Verfahren zur Gewinnung von Psiloeybin und Psiloein Die vorliegende Erfindung betrifft ein Verfahren zur Isolierung der psychotrop wirksamen Verbindun gen Psilocybin und Psilocin aus Pilzarten der Gattun gen Stropharia, Conocybe, Panaeolus, Amanita, Russula sowie folgender Pilzarten der Gattung Psilo- cybe:
Psilocybe semperviva Heim et Cailleux, Psilo- cybe caerulescens Murrill var. Mazatecorum Heim, Psilocybe Zapotecorum Heim, Psilocybe Aztecorum Heim.
Es war schon bekannt (R. Heim, C. r, hebd. Seances Acad. Sei. 242, 965 [1956]), dass eine Reihe von natürlich vorkommenden Pilzarten in Mexiko, Ostsibirien, Kamtschatka und in andern Gegenden Asiens und Amerikas von den Eingeborenen zu ri tuellen Zwecken oder als Genuss- oder Rauschmittel verwendet werden, da ihre Einnahme eigenartige Wirkungen auf den Körper und vor allem auf die Psyche des Menschen hervorruft.
Bei diesen soge nannten halluzinogenen Pilzen handelt es sich um folgende Gattungen: Psiloeybe, Stropharia, Panaeolus, Conocybe, Amanita, Russula.
Es war aber bisher nicht gelungen, aus Pilz mustern natürlicher Herkunft oder aus künstlich ge züchtetem Pilzmaterial der Gattungen Stropharia, Conocybe, Panaeolus, Amanita, Russula sowie fol gender Pilzarten der Gattung Psilocybe:
Psilocybe semperviva Heim et Cailleux, Psilocybe caerulescens Murrill var. Mazatecorum Heim, Psilocybe Zapote- corum Heim, Psilocybe Aztecorum Heim, psychotrop wirksame Verbindungen zu isolieren.
Es wurde nun ein Verfahren zur Gewinnung der Wirkstoffe Psilocybin und Psilocin gefunden, welches dadurch gekennzeichnet ist, dass man aus Pilzmaterial natürlicher Herkunft der Gattungen Stropharia, Conocybe, Panaeolus, Amanita, Russula sowie fol gender Pilzarten der Gattung Psilocybe:
Psilocybe semperviva Heim et Cailleux, Psilocybe caerulescens Murrill var. Mazatecorum Heim, Psilocybe Zapote- corum Heim, Psilocybe Aztecorum Heim oder aus künstlich gezüchtetem Pilzmaterial dieser Gattungen die Wirkstoffe extrahiert, reinigt,
voneinander trennt und die erhaltenen halogenhaltigen Wirkstoffe vor oder nach ihrer Auftrennung durch Behandlung mit Silbercarbonat von Halogen befreit. Die Darstellung des als Ausgangsmaterial verwendeten künstlich gezüchteten Pilzmaterials wird in den nachfolgenden Beispielen beschrieben.
Das Verfahren wird vorzugsweise folgender massen ausgeführt: Das aktive Pilzmaterial (Frucht körper, Sklerotien oder Mycel) wird im Luftstrom oder im Vakuum bei 20-40 vorsichtig getrocknet, feinst vermahlen und mit Wasser, einem niederen aliphatischen Alkohol oder einem Gemisch von Wasser und einem mit Wasser mischbaren organi schen Lösungsmittel bei Raumtemperatur erschöp fend extrahiert.
Die Extrakte werden im Vakuum bei tiefer Temperatur eingedampft, und der Rück stand mit Petroläther entfettet und zur Entfernung inaktiver Begleitstoffe mit Aceton oder Chloroform Alkohol extrahiert. Weitere Ballaststoffe werden durch Auflösen des Rückstandes in möglichst wenig Wasser und wiederholtes Fällen mit absolutem Athanol oder Aceton abgetrennt; das Filtrat wird im Vakuum bei tiefer Temperatur eingedampft.
Die weitere Reinigung erfolgt durch Chromato- graphie an Cellulosepulver nach dem Durchlauf verfahren; die Elution erfolgt mit wassergesättigtem Butanol oder einem andern mit Wasser nicht misch baren Alkohol. Die aufgefangenen Fraktionen wer den mit dem Keller-Reagens (eisenchloridhaltiger Eisessig und konz. Schwefelsäure) auf Wirkstoff gehalt geprüft.
