worin X eine normale oder verzweigte Kohlenwasser stoffkette mit höchstens 5 C-Atomen, Y eine di- alkylierte Aminogruppe, beispielsweise eine Dimethyl- amino- oder Diäthylaminogruppe,
eine gegebenen falls weitere Heteroatome enthaltende cyclische Alkyleniminogruppe, z. B. eine Pyrrolidino-, Pi- peridino-, Morpholino- oder N'-Methyl-piperazino- gruppe, oder eine diesen Gruppen entsprechende quaternäre Ammoniumgruppe, und R Wasserstoff, eine Alkylgruppe mit höchstens 5 C-Atomen oder eine gegebenenfalls substituierte Aralkylgruppe be deuten.
Diese Verbindungen erhält man erfindungsgemäss durch Hydrierung von entsprechenden in den Benzol- ring.-n gegebenenfalls substituierten Diazepinderi- vaten der Formel

Als Substituenten in den Benzolringen können z. B. Halogenatome oder 1 bis 3 C-Atome enthal- tende Alkyl-, Alkoxy- oder Alkylmercaptogruppen auftreten.
Die Hydrierung erfolgt zweckmässig mit Wasser stoff und Edelmetallkatalysatoren, z. B. Nickel- oder Kupferchromitkatalysatoren, oder unter Verwendung von komplexen Hydriden, vorzugsweise Lithiumalu- miniumhydrid.
Das Hydrierungsprodukt kann anschliessend in 10-Stellung alkyliert, aralkyliert oder acyliert wer den, beispielsweise indem man es nach voraus gegangener Metallisierung mit einem Alkylhalogenid mit 1 bis 5 C-Atomen, einem gegebenenfalls substi tuierten Aralkylhalogenid oder einem Carbonsäure- halogenid behandelt.
Soweit nicht von einem quater- nären Ammoniumderivat ausgegangen wurde, kann das Produkt auch nachträglich in das quaternäre Ammoniumderivat übergeführt werden, beispielsweise durch Behandeln mit Dialkylsulfaten, Alkylhalogeni- den oder Sulfonsäurealkylestern.
Die Verbindungen entsprechend Formel 1 können sowohl in Form der freien Basen als auch in Form ihrer Salze mit geeigneten Säuren, wie Halogen wasserstoffsäuren, Schwefelsäure, Salpetersäure, Phosphorsäure, Essigsäure, Oxalsäure, Malonsäure, Bernsteinsäure, Äpfelsäure, Maleinsäure, Weinsäure oder Toluolsulfonsäure, gewonnen werden.
Die Ausgangssubstanzen (11) gewinnt man z. B. durch Cyclodehydratisierung von N(-X-Y)-o-Acyl- amino-diphenylaminen in Gegenwart von Polyphos phorsäure.
Die in der beschriebenen Weise erhaltenen Sub stanzen sind neue Verbindungen, welche als Wirk- stoffe in Arzneimitteln Verwendung finden.
<I>Beispiel 1</I> Zu 1,0 g Lithiumaluminiumhydrid in 30 ml abso lutem Tetrahydrofuran liess man unter Rühren innert 20 Minuten eine Lösung von 1,40 g 5-(y -Dimethylamino-propyl)- 5H-dibenzo[b,e] [1,4]diazepin in 35 ml absolutem Tetrahydrofuran zutropfen. Unter Rühren wurde dann 3 Stunden auf Rückfluss erhitzt, worauf man überschüssiges Lithiumalumi- niumhydrid mit Essigester zerstörte und das Reak tionsgemisch im Vakuum zur Trockne brachte. Der Rückstand wurde zwischen Wasser und Äther ver teilt.
Nach Abtrennen von anorganischen Hydroxyden durch Filtrieren wurde das basische Produkt durch Extraktion mit Essigsäure, Fällung mit Ammoniak und Extraktion der freien Base mit Äther isoliert. Man erhielt 1,02 g (72<B>%</B> der Theorie) 5-(y-Dimethylamino-propyl)-10,11-dihydro- 5H-dibenzo[b,e] [1,4]diazepin mit dem Schmelzpunkt 99-101 C.
<I>Beispiel 2</I> Bei gleichem Vorgehen wie in Beispiel 1 erhielt man aus 6,3 g 5-()'-Dimethylamino-äthyl)-7-chlor- 5H-dibenzo[b,e] [1,4]diazepin 4,7 g 5-((3-Dimethylamino-äthyl)-7-chlor-10,11-dihydro- 5H-dibenzo[b,e] [1,4]diazepin in Form von massiven Prismen vom Schmelzpunkt 114-116 C (aus Äther Petroläther).
<I>Beispiel 3</I> Bei gleichem Vorgehen wie in Beispiel 1 erhielt man aus 5,8 g 5-(y-Dimethylamino-propyl)-7-chlor- 5H-dibenzo[b,e] [1,4]diazepin 4,5 g 5-(y-Dimethylamino-propy l)-7-chlor-10,11- dihydro-5H-dibenzo[b,e][1,4]diazepin in Form von körnigen Kristallen vom Schmelzpunkt 114-116 C (aus Äther/Petroläther).
