Die neuen Verbindungen haben wertvolle pharmakologische Eigenschaften, insbesondere eine blut druckerniedrigende und eine spasmolytische Wirkung.
Bei der Durchführung des erfindungsgemässen Verfahrens lässt man z. B. 1 mol der Verbindung der Formel III mit mindestens 1 mol der freien Base der Formel II reagieren. Vorzugsweise wählt man zwei Äquivalente letzterer Verbindung, wobei 1 mol als Säurebindemittel wirksam ist. Man kann auch 1 Äquivalent der Verbindung II und weiter 1 Äquivalent Säurebindemittel, z. B. ein Tertiäramin, wie z. B. Pyridin, Kollidin oder Tri-Äthanolamin, verwenden.
Die Reaktion kann ohne oder vorzugsweise in Anwesenheit eines Lösungsmittels ausgeführt werden.
Dazu eignen sich beispielsweise aromatische Kohlenwasserstoffe, wie z. B. Benzol, Toluen, Xylen, oder Chloroform, Tetrachlorkohlenstoff, Aceton und Acetonitril.
Auch Äther wie z. B. Diäthyläther, Diisopropyl äther, sind hierzu als Lösungsmittel anwendbar.
Weiter können grundsätzlich auch aliphatische Kohlenwasserstoffe als Lösungsmittel verwendet werden. Diese sind jedoch sehr wenig polar, wodurch die Kopplungsreaktion gewöhnlich langsam vollzogen wird.
Es empfiehlt sich, die Reaktion bei höherer Temperatur durchzuführen. Bei Anwendung eines Lösungsmittels ist es am einfachsten, die Reaktion bei der Siedetemperatur des Lösungsmittels stattfinden zu lassen. Eine Temperatur zwischen 40 und 130 C liefert gute Ergebnisse.
Die Dauer der Reaktion hängt von der Wahl des Lösungsmittels und der Reaktionstemperatur ab. Mit Rücksicht darauf kann die Reaktionsdauer zwischen einigen Stunden und etwa zwei Tagen schwanken.
Verbindungen der Formel III können auf verschiedene Weise hergestellt werden. Man kann z. B. eine substituierte Benzoesäure der Formel

oder ein funktionelles Derivat davon mit einem Halogenalkohol der Formel Hlg-Y-OH, worin Hlg Halogen bedeutet, verestern.
Zur Herstellung der Verbindungen der Formel III kann man auch eine Säure der obengenannten Formel mit einem Dialkohol der Formel HO-Y-OH verestern. Es empfiehlt sich, die Reaktion mit mindestens einer äquimolaren Menge des Dialkohols durchzuführen, um zu verhüten, dass Verbindungen hergestellt werden, bei denen beide Alkoholgruppen verestert sind. Die erhaltenen Hydroxy-Ester können dann durch Halogenierungsmittel, die die Hydroxyl Gruppe durch ein Halogenatom ersetzen, in die ge wünschten Verbindungen der Formel III umgewandelt werden. Als Halogenierungsmittel kommen hierbei z. B. Thionylchlorid, Phosphorpentachlorid, Phosphoroxychlorid, Phosphortribromid oder Thio nylbromid in Betracht.
Gemäss einem dritten Verfahren können Verbindungen der Formel III auch dadurch hergestellt werden dass man ein Metallsalz, vorzugsweise ein Alkalisalz, z. B. das Natriumsalz einer Benzoesäure der obengenannten Formel, mit einer aliphatischen Dihalogenverbindung der Formel Hlg-Y-Hlg verestert. Die Reaktion wird wieder vorzugsweise mit mindestens einer äquimolaren Menge der Dihalogenverbindung durchgeführt, um zu verhüten, dass auch das zweite Halogenatom sich an der Veresterung beteiligt. Als Halogenverbindung wird vorzugsweise die entsprechende Dichlor-oder Dibromverbindung angewandt.
Aus den nach dem erfindungsgemässen Verfahren hergestellten Verbindungen können auch Salze hergestellt werden, wie z. B. das Salzsäure-, Brom wasserstoffsäure-, Schwefelsäure-, Phosphorsäure-, Essigsäure-oder Zitronensäuresalz.
