Verfahren zur Herstellung von Reserpsäureestern Im Schweizer Patent Nr. 334 151 wird unter an derem die Herstellung von Reserpsäurealkylestern, deren Hydroxylgruppe mit organischen Carbon- säuren verestert ist, und deren Salze beschrieben.
Gegenstand der Erfindung ist nun die Herstellung von Reserpsäurealkylestern der Formel

worin R1 für eine Alkylgruppe mit 1-5 Kohlenstoff atomen und R, und R3 für Wasserstoff oder Alkyl- gruppen mit 1-5 Kohlenstoffatomen und einer der Reste R4 und R", für Wasserstoff und der andere für eine Alkoxygruppe mit 1-5 Kohlenstoffatomen oder eine Dialkylaminogruppe,
deren Alkylreste 1-5 Kohlenstoffatome enthalten, stehen.
Die neuen Ester besitzen eine dem Reserpin vergleichbare ausgeprägte sedativ-hypnotische Wirk- samkeit, jedoch sind sie weniger blutdrucksenkend wirksam.
Die neuen Ester lassen sich nach an sich be kannten Methoden erfindungsgemäss herstellen, indem man einen Reserpsäurealkylester mit einer freien Hydroxylgruppe, dessen Aikylrest 1-5 Kohlen stoffatome besitzt, besonders den Reserpsäuremethyl- ester, mit einer Benzoesäure der Formel

worin X eine Dialkylaminogruppe, deren Alkylreste 1-5 Kohlenstoffatome besitzen, oder einen durch Reduktion in die NH2 Gruppe überführbaren Rest bedeutet,
oder einem ihrer funktionellen Säure- derivate, wie dem Anhydrid oder dem Halogenid, z. B. dem Chlorid, verestert und, falls man einen Di- ester mit einem in die NH2 Gruppe überführbaren Rest erhält, diesen Rest zur NH2 Gruppe reduziert. Die Veresterung wird zweckmässig in Anwesen heit von Verdünnungs- und bzw. oder Veresterungs- mitteln durchgeführt, wie in Gegenwart säurebinden der Mittel, z. B.
Alkali- oder Erdalkalicarbonaten oder starken organischen Basen, z. B. tert. Aminen, wie Pyridin oder Collidin. Dabei kann man die Aus gangsstoffe auch in Form ihrer Salze verwenden.
Die nachträgliche Umwandlung der durch Re duktion in die NH.- Gruppe überführbaren Reste muss natürlich unter solchen Bedingungen durch geführt werden, die weder die Carboalkoxygruppe noch die Benzoyloxygruppe spalten oder eine Um lagerung besonders in 3-Stellung des Reserpsäure- skeletts bewirken.
Als durch Reduktion in die pri märe Aminogruppe umwandelbarer Rest sollen in erster Linie die Nitrogruppe, ferner die Alkoxy- carbonyl- oder die Benzyloxycarbonylaminogruppen, z. B. die Methoxy- oder Äthoxycarbonylamino- gruppe, genannt werden. So kann man die Nitro- gruppe mit katalytisch aktiviertem Wasserstoff in an sich bekannter Weise reduzieren.
Nimmt man diese Reduktion in Gegenwart eines aliphatischen Aldehyds oder aliphatischen Ketons vor, so erhält man eine alkylierte Aminogruppe. Als Aldehyde lassen sich Alkanale mit 1-5 Kohlenstoffatomen, wie Form aldehyd, vorzugsweise in einer wässrigen, 20 bis 40%igen, in erster Linie ungefähr 37%igen Lö- sung,
Acetaldehyd oder Propionaldehyd verwenden. Dabei erhält man direkt tertiäre Aminogruppen, wie die Dimethylamino- oder Diäthylaminogruppe. Ver wendet man jedoch bei dieser Reduktion Dialkyl- ketone, z. B. Aceton oder Methyläthylketon, erhält man vorzugsweise sekundäre Aminogruppen. Man kann aber auch Aminogruppen nachträglich in eine sekundäre oder tertiäre Aminogruppe umwandeln.
Dies lässt sich in an sich bekannter Weise mit Alky- lierungsverfahren durchführen. So lässt sich eine unsubstituierte Arninogruppe durch katalytische Hy drierung in Gegenwart einer wässrigen Formaldehyd lösung in die Dimethylaminogruppe überführen. Ver wendet man Aceton, so erhält man auf die nämliche Weise eine Isopropylaminogruppe. Man kann aber auch mit einem Aldehyd die Schiffsche Base herstel len, welche sich dann reduzieren lässt.
Je nach der Arbeitsweise erhält man die neuen Diester in freier Form oder als Salze. Letztere lassen sich aus den freien Estern durch Umsetzung mit an organischen oder organischen Säuren, wie Halogen wasserstoffsäuren, Schwefelsäure, Phosphorsäure, Salpetersäure, Essigsäure, Propionsäure, Zitronen säure, Milchsäure, Oxalsäure, Bernsteinsäure, Äpfel säure, Weinsäure, Ascorbinsäure, Methansulfonsäure, Äthansulfonsäure, Oxyäthansulfonsäure, Benzoe- säure, Salicylsäure,
p-Aminobenzoesäure oder To- luolsulfonsäure, gewinnen. Aus den Salzen können die neuen Ester in freier Form, z. B. durch Behan deln ihrer Lösung mit Silbercarbonat, wieder erhalten werden. Dabei können je nach den Bedingungen der Salzbildung und(oder der Anzahl der salzbildenden Gruppen Mono-, Bis- oder Polysalze erhalten wer den. Die neuen Salze können auch als Hydrate, z. B.
Hemihydrate, Monohydrate, Sesquihydrate oder Poly hydrate, kristallisieren.
In den nachfolgenden Beispielen besteht zwischen Gewichtsteil und Volumteil die gleiche Beziehung wie zwischen Gramm und Kubikzentimeter. Die Temperaturen sind in Celsiusgraden angegeben. <I>Beispiel 1</I> Eine Lösung von 9,68 Gewichtsteilen 2-Methoxy- 5-nitrobenzoesäure in 100 Volumteilen Thionyl- chlorid wird 1 Stunde am Rückfluss gekocht.
Dann destilliert man das überschüssige Thionylchlorid unter vermindertem Druck ab und fügt unter fort laufendem Abdestillieren verschiedene Male je 75 Volumteile Toluol zu, wodurch auch die letzten Reste Thionylchlorid entfernt werden. Das so erhaltene rohe 2-Methoxy-5-nitrobenzoylchlorid schmilzt bei 85,5-87,5 .
Man fügt dieses Säurechlorid zu einer gesättigten Lösung von 16,59 Gewichtsteilen Reserp- säuremethylester in 100 Volumteilen trockenem Pyridin und rührt, bis sich alle festen Teile gelöst haben, lässt bei Raumtemperatur über Nacht stehen, giesst die Pyridinlösung in 1200 Volumteile Wasser, dekantiert das Wasser von den ausgefallenen gummi artigen Anteilen ab und fügt zur Kristallisation Methanol zu.
Nach dem Umkristallisieren aus Me thanol schmilzt der 18 - O - (2' - Methoxy - 5'-nitro- benzoyl)-reserpsäuremethylester bei 242-244 .
Eine Lösung von 2,59 Gewichtsteilen dieses Esters in 300 Volumteilen 95Q/oigem Äthanol wird in Gegenwart von 0,5 Gewichtsteilen 10%igem Palladium auf Tierkohle-Katalysator mit Wasser stoff bei atmosphärischem Druck reduziert. Die Re aktionsmischung wird filtriert,
das Filtrat unter ver mindertem Druck eingedampft und der gummiartige Rückstand in Methanol aufgenommen. Man lässt die Lösung bei Raumtemperatur über Nacht stehen, filtriert die Kristalle ab und kristallisiert sie aus Methanol um. Der so erhaltene kristalline 18-0-(2'- Methoxy - 5'- aminobenzoyl) - reserpsäuremethylester enthält 1 Mol Methanol und schmilzt bei 148-151 .
<I>Beispiel 2</I> Zu einer Mischung von 27,5 Gewichtsteilen Reserpsäuremethylester in 275 Volumteilen trocke nem Pyridin gibt man 20,6 Gewichtsteile (200/0 Lberschuss) 3 - Dimethylamino - 6 - methoxybenzoyl- chlorid-hydrochlorid unter Rühren und in einer Stick stoffatmosphäre zu. Man lässt die Reaktionsmischung 18 Stunden bei 20-25 stehen, giesst unter gutem Rühren langsam in 2750 Volumteile Eiswasser und filtriert.
Den Rückstand wäscht man mit Wasser, nimmt ihn in 126 Volumteile Methylenchlorid auf und filtriert die Lösung über eine Kolonne von 25 Gewichtsteilen Magnesium-Aluminiumsilikat. Man wäscht die Kolonne mit ungefähr 190 Volumteilen Methylenchlorid nach, dampft die vereinigten Methy- lenchloridlösungen zur Trockne ein und nimmt in 100 Volumteile Methanol auf.
Der so erhaltene 18-0-(2'-Methoxy-5-dimethylamino-benzoyl)-reserp- säuremethylester schmilzt bei 145-150 .
In analoger Weise, ausgehend von 3,5-Bis-di- methylamino-benzoylchlorid-hydrochlorid, erhält man den 18-0-(3',5'-Bis-dimethylamino-benzoyl)-reserp- säuremethylester vom F. 260-261 .

