Verfahren zur Herstellung von 9a-Fluor-llss, 17a, 21-trihydroxy-16a-methyl-pregnen- (oder 1,4-pregnadien)-3,20-dion-21-acylaten Gegenstand vorliegender Erfindung ist ein Ver fahren zur Herstellung von 9a-Fluor-llss,17a,21-tri- hydroxy-16a-methyl-4-pregnen-(oder 1,4-pregnadien)- 3,20-dion-21-acylaten, das dadurch gekennzeichnet ist, dass man ein l 1/,0)-Hydroxy-steroid der allgemeinen Formel

in der die punktierte Linie einer Einfach- oder einer Doppelbindung entspricht,
und in der Ac den Acyl- rest einer Carbonsäure darstellt, zur entsprechenden _J91I1j-Verbindung dehydratisiert, diese 49(I1)-Verbin- dung mit unterchloriger, unterbromiger oder unter- jodiger Säure zum entsprechenden 9a-Halogen-llss- hydroxysteroid umsetzt, dieses mit einer Base zu der entsprechenden 9,8,11 ss-Epoxyverbindung umsetzt,
worauf diese mit Fluorwasserstoff in die 9a-Fluorver- bindung übergeführt wird.

In obigen Formeln bedeutet Ac einen Acylrest einer Carbonsäure, vorzugsweise mit 1 bis 12 Kohlen stoffatomen, und X' Chlor, Brom oder Jod. Die punk tierten Linien stellen Einfach- oder A'-Doppelbindun- gen dar.
Die erfindungsgemäss erhältlichen Verbindungen sind äusserst wirksam gegen Entzündungen und zeigen keinerlei unerwünschte Nebenwirkungen, wie Magen- Darmstörungen, Salz-Zurückhaltung, Ödeme und der gleichen, also Erscheinungen, die bekanntlich bei ähnlich wirkenden Steroiden auftreten. Viele der Ester mit höherem Molekulargewicht, besonders die gegen Hydrolyse beständigen und/oder die in Körper flüssigkeiten unlöslichen, ergeben über die entspre chenden 21-Hydroxy-Verbindungen Stoffe mit verlän gerter Wirkungsdauer. Die obigen Verbindungen eig nen sich für die Behandlung von Entzündungen z. B.
der Haut, der Augen, der Atmungswege, der Knochen oder inneren Organe, die durch bakterielle oder Virusinfektionen hervorgerufen werden. Die Verbin dungen sind auch bei Kontakt-Dermatitis und allergi schen Erscheinungen wirksam. Sie besitzen einen ver besserten therapeutischen Quotienten, verglichen mit den keine Methylgruppe in 16-Stellung aufweisenden Verbindungen. Für die erwähnten Zwecke können sie in bekannter Weise Salben, Cremen, Lotionen, Sprays und dergleichen einverleibt werden.
Auch kön nen sie irgendwelchen Antibiotika, besonders Peni- cillinen, Neomycin, Tetracyclin, Chloromycetin und Novobiocin zugesetzt werden.
Die erfindungsgemäss erhältlichen Verbindungen eignen sich auch als Zwischenprodukte zur Herstel lung anderer Steroide, sie besitzen ferner Glucocorti- coide, mineralcorticoide sowie progestationale Wir kung.
Die Ausgangsmaterialien des erfindungsgemässen Verfahrens können aus dem bekannten 11-Keto-16- dehydroprogesteron durch folgende Synthese herge stellt werden: Die 3-Ketogruppe wird selektiv durch überführung in ein 3-Ketal oder 3-Enamin geschützt. Während sich die Pyrrolidyl-3-Enamine selektiv bil den, muss die 20-Ketogruppe bei der Ketalisierung zuerst geschützt werden. Deswegen ist die Herstellung der 3-Enamine bevorzugt.
Das in 3-Stellung ge schützte 11-Keto-16-dehydroprogesteron wird dann mit einem Methyl-Grignard-Reagens in Gegenwart von Kupfer(I)-chlorid umgesetzt [vergleiche Grignard Reactions , Kharasch und Reinmuth, Preutice Hall, Inc. Publishers (1954), Seite 219], wobei man 11- Keto-16a-methylprogesteron erhält.
Die die 3-Stellung schützende Gruppe wird dann hydrolysiert, worauf man das freie 11-Keto-16a-methylprogesteron erhält.
Diese Verbindung wird in den 3,11-Diketo-16a methyl-4,17(20)-[cis]-pregnadien-21-carbonsäurealkyl- ester übergeführt, z. B. nach der Methode gemäss US-Patent Nr. 2 790 814. Dabei wird das 11-Keto- 16a-methylprogesteron mit jeweils mehr als 2-Mol- Äquivalenten eines Oxalsäuredialkylesters und einer Base umgesetzt, wobei man das Alkalimetall-dienolat des 2,21-Dialkoxyoxalyl-11-keto-16a-methylprogeste- rons erhält.
Diese Verbindung oder das durch Um setzung mit Essigsäure erhaltene freie Enol wird dann mit Chlor oder vorzugsweise Brom trihalogeniert, und man erhält so das 2,21-Dialkoxyoxalyl-2,21,21-tri- halo-11-keto-16a-methylprogesteron. In Gegenwart starker Basen lagert sich diese Verbindung um und es bildet sich der 2-Halogen-3,11-diketo-16a-methyl- 4,17(20)
-[cis]-pregnadien-21- alkylester. Das 2stän- dige Halogen wird mit Zink und Essigsäure oder an deren für den Halogenentzug geeigneten Mitteln ent fernt, und man gelangt zu dem 3,11-Diketo-16a-me- thyl - 4,17 (20)-[cis]-pregnadien - 21- carbonsäurealkyl- ester. Die Umwandlung der 3,11-Diketo-16a-methyl- 4,17(20)-[cis]-pregnadien-21-ester in das 11ss,21-Di- hydroxy-16a-methyl- 4,17(20)
-[cis]-pregnadien-3-on kann auf die in den US-Patenten Nrn. 2 7l5 621, <B>2781343</B> beschriebene Weise durchgeführt werden. In der US-Patentschrift Nr. 2 790 814 ist die Um wandlung des 3,11-Diketo-4,17(20)-[cis]-pregnadien- 21-esters in das llss,21-Dihydroxy-4,17(20)-[cis]- pregnadien-3-on,
der Schutz der 3-Ketogruppe durch Ketalisierung oder Enaminierung sowie die Reduktion der 11-Ketogruppe und der 21-Carboxyalkylgruppe mit Natrium- oder Lithiumaluminiumhydrid oder der gleichen, die die C-C-Doppelbindungen nicht beein trächtigen, beschrieben, nach dieser Methode erhält man I ll,21-Dihydroxy-16a-methyl-4,17(20)-[cis]- pregnadien-3-on, welches in 3-Stellung noch geschützt ist.
Die die 3-Ketostellung schützende Gruppe wird durch Hydrolyse entfernt, und man erhält 11f,21-Di- hydroxy -16a- methyl- 4,17(20)-[cis]-pregnadien-3-on. Diese Verbindung kann alsdann selektiv mit Selen dioxyd oder auf enzymatischem Wege in die entspre chende 11.4-Verbindung übergeführt werden.
Die 21-Oxygruppe der Al- oder 41,4-Verbindung wird dann vorzugsweise verestert, um sie während der folgenden oxydativen Hydroxylierung zu schützen. Die Veresterung wird in üblicher Weise mit Säure anhydriden oder Säurehalogeniden durchgeführt. Die so hergestellten Ester werden dann oxydativ mit 0s04 und einem Peroxyd, gemäss den in den US-Patenten Nrn. 2 769 825, 2 769 823 oder in J.
Am. Chem. Soc. 77,4436 (1955) beschriebenen Methoden in die ent sprechenden 17a-Hydroxy-20-keto-21-acyloxy-Ver- bindungen (I) übergeführt.
Diese Verbindungen werden sodann erfindungs gemäss in die entsprechenden 9a-Halogenverbindun- gen umgewandelt, indem man die 11fl'-Hydroxy- gruppe in an sich bekannter Weise, z. B. mit einem N-Halogenamid oder N-Halogenimid in Gegenwart von wasserfreiem S02 oder von Thionylchlorid oder Methansulfonylchlorid in Pyridin oder durch Erhitzen mit Toluolsulfonsäure in einem organischen Lösungs mittel wie z. B.
