Verfahren zur Herstellung von neuen N-heterocyclischen Verbindungen Die vorliegende Erfindung bezieht sich auf ein Verfahren zur -Herstellung neuer N-heterocyclischer Verbindungen mit wertvollen pharmakologischen Eigenschaften.
Es wurde überraschenderweise gefunden, dass Derivate des Iminodibenzyls (10,11-Dihydro-5H- dibenzo[b,f]azepin) der Formel

worin R einen niedermolekularen Alkylrest oder einen Cycloalkylrest und alkylen einen geradkettigen oder verzweigten Alkylenrest mit 2-6 Kohlenstoffatomen bedeuten, wertvolle pharmakologische Eigenschaften, insbesondere antiallergische, antiemetische,
antikon vulsive und sedative Wirksamkeit besitzen.
Zur Herstellung der neuen Verbindungen der Formel I lässt man erfindungsgemäss auf Verbindun gen der Formel

worin Rl einen Arylmethyl- oder Diarylmethylrest und R, Wasserstoff oder einen Arylmethyl- oder Diarylmethylrest bedeuten, katalytisch aktivierten Wasserstoff bis zur Aufnahme von im wesentlichen einem bzw. zwei Mol je nach der Bedeutung von R, einwirken (das heisst zwei Mol, wenn R2 nicht Wasserstoff bedeutet). Die Hydrogenolyse des Restes R1 kann z.
B., falls R2 Wasserstoff bedeutet, in einer organischen oder vorzugsweise in saurer, wässrig organischer Lösung, z. B. in methanohscher Salz säure, in Gegenwart eines Nickel- oder Edelmetall- katalysators erfolgen. So wird z.
B. die Hydrogeno- lyse in Gegenwart von Palladiumkohle bei Raum temperatur und Niederdruck bis zur Aufnahme von ungefähr der molaren Menge Wasserstoff durchge- führt. Zur gleichzeitigen Abspaltung eines von Wasserstoff verschiedenen Restes R2 sind meist etwas energischere Reaktionsbedingungen, z. B.
Hydrierung in Gegenwart von Raney-Nickel bei Temperaturen bis etwa 80 und Drücken bis etwa 50 Atmosphären notwendig; die Hydrierung wird in diesem Falle erst nach Aufnahme von ungefähr der doppeltmolaren Menge Wasserstoff abgebrochen.
In den Ausgangsstoffen der allgemeinen Formel II bedeutet R1 vorzugsweise ein Benzyl- oder Benz hydrykest und R2 Wasserstoff oder ein Benzylrest. Als Reste R kommen dabei z.
B. der Methyl-, Äthyl-, n-Propyl-, Isopropyl-, n-Butyl-, Isobutyl-, sek.-Butyl-, n-Amyl-, Isoamyl-, n-Hexyl-, Cyclopentyl-, Cyclo- hexyl-, Cyclohexylmethyl-, Cycloheptyl- und Cyclo- octylrest in Betracht.

