Verfahren zur Herstellung von Abkömmlingen der 2-Cyano-4-acetylglutarsäure
Es ist bekannt, daß sich Aldehyde sowohl mit Acetessigester als auch mit Abkömmlingen
der Cyanessigsäure in Gegenwart von sekundären Aminen unter Wasseraustritt vereinigen
(Berichte der Deutschen .Chemischen Gesellschaft 29 [t896] S. 172; 31 [r898)
S. 730; Gazzetta Chimica Italiana 31, I [igoi] S.265). Dabei entstehen jedoch
Kondensationsprodukte aus Aldehyden und Acetessigester bzw. aus Aldehyden und den
Abkömmlingen der Cyanessigsäure. Zaun Beispiel erhält man aus Benzaldehyd und AceteSsigester
Benzalacetessigester und aus Benzaldehyd und Cyanessigester Benzalcyanessigester.
Es war zu erwarten, daB bei gleichzeitiger Einwirkung eines Aldehyds auf ein Gemisch
von Acetessigester und einen Abkömmling der Cyanessigsäure auch ein Gemisch der
Kondensationsprodukte entsteht. Wie nun gefunden wurde, erhält man, in Anlehnung
an die bekannte Arbeitsweise, aber bei gleichzeitiger Einwirkung von Aldehyden bzw.
Aldehydhydraten auf Acetessigester und Verbindungen der allgemeinen Formel N C-
C H2-R, wobei R die Nitrilgruppe, die veresterte Carboxylgruppe, die Säureamidgruppe
oder die in der Amidgruppe substituierte Säureamidgruppe bedeutet, in Gegenwart
eines-sekundären Amins, wie Piperidin, als Katalysator bei erhöhter Temperatur Ab-Icömmlinge
der z-Cyano-,I-acetylglutarsäure. Bei der Umsetzung tritt unter Austritt von i Molekül
Wasser Kondensation aller drei Komponenten ein. Der Vorgang wird z. B. für die Umsetzung
eines Aldehyds, wobei der Rest R' das Radikal des Aldehyds bedeutet, mit Acetessigester
und Cyanacetamid durch folgendes Formelschema veranschaulicht:
Bei der Verwendung anderer Verbindungen der Al lgemeinen Formel N C-C H2 R braucht
das Cyanacetamid nur sinngemäß durch jene ersetzt zu werden. Es ist überraschend,
daß
nicht ein Gemisch von Kondensationsprodukten, sondern ein einheitliches
Kondensationsprodukt, selbst wenn eine der nicht aldehydischen Komponenten im Überschuß

Die Kondensation wird zweckmäßig in Gegenwart eines indifferenten
Lösungsmittels ausgeführt, ihr Gelingen ist jedoch an die Natur und an die Anwesenheit
des Lösungsmittels nicht gebunden. Zweckmäßig verwendet man ein Lösungsmittel, in
welchem die Reaktionsteilnehmer genügend löslich sind und aus dem sich das Reaktionsprodukt

Beispiel i fob Gewichtsteile Benzaldehyd werden gemeinsam mit 8.a_
Gewichtsteilen Cyanacetamid, 13o Gewichtsteilen Acetessigester und 80o Volumteilen
Alkohol bis zur Lösung des Cyanacetamids auf 55 bis 6o° erwärmt. Nun setzt man 2
Gewichtsteile Piperidin zu, wobei sich die Lösung dunkler färbt und die Temperatur
spontan auf 7o bis 75° steigt. Dann wird 30 Minuten auf dieser Temperatur
gehalten und über :acht erkalten gelassen. Der entstandene Kristallbrei wird abgesaugt.
Das 2-Cyano-3-phenyl-d.-acetylglutarestersäureamid kristallisiert aus Methanol in
langen Nadeln, die beim Trocknen verwittern und bei I26° schmelzen. Die Ausbeute

