Verfahren zur Herstellung von N-heterocyclischen Verbindungen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von N-heterocyclischen Verbindungen mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass man Derivate des Iminodibenzyls (10, 11-Dihydro- 5H-dibenz [b, f] azepin) und Iminostilbens (5H-Dibenz- [b, f] azepin der Formel I

Die Umsetzungen von Verbindungen der Formel II mit organischen Säurehalogeniden, z. B. mit Chlor ameisensäure-methylester,-äthylester oder-benzylester, Benzoylchlorid oder Phosgen kann in An-oder Abwesenheit eines geeigneten organischen Lösungs- mittels, wie z. B. Benzol, Toluol, Diäthyläther, Diisopropyläther oder Tetrahydrofuran bei Raumtemperatur oder erhöhter Temperatur durchgeführt werden.
Die Säurehalogenide können in äquimolarer Menge oder in einem Überschuss verwendet werden und in letzterem Falle als einziges Reaktionsmedium dienen.
Beim Zusammengeben der Reaktionskomponenten erfolgt oft bereits die Umsetzung unter Wärmeentwicklung und Freisetzung des den Rest R enthaltenden Alkylhalogenids. Die Verbindungen der allgemeinen Formel III werden durch Hydrolyse, z. B.
Behandlung mit Alkalimetallhydroxyden bei erhöhter Temperatur in höhersiedenden hydroxylhalti- gen organischen Lösungsmitteln, wie z. B. Äthylen- glykol oder Diäthylenglykol bzw. deren niederen Monoalkyläther, oder in niederen Alkoholen vorzugsweise im geschlossenen Gefäss, in Verbindungen der Formel I übergeführt. Sofern der Rest Ac in den Verbindungen der Formel III durch einen Carbobenzyloxyrest oder ringsubstituierten Carbobenzyloxyrest verkörpert ist, kommt auch die Hydrogenolyse von Verbindungen der Formel III zu Verbindungen der Formel I, z. B. durch Einwirkung von Wasserstoff in Gegenwart von Edelmetallkatalysa- toren, wie z. B. Palladium-Kohle in einem geeigneten inerten organischen Lösungsmittel, wie z.
B. niederen, gegebenenfalls wasserhaltigen Alkanolen, in Betracht.
Zahlreiche Ausgangsstoffe der Formel II sind bereits bekannt und weitere in analoger Weise wie die bekannten Verbindungen herstellbar.
Zum Beispiel wird zur Herstellung von Verbindungen der Formel II ein gegebenenfalls substituiertes Iminodibenzyl oder Iminostilben der Formel IV,