können auch Alkalimetallverbindungen von N-Acylderivaten von primären Aminen der Formel IV, z. B. von niedermolekularen N-Alkyl-acetamiden, N-Alkyl-formamiden,
N-Cycloalkyl-acetamiden oder N-Cycloalkyl-formamiden mit reaktionsfähigen Estern von Verbindungen der Formel III umgesetzt und die unmittelbar erhaltenen N-Acylderivate zu Verbindungen der Formel 1I hy- drolysiert werden.
Zu reaktionsfähigen Estern von Verbindungen der Formel III gelangt man beispielsweise durch Umsetzung von Alkalimetallverbindungen von 2 Arylmethoxy-iminodibenzyl oder 2 Diaryhnethoxy-iminodibenzyl mit reaktionsfähigen, vorzugsweise gemischten Di- estern von Alkylenglykolen, wie z. B.
1-Chlor-2-brom-äthan, 1-Chlor-3-brom-propan, 1,2-Dichlor-propan, 1,2-Dibrom-propan, 1-Chlor-4-brom-n-butan, 1,4-Dibrom-butan, 1-Brom-2-methyl-3-chlor-propan oder oi- Halogenalkylestern der p-Toluolsulfonsäure, 2,4 Dinitro-benzolsulfonsäure oder Methansulfonsäure. Ferner erhält man solche reaktionsfähige Ester z. B.
auch durch Umsetzung der obengenannten Alkali- metallverbindungen mit Alkylenoxyden und Behand lung der entstandenen Hydroxyalkylverbindungen mit anorganischen Säurehalogeniden, Arylsulfonsäure- chloriden oder Methansulfochlorid, wobei man 5-Halogenalkyl-, 5-Arylsulfonyloxy-alkyl- bzw.
Me- thansulfonyloxyalkyl-derivate von 2-Arylmethoxy- oder 2 Diarylmethoxy-iminodibenzyl erhält.
2 - Arylmethoxy- und 2 - Diarylmethoxy - imino- dibenzyle, z. B. das 2 Benzyloxy-iminodibenzyl, er hält man beispielsweise durch Oxydation von Imino- dibenzyl mittels einer Lösung von Kaliumnitroso- disulfonat (Fremysches Salz), Reduktion oder Hy drierung, z.
B. in Methanollösung in Gegenwart von Lindlar-Katalysator, des erhaltenen 2-Oxo-10,11- dihydro-2H-dibenzo[b,f]azepin und Verätherung des, entstandenen 2-Hydroxy-iminodibenzyls durch Um setzung mit reaktionsfähigen Estern von Arylmetha- nolen oder Diarylmethanolen in Gegenwart säure- bindender Mittel, z.
B. mit Benzylehlorid in alkoho- lischer Kalilauge.
Mit anorganischen oder organischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Äthandisulfon- säure, Essigsäure, Citronensäure, Äpfelsäure, Bern steinsäure, Maleinsäure, Fumarsäure, Weinsäure,
Benzoesäure und Phthalsäure bilden die erfindungs- gemäss erhaltenen sekundären Basen wasserlösliche Salze.
Im nachfolgenden Beispiel bedeuten Teile Ge wichtsteile; diese verhalten sich zu Volumteilen wie g zu cm3. Die Temperaturen sind in Celsiusgraden angegeben.
<I>Beispiel</I> a) 7,5 Teile 2 Benzyloxy-iminodibenzyl und 5,5 Teile 1-Chlor-3-brom-propan werden in 150 Volum- teilen abs. Benzol gelöst und bei 58-60 mit einer Suspension von 1,5 Teilen Natriumamid in Toluol portionenweise versetzt. Man rührt das Reaktions gemisch anschliessend 16 Stunden bei dieser Tempe ratur.
Nach dem Abkühlen wird es mit Wasser ver setzt, die Benzollösung abgehoben, gründlich mit Wasser gewaschen, getrocknet und eingedampft, wo bei das 5-(y-Chlor-propyl)-2-benzyloxy-iminodibenzyl als viskoses<B>öl</B> zurückbleibt.
b) 8,5 Teile der obigen Chlorpropylverbindung werden in 25 Volumteilen abs. Benzol gelöst und im Autoklaven mit 40 Volumteilen bei 5 mit Me- thylamin gesättigtem Methanol 12 Stunden auf 80 bis 90 erhitzt.
Nach dem Abkühlen wird das Lö sungsmittel abgedampft und der Rückstand mit Wasser versetzt und erschöpfend ausgeäthert. Durch dreimaliges Schütteln mit verdünnter Essigsäure wer den der ätherischen Lösung die basischen Anteile entzogen.
Die vereinigten essigsauren Auszüge werden alkalisch gestellt und ausgeäthert. Die ätherische Lö sung wird gewaschen, getrocknet und eingedampft. Der ölige Rückstand wird im Hochvakuum destilliert, wobei das 5 - (y -Methylamino-propyl)-2-benzyloxy- iminodibenzyl unter 0,02 mm Druck bei 225 über geht.
c) 3,4 Teile des obigen Destillats werden in 9,2 Volumteilen 1n Salzsäure gelöst und mit Methanol auf 100 Volumteile ergänzt. Diese Lösung wird in Gegenwart von 1 Teil 5a/miger Palladiumkohle bei Raumtemperatur und Niederdruck hydriert, bis die berechnete Menge Wasserstoff aufgenommen ist. Man filtriert vom Katalysator ab, engt das Filtrat ein und versetzt den Rückstand mit verdünnter Ammoniaklösung bis zur alkalischen Reaktion.
Die ausgeschiedenen Kristalle werden abgesaugt und aus Aceton umkristallisiert. Das so erhaltene 5-(y-Methyl- amino-propyl)-2-hydroxy-iminodibenzyl schmilzt bei 160 .