Die erfindungsgemäss erhaltenen Verbindungen können in an sich bekannter Weise zu Heilmitteln, z. B. in der Form von Tabletten, Dragées, Injektionsflüssigkeiten und dergleichen verarbeitet werden.
In den nachstehenden Ausführungsbeispielen bedeutet Ather stets Diäthyläther und Alkohol stets 96 böiges Äthanol.
Beispiel I 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-6- [N-äthyl- N-(2"-phenyl-äthyl)-amino]-hexan a) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-6-brom- hexan.
Diese Verbindung wurde auf zwei Weisen hergestellt, die weiter unten unter 1 und 2 beschrieben werden.
1. Eine Suspension von 23, 5 g (0, 1 mol) des Natriumsalzes von 3, 4, 5-Trimethoxy-benzoesäure in einer Lösung von 24, 5 g (0, 1 mol) 1, 6-Dibrom-hexan in 50 ml Xylen wurde während 60 Stunden in einem Bad von 154 C aufbewahrt. Nach Abkühlung wurde der Niederschlag abgesaugt ; das Xylen wurde von dem Filtrat in Vakuum verdampft und das Residuum in Vakuum destilliert. Die Ausbeute betrug 10, 5 g (29"/o). Siedepunkt 187 bis 189 C ; 0, 1 mm. Bromgehalt 22, 22"/ (berechnet 21, 33"/o). Bei der Destillierung wurde ausserdem 14, 4 g 1, 6-Dibromhexan zurückgewonnen.
2. 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-6-brom- hexan.
23, 5 g (0, 1 mol) des Natriumsalzes von 3, 4, 5 Trimethoxy-benzoesäure und 30 g (0, 12 mol) 1, 6 Dibrom-hexan wurden 300 ml wasserfreies Acetonitril zugesetzt, und das Gemisch wurde während 43 Stunden gekocht. Der Niederschlag wurde abgesaugt und mit Äther gewaschen. Das Filtrat wurde in Vakuum eingedampft, und dem Residuum wurde 150 ml Atherzugesetzt. DerNiederschlagwurde abgesaugtund der Äther aus dem Filtrat verdampft. Das Residuum wurde in Vakuum destilliert. Die Ausbeute war 16, 5 g (44"/o). Siedepunkt 186 bis 191"C'0, 08 mm.
Bromgehalt 24, 20/o (berechnet 21, 3'Vo). b) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-6- [N- äthyl-N- (2"-phenyl-äthyl)-amino]-hexan.
Eine Lösung von 10, 3 g (0, 03 mol) 1- (3', 4', 5'- Trimethoxy-benzoyloxy)-6-brom-hexan und 9 g (0, 06 mol) N-Äthyl-N- (2-phenyl-äthyl)-amin in 75 ml trockenem Benzol wurde während 70 Stunden gekocht. Darauf wurde das Benzol in Vakuum verdampft und dem Residuum 100 ml trockener Äther zugesetzt. Der entstandene Niederschlag wurde abgesaugt und der Äther aus dem Filtrat verdampft.
Das ölartige Residuum hatte ein Gewicht von 11, 9 g und ein Äquivalentgewicht von 376 (berechnet 443).
Durch Zusatz von ln alkoholischer Salzsäurelösung wurde der Stoff in das Hydrochlorid umgewandelt.
Versuche, dieses Hydrochlorid aus Methyläthyl keton-Ather und Alkohol-Äther zu kristallisieren, hatten keine Resultate. Das Hydrochlorid wurde darauf durch Verdampfung der Lösungsmittel in Form eines ) ls erhalten und in 150 ml Wasser gelöst. Diese Lösung wurde zweimal mit 40 ml Äther extrahiert und darauf mit 10 ml konzentriertem Ammoniak alkalisch gemacht, worauf wieder dreimal mit 50 ml Äther extrahiert wurde. Die Lösung in Äther wurde auf Natriumsulfat getrocknet, filtriert und der Äther aus dem Filtrat verdampft. Das Residuum hatte noch N-Äthyl-N- (2-phenyl-äthyl)-amin und wurde daher während drei Stunden bei 1 mm auf 120 C aufbewahrt.