Die Fraktionen mit positiver Farb- reaktion werden vereinigt und nötigenfalls nochmals einer chromatographischen Aufteilung an einer Säule aus Cellulosepulver unterworfen. Aus dem Durchlauf-Chromatogramm wird eine rascher wan dernde Zone eluiert, welche einen durch eine rein blaue Keller-Farbreaktion gekennzeichneten, Psilo- cin genannten Wirkstoff liefert, während aus einer langsamer wandernden Zone ein zweiter,
mengen mässig vorherrschender Wirkstoff mit violetter Farb- reaktion, das Psilocybin erhalten wird.
Die aus der Säule schon in ziemlich reiner Form erhaltenen Wirkstoffe sind halogenhaltig und kristal lisieren als solche nicht: Erst eine chemische Be handlung kann das Halogen entfernen, worauf man kristallisierte Verbindungen erhält. Zu diesem Zweck wird eine wässerige Lösung der Wirkstoffe mit Silber- carbonat behandelt, das Filtrat von den überschüssi gen Silber-Ionen mit Schwefelwasserstoff befreit und im Vakuum bei tiefer Temperatur eingeengt, wobei die Substanzen aus der konzentrierten Lösung kri stallisieren.
Für die Analyse wird das Psilocybin nochmals aus Methanol oder Wasser umkristallisiert. Aus Wasser werden feine, schneeweisse Nadeln, aus Me thanol farblose, methanolhaltige sechseckige Platten oder Prismen erhalten, die bei 195-220 unter Zer setzung schmelzen. Die Verbindung löst sich in 120 Teilen siedendem Methanol oder in 20 Teilen sie dendem Wasser; in höheren Alkoholen oder andern organischen Lösungsmitteln ist sie sehr schwer löslich oder unlöslich.
Zur Analyse wird im Hochvakuum bei 100 ge- trocknet, wobei eine Gewichtsabnahme von 10,4% eintritt. Die Resultate der Elementaranalyse stimmen auf die Bruttoformel C1,H1704N,P (MG 284,2). Psilocybin ist eine optisch inaktive, amphotere Ver bindung und löst sich leicht unter Salzbildung in ver dünnten wässrigen Mineralsäuren und in verdünnten wässrigen Alkalien.
Die Lösung von Psilocybin in 80 0/migem wässrigem Äthanol reagiert schwach sauer (pH 5,2). Das UV-Spektrum in methanolischer Lö sung zeigt Maxima bei 222, 267 und 290 mY.
Das Psilocin ist charakterisiert durch das UV-Spektrum (in methanolischer Lösung) mit den Maxima bei 222, 260, 267, 283 und 293 m,u einer seits und durch die Keller-Farbreaktion, bei welcher - im Gegensatz zu Psilocybin - eine reinblaue Farbe entsteht.
Bei peroraler Verabreichung in Dosen von 4 bis 8 mg ruft das Psilocybin beim Menschen die gleichen Symptome hervor, die bei Einnahme der Pilze auf treten und bereits eingehend beschrieben sind (R. Heim, C. r. hebd. Seances Acad. Sei. 245, 597 [1957]). Die Verbindung soll zur Erforschung von Geisteskrankheiten und Psychosen verschiedenster Genese verwendet werden.
<I>Beispiel 1</I> Gewinnung von Psilocybin und Psilocin aus Stropharia cubensis Earle natürlicher Provenienz Die in Mexiko gesammelten Fruchtkörper von Stropharia cubensis Earle werden an der Luft bei Raumtemperatur im Schatten vorsichtig getrocknet. 24,2 g getrocknete Fruchtkörper werden feinst ge mahlen und bei Zimmertemperatur einmal mit 300 cm-' und dreimal mit je<B>150</B> cm Methanol je 1/2 Stunde ausgeschüttelt. Die Extrakte werden ver einigt und im Vakuum zur Trockne eingedampft.
Der Rückstand (6 g) wird zur Entfettung viermal mit je 125 cm3 Petroläther und noch dreimal mit je 50 cm3 Chloroform verrieben, das 10% Athanol enthält. Der noch ungelöste Rest von etwa 5 g wird in 5 cm- Wasser gelöst, und aus dieser Lösung wer den durch langsames Versetzen mit 50 cm?, abs. Athanol weitere Begleitstoffe ausgefällt, wobei die Wirkstoffe in der Lösung angereichert werden. Die Reinigung wird in gleicher Weise noch zwei- bis drei mal wiederholt.