mit einer Verbindung der Formel V,

in der Hal Chlor oder Brom bedeutet, und X, Y, Z, R, m, n und n'die oben angegebene Bedeutung haben, mittels eines alkalischen Kondensationsmittels, wie z. B. Natriumamid, in einem geeigneten inerten organischen Lösungsmittel, wie z. B. Benzol oder Toluol, kondensiert.
Die erfindungsgemäss herstellbaren Verbindungen der Formel I bilden mit anorganischen und organischen Säuren, wie
Salzsäure, Bromwasserstoffsäure, Schwefelsäure,
Phosphorsäure, Methansulfonsäure, Äthandisulfonsäure, ss-Hydroxy-äthansulfonsäure,
Essigsäure, Bernsteinsäure, Fumarsäure,
Maleinsäure, Äpfelsäure, Weinsäure,
Citronensäure, Benzoesäure, Salicylsäure und
Mandelsäure, Salze, die zum Teil wasserlöslich sind.
Die Verbindungen der Formel I besitzen wertvolle pharmakologische Eigenschaften, insbesondere antiallergische, spasmolytische, sedative und antiemetische Wirksamkeit. Sie antagonisieren die Wirkung von Serotonin und Reserpin, und potenzieren die Wirkung von Narkotika. Sie sind überdies als Antioxydantien sowie als Zwischenprodukte zur Herstellung weiterer pharmakologisch wertvoller Stoffe verwendbar. Auch Verbindungen der Formel III, insbesondere solche mit einem niederen Carbalkoxyrest als Ac, besitzen teilweise wertvolle pharmakologische Eigenschaften.
In den nachfolgenden Beispielen bedeuten Teile Gewichtsteile, diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
Beispiel a) 32 Teile 5- [i (1'-Methyl-2'-piperidinyl)-äthyl]- iminodibenzyl werden in 150 Volumteilen abs. Benzol gelöst und zu 27, 5 Volumteilen Chlorameisensäure- äthylester unter Rühren zugetropft. Die Reaktion ist exotherm, wobei Methylchlorid entweicht. Nach etwa einer halben Stunde ist die Reaktion beendet. Dann wird die Reaktionslösung filtriert, mit 2n Salzsäure gewaschen, hierauf getrocknet und eingedampft.
Das 5- [ss- (1'-Carbäthoxy-2'-piperidinyl)-äthyl]-iminodi- benzyl bleibt als 01 zurück. b) Das gemäss a) erhaltene Rohprodukt wird in einer Lösung von 300 Volumteilen Diäthylenglykol- monoäthyläther, 40 Volumteilen Wasser und 40 Teilen Kaliumhydroxyd 20 Stunden bei 160 erhitzt.
Anschliessend wird das Reaktionsgemisch am Wasserstrahlvakuum auf etwa das halbe Volumen eingeengt.
Die Lösung wird mit 150 Volumteilen Wasser verdünnt und mit konz. Salzsäure angesäuert. Nach kurzem Erhitzen wird sie mit verdünnter Natronlauge wiederum alkalisch gestellt und gründlich ausge äthert. Aus der getrockneten Ätherlösung erhält man nach dem Einengen ein nahezu farbloses 01, aus welchem man bei der Destillation im Hochvakuum das 5-[ss-(2'-Piperidinyl)-äthyl]-iminodibenzyl vom Siedepunkt 200 /0, 05 Torr erhält. Durch Zugabe von der berechneten Menge Fumarsäure zur äthanolischen Lösung der Base erhält man deren Fumarat vom Smp. 221-224 .
Analog werden die folgenden Verbindungen als im Hochvakuum destillierbare Öle erhalten : ausgehend vom 5- (1'-Methyl-3'-piperidinyl)- iminodibenzyl vom Kp. 0, 02 165-167 das 5- (3'-Piperidinyl)-iminodibenzyl ; ausgehend von 5-(1'-Methyl-4'-piperidmyl) iminodibenzyl vom Kp. 0, os 151-154 das 5- (4'-Piperidinyl)-iminodibenzyl ; ausgehend vom 5- [ (1'-Methyl-3'-piperidinyl)-methyl]- iminodibenzyl vom Kp. o, ol 157-160 das 5- [ (3'-Piperidinyl)-methyl]-iminodibenzyl ;

The compounds of the formula I have valuable pharmacological properties, in particular antiallergic, spasmolytic, sedative and antiemetic activity. They antagonize the effects of serotonin and reserpine, and potentiate the effects of narcotics. They can also be used as antioxidants and as intermediates for the production of other pharmacologically valuable substances. Compounds of the formula III, in particular those with a lower carbalkoxy radical than Ac, also have valuable pharmacological properties in some cases.
In the examples below, parts mean parts by weight; these relate to parts by volume as g to cm3. The temperatures are given in degrees Celsius.
Example a) 32 parts of 5- [i (1'-methyl-2'-piperidinyl) ethyl] - iminodibenzyl are abs in 150 parts by volume. Dissolved benzene and added dropwise to 27.5 parts by volume of ethyl chloroformate with stirring. The reaction is exothermic, with methyl chloride escaping. The reaction has ended after about half an hour. The reaction solution is then filtered, washed with 2N hydrochloric acid, then dried and evaporated.
The 5- [ss- (1'-carbethoxy-2'-piperidinyl) -ethyl] -iminodibenzyl remains as 01. b) The crude product obtained according to a) is heated at 160 for 20 hours in a solution of 300 parts by volume of diethylene glycol monoethyl ether, 40 parts by volume of water and 40 parts of potassium hydroxide.
The reaction mixture is then concentrated to about half its volume in a water jet vacuum.
The solution is diluted with 150 parts by volume of water and treated with conc. Acidified hydrochloric acid. After brief heating, it is made alkaline again with dilute sodium hydroxide solution and thoroughly etherified. After concentration, the dried ether solution gives an almost colorless oil, from which 5- [ss- (2'-piperidinyl) -ethyl] -iminodibenzyl with a boiling point of 200/0.05 Torr is obtained by distillation in a high vacuum. By adding the calculated amount of fumaric acid to the ethanolic solution of the base, the fumarate thereof is obtained with a melting point of 221-224.
