ml 2,4,6-Trichlor-pyrimidin versetzt. Unter gutem Rühren wird innert 20 Minuten auf 80 erhitzt (Feuchtigkeitsausschluss). Nach 11/2 Stunden Erhitzen (80-85 ) wird das Dioxan im Va kuum abdestilliert, der Rückstand in 75 ml Wasser gelöst und mit Kohlendioxyd neutralisiert.
Nach 1 Stunde Stehen bei 0 werden 2,7 g N4 Acetyl-sulfa- nilamid abfiltriert. Das Filtrat ergibt beim Ansäuern 2,6 - Dichlor - 4 - (N4 - acetyl-sulfanilamido)-pyrimidin vom Schmelzpunkt 240-245 , das gemäss Beispiel 3 in 2,6 - Dimethoxy - 4 - sulfanilamido-pyrimidin über geführt werden kann. <I>Beispiel 6</I> 11,64 g (2 Mol) trockenes Sulfanilamidnatrium werden mit 20 ml Dimethylformamid vermischt und auf 100 erwärmt.
Das Natriumsalz bleibt zum gro ssen Teil ungelöst. Man fügt nun langsam (etwa 15 Minuten) 5,37 g 2-Methoxy-4,6-dichlor-pyrimidin unter Rühren hinzu. Man sorgt dafür, dass die stei gende Temperatur 110 nicht überschreitet. Nach Abklingen der Reaktion (Temperaturabfall) lässt man das nunmehr sirupartige Gemisch 1/@ Stunde auf dem siedenden Wasserbad unter Feuchtigkeitsausschluss stehen. Anschliessend wird im Vakuum (12 mm) Di- methylformamid abdestilliert. Der Rückstand wird mit 40 ml Wasser verrührt; dabei scheidet sich das überschüssige Sulfanilamid ab.
Man lässt 1/2 Stunde im Eisschrank stehen und saugt ab. Das Filtrat wird mit 50 % igem Eisessig angesäuert, wobei sich 2- Methoxy - 4-sulfanilamido-6-chlor-pyrimidin abschei det. Der Niederschlag wird abgesaugt, in Soda gelöst und nochmals mit 50 % igem Eisessig ausgefällt. 8,3 g Rohausbeute, Schmelzpunkt 169-1711. Nach Umkri- stallisieren aus 20 ml Methanol steigt der Schmelz punkt auf 172-174 .

Man säuert mit 50 % iger Essigsäure an und erhält 2,7 g von rohem 2,6-Di- methoxy - 4 - sulfanilamido-pyrimidin vom Schmelz punkt 201'.
Durch Umkristallisieren aus 80%igem Äthanol lässt sich daraus 2,5 g reines Produkt vom Schmelzpunkt 202-203 gewinnen.
<I>Beispiel 7</I> 9,6 g (2 Mol) trockenes Sulfanilamidnatrium wer den mit 20 ml Dimethylformamid vermischt und auf 100 erwärmt, wobei das Natriumsalz zum grossen Teil ungelöst bleibt. Anteilweise fügt man 4,8 g 2- Methylthio-4,6-dichlor-pyrimidin unter Rühren hinzu. Dabei verschwindet das Natriumsalz und ein homo genes, sirupartiges Gemisch entsteht. Nach beendetem Eintragen wird noch 15 Minuten bei 100 gerührt und schliesslich zur Vervollständigung der Reaktion 45 Minuten auf dem Wasserbad bei 98 belassen.
Danach wird das Dimethylformamid im Vakuum (12 mm) abdestilliert. Der Rückstand wird mit 35 ml Wasser aufgenommen. Anfangs geht alles in Lösung, doch findet bald darauf bei Rühren und Kratzen mit dem Glasstab eine Abscheidung von überschüssigem Sulfanilamid statt. Nach 30 Minuten Stehenlassen bei 0' im Eisschrank wird der Niederschlag abgenutscht und das Filtrat tropfenweise mit 50 % iger Essigsäure angesäuert. Es scheidet sich eine harzartige Masse aus, die abgetrennt und in wässeriger Sodalösung ge löst wird.
Bei erneutem Ansäuern mit Essigsäure be ginnt das ausgefallene Harz bald zu einer harten, kri stallinen Masse zu erstarren, die in der Reibschale zerrieben und darauf abgenutscht wird. Man kristal lisiert aus 10 ml Methanol um, dann nochmals 40 ml in absolutem Benzol, indem man das Produkt 20 Mi nuten darin zum Sieden unter Rückfluss erwärmt. Die nach dem Abnutschen erhaltenen Kristalle von 2- Methylthio-4-sulfanilamido-6-chlor-pyrimidin schmel zen bei 156-157". Ausbeute an Rohprodukt 7,4 g; nach Umkristallisieren und Umlösen 5,5 g.
Die Ver bindung ist in Wasser wenig löslich, dagegen leicht in Methanol, Essigester, Aceton und weniger leicht in Äther und Benzol. In Natriumbicarbonatlösung ist die neue Verbindung löslich. Sie lässt sich durch Um setzung mit Natriummethylat in 2,6-Dimethoxy-4- sulfanilamido-pyrimidin überführen.

At first everything goes into solution, but soon afterwards, when stirring and scratching with the glass rod, excess sulfanilamide is deposited. After standing for 30 minutes at 0 'in the refrigerator, the precipitate is filtered off with suction and the filtrate is acidified dropwise with 50% acetic acid. A resin-like mass separates out and is separated and dissolved in an aqueous soda solution.
When acidifying again with acetic acid, the precipitated resin soon begins to solidify into a hard, crystalline mass, which is ground in the mortar and then sucked off. It crystallizes from 10 ml of methanol, then another 40 ml in absolute benzene by heating the product to reflux for 20 minutes. The crystals of 2-methylthio-4-sulfanilamido-6-chloro-pyrimidine obtained after suction filtration melt at 156-157 ". Yield of crude product 7.4 g; after recrystallization and redissolving, 5.5 g.
The compound is sparingly soluble in water, but easily in methanol, ethyl acetate, acetone and less readily in ether and benzene. The new compound is soluble in sodium bicarbonate solution. It can be converted into 2,6-dimethoxy-4-sulfanilamido-pyrimidine by reaction with sodium methylate.
<I> Example 8 </I> 3.7 g of 2-methylthio-4- (N4 acetyl-sulfanilamido) pyrimidine (prepared according to Example 7) are mixed with 1.15 g of sodium in 20 ml of absolute methanol in a glass autoclave for 5 hours l20 treated in an oil bath. Sodium chloride separates out on cooling. The methanol is distilled off in vacuo and the residue is dissolved in 20 ml of water. 50% acetic acid is added to the aqueous solution, and the smell of methyl mercaptan occurs.
The precipitated crystals of 2,6-dimethoxy-4- (N4 acetylsulfanilamido) pyrimidine have a melting point of 222 to 224 after recrystallization from methanol. Yield 3.2g.
1 g of the 2,6-dimethoxy-4- (N4 acetylsulfanilamido) pyrimidine obtained is dissolved in 10 ml in sodium hydroxide solution and saponified by heating on a water bath at 58 for 2 hours. After acidification with acetic acid, 0.75 g of crude product with a melting point of 188-192 are obtained. After recrystallizing twice from methanol, the melting point of the 2,6-dimethoxy-4-sulfanilamido-pyrimidine thus obtained is 197-199.