Die dekantierten Lösungen werden vereinigt und im Vakuum zur Trockne eingedampft. Der Rückstand wird in Methanol aufgenommen und mit 10 g Cellulosepulver, dem 50* /o Wasser zugefügt wurden, versetzt. Das Methanol wird im Vakuum abgedampft, und das mit der Wirksubstanz behaftete Cellulosepulver auf eine Säule von 50 g Cellulose- pulver gegeben, das 5010. Wasser enthält; vorher wird die Säule mit wassergesättigtem Butanol rein gewa schen. Man eluiert mit wassergesättigtem Butanol und fängt Fraktionen von 10 cm3 auf.
Die einzelnen Fraktionen werden im Hochvakuum bei höchstens 50 Badtemperatur eingedampft und mit eisen- chloridhaltigem Eisessig und konz. Schwefelsäure auf Wirkstoffgehalt geprüft. Dazu werden Proben von 0,25 mg Rückstand mit 2 cm- Keller-Reagens an gesetzt. Die wirksamen Fraktionen sind durch eine violette (Psilocybin) oder rein blaue (Psilocin) Keller- Reaktion gekennzeichnet.
Die Fraktionen mit positiver Farbreaktion der gleichen Nuance werden vereinigt. Man löst das amorphe Pulver der obigen Eindampfrückstände in 2 cm-' Wasser und schüttelt mit 0,25 g Silbercarbo- nat. Nach der Filtration werden die überschüssigen Silberionen mit Schwefelwasserstoff entfernt. Aus der schonend eingeengten Lösung kristallisiert das Psilo- cybin in farblosen feinen Nadeln.
Man erhält 58 mg kristallisiertes Psilocybin, entsprechend einer Aus beute von 0,24 /o - bezogen auf die getrockneten Fruchtkörper - und eine Spur Psilocin. <I>Beispiel 2</I> Gewinnung von Psilocybin und Psilocin aus Reinkulturen von Psilocybe semperviva Heim et Cailleux a) Herstellung des Impfstoffes:
Man legt von Basidiosporen des mexikanisch--n Hutpilzes Psilocybe semperviva Heim et Cailleux Reinkulturen auf Bierwürze-Agar an. Zu diesem Zweck werden die von den Lamellen eines reifen Fruchtkörpers abfallenden Sporen auf einer sterilen Unterlage aufgefangen und auf Bierwürze-Agar ge bracht und bebrütet.
Aus den so entstandenen Pri märkulturen wird das Impfungsmaterial in genügen der Menge folgendermassen hergestellt: Das Mycel wird unter sterilem Leitungswasser mit einem rauhen Spatel abgeschabt, so dass eine Suspension feiner Mycel-Flocken entsteht. Mit dieser Suspension werden Erlenmeyer-Kolben beimpft, welche eine Nährlösung und sattelförmige Porzellan füllkörper, wie sie zur Füllung von Destillations- kolonnen gebräuchlich sind, enthalten.
Beste Er fahrungen machten wird mit 300-cm3-Erlenmeyern, enthaltend 50 g Sattelfüllkörper (Grösse 1 cm, Ge wicht etwa 0,9 g) und 80 cm3 Nährlösung, be stehend aus Bierwürze von etwa 4a/11 Trockensubstanz und 0,21/o Agar.
Die Kultur wird bei einer Temperatur von 241 bebrütet; nach zwei Wochen hat sich auf der Ober fläche eine kompakte Myceldecke gebildet. Die ganze Kultur wird 30 Minuten lang auf der rotierenden Schüttelmaschine geschüttelt, wobei die Sattelfüll- körper durch ihre scharfen Kanten das Mycel zer mahlen: Es entsteht dabei eine feine Suspension von Mycelflocken. Der so erhaltene Impfstoff genügt zur Beimpfung von 50 Liter Nährlösung.
b) Herstellung der Kultur: Frische, helle Bierwürze ohne Hopfenzusatz wird mit Leitungswasser auf einen Gehalt von 811/9 an Trockensubstanz verdünnt. Zu jedem Liter dieser Lösung fügt man:

<tb> Agar-Agar <SEP> 2,0 <SEP> g Diese Nährlösung wird in Portionen von je 1 Liter in Penicillinkolben abgefüllt und während 25 Minuten im Autoklaven bei 108 sterilisiert.