Beispiel e Verwendet man im Beispiel i an Stelle des Cyanacetamids
66 Gewichtsteile Cyanacetonitril, so erhält man in analoger Weise das 2 - Cyano
- 3 - phenyl -4- acetylglutarestersäurenitril,welches aus Alkohol in ziemlich schwer
löslichen weißen Prismen vom Schmelzpunkt 186 bis I87° korr. kristallisiert. Die
Ausbeute beträgt 68 % der Theorie. Beispiel 3 151 Teile p-N itrobenzaldehyd,
8.4 Teile Cyanacetamid, i30 Teile Acetessigester werden mit 25o Volumteilen Alkohol
bis nahe zum Siedepunkt der Mischung erwärmt. Nun setzt man 2 Teile Piperidin zu,
wobei die Mischung infolge der frei werdenden Reaktionswärme ins Sieden gerät. Man
erhält io Minuten im Kochen, setzt das doppelte Volumen Wasser hinzu und läßt zur
Kristallisation erkalten. Nach mehrstündigem Stelien wird der entstandene dicke
Kristallbrei abgesaugt und nötigenfalls aus Alkohol umkristallisiert. Das a-Cyano-3-nitrophenyl-:l.-acetylglutarestersäureamid
kristallisiert in weißen nadeligen Prismen vom Schmelzpunkt I54.". DieAusbeute beträgt
3o0`0 derTheorie. Beispiel 7a Gewichtsteile Isobutyraldehyd, 8d. Gewichtsteile
Cyanacetamid, r30 Gewichtsteile Acetessigester werden in 400 Voluinteilen Alkohol
in der Wärme gelöst. Man läßt auf etwa .1o'' erkalten und setzt dann 2 Gewichtsteile
Piperidin zu. Die Mischung zeigt nach 5tägigem Stehen Kristallabscheidung, die sich
bei Zusatz eines gleichen Volumens Wasser noch vermehrt. Man erhält das 2-Cyano-3-i
sopropy l-4-acety l glutar estersätireamid in schönen weißen Prismen vom Schmelzpunkt
14.3°. Die Ausbeute beträgt 27 °/o der Theorie. Beispiel s 72 Gewichtsteile Isobutyraldehyd,
66 Gewichtsteile Malondinitril und i30 Gewichtsteile Acetessigester geben beim 3o
Minuten langen Erwärmen in Gegenwart von 2 Gewichtsteilen Piperidin auf etwa 7o
bis 8o° das 2-Cyano-3-isopropyl-.t-acetylglutarestersäurenitril in schönen weißen
Prismen. Man saugt ab und wäscht mit Methylalkohol nach. Die Verbindung schmilzt
bei 188 bis i89°. Die Ausbeute beträgt :251/, der Theorie. Beispiel 6 155 Gewichtsteile
Phenoxyacetaldehydhydrat, 8.4 Gewichtsteile Cyanacetamid, 130
Gewichtsteile
Acetessigester werden unter den gleichen Bedingungen wie in Beispie13 umgesetzt.
Man erhält das 2-Cyano-3-phenoxymethyl-.4.-acetylglutarestersäureamid in einer Ausbeute
von 63 °/o der Theorie. Es kristallisiert aus Methanol in glänzenden Platten vom
Schmelzpunkt 1.a.9°. Beispiel ? Verwendet man in Beispiel 6 an Stelle des Cyanacetamids
66 Gewichtsteile Malondinitril, so entsteht in einer Ausbeute von 6o °/o der Theorie
das 2-Cyano-3-phenoxymethyl-4.-acetylglutarestersäurenitril. Es kristallisiert aus
Alkohol in weißen Prismen. die ebenfalls wie das Amid bei id.g° schmelzen. Beispiel
8 132 Gewichtsteile Zimtaldehyd, 84 Gewichtsteile Cyanacetamid und 13o Gewichtsteile
Acetessigester werden mit Zoo Voluinteilen Alkohol und 2 Gewichtsteilen Piperidin
i Stunde auf dem Dampfbad unter Rückfluß erwärmt. Es entsteht eine hellgelbe, homogene
Lösung, aus der sich das sehr langsam kristallisierende 2-Cyano-3-styryl-.l-acetylglutarestersäureamid
im Laufe mehrerer Tage
abscheidet. Man kristallisiert aus Eisessig
um.