Benzol, dehydratisiert, wobei man die entsprechende 9(11)-Dehydroverbindung (II) erhält. Diese Verbindung wird dann mit HOCl, HOBr oder HOJ, die in üblicher Weise aus N-Halogenamiden oder -imiden in Gegenwart wässriger HC104 fabriziert werden, umgesetzt und in die entsprechende 9a-Halo- gen-llss-hydroxy-Verbindung (1I1) übergeführt. Um setzung der zuletzt genannten Verbindung mit einer Base wie z.
B. CH.COOK führt zu den entsprechen den Epoxyverbindungen IV. Diese Epoxyverbindung wird dann mit wässrigem oder wasserfreiem Fluor wasserstoff in die 9a-Fluor-Ilss-hydroxy-Verbindung V übergeführt.
Letztere kann zur entsprechendem 21- Hydroxyverbindung, d. h. 9a-Fluor-lIss,17a,21-tri- hydroxy-16a-methyl-4-pregnen-3,20-dion oder 9a Fluor-11,B,17a,21-trihydroxy-16a-methyl-1,4-pregna- dien-3,20-dion hydrolysiert werden; sie kann ferner zu der entsprechenden 11-Ketoverbindung oxydiert werden.
Präparation <I>1</I> 11-Keto-16a-methylprogesteron. Zu einer heissen Lösung aus 4,56 g (0,014 Mol) 11-Keto-16-dehydroprogesteron in 45 ml Methanol wurden 2,25 ml Pyrolydin unter Bewegung der Flüs sigkeiten zugegeben. Das Reaktionsprodukt, das 3- Pyrrolydin-enamin des 11-Keto-16-dehydroprogeste- rons schied sich bald danach in gelben Kristallen aus. Es wurde sodann auf 5 abgekühlt. Die Mischung wurde filtriert, der Filterkuchen gewaschen (mit kal tem Methanol) und im Vakuum getrocknet.
Man er hielt so 4,56 g des 3-Pyrrolydyl-3,5,16-pregnatrien- 11,20-dions, das bei 154-169 schmilzt.
Eine Lösung des so erhaltenen Enamins in 110 ml Tetrahydrofuran wurde langsam und unter Rühren einer Suspension aus 45 ml käuflichen 3-molaren Methylmagnesiumbromids zugefügt und sodann auch noch 0,90 g Cupro-chlorid. Die Mischung wurde auf Raumtemperatur abgekühlt und unter Stickstoff atmosphäre noch 3 Std. gerührt. Das überschüssige Grignard-Reagens wurde durch tropfenweise Zugabe von 6,5 ml Wasser vorsichtig zerstört.
Zu der Mi schung, die 3-Pyrrolydyl-16a-methyl-3,5-pregnadien- 11,20-dion enthält, wurden 4 ml Essigsäure und 16 ml Methanol zugegeben, wobei sie so lange erwärmt wurde, bis eine klare gelbe Lösung vorlag. 16 ml einer 10 % igen wässrigen Lösung aus Natrium- hydroxyd wurde zugegeben, wobei sich ein pH von 8 einstellte.
Sodann erhitzte man die Mischung unter Rückfluss noch 45 Min. und gab dann 1 ml Essig säure zu und 250 ml Wasser, worauf man die gekühlte Mischung 3mal mit Methylenchlorid extrahierte. Die vereinigten Extrakte wurden gewaschen (mit einer wässrigen Bicarbonatlösung) und sodann mit Natrium sulfat getrocknet.
Die getrocknete Lösung wurde zur Trockne eingedampft und der Rückstand - im we sentlichen 11-Keto-16a-methylprogesteron - wurde in 400 ml Methylenchlorid aufgelöst und durch eine 200-g-Kolonne von Magnesiumsilikat (Florisil) chro- matographiert. Die Kolonne wurde mit Mengen von je 400 ml Lösungsmittel entwickelt, und zwar in fol gender Zusammensetzung und Reihenfolge:
Fünf aus Hexanen (Skellysolve B) + 3 % Aceton und dann mit + 5% Aceton, acht Teile Hexane + 7% Aceton, 5 Teile Hexane + 104/o Aceton,
4 Teile Hexane + 15 % Aceton und schliesslich eine aus Aceton. Die Fraktionen 9-25 (wobei die Methylenfraktion mitge rechnet wurde) wurden kombiniert und aus einer Mi schung von Aceton und Wasser umkristallisiert. Auf diese Weise erhielt man 2,03 g 11-Keto-16a-methyl- progesteron, dessen Schmelzpunkt zwischen 179 und l82 lag.
Eine aus Acetylacetat und Hexanen umkristalli sierte Probe schmolz zwischen 183 und 185 , besass ein (a)D von + 255 (CHC13) und ein A, m ä;. von 238 ,a und ein am von<B>15,850</B> und folgende Analyse:

Eine Lösung aus 2,00 g (5,8 Millimol) 11-Keto- 16a-methylprogesteron in 30 ml trockenen Tert.- Butylalkohols wurde unter Stickstoffatmosphäre auf 50 erwärmt.
Zu dieser Lösung gab man 3,2 ml Äthyloxalat und 3,03 g einer 25 %igen methanoli- schen Lösung von Natriumhydroxyd. Daraufhin bil dete sich sofort ein grünlichgelber Niederschlag des Natriumdienolats des 2,21-Diäthoxyoxalyl-11-keto- 16a-methylprogesterons.
Man rührte die Mischung noch 20 Min. und gab sodann eine gekühlte Lösung von 0,98 g trockenen Natriumacetats und 0,84 ml Essigsäure in 40 ml Methanol zu und erzeugte so das freie Dienol. Die Lösung wurde dann auf 0 abgekühlt, worauf man ihr tropfenweise eine gleichfalls gekühlte Lösung von 2,0 g Brom in Methanol während einer Zeit von 10 Min. zugab. Man erhielt auf diese Weise 2,21,21 Tribromo-2,21-Diäthoxyoxalyl-1-keto-16a-methyl- progesteron.
Das Kühlbad wurde entfernt, worauf man der Lösung 5,72 g einer 25%igen methanolischen Na- triummethoxydlösung zugab. Das Rühren wurde noch 2,5 Std. fortgesetzt, und man erhielt so 2-Bromo 2,11-diketo-16a-methyl-4,17(20)-[cis]-pregnadien-21- oin-säuremethylester.
Zu der so erhaltenen Lösung gab man dann 5 ml Essigsäure und 1 g Zinkstaub, wonach man das Rüh ren noch eine halbe Stunde fortsetzte. Die Mischung wurde daraufhin mit Wasser verdünnt, und die festen Stoffe wurden abfiltriert. Das Filtrat wurde mit Me- thylenchlorid erschöpfend extrahiert und das Extrakt mit Natriumsulfat getrocknet und eingedampft. Den Rückstand löste man in 400 ml Methylenchlorid, worauf er in eine Kolonne, die 200 g Magnesium silikat (Florisil) enthielt, gegossen wurde.
Die Ko lonne wurde mit 400-ml-Portionen eines Lösungs mittels folgender Zusammensetzung in folgender Rei henfolge entwickelt: 4 Hexane (Skellysolve B) + 5 0.1o Aceton, 10 Hexane + 7% Aceton, 10 Hexane + 10,% Aceton,
und schliesslich 1 Teil nur Aceton.
Die Fraktionen 14-23 (mit der Methylenfraktion) enthielt das Ausgangs-1 1-keto-16a-methylprogesteron. Die Fraktionen 7-12 enthielten 3,11-Diketo-16a-me- thyl- 4,17(20) - [cis] -pregnadien - 21- oin-säuremethyl- ester, die nach dem Umkristallisieren aus Methanol- Wasser und dann nur aus Methanol zwischen 177 bis 184 schmolz.
Es hatte ein i mäX 232,5 #I, ein am von 26,200 und ein [a]n von<B>+137</B> (CHCI3) und zeigte folgende Analyse:

Präparation <I>3</I> 11 ss,21-Dihydroxy-16a-methyl-4,17(20)-[cis]- pregnadien-3-on.