Nach dem Erkalten werden die Kolben mit je 2 cm3 einer Suspension von P. semperviva Heim et Cailleux be- impft. Die erhaltenen Kulturen werden im Dunkeln bei 24-261 bebrütet. Es bildet sich die unter 2 a) be schriebene Myceldecke.
c) Isolierung des Pilzmaterials: Nach sieben Wochen wird die reife Kultur durch ein Gazetuch filtriert, das Pilzmaterial ausgepresst und im Vakuum bei 301 getrocknet. Aus einem An satz mit 100 Penicillinkolben, enthaltend 100 Liter Nährlösung, werden bei der Ernte nach 49 Tagen 2540 g Trockenmaterial erhalten, das heisst 25,4 g pro Liter angesetzter Nährlösung. Das Kulturfiltrat, das auch gewisse Mengen Wirkstoffe enthält, wird nach dem gleichen Verfahren aufgearbeitet wie im folgenden Abschnitt für das Pilzmaterial beschrieben wird.
(1) Gewinnung der Wirkstoffe: 306 g getrocknetes Pilzmaterial werden feinst pulverisiert, dreimal mit je 500 cm3 Chloroform und noch dreimal mit je 500 cm3 Chloroform, das 10% Athanol enthält, ausgeschüttelt. Dabei gehen 2,8 g inaktive Begleitstoffe in Lösung.
Das vorextrahierte Pilzmaterial wird einmal mit 3 Liter und dreimal mit je 1,5 Liter Methanol erschöpfend ausgezogen. Die vereinigten Auszüge werden unter vermindertem Druck zur Trockne eingedampft, wobei ein hell brauner Rückstand von 17,5 g verbleibt. Zur Ent fernung fettartiger Verunreinigungen wird dieser Rückstand in 17,5 cm3 Wasser aufgenommen und die Suspension einmal mit 500 em3 und zweimal mit je 250 cm3 Petroläther ausgeschüttelt. Die Petrol- ätherlösung enthält 0,75 g inaktive Begleitstoffe.
Die zurückbleibende wässrige Lösung wird im Vakuum auf etwa 25 cm3 eingeengt und unter kräftigem Rühren -langsam mit 250 cm3 abs. Athanol versetzt. Von der dabei entstehenden klebrigen Fällung wird die wirkstoffhaltige Lösung durch Dekantieren abge trennt. Die Fällung wird nochmals in wenig Wasser gelöst und mit der zehnfachen Menge abs. Äthanol behandelt. Diese Reinigung durch Fällung wird mit dem Rückstand noch zweimal wiederholt. Die Lö sungen werden vereinigt und im Vakuum zur Trockne eingedampft.
Es verbleibt ein fester Rückstand von 11,7 g, der die gesamte Menge der Wirkstoffe ent hält.
Für die Chromatographie an der Cellulosesäule wird der Rückstand in möglichst wenig 50%iges Methanol gelöst, die Lösung mit 40 g Cellulosepulver gut vermischt, und das Material im Vakuum getrock net.
Das auf diese Weise mit Substanz beladene Cellulosepulver wird auf eine Cellulosesäule gegeben, die durch Aufschlämmen von 350 g Cellulosepulver mit wassergesättigtem Butanol hergestellt wird. Man entwickelt im Durchlaufverfahren mit wassergesättig tem Butanol, wobei Fraktionen von je 100 cm3 ab getrennt und im Hochvakuum bei einer 501 nicht übersteigenden Badtemperatur eingedampft werden.
Die mittleren Fraktionen (3,35 g) geben eine positive Kellersche Farbreaktion und werden zur weiteren Reinigung nochmals nach dem gleichen Verfahren auf Cellulosepulver chromatographiert. Bei der Entwicklung mit wassergesättigtem Buta- nol werden Fraktionen von je 50 cm3 abgetrennt und nach Eindampfen im Hochvakuum bei höchstens 501 Badtemperatur mit der Kellersehen Farbreaktion geprüft.