Die Ausbeute war 2, 4 g (18"/.). Äq. Gewicht 440 (berechnet 443). UV-Ab- sorptionsspektrum : ?max = 265mÁ.?max = 11 000.

350 mg der Base wurde mit alkoholischer Salzsäure in das Hydrochlorid umgewandelt, das aus einem Gemisch von Methyläthylketon und Äther kri- stallisiert wurde. Ausbeute 270 mg mit einem Schmelzpunkt von 65 bis 67 C. Chlorgehalt : 7, 43 1/o (berechnet 7, 41"/e). W. D. K. 31. Spasm. Akt. 20-40.
Beispiel 11 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-6- [ l"-methyl 2"-(4"-methoxy-phenyl)-äthyl}-amino]- hexanhydrochlorid
Eine Lösung von 7, 7 g (0, 04 mol) N-Athyl-N [l-methyl-2-(4'-methoxy-phenyl)-äthyl]-amin und 6, 6 g (0, 02 mol) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-6brom-hexan in 50 ml trockenem Acetonitril wurde während 90 Stunden gekocht. Das Acetonitril wurde in Vakuum verdampft und dem Residuum 50 ml trockener Äther zugesetzt. Der Niederschlag wurde abgesaugt und der Äther aus dem Filtrat verdampft.
Das Residuum wurde in 45 ml 0, 5n Salzsäure gelöst, und diese Lösung wurde zweimal mit 25 ml Äther gewaschen. Die Wasserschicht wurde alkalisch gemacht mit 5 ml konzentriertem Ammoniak und dreimal mit 30 ml Äther extrahiert. Die Ätherextrakte wurden auf Na2SO4 getrocknet, filtriert und der Äther verdampft. Das Residuum wurde während drei Stunden in Vakuum (0, 1 mm) auf 90 C gehalten und darauf in 24 ml 0, 76n alkoholischer Salzsäure gelöst. Der Alkohol wurde verdampft und das Residuum aus einem Gemisch von 15 ml Methyläthyl- keton und 10 ml Äther kristallisiert. Ausbeute : 6, 0 g (60"/ (t). Schmelzpunkt 137 bis 140 C.
Chlorgehalt 7, 03% (berechnet 6, 78 /o) UV-Absorptionsspektrum ?max = 267,5 mÁ, ?max = 12 600.
Beispiel III 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-3- [N= äthyl-
N-l"-methyl-2"-(4"'-methoxy-phenyl)-äthyl}- aminol-propan a) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-3-brom propan.
Der Stoff wurde gemäss dem im Beispiel I a) beschriebenen Verfahren durch Veresterung des Natriumsalzes von 3, 4, 5-Trimethoxy-benzoesäure mit 1, 3-Dibrom-propan hergestellt. Ausbeute : 5211/o.
Siedepunkt 165 bis 169¯ C/O, 3 mm. Bromgehalt : 23, 7"/o (berechnet 24%). b) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-3- [N- Ïthyl-N-{1"-methyl-2"-(4"-methoxy-phenyl) äthyl}-amino]-propan.
Der Stoff wurde wie in Beispiel II beschrieben durch Kopplung der unter a) beschriebenen Verbindung mit N-Äthyl-N- [1-methyl-2- (4'-methoxy phenyl)-äthyl]-amin hergestellt. Ausbeute 54"/ Äq.
Gewicht 432 (berechnet 445). UV-Absorptions spektrum 265, 5 mut. 11600. Spasm.

Beispiel IV
1-(3', 4', 5'-Trimethoxy-benzoyloxy)-6-[N-äthyl-
N- l"-methyl-2"- (3"', 4"'-dimethoxy-phenyl)-äthyl}- amino]-hexan
Die Herstellung fand gemäss dem in Beispiel 1 b) beschriebenen Verfahren statt, indem 1-(3', 4', 5'-Tri- methoxy-benzoyloxy)-6-brom-hexan (hergestellt nach Beispiel I a) in Toluen als Lösungsmittel während 100 Stunden mit 1- (3', 4'-Dimethoxy-phenyl)-2- (N- äthyl-amino)-propan gekocht wurde. Ausbeute 50 /o.
¯q. Gewicht 485 (berechnet 517). UV-Absorptions spektrum 270 mu,. 12 100. Spasm.