Eine Lösung aus 6,0 g 3,11-Diketo-16a-methyl- 4,17(20) - [cis] - pregnadien - 21- oin -säuremethylester, 6 ml Pyrrolydins und 180 ml p-Toluol-sulfonsäure in 120 ml Benzol wurden unter Rückfluss 1 Stdt. lang erhitzt, wobei das Reaktionswasser dauernd entfernt wurde. Die Lösung wurde destilliert, und man erhielt einen Rückstand, der im wesentlichen aus 3-Pyrro lidyl-16a-methyl-3,5,17(20)-[cis]-pregnatrien-21-me- thylester bestand.
Man löste daraufhin den Rückstand in 30 ml Benzol und 10 ml Äther und gab eine Suspension von 2,2 g Lithiumaluminiumhydrid in 350 ml Äther hinzu, und zwar während eines Zeitraumes von 5 Min. Diese Mischung wurde bei Raumtemperatur noch 90 Min. gerührt. Man gab 20 ml Äthylacetat hinzu und um das überschüssige Lithiumaluminiumhydrid zu zerstören sodann noch 30 ml Wasser.
Der Äther und das Benzol wurden bei vermindertem Druck destilliert, worauf man einen im wesentlichen aus 3-Pyrrolidyl-11 f,21-dihydroxy-16a-methyl-3,5,17(20)- [cis]-pregnatrien-21-oin-säuremethylester bestehenden Rückstand erhielt, der mit etwas anorganischen Stof fen vermischt war.
Dieser rohe Destillationsrückstand wurde mit 400 ml Methanol vermischt und auf 40 erwärmt, bis eine Lösung vorlag. 70 ml einer wässrigen 5%igen Natriumhydroxydlösung wurde zugegeben, wobei die Mischung noch 10 Min. lang auf 40 erwärmt wurde. Die Lösung wurde neutralisiert (mit Essigsäure), das Lösungsmittel bei vermindertem Druck abdestilliert, und zwar bei einer Temperatur, die unter 45 lag.
Der Rückstand wurde mit 100 ml Wasser, das 10 ml konzentrierte Salzsäure enthielt, vermischt, filtriert und gewaschen, worauf man I1/3,21-Dihydroxy-16a- methyl-4,17(20)-[cis]-pregnadien-3-on erhielt. Präparation <I>4</I> 11,B,21-Dihydroxy-16a-methyl-1,4,17(20)-[cis]- pregnatrien-3-on.
Septomyxa affinis wurde auf einem Malz-Agar- boden gezüchtet, der aus 50 g trockenem Malzextrakt, 5 g Edamin (enzymatische Verdauung des Laktalbu- mins) und 20 g Agar gelöst in 1 Liter Leitungswasser bestand, wobei ein pH von 6,5-7,0 eingestellt wurde.
Das Inokulum des Agarbodens wurde in einen 1-Liter-Erlenmeyerkolben, der 100 ml Malzextrakt enthielt, bei Raumtemperatur 4-7 Tage lang zuge- brütet, damit sich Sporen bilden.
Die Sporen wurden in 100 ml einer sterilen 1%igen Salzlösung suspen- diert. 5 ml dieses Sporeninokulums wurden in jeweils 250 ml fassende Kolben, die 100 ml des Mediums enthielten,
gegeben. Das Medium bestand aus 1%iger Cerelos Dextrose und 2'0/0, Getreideweiche aus 60% Feststoffen, die mit Natriumhydroxyd auf einen pH zwischen 4,8 und 5 gebracht wurde. Nun wurde unter Schütteln 48-72 Std. bei Raumtemperatur angebrütet, worauf das Ganze zu 61 des obigen Gärungsmediums zugegeben wurde.
Diese Kultur wurde bei Raum temperatur 24 Std. lang stehengelassen und wurde hierbei gelüftet<B>(11</B> Luft pro Std.). Diese 6 1 der Brut kultur wurde dann 100 Litern des Mediums zuge geben und 24 Std. lang bei Raumtemperatur und un ter Rühren und Lüften (2 1 pro Min.) stehengelassen. Zu dieser 24-Std.-Kultur wurden 25 g 11 ss,21-Di- hydroxy-16a-methyl-4,17(20)-[cis]-pregnadien-3-on, aufgelöst in 500 ml Aceton, hinzugefügt. Das Rühren und Lüften wurde (1 1 pro Min.) 24 Std. lang fort gesetzt.
Das Mycel wurde aus dem Gemisch durch Filtra tion entfernt und mit 20 1 Wasser gewaschen, wobei das Waschwasser dem zuvor erhaltenen Filtrat zuge geben wurde. Das gewaschene Mycel wurde suspen diert und aufgeschlämmt, und zwar je 2mal in 12 1 Aceton und dann wiederum je 2mal in 121 Methylen- chlorid suspendiert. Die so erhaltenen Aceton- und Methylenchlorid-Mycelextrakte wurden vereint und dem Extrakt zugegeben, der aus 4 Extraktionen des Filtrats und des Waschwassers erhalten wurde, wobei jede Extraktion mit 24 Litern Methylenchlorid durch geführt wurde.
Die Extrakte des gefilterten Filtrats und des Waschwassers und die des Mycels wurden kombiniert und 2mal mit jeweils 12 Litern einer 20/aigen Natriumbikarbonatlösung und dann 2mal mit 12 Litern Wasser gewaschen. Die gewaschenen Ex trakte wurden konzentriert und ergaben daraufhin 3 Liter eines Konzentrats, welches im Anschluss daran zur Trockne eingedampft wurde (Dampfbad), worauf man einen rohen kristallinen Rückstand erhielt. Die ser Rückstand wurde 6mal mit 25 ml Diäthyläther behandelt.
Die zurückbleibenden Kristalle wurden in 225 ml heissen Methanols aufgelöst, filtriert und ge kühlt und ergaben Kristalle des llss,21-Dihydroxy- 16a-methyl-1,4,17(20)-[cis]-pregnatrien-3-ons. Präparation <I>5</I> 11 ss,21-Dihydroxy-16a-methyl-4,17(20)-[cis]- pregnadien-3-on-21-acetat. Eine Lösung von 3 g 3,11-Diketo-16ct7methyl- 1,4,17(20) - [cis]-pregnatrien - 21- oin-säuremethylester und 3 ml Pyrrolidin in 16 ml Benzol wurden 1 Std.
unter Rückfluss erhitzt, wobei gleichzeitig das Reak tionswasser entfernt wurde. Auf diese Weise erhielt man 3-Pyrrolidyl-11-keto-16a-methyl-3,5,17(20)- [cis]-pregnatrien-21-oin-säuremethylester. Das Lö sungsmittel wurde von der so erhaltenen Mischung bei vermindertem Druck abdestilliert, worauf der Rückstand mit einer Mischung aus 15 ml Benzol und 10 ml Äther aufgelöst wurde. Dieser Lösung gab man 1,1g Lithiumaluminiumhydrid zu und suspendierte dies in 175 ml Äther. Die erhaltene Lösung wurde bei 26 1,5 Std. lang gerührt.
Dieser Lösung gab man 10 ml Äthylacetat und dann 15 ml Wasser zu, wo nach das Lösungsmittel bei vermindertem Druck ab destilliert wurde. Der im wesentlichen aus anorgani schen Salzen und 3-Pyrrolidyl-llss,21-dihydroxy-16a- methyl-3,5,17(20)-[cis]-pregnatrien bestehende Rück stand wurde in 200 ml Methanol suspendiert, worauf 35 ml einer 5 % igen wässrigen Natriumhydroxyd- lösung der Suspension zugefügt wurden.
Das Ganze wurde dann 10 Min. lang auf 40-45 erhitzt. Die gekühlte Mischung wurde mit Essigsäure und Metha nol neutralisiert und dann destilliert. Der Rückstand wurde mit 50 ml Methylenchlorid und 100 ml Was ser, welches 15 ml konzentrierte Salzsäure enthielt, gerührt. Die Methylenchlorid-Lösung wurde von der wässrigen Schicht abgetrennt, die sodann mit einer weiteren Portion Methylenchlorid extrahiert worden war.