Dabei wird folgende Verteilung erhalten:

<tb> 31-43 <SEP> 758 <SEP> negativ Der Rückstand der Fraktionen 8-l6 (87 mg) liefert nach der Behandlung mit Silbercarbonat, wie im Beispiel 1 beschrieben, 45 mg reines Psilocin. Die Fraktionen 21-30 (l053 mg) liefern nach der Behandlung mit Silbercarbonat 765 mg reines Psilo- cybin.
Aus dem Kulturfihrat werden die Wirkstoffe nach dem gleichen Verfahren gewonnen. Zu diesem Zweck wird das Filtrat im Vakuum auf etwa 1(1o seines Vo lumens konzentriert. Man fällt mit dem zehnfachen Volumen Methanol, filtriert von den ausgefällten Begleitstoffen ab, dampft die Lösung im Vakuum zur Trockne ein und extrahiert den Rückstand drei mal mit der zehnfachen Menge Methanol. Der Ein dampfrückstand der Methanolextrakte wird wie oben weiter verarbeitet. Aus 12 Liter Kulturfiltrat werden 80 mg Psilocybin und 6 mg Psilocin erhalten.
Ge samtausbeute: 845 mg Psilocybin und 51 mg Psilocin, entsprechend einem Gehalt von 0,28 bzw. 0,0171/o, bezogen auf das getrocknete Pilzmaterial. <I>Beispiel 3</I> Psilocybin und Psilocin aus Reinkulturen von Stropharia cubensis Earle Eine Nährlösung wird wie folgt hergestellt:
Frische, helle Bierwürze ohne Hopfenzusatz wird mit Leitungswasser auf einen Gehalt von 6% an Trockensubstanz verdünnt. Zu jedem Liter dieser Lösung werden zugefügt:

Nach 52- tägiger Bebrütung im Dunkeln bei 24 erhält man aus einem Ansatz von 20 Liter 452 g getrocknetes Pilzmaterial, das heisst 22,6 g pro Liter. Die Gewin nung der Wirkstoffe aus dem Pilzmaterial erfolgt nach dem im Beispiel 2 beschriebenen Verfahren.
Ausbeute: 1083 mg reines kristallisiertes Psilocybin und 45 mg Psilocin, entsprechend einer Ausbeute von 0,24 bzw. 0,010%, bezogen auf das getrocknete Pilzmaterial.
<I>Beispiel 4</I> Psilocybin und Psilocin aus Psilocybe semperviva Heim et Cailleux natürlicher Provenienz Die durch künstliche Züchtung auf natürlichen Nährböden (Kompost) gewonnenen reifen Frucht körper von Psilocybe semperviva Heim et Cailleux werden im Luftstrom bei 30 vorsichtig getrocknet.
32 g getrocknete Fruchtkörper werden feinst ge mahlen und bei Zimmertemperatur einmal mit 300 cm-' und dreimal mit je 150 cm3 Methanol je ?% Stunde ausgeschüttelt. Die vereinigten Extrakte werden im Vakuum zur Trockne eingedampft.
Der Rückstand (8,3 g) wird zur Entfettung viermal mit je 125 ein?- Petroläther und dreimal mit je 50 cm3 Chloroform verrieben, das 10% Äthanol enthält. Es verbleibt ein Rest von 6,5 g.
Dieser Rückstand wird in 6 cm?, Wasser gelöst und die Lösung langsam mit 60 cm3 abs. Äthanol zur Ausfällung weiterer Begleit- stoffe versetzt, wobei die Wirkstoffe in der Lösung angereichert werden. Die Reinigung wird in gleicher Weise noch zweimal wiederholt. Die Lösungen wer den dekantiert, vereinigt und im Vakuum zur Trockne eingedampft. Der Rückstand wird in Me thanol aufgenommen und wie im Beispiel 1 be schrieben auf Cellulosepulver chromatographiert. Den so erhaltenen Wirkstoffen wird durch Behand lung mit Silbercarbonat das Halogen entzogen. Nach .
Umkristallisieren erhält man 160 mg kristallisiertes Psilocybin und 32 mg Psilocin, entsprechend einem Gehalt von 0,5 bzw. 0,10/0, bezogen auf das getrock nete Pilzmaterial.
