Procédé de préparation de dicarbamates
La présente invention a pour objet un procédé de préparation de nouveaux dicarbamates de 2, 2 dialcoyl-1, 3-propane-diols N-monosubstitués de formule :

dans laquelle R est un radical méthyle ou éthyle, R'est un radical éthyle, n-butyle, sec.-butyle ou isopropyle et X est un radical méthyle, n-butyle, propyle, isopropyle, allyle ou propargyle.
Ce procédé est caractérisé en ce que l'on fait réagir un monocarbamate de formule :

avec un ester carbamique d'un alcool à bas point d'ébullition, tel que l'uréthane.
Le monocarbamate de départ peut être préparé par réaction d'un 2-R-2-R'-1, 3-propane-diol avec du carbonate de diéthyle, puis réaction de l'ester cyclique ainsi formé avec une amine de formule NH2X, selon le schéma suivant :

Le monocarbamate de départ peut également être préparé par réaction d'un 2-R-2-R'-1, 3-propanediol avec du phosgène, puis réaction du monoester chloroformique ainsi obtenu avec une amine de formule NH2X, selon le schéma suivant :

Les composes obtenus par le procédé selon l'in- vention sont des liquides incolores à point d'ébulli- tion élevé ou des solides à bas point de fusion.
Ils sont solubles dans la plupart des solvants organiques mais ne sont que peu solubles dans l'eau à la température ordinaire. Ils sont facilement hydrolyses par les solutions caustiques ou d'acides forts chaudes en donnant le diol correspondant, de l'ammoniaque,
Famine primaire correspondante et de l'anhydride carbonique.
Ces nouveaux dicarbamates possèdent un pouvoir de relaxation musculaire exceptionnel, de sorte qu'ils sont des médicaments très intéressants contre les spasmes musculaires. Ils possèdent en outre un pouvoir marqué de dépression du système nerveux central et une action hypotensive.
Les matières de départ sont de préférence choisies de manière que les dicarbamates obtenus soient ceux identifiés dans le tableau ci-après par leurs substituants X, R et R', leur formule brute, leur teneur en azote, leur indice de réfraction (n25) et leur point de fusion ou leur température d'ébullition sous la pression indiquée entre parenthèses. Le tableau donne en outre pour chaque composé la dose paralysante moyenne (DPÏO) et la dose létale moyenne (DL50) chez la souris, par voie intrapéri- tonale, toutes deux exprimées en mg de composé par kg de souris.

Dans l'exemple qui suit, les parties sont exprimées en poids.
Exemple
Préparation du N-propargyl-dicarbamate de 2-méthyl-
2-sec. butyl-1, 3-propane-diol
On distille pour éliminer toutes les traces d'eau résiduelle un mélange de 22, 7 parties de N-propargyl-carbamate de 2-méthyl-2-sec.-butyl-3-hydroxy- propyle, 10, 7 parties d'uréthane et 200 parties de xylène. On laisse refroidir le mélange et on ajoute 1 partie d'isopropylate d'aluminium. On distille le mélange jusqu'à élimination de sensiblement la quantité théorique d'Úthanol sous la forme de son azéotrope avec le xylène. Le xylène est chassé par distillation sous pression réduite et 100 parties d'eau sont ajoutées. On distille l'eau sous pression réduite et le résidu est dissous dans 150 parties d'isopropanol.
La matière insoluble est séparée par filtration et l'isopropanol éliminé par distillation. Après refroidissement, on obtient 15 parties de produit purifié ayant les constantes physiques indiquées dans le tableau I.
Le N-propargyl-carbamate de 2-méthyl-2-sec. butyl-3-hydroxy-propyle de départ peut être préparé comme suit :
On ajoute en agitant une solution refroidie à 10 % de 1 mole de phosgène dans du toluène à une solution refroidie de 1 mole de 2-méthyl-2-sec. butyl1, 3-propane-diol et de 2 moles de diméthylaniline, également dissoutes dans du toluène, à une vitesse telle que la température du mélange soit maintenue à environ 250 C. On laisse séjourner le mélange à cette température pendant plusieurs heures, puis on le refroidit et on l'extrait par une solution froide d'acide chlorhydrique à 5 % pour éliminer la diméthylaniline.
La couche toluénique est desséchée à l'aide d'un agent déshydratant approprié et le chlorocarbonate de 2-méthyl-2-sec. butyl-3-hydroxy-propyle ainsi obtenu est utilisé dans la réaction suivante sous la forme de sa solution dans le toluène anhydre.
On traite 0, 1 mole de chlorocarbonate de 2-méthyl-2-sec. butyl-3-hydroxy-propyle, préparé comme décrit ci-dessus, par 0, 2 mole de propargylamine anhydre. On laisse la réaction se dérouler à la température ambiante, la solution est refroidie, extraite par de l'acide chlorhydrique étendu et la couche organique est concentrée par évaporation du solvant. Le résidu, constitué par du monocarbamate brut, est purifié par distillation sous pression réduite, et la fraction distillant à 123-126 C sous 0, 04 mm est recueillie. nD =1, 4790. Rendement environ 65 % du rendement théorique.

