Verfahren zur Herstellung von neuen Aminen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Amine mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Derivate der 5H-Dibenzo [a, d] cycloheptapolyene entsprechend der Formel I,

in der Xt und X2 entweder je ein Wasserstoffatom oder zusammen eine zusätzliche Bindung und RJ einen niederen Alkylrest bedeuten, und ihre Salze mit anorganischen und organischen Säuren wertvolle pharmakologische Eigenschaften, insbesondere antihistaminische, dekontrahierende und spasmolytische, sedative und antiemetische Wirksamkeit besitzen. Sie antagonisieren die Wirkung von Serotonin und Reserpin und potenzieren die Wirkung von Narkotica. Sie können z.
B. zur Behandlung von Geisteskrankheiten, insbesondere Gemütsdepressionen, dienen und sind überdies insbesondere infolge Anwesenheit einer sekundären Aminogruppe als Zwischenprodukte zur Herstellung weiterer pharmakologisch wertvoller Stoffe verwendbar.
In den Verbindungen der Formel I ist R1 beispielsweise ein Methyl-, Athyl-, n-Propyl- oder Isopropylrest.
Die neuen Verbindungen der Formel I werden erfindungsgemäss hergestellt, indem man eine Verbindung der Formel II,

in der Ac einen durch Hydrolyse bzw. Hydrogenolyse abspaltbaren Rest, insbesondere einen Carbalkoxyrest, Carbobenzyloxyrest, Chlorcarbonylrest oder Cyanorest bedeutet, hydrolysiert oder bzw. hydrogenolysiert.
Die Hydrolyse von Verbindungen der Formel II kann z. B. durch Behandlung derselben bei erhöhter Temperatur entweder in höhersiedenden hydroxylhaltigen organischen Lösungsmitteln wie z. B. Athylenglykol, Diäthylenglykol oder deren niederen Monoalkyläthern oder in niederen Alkanolen, vorzugsweise im geschlossenen Gefäss, erfolgen. Sofern in Verbindungen der Formel II der Rest Ac einen Carbobenzyloxyrest oder ringsubstituierten Carbobenzyloxyrest und Xt und X2 Wasserstoff bedeuten, kommt auch die Hydrogenolyse von Verbindungen der Formel II zu Verbindungen der Formel I, z. B. durch Einwirkung von Wasserstoff in Gegenwart von Edelmetallkatalysatoren, wie z. B. Palladium-Kohle, in einem geeigneten inerten organischen Lösungsmittel, wie z. B. niederen, gegebenenfalls wasserhaltigen Alkanolen, in Betracht.
Die Ausgangsstoffe der Formel II sind ihrerseits beispielsweise durch Umsetzung einer Verbindung der Formel III,