<I>Beispiel 5</I> Psilocybin aus Psilocybe caerulescens Murrill var. Mazatecorum Heim natürlicher Provenienz Die durch künstliche Züchtung auf natürlichen Nährböden (Kompost) gewonnenen reifen Frucht körper von Psilocybe caerulescens Murrill var. Maza- tecorum Heim werden vorsichtig getrocknet, worauf das getrocknete Pilzmaterial (7,3 g)
feinst gemahlen und mit Methanol erschöpfend extrahiert wird, wie im Beispiel 1 beschrieben. Die Extrakte werden ver einigt und im Vakuum zur Trockne eingedampft. Der Rückstand wird wie im Beispiel 1 angegeben auf die Wirkstoffe weiter verarbeitet. Man erhält 14,6 mg reines Psilocybin, entsprechend einem Ge halt von 0,20/a Psilocybin (bezogen auf das getrock nete Pilzmaterial). Aus dem aufgearbeiteten Pilz muster wurde kein Psilocin erhalten.
<I>Beispiel 6</I> Psilocybin aus Reinkulturen von Psilocybe caerulescens Murrill var. Mazatecorum Heim Das Kulturmedium wird vorbereitet, indem man frische, helle Bierwürze ohne Hopfenzusatz mit Lei tungswasser auf einen Gehalt von 4,00/9 an Trocken substanz verdünnt. Zu jedem Liter dieser Lösung fügt man:

Diese Nährlösung wird wie im Beispiel 2 ange geben sterilisiert, mit der Mycel-Suspension einer Reinkultur von Psilocybe caerulescens Murrill var. Mazatecorum Heim aus Mexiko beimpft, und die entstandenen Kulturen im Dunkeln bei 26 bebrütet. Nach 48 Tagen werden pro Liter Lösung 20;4 g ge trocknetes Pilzmaterial geerntet. Die Isolierung und Reinigung der Wirkstoffe aus diesem Material erfolgt wie im Beispiel 2 angegeben.

86,5 g getrocknetes Mycel. Das feinst gemahlene Mycel wird 1/2 Stunde mit 1 Liter 80%igem wässrigem Äthanol bei Raumtemperatur ausgeschüttelt. Nach dem Ab filtrieren des Rückstandes wird dieser noch dreimal auf die gleiche Weise extrahiert.
Der Eindampfrück- stand der vereinigten Extrakte (8,9 g) wird zur Ent fernung von fettartigen Begleitstoffen und inaktiven Pallaststoffen nacheinander zweimal mit 100 cm3 Petroläther, zweimal mit 80 cm3 Chloroform und zweimal mit 50 em 3 Aceton ausgezogen. Es verblei ben 5,6 g eines wasserlöslichen Pulvers, das in 6 em3 Wasser gelöst wird.
Unter gutem Rühren versetzt man die Lösung langsam mit 60 em3 Aceton, trennt die entstandene Fällung ab, löst sie wieder in wenig Wasser und fällt nochmals mit der zehnfachen Menge Aceton. Diese Reinigung durch Fällung wird mit dem in Aceton unlöslichen Anteil noch zweimal wiederholt, wonach die gesamte Menge der Wirk stoffe in die wässrig-acetonischen Extrakte über gegangen ist. Die Extrakte werden im Vakuum ein gedampft, und der Rückstand (2,8 g), wie im Bei spiel 2 beschrieben, durch Chromatographie an einer Säule aus Cellulosepulver weiter gereinigt und auf geteilt.
Nach zweimaliger Chromatographie erhält man 225 mg kristallisiertes Psiloeybin und 15 mg Psilocin, entsprechend einer Ausbeute von 0,26 bzw. 0,0170/0, bezogen auf das getrocknete Pilzmaterial.

Sci. 242, 965 [1956]) that a number of naturally occurring species of mushrooms in Mexico, Eastern Siberia, Kamchatka and in other regions of Asia and America from the Indigenous people are used for ritual purposes or as stimulants or intoxicants, as their ingestion has peculiar effects on the body and above all on the psyche of humans.
These so-called hallucinogenic mushrooms are of the following genera: Psiloeybe, Stropharia, Panaeolus, Conocybe, Amanita, Russula.