This process is characterized in that a monocarbamate of formula is reacted:

The starting monocarbamate can be prepared by reacting a 2-R-2-R'-1, 3-propanediol with diethyl carbonate, then reacting the cyclic ester thus formed with an amine of formula NH2X, according to the following diagram:

The starting monocarbamate can also be prepared by reacting a 2-R-2-R'-1, 3-propanediol with phosgene, then reacting the chloroform monoester thus obtained with an amine of formula NH2X, according to the following scheme:

The compounds obtained by the process according to the invention are colorless liquids with a high boiling point or solids with a low melting point.
They are soluble in most organic solvents but are only slightly soluble in water at room temperature. They are easily hydrolyzed by caustic solutions or hot strong acids giving the corresponding diol, ammonia,
Corresponding primary starvation and carbon dioxide.
These new dicarbamates have an exceptional muscle relaxation power, so they are very interesting drugs against muscle spasms. They also have a marked power of central nervous system depression and hypotensive action.
The starting materials are preferably chosen so that the dicarbamates obtained are those identified in the table below by their substituents X, R and R ', their crude formula, their nitrogen content, their refractive index (n25) and their melting point or their boiling point under the pressure indicated in parentheses. The table further gives for each compound the mean paralyzing dose (DP10) and the mean lethal dose (LD50) in mice, intraperitoneally, both expressed in mg of compound per kg of mouse.

Example
Preparation of 2-methyl- N-propargyl-dicarbamate
2-sec. butyl-1,3-propanediol
A mixture of 22.7 parts of 2-methyl-2-sec.-butyl-3-hydroxy-propyl N-propargyl-carbamate, 10.7 parts of urethane and 10.7 parts of urethane is distilled to remove all traces of residual water. 200 parts of xylene. The mixture is allowed to cool and 1 part of aluminum isopropylate is added. The mixture is distilled until substantially the theoretical amount of ethanol in the form of its azeotrope is removed with xylene. The xylene is removed by distillation under reduced pressure and 100 parts of water are added. The water is distilled off under reduced pressure and the residue is dissolved in 150 parts of isopropanol.
Insoluble material is separated by filtration and the isopropanol removed by distillation. After cooling, 15 parts of purified product are obtained having the physical constants shown in Table I.
2-Methyl-2-sec N-propargyl-carbamate. The starting butyl-3-hydroxy-propyl can be prepared as follows:
A cooled 10% solution of 1 mole of phosgene in toluene is added with stirring to a cooled solution of 1 mole of 2-methyl-2-sec. butyl1,3-propane-diol and 2 moles of dimethylaniline, also dissolved in toluene, at a rate such that the temperature of the mixture is maintained at about 250 C. The mixture is left to stand at this temperature for several hours, then It is cooled and extracted with a cold 5% hydrochloric acid solution to remove the dimethylaniline.
The toluene layer is dried using a suitable dehydrating agent and sec 2-methyl-2-chlorocarbonate. Butyl-3-hydroxy-propyl thus obtained is used in the following reaction in the form of its solution in anhydrous toluene.
0.1 mole of 2-methyl-2-sec chlorocarbonate is treated. butyl-3-hydroxy-propyl, prepared as described above, with 0.2 moles of anhydrous propargylamine. The reaction is allowed to proceed at room temperature, the solution is cooled, extracted with extended hydrochloric acid and the organic layer is concentrated by evaporation of the solvent. The residue, consisting of crude monocarbamate, is purified by distillation under reduced pressure, and the fraction distilling at 123-126 C under 0.04 mm is collected. nD = 1.4790. Yield about 65% of theoretical yield.