Beispiel V 1-(3', 4', 5'-Trimethoxy-benzoyloxy)-3-[N-äthyl-
N-(2"-phenyl-Ïthyl)-amino]-propan a) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-3-chlor- propan.
Eine Lösung von 13 g (0, 056 mol) 3, 4, 5-Trimethoxy-benzoyl-chlorid und 25 ml (0, 23 mol) 3-Chlor-propanol-1 in 25 ml trockenem Toluen wurde während 6 Stunden gekocht. Nachdem das Toluen und der ¯berschu¯ an 3-Chlor-propanol-1 in Vakuum verdampft worden war, wurde das Residuum in Vakuum destilliert. Die Ausbeute war 14 g (87 /o). Siedepunkt 179 bis 180 Cl0, 7 mm. Chlorgehalt 11, 0 /o (berechnet 12, 3%).
UV-Absorptionsspektrum imax = 266 m, u. ?max = 10 250. b) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-3- [N- äthyl-N- (2"-phenyl-äthyl)-amino]-propan.
Eine Lösung von 3, 75 g (0, 025 mol) N-Äthyl N- (2-phenyl-äthyl)-amin und 4, 75 g (0, 016 mol) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-3-chlor-propan in 50 ml Acetonitril wurde während 230 Stunden gekocht. Nach Verdampfung des Acetonitrils wurde dem Residuum 25 ml Äther zugesetzt. Es entstand ein Niederschlag, der abgesaugt und mit 20 bzw.
30 ml Äther gewaschen wurde. Das Ätherfiltrat wurde mit 10 ml 2n Salzsäurelösung und darauf mit 15 ml Wasser extrahiert. Die Gesamtextrakte wurden mit 20 ml Äther gewaschen, und darauf wurde 15 ml 2N-NaOH zugesetzt und dreimal mit 25 ml Äther extrahiert. Die Gesamtätherextrakte wurden auf Natriumsulfat getrocknet, filtriert, der Äther verdampft und das Residuum während 10 Stunden auf 100 C unter einem Druck von 0, 25 mm gehalten.
Die Ausbeute war 1, 36 g (21a/o). Aquivalentgewicht 408 (berechnet 401). UV-Absorptionsspektrum x", a3 = 267m, u. emaX = 19 550. Von dieser freien Base wurden 100 mg mit 67 mg Pikrinsäure in 25 ml Alkohol gelöst. Nach einigen Tagen Aufbewahrung im Eisschrank kristallisierte sich das Pikrat. Schmelzpunkt 135 bis 137 C. W. D. K. 600 Sp. Akt. 13.

Die Herstellung wurde gemäss dem in Beispiel I a) beschriebenen Verfahren durchgeführt, indem das Natriumsalz von 3, 4, 5-Trimethoxy-benzoesäure mit 1, 5-Dibrom-pentan verestert wurde. Die Ausbeute war 56 /o. Siedepunkt 182 bis 196 C ; 0, 05 mm.
Bromgehalt : 22, 16 /o (berechnet 22, 14%). Schmelzpunkt 79 bis 81 C. b) 1- (3', 4', 5'-Trimethoxy-benzoyloxy)-5- [N- äthyl-l"-methyl-2"- (4"'-methoxy-phenyl) äthyl-amino]-pentan.
Die Herstellung fand gemäss dem in Beispiel II beschriebenen Verfahren statt, indem die unter a) beschriebene Verbindung mit N-Äthyl-N-[l-methyl- 2-(4'-methoxy-phenyl)-äthyl]-amin gekoppelt wurde.
Die Ausbeute war 54"/ & . Äq. Gewicht 475 (berechnet 473). UV-Absorptionsspektrum = 276 m, u.

The new compounds have valuable pharmacological properties, in particular a blood pressure-lowering and a spasmolytic effect.
When carrying out the inventive method, z. B. react 1 mol of the compound of formula III with at least 1 mol of the free base of formula II. It is preferred to choose two equivalents of the latter compound, 1 mol being effective as an acid binder. The reaction can be carried out without or, preferably, in the presence of a solvent.
For example, aromatic hydrocarbons, such as. B. benzene, toluene, xylene, or chloroform, carbon tetrachloride, acetone and acetonitrile.