in der R2 einen Benzylrest, einen niederen Alkenylrest oder einen vorzugsweise mit R1 identischen niederen Alkylrest bedeutet, mit einem organischen Säurehalogenid oder -anhydrid, insbesondere einem Kohlensäureesterchlorid (Chlorameisensäureester), Phosgen oder Bromcyan herstellbar. Die Umsetzung von Verbindungen der Formel III mit organischen Säurehalogeniden wie z. B. Chlorameisensäuremethylester, -äthylester oder benzylester, Benzoylchlorid, Phosgen oder Bromcyan oder mit Anhydriden, insbesondere Acetanhydrid bei Anwesenheit eines Benzylrestes als R2,kann in An- oderAbwesenheit eines geeigneten organischen Lösungsmittels wie z. B.
Benzol, Toluol, Diäthyläther, Diisopropyläther oder Tetrahydrofuran bei Raumtemperatur oder erhöhter Temperatur durchgeführt werden. Die Säurehalogenide oder Anhydride können in äquimolarer Menge oder in einem erheblichen Überschuss verwendet werden und insbesondere im letzteren Falle als einziges Reaktionsmedium dienen. Die Umsetzung kann bereits beim Zusammengeben der Reaktionskomponenten unter Wärmeentwicklung und Freisetzung des den Rest R2 enthaltenden Benzyl-, Alkenyl- oder Alkylhalogenids eintreten und wird nötigenfalls durch Erwärmen vervollständigt bzw. eingeleitet und durchgeführt.
Zu Verbindungen der Formel III gelangt man beispielsweise durch Umsetzung von 5H-Dibenzo [a,dlcyclohepten-5-on oder
10,1 1-Dihydro-5H-dibenzo[a,d] cyclohepten-5-on mit einer 0-Dialkylamino-propyl-metallverbindung, r-(N-Alkyl-alkenylamino)-propyl- metallverbindung, y-Dialkenylamino-propyl-metallverbindung oder y-(N-Alkyl-benzylamino)-propyl-metallverbindung und anschliessende Wasserabspaltung. Als dazu geeignete metallorganische Verbindungen kommen insbesondere magnesiumorganische, lithiumorganische und zinkorganische Verbindungen in Betracht.
Vorzugsweise werden die entsprechenden organischen Magnesiumhalogenidverbindungen verwendet, welche aus den N,N-disubstituierten y-Amino-propylhalogeniden in Diäthyläther, Tetrahydrofuran oder einem anderen ätherartigen Lösungsmittel in bekannter Weise hergestellt werden. Die Wasserabspaltung als zweite Reaktionsstufe geht sehr leicht vor sich, z. B. bereits beim Stehenlassen der tertiären Hydroxylverbindungen in konz. wässrig-alkoholischer Salzsäure bei Raumtemperatur.
Die erfindungsgemäss erhaltenen Verbindungen der Formel I bilden mit anorganischen und organischen Säuren, wie
Salzsäure, Bromwasserstoffsäure, Schwefelsäure,
Phosphorsäure, Methansulfonsäure, Äthandisulfonsäure, ss-Hydroxy-äthansulfonsäure,
Essigsäure, Bernsteinsäure, Fumarsäure,
Maleinsäure, Äpfel säure, Weinsäure,
Citronensäure, Benzoesäure, Salicylsäure und
Mandelsäure, Salze, die zum Teil wasserlöslich sind.
Zu den nachfolgenden Beispielen bedeuten Teile Gewichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel 1
13,0 Teile 5-(y-Dimethylamino-propyliden)
10,11 -dihydro-5H-dibenzo[a,d]cycloheptan löst man in 100 Volumteilen abs. Benzol und tropft diese Lösung unter starkem Rühren zu 18 Teilen Chlorameisensäureäthylester. Die Temperatur steigt auf etwa 350, und es entweicht Methylenchlorid. Anschliessend rührt man noch 1 y2 Stunden bei 400 weiter. Nach dem Abkühlen wird die Reaktionslösung vorerst mit Wasser und anschliessend mit 2n Salzsäure gewaschen, getrocknet und eingedampft. Der Rückstand wird im Hochvakuum destilliert. Das
5-[y-(N-Carbäthoxy-methylamino)-propyliden]-
10,1 l-dihydro-5H-dibenzo[a,d]cyclohepten geht unter 0,006 Torr bei 164" über.
14,3 Teile des obigen Urethans werden in einer Lösung von 7,0 Teilen Kaliumhydroxyd, 7,0 Teilen Wasser und 50 Teilen Diäthylenglykolmonoäthyläther 20 Stunden unter Rückfluss gekocht. Nach dem Abkühlen giesst man die Reaktionslösung in 250 Teile Wasser und extrahiert sie erschöpfend mit Äther. Der ätherischen Lösung werden die basischen Anteile durch viermaliges Extrahieren mit verdünnter Essigsäure entzogen.
Die vereinigten sauren Auszüge werden alkalisch gestellt und die ausgeschiedene Base in Äther aufgenommen. Nach dem Trocknen wird die Ätherlösung eingedampft und der Rückstand aus Pentan umkristallisiert. Das so erhaltene
5-(y-Methylamino-propyliden)-l O,ll-dihydro-
5H-dibenzo[a,d] cyclohepten schmilzt bei 580. Das mit alkoholischer Salzsäure bereitete Hydrochlorid schmilzt bei 217".
Beispiel 2
In gleicher Weise erhält man, ausgehend vom
5-(y-Dimethylamino-propyliden)-5H dibenzo[a,d]cyclohepten, das 5-[y-(N-Carbäthoxy-methylamino)-propyliden]- 5H-dibenzo[a,d]cyclohepten vom Kr.0,02 185 und aus diesem das 5-(y-Methylamino-propyliden) SH-dibenzoCa,dlcycloheptan, dessen Hydrochlorid nach Kristallisation aus wasserfreiem Äthanol-Äther bei 182-184 schmilzt.