<I>Beispiel 4</I> Bei gleichem Vorgehen wie in Beispiel 1 erhielt man aus 4,3 g 5-(/3-Dimethylamino-äthyl)-7-chlor-11-methyl- 5H-d.ibenzo[b,e] [1,4]diazepin 2,9 a <B>b</B> d,1-5-(/)'-Dimethylamino-äthyl)-7-chlor-10,11- dih,ydro-11-me.thyl-5H-dibenzo[b,e] [1,4]diazepin in Form von massiven Körnern vom Schmelzpunkt 104-106 C (aus Äther/Petroläther). <I>Beispiel 5</I> 20 g 5-(/)-Dimethylamino-äthyl)-11-methyl- 5H-dibenzo[b,e] [1,4]diazepin wurden in Gegenwart von 4 g Raney-Nickel,
1 g Palladium auf Kohle (5 %) und 9 g Kaiiumhydroxyd- Perlen in 140 ml Methanol bei Zimmertemperatur unter leichtem überdruck in Wasserstoffatmosphäre ,bis zum Stillstand der Wasserstoffaufnahme geschüt telt. Nach Abnutschen vom Katalysator und Ein engen im Vakuum verteilte man den Rückstand zwi- sehen Äther und Wasser, wusch die ätherische Lö sung mit Wasser und arbeitete sie in üblicher Weise auf.
Beim Umkristallisieren aus Äther/Petroläther unter Klärung mit Aluminiumoxyd erhielt man 16 g d,1-5-(l'-Dimethylamino-äthyl)-10,11-dihydro- 11-methyl-5H-dibenzo[b,e] [1,4]diazepin in Form von weissen Prismen mit dem Schmelzpunkt 75-76 C.
Durch Umsetzung mit Dimethylsulfat kann man daraus das entsprechende Methosulfat in Form von prismatischen Nadeln mit dem Schmelzpunkt 204 bis 208v C erhalten.
<I>Beispiel 6</I> 4,5 g des gemäss Beispiel 5 erhaltenen d,1-5-(/3-Dimethylamino-äthyl)-10,11-dihydro- 11-methyl-5H-dibenzo[b,e] [ 1,4]diazepins wurden mit 30 cm3 Acetanhydrid 4 Stunden lang auf Rückfluss erhitzt. Nach Eindampfen am Vakuum und Auflösen des Rückstandes in Wasser wurde die Base mit Ammoniak freigesetzt und in Äther auf genommen. Die ätherische Lösung wurde in üblicher Weise aufgearbeitet.
Durch Umkristallisieren aus Aceton/Petroläther unter Klärung mit Aluminium oxyd erhielt man 3,8 g der entsprechenden 10-Acetyl- Verbindung in Form von schwach gelblichen Prismen mit dem Schmelzpunkt 126-128 C.

Man erhielt 1,8 g 5-(ss-Dimethylamino-äthyl)-10-methyl- 10,11-dihydro-5H-dibenzo[b;e] [1,4]diazepin als dickes Öl vom Siedepunkt 165-167 C/0,02 Torr. Das Methosulfat konnte aus Aceton kristallin gewon nen werden, Schmelzpunkt 177-182 C.
<I>Beispiel 19</I> Bei gleichem Vorgehen wie in Beispiel 18, jedoch unter Verwendung von Benzylchlorid an Stelle von Monochlormethan, erhielt man 5-(/3-Dimethylamino-äthyl)-10-benzyl- 10,11-dihydro-5H-dibenzo[b;e] [1,4]diazepin als dickes Öl vom Siedepunkt 212-214 Ci0,08 Tarr. in einer Ausbeute von 63 0/0 der Theorie. Das Metho- sulfat, welches aus Aceton zur Kristallisation gebracht werden konnte, zeigte den Schmelzpunkt 138-143 C.
<I>Beispiel 20</I> Das Produkt von Beispiel 1 wurde gemäss Bei spiel 18 methyliert, wobei man 5-(y-Dimethylamino-propyl)-10-methyl- 10,11-dihydro-5H-dibenzo[b,e] [1,4]diazepin als dickes Öl vom Siedepunkt 164-165 C/0,01 Torr. in einer Ausbeute von 68 % der Theorie erhielt. <I>Beispiel 21</I> Das Produkt von Beispiel 1 wurde gemäss Bei spiel 19 benzyliert,
wobei man 5-(y-Dimethylamino-propyl)-10-benzyl- 10,11-dihydro-5H-dibenzo[b,e][1,4]diazepin in einer Ausbeute von 64 % der Theorie in Form eines dicken Öls vom Siedepunkt 207 C/0,01 Torr. erhielt, welches beim Stehen kristallin erstarrte; Schmelzpunkt 45-47 C (aus kaltem Petroläther).
Bei gleichem Vorgehen wie in den vorausgehen den Beispielen erhielt man ferner die in der nach folgenden Tabelle aufgeführten Verbindungen.

As substituents in the benzene rings, for. B. halogen atoms or 1 to 3 carbon atoms containing alkyl, alkoxy or alkyl mercapto groups occur.