Ether such as B. diethyl ether, diisopropyl ether, can be used as solvents for this purpose.

To prepare the compounds of the formula III, an acid of the above formula can also be esterified with a dialcohol of the formula HO-Y-OH. It is advisable to carry out the reaction with at least an equimolar amount of the dialcohol in order to prevent compounds from being produced in which both alcohol groups are esterified. The hydroxy esters obtained can then be converted into the desired compounds of the formula III by halogenating agents which replace the hydroxyl group with a halogen atom. As halogenating agents here come, for. B. thionyl chloride, phosphorus pentachloride, phosphorus oxychloride, phosphorus tribromide or Thio nylbromid into consideration.

The compounds obtained according to the invention can be used in a manner known per se for medicinal products such. B. processed in the form of tablets, dragees, injection liquids and the like.
In the examples below, ether always means diethyl ether and alcohol always means gusty ethanol.
Example I 1- (3 ', 4', 5'-Trimethoxy-benzoyloxy) -6- [N-ethyl-N- (2 "-phenyl-ethyl) -amino] -hexane a) 1- (3 ', 4 ', 5'-trimethoxy-benzoyloxy) -6-bromo-hexane.
This connection was made in two ways, which are described below under 1 and 2.
1. A suspension of 23.5 g (0.1 mol) of the sodium salt of 3, 4, 5-trimethoxy-benzoic acid in a solution of 24.5 g (0.1 mol) of 1,6-dibromo-hexane in 50 ml of xylene was stored in a 154 ° C bath for 60 hours. After cooling, the precipitate was filtered off with suction; the xylene was evaporated from the filtrate in vacuo and the residue distilled in vacuo. The yield was 10.5 g (29 "/ o). Boiling point 187 to 189 ° C.; 0.1 mm. Bromine content 22.22" / (calculated 21.33 "/ o). The distillation also gave 14.4 g 1,6-dibromohexane recovered.
2. 1- (3 ', 4', 5'-trimethoxy-benzoyloxy) -6-bromo-hexane.
23.5 g (0.1 mol) of the sodium salt of 3.4.5 trimethoxybenzoic acid and 30 g (0.12 mol) 1.6 dibromohexane were added to 300 ml of anhydrous acetonitrile and the mixture was stirred for 43 hours cooked. The precipitate was filtered off with suction and washed with ether. The filtrate was evaporated in vacuo and 150 ml of ether was added to the residue. The precipitate was sucked off and the ether evaporated from the filtrate. The residue was distilled in vacuo. The yield was 16.5 g (44 "/ o). Boiling point 186 to 191" C'0.08 mm.
Bromine content 24.20 / o (calculated 21.3'Vo). b) 1- (3 ', 4', 5'-trimethoxy-benzoyloxy) -6- [N-ethyl-N- (2 "-phenyl-ethyl) -amino] -hexane.
A solution of 10.3 g (0.03 mol) 1- (3 ', 4', 5'-trimethoxy-benzoyloxy) -6-bromo-hexane and 9 g (0.06 mol) N-ethyl-N- (2-phenyl-ethyl) -amine in 75 ml of dry benzene was boiled for 70 hours. The benzene was then evaporated in vacuo and 100 ml of dry ether was added to the residue. The resulting precipitate was filtered off with suction and the ether evaporated from the filtrate.
The oily residue had a weight of 11.9 g and an equivalent weight of 376 (calculated 443).
The substance was converted into the hydrochloride by adding an alcoholic hydrochloric acid solution.
Attempts to crystallize this hydrochloride from methyl ethyl ketone ether and alcohol ether had no results. The hydrochloride was then obtained in the form of an oil by evaporation of the solvents and dissolved in 150 ml of water. This solution was extracted twice with 40 ml of ether and then made alkaline with 10 ml of concentrated ammonia, whereupon it was extracted three times with 50 ml of ether. A solution of 7.7 g (0.04 mol) of N-ethyl-N [1-methyl-2- (4'-methoxyphenyl) ethyl] amine and 6.6 g (0.02 mol) of 1 - (3 ', 4', 5'-trimethoxy-benzoyloxy) -6bromo-hexane in 50 ml of dry acetonitrile was boiled for 90 hours.