The hydrolysis of compounds of formula II can, for. B. by treating the same at elevated temperature either in higher-boiling hydroxyl-containing organic solvents such as. B. ethylene glycol, diethylene glycol or their lower monoalkyl ethers or in lower alkanols, preferably in a closed vessel. If, in compounds of the formula II, the radical Ac is a carbobenzyloxy radical or ring-substituted carbobenzyloxy radical and Xt and X2 are hydrogen, the hydrogenolysis of compounds of the formula II to give compounds of the formula I, e.g. B. by the action of hydrogen in the presence of noble metal catalysts, such as. 10.1 1-Dihydro-5H-dibenzo [a, d] cyclohepten-5-one with a 0-dialkylamino-propyl-metal compound, r- (N-alkyl-alkenylamino) -propyl-metal compound, y-dialkenylamino-propyl- metal compound or y- (N-alkyl-benzylamino) -propyl-metal compound and subsequent elimination of water. Organometallic compounds suitable for this purpose are, in particular, organomagnesium, organolithium and organozinc compounds.
The corresponding organic magnesium halide compounds are preferably used, which are prepared in a known manner from the N, N-disubstituted γ-amino-propyl halides in diethyl ether, tetrahydrofuran or another ethereal solvent. The elimination of water as the second reaction stage is very easy to do, e.g. B. already when standing the tertiary hydroxyl compounds in conc. aqueous-alcoholic hydrochloric acid at room temperature.

In the following examples, parts are parts by weight; these are related to parts of volume as g to cm3. The temperatures are given in degrees Celsius.

13.0 parts of 5- (γ-dimethylamino-propylidene)
10,11 -dihydro-5H-dibenzo [a, d] cycloheptane is dissolved in 100 parts by volume of abs. Benzene and this solution is added dropwise to 18 parts of ethyl chloroformate with vigorous stirring. The temperature rises to about 350 and methylene chloride escapes. The mixture is then stirred at 400 for a further 1 y2 hours. After cooling, the reaction solution is first washed with water and then with 2N hydrochloric acid, dried and evaporated. The residue is distilled in a high vacuum. The

10.1 l-dihydro-5H-dibenzo [a, d] cycloheptene goes over below 0.006 Torr at 164 ".
14.3 parts of the above urethane are refluxed for 20 hours in a solution of 7.0 parts of potassium hydroxide, 7.0 parts of water and 50 parts of diethylene glycol monoethyl ether. After cooling, the reaction solution is poured into 250 parts of water and extracted exhaustively with ether. The basic components are removed from the ethereal solution by extracting it four times with dilute acetic acid.
The combined acidic extracts are made alkaline and the precipitated base is taken up in ether. After drying, the ether solution is evaporated and the residue is recrystallized from pentane. The thus obtained
5- (y-methylamino-propylidene) -l O, ll-dihydro-
5H-dibenzo [a, d] cycloheptene melts at 580. The hydrochloride prepared with alcoholic hydrochloric acid melts at 217 ".
Example 2
In the same way, starting from
5- (y-Dimethylamino-propylidene) -5H dibenzo [a, d] cycloheptene, the 5- [y- (N-carbethoxymethylamino) -propylidene] -5H-dibenzo [a, d] cycloheptene of Kr.0, 02 185 and from this 5- (γ-methylamino-propylidene) SH-dibenzoCa, dlcycloheptane, the hydrochloride of which melts at 182-184 after crystallization from anhydrous ethanol ether.