Dieser Extrakt wurde dann zu der ursprüng lichen Methylenchloridlösung zugegeben. Nach Ab- dampfung des Lösungsmittels aus den Extrakten er hielt man rohes 11fl,21-Dihydroxy-16a-methyl-4,17- (20)-[cis]-pregnadien-3-on.
Das rohe 11 ss,21-Dihydroxy-16a-methyl-4,17(20)- [cis]-pregnadien-3-on wurde dann mit 5 ml Essig säureanhydrid mit 10 ml Pyridin bei 26 4 Std. lang behandelt. Die erhaltene Mischung wurde mit 10 ml Wasser vermischt und gekühlt, um das überschüssige Essigsäureanhydrid zu hydrolysieren. Sodann wurde die Lösung mit Methylenchlorid extrahiert. Der Ex trakt wurde erst mit verdünnter Salzsäure, mit Was ser und dann mit einer gesättigten wässrigen Natrium bikarbonatlösung und schliesslich nochmals mit Was ser gewaschen.
Das Methylenchlorid wurde von der Lösung abdestilliert, worauf man den Rückstand aus einer Mischung aus Aceton und Skellysolve B Hexan- kohlenwasserstoffen umkristallisierte.
Auf diese Weise erhielt man reines 11 ss,21-Dihydroxy-16a-methyl- 4,17(20)-[cis]-pregnadien-3-on-acetat. Präparation <I>6</I> 11 ss,21-Dihydroxy-16a-methyl-1,4,17(20)- [cis]-pregnadien-3-on-21-acetat. 11 ss,21- Dihydroxy -16a - methyl -1,4,17 (20)-[cis]- pregnadien-3-on wird in 20 ml Pyridin und 15 ml Acetanhydrid gelöst und 4 Std. lang auf 40 erwärmt.
Die Lösung wird abgekühlt und dann langsam mit Wasser verdünnt. Das ausgefällte Steroid wird ab filtriert, mit Wasser gewaschen und getrocknet.
l lss,21-Dihydroxy-16a-methyl-1,4,17(20)-[cis]- pregnatrien-3-on kann zu anderen 21-Estern umge wandelt werden. Man nimmt hierzu am besten ein geeignetes Säureanhydrid oder Säurehalogenid und kann im übrigen auch nach anderen bekannten Me thoden wie beispielsweise durch Umesterung die obige Verbindung herstellen.
Präparation <I>7</I> 11 ss,17a,21-Trihydroxy-16a-methyl- 1,4-pregnadien-3,20-dion-21-acetat. Zu einer Lösung von<B>100</B> mg 11 ss,21-Dihydroxy- 16a-methyl-1,4,17(20) - [cis] -pregnatrien- 3 -on- 21- acetat in 5 ml trockenen tert. Butylalkohols gab man nacheinander 0,065 ml Pyridin, 0,
43 ml N-Methyl- morpholinoxydperoxyd entsprechend einem Titer von 44 ml 0,01 normaler Natriumtiosulfatlösung pro ml, die durch Reaktion des N-Methylmorpholins mit 2 molaren Äquivalenten von Wasserstoffsuperoxyd in tert. Butylalkohol und 0,2 mg Osmiumtetroxyd in 0,043 ml tert. Butylalkohol. Man liess 24 Std. lang bei Raumtemperatur stehen und verdünnte die Lö sung mit Wasser, worauf sie mit Methylenchlorid extrahiert wurde.
Die Extrakte wurden mit verdünn ter Salzsäure gewaschen, dann mit verdünntem Na triumbikarbonat und schliesslich noch einmal mit Wasser. Nach dem Trocknen wurde die Lösung destil liert und der Rückstand über eine 10 g Fluorisihna- gnesiumsilikatkolonne chromatographiert. Die Ko lonne wurde mit Skellysolve B Hexankohlenwasser- stoffen, die jeweils sich steigernde Acetonmengen ent hielt, entwickelt.
Auf diese Weise eluierte man l Iss,17a,21-Trihydroxy-16a-methyl-1,4-pregnadien- 3,20-dion-21-acetat, welches durch Verdampfen des Lösungsmittels eingeengt wurde.
In den folgenden Beispielen wird je eine Stufe des erfindungsgemässen Verfahrens beschrieben. <I>Beispiel 1</I> 16a.-Methyl-17a,21-dihydroxy-1,4,9(11)-pregnatrien- 3,20-dion-21-acetat.
Zu einer Lösung von 8,5 g 11ss,17a,21-Tri hydroxy-16a-methyl-1,4-pregnadien-3,20-dions-21- acetat in 42,5 ml Pyridin gab man 5,63g N-Brom- acetamid. Man liess bei Raumtemperatur eine Viertel stunde stehen, kühlte daraufhin die Reaktionslösung auf 5-10 ab und leitete dann unter Schütteln Schwe feldioxyd über die Oberfläche der Lösung, so lange, bis dieses Iodstärkepapier nicht mehr färbte. Während der Zugabe des Schwefeldioxydes erwärmte sich die Reaktionsmischung.
Man hielt jedoch die Temperatur unter 30 , indem man von aussen kühlte und die Menge des zugeführten Schwefeldioxydes entspre chend kontrollierte. Sodann gab man zu dieser Reak tionsmischung 400 ml Eiswasser und sammelte den erhaltenen Niederschlag durch Filtration. Das Mate rial wurde aus Aceton-Skellysolve-B-Hexanen um kristallisiert, und man erhielt 16a-Methyl-17a,21-di- hydroxy-1,4,9(11)-pregnatrien-3,20-dion-21-acetat. <I>Beispiel 2</I> 16a-Methyl-17a,21-dihydroxy-4,9(11)-pregnadien- 3,
20-dion-21-acetat. Geht man wie in Beispiel 1 vor und nimmt als Ausgangsmaterial 11 ss,17a,21-Trihydroxy -16a-me- thyl-4-pregnen-3,20-dion-21-acetat, so erhält man 16a-Methyl-17a,21-dihydroxy-4,9(11)-pregnadien- 3,20-dion-21-acetat.
Nimmt man als Ausgangsmaterial ein anderes 21-Acylat des 11ss,17a,21-Trihydroxy-16a-methyl- 1,4-pregnadien-3,20-dion oder ein 21-Acylat des 11 ss,17a,21-Trihydroxy-16a-methyl -4-pregnen-3,20- dions und führt die Dehydration so aus wie in Bei spiel 1, so erhält man das entsprechende 21-Acylat des 16a-Methyl-16a,21-dihydroxy-1,4,9(11)-pregna- trien-3,20-dions bzw.
des 16a-Methyl-17a,21-di- hydroxy-4,9(11)-pregnadien-3,20-dions.
<I>Beispiel 3</I> 9a-Brom-11,B,17a,21-trihydroxy-16a-methyl- 1,4-pregnadien-3,20-dion-21-acetat.
Zu einer Lösung von 5,68 g 16a-Methyl-17a,21- dihydroxy-1,4,9(11)-pregnatrien-3,20-trion-21-acetat in 100 ml Methylenchlorid und 250 ml tert. Butyl- alkohol gab man eine Lösung von 14 ml 721/oiger Perchlorsäure in 100 ml Wasser und dann noch eine Lösung von 2,34 g N-Bromacetamid in 60 m1 tert.- Butylalkohol hinzu.
Nachdem man die Reaktions mischung noch 15 Min. lang gerührt hatte, wurde eine Lösung von 2,8g Natriumsulfit in 140 ml Wasser zugegeben, worauf die Reaktionsmischung auf ein Volumen von 500 ml unter vermindertem Druck und bei 50 eingedampft wurde. Bei diesem Punkt fing die Kristallisation an. Das Konzentrat wurde abgekühlt (Eisbad), während man unter Rühren 500 ml Wasser zugab. Man rührte 1 Std., worauf man das kristalline Produkt vermittels Filtration isolierte und dann die Kristalle mit Wasser wusch und an der Luft trock nete. Auf diese Weise erhielt man 9a-Brom-11ss,17a 21-trihydroxy-16a-methyl-1,4-pregnadien-3,20-dion- 21-acetat.