The hydrogenation is conveniently carried out with hydrogen and noble metal catalysts, eg. B. nickel or copper chromite catalysts, or using complex hydrides, preferably lithium aluminum hydride.
The hydrogenation product can then be alkylated, aralkylated or acylated in the 10-position, for example by treating it, after previous metallization, with an alkyl halide having 1 to 5 carbon atoms, an optionally substituted aralkyl halide or a carboxylic acid halide.
Unless a quaternary ammonium derivative was assumed, the product can also be converted subsequently into the quaternary ammonium derivative, for example by treatment with dialkyl sulfates, alkyl halides or sulfonic acid alkyl esters.
The compounds according to formula 1 can be obtained both in the form of the free bases and in the form of their salts with suitable acids, such as hydrohalic acids, sulfuric acid, nitric acid, phosphoric acid, acetic acid, oxalic acid, malonic acid, succinic acid, malic acid, maleic acid, tartaric acid or toluenesulfonic acid .
The starting substances (11) are obtained, for. B. by cyclodehydration of N (-X-Y) -o-acylamino-diphenylamines in the presence of polyphosphoric acid.
The substances obtained in the manner described are new compounds which are used as active ingredients in drugs.
<I> Example 1 </I> To 1.0 g of lithium aluminum hydride in 30 ml of absolute tetrahydrofuran, a solution of 1.40 g of 5- (γ-dimethylamino-propyl) -5H-dibenzo [b , e] [1,4] diazepine in 35 ml of absolute tetrahydrofuran is added dropwise. The mixture was then heated to reflux for 3 hours while stirring, whereupon excess lithium aluminum hydride was destroyed with ethyl acetate and the reaction mixture was brought to dryness in vacuo. The residue was divided between water and ether.
After removing inorganic hydroxides by filtration, the basic product was isolated by extraction with acetic acid, precipitation with ammonia and extraction of the free base with ether. 1.02 g (72% of theory) of 5- (γ-dimethylamino-propyl) -10,11-dihydro-5H-dibenzo [b, e] [1,4] diazepine were obtained the melting point 99-101 C.
<I> Example 2 </I> Using the same procedure as in Example 1, 6.3 g of 5 - () '- dimethylamino-ethyl) -7-chloro-5H-dibenzo [b, e] [1,4 ] diazepine 4.7 g of 5 - ((3-dimethylamino-ethyl) -7-chloro-10,11-dihydro-5H-dibenzo [b, e] [1,4] diazepine in the form of massive prisms with a melting point of 114- 116 C (from ether petroleum ether).
<I> Example 3 </I> Using the same procedure as in Example 1, 5.8 g of 5- (γ-dimethylamino-propyl) -7-chloro-5H-dibenzo [b, e] [1,4] diazepine 4.5 g of 5- (γ-dimethylamino-propy l) -7-chloro-10,11-dihydro-5H-dibenzo [b, e] [1,4] diazepine in the form of granular crystals with a melting point of 114-116 C (from ether / petroleum ether).
<I> Example 4 </I> Using the same procedure as in Example 1, 4.3 g of 5 - (/ 3-dimethylamino-ethyl) -7-chloro-11-methyl-5H-dibenzo [b, e] [1,4] diazepine 2.9 a <B> b </B> d, 1-5 - (/) '- dimethylamino-ethyl) -7-chloro-10,11- dih, ydro-11- me.thyl-5H-dibenzo [b, e] [1,4] diazepine in the form of massive grains with a melting point of 104-106 C (from ether / petroleum ether). <I> Example 5 </I> 20 g of 5 - (/) - dimethylamino-ethyl) -11-methyl-5H-dibenzo [b, e] [1,4] diazepine were in the presence of 4 g of Raney nickel,
1 g of palladium on charcoal (5%) and 9 g of potassium hydroxide beads in 140 ml of methanol at room temperature under a slight excess pressure in a hydrogen atmosphere, until the hydrogen uptake stops. After filtering off the catalyst with suction and concentrating in vacuo, the residue was distributed between ether and water, the ethereal solution was washed with water and worked up in the usual way.
Recrystallization from ether / petroleum ether with clarification with aluminum oxide gave 16 gd, 1-5- (l'-dimethylamino-ethyl) -10,11-dihydro-11-methyl-5H-dibenzo [b, e] [1,4 ] diazepine in the form of white prisms with a melting point of 75-76 C.
The corresponding methosulfate can be obtained therefrom in the form of prismatic needles with a melting point of 204 to 208 ° C. by reaction with dimethyl sulfate.
<I> Example 6 </I> 4.5 g of the d, 1-5 - (/ 3-dimethylamino-ethyl) -10,11-dihydro-11-methyl-5H-dibenzo [b, e ] [1,4] diazepines were refluxed with 30 cm3 of acetic anhydride for 4 hours. After evaporation in vacuo and the residue dissolved in water, the base was released with ammonia and taken up in ether. The ethereal solution was worked up in the usual way.
Recrystallization from acetone / petroleum ether with clarification with aluminum oxide gave 3.8 g of the corresponding 10-acetyl compound in the form of pale yellowish prisms with a melting point of 126-128 C.