A solution of 2.59 parts by weight of this ester in 300 parts by volume of 95Q / o strength ethanol is reduced in the presence of 0.5 parts by weight of 10% palladium on charcoal catalyst with hydrogen at atmospheric pressure. The reaction mixture is filtered,
the filtrate was evaporated under reduced pressure and the gummy residue was taken up in methanol. The solution is left to stand at room temperature overnight, the crystals are filtered off and recrystallized from methanol. The crystalline 18-0- (2'-methoxy-5'-aminobenzoyl) -reserpsäuremethylester thus obtained contains 1 mol of methanol and melts at 148-151.
<I> Example 2 </I> 20.6 parts by weight (200/0 excess) of 3-dimethylamino-6-methoxybenzoyl chloride hydrochloride are added to a mixture of 27.5 parts by weight of methyl reserpate in 275 parts by volume of dry pyridine, with stirring and in a nitrogen atmosphere. The reaction mixture is left to stand for 18 hours at 20-25, slowly poured into 2750 parts by volume of ice-water with thorough stirring and filtered.
The residue is washed with water, taken up in 126 parts by volume of methylene chloride and the solution is filtered through a column of 25 parts by weight of magnesium aluminum silicate. The column is washed with about 190 parts by volume of methylene chloride, the combined methylene chloride solutions are evaporated to dryness and taken up in 100 parts by volume of methanol.
The 18-0- (2'-methoxy-5-dimethylaminobenzoyl) -reserp- acid methyl ester obtained in this way melts at 145-150.
In an analogous manner, starting from 3,5-bis-dimethylamino-benzoyl chloride hydrochloride, the 18-0- (3 ', 5'-bis-dimethylamino-benzoyl) -reserp- acid methyl ester with a melting point of 260-261 is obtained .