<I>Beispiel 4</I> 9a-Brom-11 ss,17a,21-trihydroxy-16a-methyl- 4-pregnen-3,20-dion-21-acetat. Man arbeitet wie im Beispiel 3, geht aber von 16a-Methyl-17a,21-dihydroxy-4,9(11)-pregriadien- 3,20-dion-21-acetat aus und erhält so 9a-Brom- 11 ss,17a,21-trihydroxy-16a- methyl-4-pregnen-3,20- dion-21-acetat.
Geht man von anderen 21-Acylaten des 16a-Me thyl-17a,21-dihydroxy-1,4,9(11)-pregnadien-3,20- dions oder des 17a,21-Dihydroxy-16a-methyl-4,9(11)- pregnadien-3,20-dions aus und verfährt im übrigen wie in Beispiel 3, so erhält man die entsprechenden 21-Acylate des 9a-Brom-11 ss,17a,21-trihydroxy-16a- methyl-1,4-pregnadien-3,20-dions sowie des 9a-Brom- 11 ss,17a,
21-trihydroxy-16a-methyl- 4-pregnen-3,20- dions.
Ersetzt man das N-Chlorsuccinimid durch N- Bromacetamid und arbeitet im übrigen wie in den Beispielen 2 und 3, so erhält man die entsprechenden 9a-Chlorverbindungen, z.
B. 9a-Chlor-17a,21-tri hydroxy-16a-methyl-1,4-pregnadien-3,20-dion-21- acetat bzw. 9a-Chlor-llss,17a,21-trihydroxy-16a-me- thyl-4-pregnen-3,20-dion-21-acetat. <I>Beispiel 5</I> Zu einer Lösung von 6,78 g 9a-Brom-11ss,17a,21 trihydroxy-16a-methyl-1,4-pregnadien-3,20-dion-21- acetat in 175 ml Aceton gab man 6,78g Kalium- acetat und erhitzte die so erhaltene Suspension unter Rückfluss etwa 17 Std. lang. Die Mischung wurde dann konzentriert, bis ihr Volumen etwa 60 ml be trug (unter vermindertem Druck) und wurde dann mit Wasser verdünnt.
Hiernach extrahierte man sie mit Methylenchlorid. Die Methylenchloridextrakte wurden kombiniert, mit Wasser gewaschen, dann mit Natriumsulfat getrocknet und eingedampft. Der Rück stand wurde wieder in Methylenchlorid aufgelöst und an 200 g Florisil chromatographiert. Die Kolonne wurde mit Einliterportionen Skellysolve B-Hexanen, die gesteigerte Mengen an Aceton enthielten, eluiert.
Auf diese Weise erhielt man 9ss,llss-Epoxy-16a methyl-17ce,21-dihydroxy-1,4-pregnadien-3,20-dion- 21-acetat, nachdem durch Eindampfen der Eluate das Lösungsmittel entfernt worden war.
<I>Beispiel 6</I> Arbeitet man wie in Beispiel 5, geht jedoch von 9(t-Brom-1 1ss,17 a,21- trihydroxy -16a - methyl-4-pre- gnen-3,20-dion-21-acetat aus, so erhält man 9ss,llss Epoxy-16a-methyl-17a,21-dihydroxy-4-pregnen-3,20- dion-21-acetat. <I>Beispiel 7</I> Zu ungefähr 1,3 g Fluorwasserstoff in einer Poly- äthylenflasche, der bei einer Temperatur von 60 gehalten wurde, gab man 2,3 ml Tetrahydrofuran und dann eine Lösung von 500 mg (0,
0012 Mol) 9ss,11 ss-Epoxy-17a,21-dihydroxy-16a-methyl-1,4-pre- gnadien-3,20-dion-21-acetat in 2 ml Methylenchlorid. Die Steroidlösung wurde mit zusätzlichen 2 ml Me- thylenchlorid eingespült. Die leicht rötlich gefärbte Lösung wurde sodann 2 Std. bei etwa -30 gehalten und dann 2 Std. bei etwa -10 .
Am Ende dieser Zeit wurde sie vorsichtig mit einem überschuss gekühlten Natriumbikarbonats, welches in Wasser aufgelöst war, versetzt, worauf das organische Material mit Hilfe von zusätzlichem Methylenchlorid extrahiert wurde. Die kombinierten Extrakte wurden mit Wasser ge waschen, mit Natriumsulfat getrocknet und sodann auf 35 ml unter vermindertem Druck eingedampft.
Die Lösung wurde mit 40 ml Florisil chromatogra- phiert. Die Kolonne wurde mit 500-ml-Portionen von Skellysolve B-Hexanen, die steigende Mengen Aceton enthielten, entwickelt. Man eluierte so das 9a-Fluor- 1 lss,17cu,21-dihydroxy-16a-methyl-1,4-pregnadien- 3,20-dion-21-acetat, welches durch Eindampfen der Eluatfraktionen vom Lösungsmittel befreit wurde.
<I>Beispiel 8</I> Man arbeitet wie in Beispiel 7, geht jedoch von 9ss,11 ss-Epoxy-17a,21-dihydroxy-16a-methyl-4-pre- gnen-3,20-dion-21-acetat aus und erhält so 9a-Fluor- 11 ss,17a,21- trihydroxy -16a - methyl - 4-pregnen-3,20- dion-21-acetat. Geht man in ähnlicher Weise von anderen 21- Acylaten des 9ss,llss-Epoxy-17a,21-dihydroxy-16a- methyl-1,4-pregnadien-3,20-dions oder des 9ss,
llss Epoxy-17a,21-dihydroxy-16a-methyl-4-pregnen-3,20- dions aus und verfährt im übrigen so wie in Bei spiel 7 beschrieben, so erhält man das entsprechende 21-Acylat des 9a-Fluor-11ss,17a,21-trihydroxy-16a- methyl-4-pregnadien-3,20-dions bzw. des 9a-Fluor- 11 ss,17a-trihydroxy-16a-methyl-4-pregnen-3,20-dions.
Die erhaltenen Steroide können folgendermassen oxydiert werden: Man stellte eine Lösung her, die 1 ml Essig säure, 5 mg 9a-Fluor-11ss,17a,21-trihydroxy-16a- methyl-1,4-pregnadien-3,20-dion-21-acetat sowie 20 mg Chromsäureanhydrid und einen Tropfen (etwa 5 mg) Wasser enthielt. Diese Mischung wurde einige Male bei Raumtemperatur geschüttelt und 4 Std. lang stehengelassen. Hiernach wurde sie in 10 ml Was ser geschüttet und 20 Std. lang auf etwa 5 abgekühlt.
Das sich von der wässrigen Mischung abtrennende Gemisch wurde abgesaugt, und man erhielt 9a-Fluor 16a-methyl-17a,21-dihydroxy-1,4-pregnadien-3,11,20- trion-21-acetat.
Geht man vom 9a-Fluor-llss,17a,21-trihydroxy- 16a-methyl-4-pregnen-3,20-dion-21-acetat aus, so er hält man das entsprechende 3,11,20-Trion-21-acetat.
Die erhaltenen Steroide können folgendermassen hydrolysiert werden: 3,25 g 9a-Fluor-llss,17a,21-trihydroxy-16a-me- thyl-1,4-pregnadien-3,20-dion-21-acetat wurden in 325 ml Methanol aufgelöst, wobei zuvor der Luft sauerstoff vermittels 10 Minuten währenden Durch leitens von Stickstoff durch die Lösung ausgetrieben wurde. Hierzu gab man eine Lösung von 1,63g Ka- liumbicarbonat in 30 ml Wasser und spülte noch mals mit Stickstoff. Man liess die Mischung während 5 Std. unter Stickstoff bei Raumtemperatur stehen und neutralisierte dann mit 2,14 ml Essigsäure in 40 ml Wasser.
Darauf wurde die Mischung auf etwa <B>1/3</B> ihres ursprünglichen Volumens eingeengt (bei vermindertem Druck und 60 Wasserbadtemperatur). Darauf gab man 250 ml Wasser zu und kühlte die Mischung. Das kristalline Produkt wurde abgesaugt, mit Wasser gewaschen und getrocknet. Es handelt sich um 9a-Fluor-llss,17a,21-trihydroxy-16a-methyl- 1,4-pregnadien-3,20-dion.