So far, however, it has not been possible to use mushroom specimens of natural origin or artificially grown mushroom material of the genera Stropharia, Conocybe, Panaeolus, Amanita, Russula and the following mushroom species of the genus Psilocybe:
Psilocybe semperviva Heim et Cailleux, Psilocybe caerulescens Murrill var. Mazatecorum Heim, Psilocybe Zapotecorum Heim, Psilocybe Aztecorum Heim, isolate psychotropically active compounds.
A process has now been found for obtaining the active ingredients psilocybin and psilocin, which is characterized in that mushroom material of natural origin of the genera Stropharia, Conocybe, Panaeolus, Amanita, Russula and the following types of fungus of the genus Psilocybe:

The preparation of the artificially grown mushroom material used as the starting material is described in the following examples.
The method is preferably carried out as follows: The active fungal material (fruit body, sclerotia or mycelium) is carefully dried in a stream of air or in a vacuum at 20-40, and then finely ground and mixed with water, a lower aliphatic alcohol or a mixture of water and one with Water-miscible organic solvents extracted exhaustively at room temperature.
The extracts are evaporated in vacuo at low temperature, and the residue was degreased with petroleum ether and extracted with acetone or chloroform alcohol to remove inactive accompanying substances. Further dietary fiber is removed by dissolving the residue in as little water as possible and repeatedly precipitating it with absolute ethanol or acetone; the filtrate is evaporated in vacuo at low temperature.
The further purification takes place by chromatography on cellulose powder according to the run-through process; the elution is carried out with water-saturated butanol or another water-immiscible alcohol. The fractions collected are checked for active ingredient content using the Keller reagent (glacial acetic acid containing iron chloride and conc. Sulfuric acid).
The fractions with a positive color reaction are combined and, if necessary, subjected again to chromatographic division on a column made of cellulose powder. A more rapidly moving zone is eluted from the continuous chromatogram, which supplies an active ingredient called psilocin characterized by a pure blue cellar color reaction, while a second,
moderately predominant active ingredient with a violet color reaction that is obtained from psilocybin.
The active ingredients already obtained from the column in fairly pure form contain halogens and do not crystallize as such: only chemical treatment can remove the halogen, which results in crystallized compounds. For this purpose, an aqueous solution of the active ingredients is treated with silver carbonate, the filtrate is freed from the excess silver ions with hydrogen sulfide and concentrated in vacuo at low temperature, the substances crystallizing from the concentrated solution.
For the analysis, the psilocybin is recrystallized again from methanol or water. Fine, snow-white needles are obtained from water, and colorless, methanol-containing hexagonal plates or prisms are obtained from methanol, which melt at 195-220 with decomposition. The compound dissolves in 120 parts of boiling methanol or 20 parts of water; it is very sparingly soluble or insoluble in higher alcohols or other organic solvents.
For analysis, drying is carried out in a high vacuum at 100, with a weight decrease of 10.4%. The results of the elemental analysis agree with the gross formula C1, H1704N, P (MW 284.2). Psilocybin is an optically inactive, amphoteric compound and dissolves easily with salt formation in dilute aqueous mineral acids and in dilute aqueous alkalis.
The solution of psilocybin in 80% aqueous ethanol is weakly acidic (pH 5.2). The UV spectrum in methanolic solution shows maxima at 222, 267 and 290 mY.

86.5 g dried mycelium. The finely ground mycelium is shaken for 1/2 hour with 1 liter of 80% aqueous ethanol at room temperature. After the residue has been filtered off, it is extracted three times in the same way.
The evaporation residue of the combined extracts (8.9 g) is extracted in succession twice with 100 cm3 of petroleum ether, twice with 80 cm3 of chloroform and twice with 50 cm3 of acetone to remove fatty accompanying substances and inactive palladium. 5.6 g of a water-soluble powder remain, which is dissolved in 6 cubic meters of water.
While stirring well, the solution is slowly mixed with 60 cubic meters of acetone, the precipitate formed is separated off, dissolved again in a little water and again precipitated with ten times the amount of acetone. This purification by precipitation is repeated twice more with the portion insoluble in acetone, after which the entire amount of active substances has passed into the aqueous-acetone extracts. The extracts are evaporated in vacuo, and the residue (2.8 g), as described in Example 2, further purified by chromatography on a column of cellulose powder and divided.
After two chromatography, 225 mg of crystallized psiloeybin and 15 mg of psilocin are obtained, corresponding to a yield of 0.26 and 0.0170 / 0, based on the dried mushroom material.