Geht man vom 21-Acetat des 9a-Fluor-11ss,17a, 21-trihydroxy-16a-methyl-4-gregnen-3,20-dions aus, so erhält man 9a-Fluor-11,B,17a,21-trihydroxy-16a- methyl-4-pregnen-3,20-dion.
Zur Veresterung stellte man eine Lösung her, die 50 mg 9a-Fluor-11ss,17a,21-trihydroxy-16a-methyl- 1,4-pregnadien-3,20-dion in 1 ml Pyridin und 1 ml Propionsäureanhydrid enthielt. Diese Lösung liess man 21 Std. lang bei Raumtemperatur stehen und schüttete sie dann in 10 ml Wasser.
Die Reaktions mischung wurde darauf mit je 10 ml Methylenchlorid 3mal extrahiert, worauf die vereinigten Methylenchlo- ridextrakte mit Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft wurden, wobei man 9a-Fluor-11 ss,17a,21-trihydroxy-16ä-methyl-1,4-pre- gnadien-3,20-dion-21-propioriat erhielt.
Geht man vom 9a-Fluor-llss.,17a,21-trihydroxy- 16a-methyl-4-pregnen-3,20-dion aus; so' erhält man 9a-Fluor-llss,17a,21-trihydroxy-16a-methyl-4-pre- gnen-3,20-dion-21-propionat.

To a hot solution of 4.56 g (0.014 mol) of 11-keto-16-dehydroprogesterone in 45 ml of methanol, 2.25 ml of pyrolydine were added with agitation of the liquids. The reaction product, the 3-pyrrolydin-enamine of 11-keto-16-dehydroprogesterone, separated out soon afterwards in yellow crystals. It was then cooled to 5. The mixture was filtered, the filter cake washed (with cold methanol) and dried in vacuo.
He obtained 4.56 g of 3-pyrrolydyl-3,5,16-pregnatriene-11,20-dione, which melts at 154-169.
A solution of the enamine thus obtained in 110 ml of tetrahydrofuran was added slowly and with stirring to a suspension of 45 ml of commercially available 3-molar methyl magnesium bromide, and then also 0.90 g of cuprous chloride. The mixture was cooled to room temperature and stirred for a further 3 hours under a nitrogen atmosphere. The excess Grignard reagent was carefully destroyed by the dropwise addition of 6.5 ml of water.
To the mixture containing 3-pyrrolydyl-16a-methyl-3,5-pregnadiene-11,20-dione, 4 ml of acetic acid and 16 ml of methanol were added, and it was heated until a clear yellow solution was obtained . 16 ml of a 10% strength aqueous solution of sodium hydroxide were added, a pH of 8 being established.
The mixture was then heated under reflux for a further 45 minutes and then 1 ml of acetic acid and 250 ml of water were added, whereupon the cooled mixture was extracted 3 times with methylene chloride. The combined extracts were washed (with an aqueous bicarbonate solution) and then dried with sodium sulfate.
The dried solution was evaporated to dryness and the residue - essentially 11-keto-16a-methylprogesterone - was dissolved in 400 ml of methylene chloride and chromatographed through a 200 g column of magnesium silicate (Florisil). The column was developed with quantities of 400 ml of solvent in the following composition and order:
Five from hexanes (Skellysolve B) + 3% acetone and then with + 5% acetone, eight parts hexanes + 7% acetone, 5 parts hexanes + 104 / o acetone,
4 parts of hexanes + 15% acetone and finally one from acetone. Fractions 9-25 (including the methylene fraction) were combined and recrystallized from a mixture of acetone and water. In this way, 2.03 g of 11-keto-16a-methyl-progesterone were obtained, the melting point of which was between 179 and 182.
A sample recrystallized from acetylacetate and hexanes melted between 183 and 185, had an (a) D of +255 (CHC13) and an A, m ä ;. from 238, a and an am from <B> 15,850 </B> and the following analysis:

A solution of 2.00 g (5.8 millimoles) of 11-keto-16a-methylprogesterone in 30 ml of dry tert-butyl alcohol was heated to 50 under a nitrogen atmosphere.
3.2 ml of ethyl oxalate and 3.03 g of a 25% strength methanolic solution of sodium hydroxide were added to this solution. A greenish-yellow precipitate of the sodium dienolate of 2,21-diethoxyoxalyl-11-keto-16a-methylprogesterone then formed immediately.
The mixture was stirred for a further 20 minutes and then a cooled solution of 0.98 g of dry sodium acetate and 0.84 ml of acetic acid in 40 ml of methanol was added, thus producing the free dienol. The solution was then cooled to 0, whereupon a likewise cooled solution of 2.0 g of bromine in methanol was added dropwise over a period of 10 minutes. In this way 2,21,21 tribromo-2,21-diethoxyoxalyl-1-keto-16a-methyl-progesterone were obtained.
The cooling bath was removed, whereupon 5.72 g of a 25% strength methanolic sodium methoxide solution were added to the solution. Stirring was continued for a further 2.5 hours, and methyl 2-bromo-2,11-diketo-16a-methyl-4,17 (20) - [cis] -pregnadiene-21-oinate was obtained.
5 ml of acetic acid and 1 g of zinc dust were then added to the resulting solution, after which the stirring was continued for half an hour. The mixture was then diluted with water and the solids were filtered off. The filtrate was extracted exhaustively with methylene chloride and the extract was dried with sodium sulfate and evaporated. The residue was dissolved in 400 ml of methylene chloride, whereupon it was poured into a column containing 200 g of magnesium silicate (Florisil).
The column was developed with 400 ml portions of a solution by means of the following composition in the following order: 4 hexanes (Skellysolve B) + 5 0.1o acetone, 10 hexanes + 7% acetone, 10 hexanes + 10% acetone,
and finally 1 part acetone only.
Fractions 14-23 (with the methylene fraction) contained the starting 1 1-keto-16a-methylprogesterone. Fractions 7-12 contained 3,11-diketo-16a-methyl-4,17 (20) - [cis] -pregnadiene-21-o-yn-acid methyl ester, which after recrystallization from methanol-water and then only melted from methanol between 177 and 184.

A solution of 6.0 g of 3,11-diketo-16a-methyl-4,17 (20) - [cis] - pregnadiene-21-oin-acid methyl ester, 6 ml of pyrrolydine and 180 ml of p-toluenesulfonic acid in 120 ml Benzene were refluxed for 1 hour. heated for a long time, the water of reaction was continuously removed. The solution was distilled, and a residue was obtained which consisted essentially of 3-pyrrolidyl-16a-methyl-3,5,17 (20) - [cis] -pregnatriene-21-methyl ester.
The residue was then dissolved in 30 ml of benzene and 10 ml of ether and a suspension of 2.2 g of lithium aluminum hydride in 350 ml of ether was added over a period of 5 minutes. This mixture was stirred at room temperature for a further 90 minutes. 20 ml of ethyl acetate were added and, in order to destroy the excess lithium aluminum hydride, 30 ml of water were then added.
The ether and benzene were distilled under reduced pressure, whereupon one consisting essentially of 3-pyrrolidyl-11f, 21-dihydroxy-16a-methyl-3,5,17 (20) - [cis] -pregnatrien-21-oin -Acid methyl ester obtained residue that was mixed with some inorganic substances.
This crude distillation residue was mixed with 400 ml of methanol and heated to 40 until a solution was obtained. 70 ml of an aqueous 5% sodium hydroxide solution was added, the mixture being warmed to 40 for a further 10 min. The solution was neutralized (with acetic acid), the solvent was distilled off under reduced pressure, namely at a temperature below 45.
The residue was mixed with 100 ml of water containing 10 ml of concentrated hydrochloric acid, filtered and washed, whereupon I1 / 3,21-dihydroxy-16a-methyl-4,17 (20) - [cis] -pregnadiene-3- on received. Preparation <I> 4 </I> 11, B, 21-dihydroxy-16a-methyl-1,4,17 (20) - [cis] - pregnatrien-3-one.
Septomyxa affinis was grown on a malt agar soil, which consisted of 50 g dry malt extract, 5 g edamin (enzymatic digestion of lactalbumin) and 20 g agar dissolved in 1 liter of tap water, with a pH of 6.5-7 , 0 was set.
The inoculum of the agar soil was incubated in a 1 liter Erlenmeyer flask containing 100 ml of malt extract at room temperature for 4-7 days so that spores could form.
The spores were suspended in 100 ml of a sterile 1% saline solution. 5 ml of this spore inoculum was in each case 250 ml flasks containing 100 ml of the medium,
given. The medium consisted of 1% Cerelos dextrose and 2'0 / 0, 60% solids soft cereal which was brought to a pH between 4.8 and 5 with sodium hydroxide. The mixture was then incubated for 48-72 hours at room temperature with shaking, after which the whole was added to 61 of the above fermentation medium.
This culture was left to stand at room temperature for 24 hours and was ventilated (11 air per hour). This 6 l of the brood culture was then added to 100 liters of the medium and left to stand for 24 hours at room temperature with stirring and ventilation (2 l per minute). 25 g of 11 ss, 21-dihydroxy-16a-methyl-4,17 (20) - [cis] -pregnadien-3-one, dissolved in 500 ml of acetone, were added to this 24-hour culture. Stirring and ventilating were continued (1 liter per minute) for 24 hours.
The mycelium was removed from the mixture by filtration and washed with 20 liters of water, the washing water being added to the filtrate obtained above. The washed mycelium was suspended and slurried, namely twice in 12 l of acetone each time and then again twice in each case in 121 of methylene chloride. The acetone and methylene chloride mycelium extracts thus obtained were combined and added to the extract obtained from 4 extractions of the filtrate and the washing water, each extraction being carried out with 24 liters of methylene chloride.
The extracts of the filtered filtrate and the washing water and those of the mycelium were combined and washed twice with 12 liters each of a 20 / ai sodium bicarbonate solution and then twice with 12 liters of water. The washed extracts were concentrated to give 3 liters of a concentrate which was then evaporated to dryness (steam bath), whereupon a crude crystalline residue was obtained. The water residue was treated 6 times with 25 ml of diethyl ether.
The remaining crystals were dissolved in 225 ml of hot methanol, filtered and cooled to give crystals of the llss, 21-dihydroxy-16a-methyl-1,4,17 (20) - [cis] -pregnatrien-3-one. Preparation <I> 5 </I> 11 ss, 21-dihydroxy-16a-methyl-4,17 (20) - [cis] - pregnadien-3-one-21-acetate. A solution of 3 g of 3,11-diketo-16ct7methyl-1,4,17 (20) - [cis] -pregnatriene-21-oin-acid methyl ester and 3 ml of pyrrolidine in 16 ml of benzene were 1 hour.
heated under reflux, the reaction water being removed at the same time. In this way, 3-pyrrolidyl-11-keto-16a-methyl-3,5,17 (20) - [cis] -pregnatrien-21-o-yn-acid methyl ester was obtained. The solvent was distilled off from the mixture thus obtained under reduced pressure, whereupon the residue was dissolved with a mixture of 15 ml of benzene and 10 ml of ether. 1.1 g of lithium aluminum hydride were added to this solution and this was suspended in 175 ml of ether. The resulting solution was stirred at 26 for 1.5 hours.
This solution was added 10 ml of ethyl acetate and then 15 ml of water, after which the solvent was distilled off under reduced pressure. The residue consisting essentially of inorganic salts and 3-pyrrolidyl-llss, 21-dihydroxy-16a-methyl-3,5,17 (20) - [cis] -pregnatrien was suspended in 200 ml of methanol, whereupon 35 ml of a 5% aqueous sodium hydroxide solution was added to the suspension.
The whole was then heated to 40-45 for 10 min. The cooled mixture was neutralized with acetic acid and methanol and then distilled. The residue was stirred with 50 ml of methylene chloride and 100 ml of what water, which contained 15 ml of concentrated hydrochloric acid. The methylene chloride solution was separated from the aqueous layer, which was then extracted with another portion of methylene chloride.
This extract was then added to the original methylene chloride solution. After evaporation of the solvent from the extracts, crude 11fl, 21-dihydroxy-16a-methyl-4,17- (20) - [cis] -pregnadien-3-one was obtained.
The crude 11ss, 21-dihydroxy-16a-methyl-4,17 (20) - [cis] -pregnadien-3-one was then treated with 5 ml of acetic anhydride with 10 ml of pyridine at 26 for 4 hours. The obtained mixture was mixed with 10 ml of water and cooled to hydrolyze the excess acetic anhydride. The solution was then extracted with methylene chloride. The extract was washed first with dilute hydrochloric acid, with water and then with a saturated aqueous sodium bicarbonate solution and finally again with water.
The methylene chloride was distilled off from the solution, whereupon the residue was recrystallized from a mixture of acetone and Skellysolve B hexane hydrocarbons.
In this way, pure 11ss, 21-dihydroxy-16a-methyl-4,17 (20) - [cis] -pregnadien-3-one acetate was obtained. Preparation <I> 6 </I> 11 ss, 21-dihydroxy-16a-methyl-1,4,17 (20) - [cis] -pregnadien-3-one-21-acetate. 11 ss, 21-dihydroxy-16a - methyl -1,4,17 (20) - [cis] - pregnadien-3-one is dissolved in 20 ml of pyridine and 15 ml of acetic anhydride and warmed to 40 for 4 hours.
The solution is cooled and then slowly diluted with water. The precipitated steroid is filtered off, washed with water and dried.
lss, 21-dihydroxy-16a-methyl-1,4,17 (20) - [cis] - pregnatrien-3-one can be converted to other 21-esters. The best way to do this is to use a suitable acid anhydride or acid halide, and the above compound can also be prepared using other known methods, for example by transesterification.
Preparation <I> 7 </I> 11 ss, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate. To a solution of <B> 100 </B> mg 11 ss, 21-dihydroxy-16a-methyl-1,4,17 (20) - [cis] -pregnatrien-3-one-21-acetate in 5 ml dry tert. Butyl alcohol was given successively 0.065 ml of pyridine, 0,
43 ml of N-methyl morpholine oxide peroxide corresponding to a titer of 44 ml of 0.01 normal sodium thiosulfate solution per ml, which is obtained by reaction of N-methyl morpholine with 2 molar equivalents of hydrogen peroxide in tert. Butyl alcohol and 0.2 mg osmium tetroxide in 0.043 ml tert. Butyl alcohol. The mixture was left to stand for 24 hours at room temperature and the solution was diluted with water, whereupon it was extracted with methylene chloride.
The extracts were washed with dilute hydrochloric acid, then with dilute sodium bicarbonate and finally again with water. After drying, the solution was distilled and the residue was chromatographed over a 10 g fluorine magnesium silicate column. The column was developed with Skellysolve B hexane hydrocarbons, each of which contained increasing amounts of acetone.
In this way, I Iss, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate was eluted, which was concentrated by evaporation of the solvent.
One stage of the process according to the invention is described in each of the following examples. <I> Example 1 </I> 16a.-Methyl-17a, 21-dihydroxy-1,4,9 (11) -pregnatriene-3,20-dione-21-acetate.
5.63 g of N-bromine were added to a solution of 8.5 g of 11ss, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate in 42.5 ml of pyridine - acetamide. The mixture was left to stand at room temperature for a quarter of an hour, the reaction solution was then cooled to 5-10 and then sulfur dioxide was passed over the surface of the solution while shaking until this iodine starch paper no longer stained. During the addition of the sulfur dioxide, the reaction mixture warmed up.
However, the temperature was kept below 30 by cooling from the outside and controlling the amount of sulfur dioxide supplied accordingly. 400 ml of ice water were then added to this reaction mixture, and the resulting precipitate was collected by filtration. The material was recrystallized from acetone-Skellysolve-B-hexanes, and 16a-methyl-17a, 21-di-hydroxy-1,4,9 (11) -pregnatriene-3,20-dione-21-acetate was obtained . <I> Example 2 </I> 16a-methyl-17a, 21-dihydroxy-4,9 (11) -pregnadiene-3,
20-dione-21-acetate. Proceeding as in Example 1 and using 11 ss, 17a, 21-trihydroxy-16a-methyl-4-pregnen-3,20-dione-21-acetate as starting material, 16a-methyl-17a, 21 is obtained -dihydroxy-4,9 (11) -pregnadiene-3,20-dione-21-acetate.

<I> Example 3 </I> 9a-Bromo-11, B, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate.
To a solution of 5.68 g of 16a-methyl-17a, 21-dihydroxy-1,4,9 (11) -pregnatriene-3,20-trione-21-acetate in 100 ml of methylene chloride and 250 ml of tert. A solution of 14 ml of 72% perchloric acid in 100 ml of water and then a solution of 2.34 g of N-bromoacetamide in 60 ml of tert-butyl alcohol were added to butyl alcohol.
After the reaction mixture had been stirred for a further 15 minutes, a solution of 2.8 g of sodium sulfite in 140 ml of water was added, whereupon the reaction mixture was evaporated to a volume of 500 ml under reduced pressure and at 50%. At this point crystallization started. The concentrate was cooled (ice bath) while 500 ml of water were added with stirring. The mixture was stirred for 1 hour, whereupon the crystalline product was isolated by means of filtration and then the crystals were washed with water and dried in the air. In this way, 9a-bromo-11ss, 17a 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate was obtained.
<I> Example 4 </I> 9a-bromo-11ss, 17a, 21-trihydroxy-16a-methyl-4-pregnen-3,20-dione-21-acetate. The procedure is as in Example 3, but starting with 16a-methyl-17a, 21-dihydroxy-4,9 (11) -pregriadiene-3,20-dione-21-acetate and thus obtaining 9a-bromo-11 ss, 17a , 21-trihydroxy-16a-methyl-4-pregnen-3,20-dione-21-acetate.
If one starts from other 21-acylates of 16a-methyl-17a, 21-dihydroxy-1,4,9 (11) -pregnadiene-3,20-dione or of 17a, 21-dihydroxy-16a-methyl-4,9 (11) -Pregnadiene-3,20-dione and if the rest of the procedure is as in Example 3, the corresponding 21-acylates of 9a-bromo-11, 17a, 21-trihydroxy-16amethyl-1,4 are obtained -pregnadiene-3,20-dione and 9a-bromine-11 ss, 17a,
21-trihydroxy-16a-methyl-4-pregnen-3,20-dione.
If the N-chlorosuccinimide is replaced by N-bromoacetamide and the rest of the procedure is as in Examples 2 and 3, the corresponding 9a-chlorine compounds are obtained, e.g.
B. 9a-chloro-17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate or 9a-chloro-llss, 17a, 21-trihydroxy-16a-me- ethyl 4-pregnene-3,20-dione-21-acetate. <I> Example 5 </I> To a solution of 6.78 g of 9a-bromo-11ss, 17a, 21 trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate in 175 ml Acetone was added to 6.78 g of potassium acetate and the suspension thus obtained was heated under reflux for about 17 hours. The mixture was then concentrated until its volume was about 60 ml (under reduced pressure) and was then diluted with water.
They were then extracted with methylene chloride. The methylene chloride extracts were combined, washed with water, then dried with sodium sulfate and evaporated. The residue was redissolved in methylene chloride and chromatographed on 200 g of Florisil. The column was eluted with one liter portions of Skellysolve B hexanes containing increased amounts of acetone.
In this way, 9ss, 11ss-epoxy-16a methyl-17ce, 21-dihydroxy-1,4-pregnadiene-3,20-dione-21-acetate was obtained after the solvent had been removed by evaporation of the eluates.
<I> Example 6 </I> If one works as in example 5, but starts from 9 (t-bromo-11ss, 17 a, 21-trihydroxy-16a - methyl-4-pregnen-3,20-dione -21-acetate, 9ss, llss epoxy-16a-methyl-17a, 21-dihydroxy-4-pregnen-3,20-dione-21-acetate is obtained. <I> Example 7 </I> About 1 , 3 g of hydrogen fluoride in a polyethylene bottle, which was kept at a temperature of 60, were given 2.3 ml of tetrahydrofuran and then a solution of 500 mg (0,
0012 mol) 9ss, 11ss-epoxy-17a, 21-dihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate in 2 ml of methylene chloride. The steroid solution was rinsed in with an additional 2 ml of methylene chloride. The slightly reddish colored solution was then kept at about -30 for 2 hours and then at about -10 for 2 hours.
At the end of this time, excess chilled sodium bicarbonate, dissolved in water, was carefully added and the organic material was extracted with the aid of additional methylene chloride. The combined extracts were washed with water, dried with sodium sulfate and then evaporated to 35 ml under reduced pressure.
The solution was chromatographed with 40 ml of Florisil. The column was developed with 500 ml portions of Skellysolve B hexanes containing increasing amounts of acetone. The 9a-fluoro-1 lss, 17cu, 21-dihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate, which was freed from the solvent by evaporation of the eluate fractions, was thus eluted.
<I> Example 8 </I> The procedure is as in Example 7, but starting with 9ss, 11ss-epoxy-17a, 21-dihydroxy-16a-methyl-4-pregnen-3,20-dione-21- acetate and thus receives 9a-fluoro-11 ss, 17a, 21-trihydroxy-16a-methyl-4-pregnen-3,20-dione-21-acetate. If one proceeds in a similar way from other 21-acylates of 9ss, llss-epoxy-17a, 21-dihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione or of 9ss,
llss epoxy-17a, 21-dihydroxy-16a-methyl-4-pregnen-3,20-dione and otherwise proceeds as described in Example 7, the corresponding 21-acylate of 9a-fluoro-11ss is obtained, 17a, 21-trihydroxy-16a-methyl-4-pregnadiene-3,20-dione or des 9a-fluoro-11ss, 17a-trihydroxy-16a-methyl-4-pregnen-3,20-dione.
The steroids obtained can be oxidized as follows: A solution was prepared containing 1 ml of acetic acid, 5 mg of 9a-fluoro-11ss, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione 21 acetate as well as 20 mg chromic anhydride and one drop (about 5 mg) water. This mixture was shaken a few times at room temperature and left to stand for 4 hours. It was then poured into 10 ml of water and cooled to about 5 for 20 hours.
The mixture separating from the aqueous mixture was filtered off with suction, and 9a-fluoro 16a-methyl-17a, 21-dihydroxy-1,4-pregnadiene-3,11,20-trione-21-acetate was obtained.
If you start from 9a-fluoro-llss, 17a, 21-trihydroxy-16a-methyl-4-pregnen-3,20-dione-21-acetate, you get the corresponding 3,11,20-trione-21- acetate.
The steroids obtained can be hydrolyzed as follows: 3.25 g of 9a-fluoro-IIss, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione-21-acetate were dissolved in 325 ml of methanol dissolved, the oxygen having previously been expelled from the air by passing nitrogen through the solution for 10 minutes. For this purpose, a solution of 1.63 g of potassium bicarbonate in 30 ml of water was added and the system was flushed again with nitrogen. The mixture was left to stand under nitrogen at room temperature for 5 hours and then neutralized with 2.14 ml of acetic acid in 40 ml of water.
The mixture was then concentrated to about 1/3 of its original volume (at reduced pressure and water bath temperature). 250 ml of water were then added and the mixture was cooled. The crystalline product was filtered off with suction, washed with water and dried. It is 9a-fluoro-llss, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione.
If one starts from the 21-acetate of 9a-fluoro-11ss, 17a, 21-trihydroxy-16a-methyl-4-gregnen-3,20-dione, then 9a-fluoro-11, B, 17a, 21-trihydroxy is obtained -16a-methyl-4-pregnen-3,20-dione.
For esterification, a solution was prepared which contained 50 mg of 9a-fluoro-11ss, 17a, 21-trihydroxy-16a-methyl-1,4-pregnadiene-3,20-dione in 1 ml of pyridine and 1 ml of propionic anhydride. This solution was left to stand for 21 hours at room temperature and then poured into 10 ml of water.
The reaction mixture was then extracted 3 times with 10 ml of methylene chloride each time, whereupon the combined methylene chloride extracts were washed with water, dried with sodium sulfate and evaporated, giving 9a-fluoro-11ss, 17a, 21-trihydroxy-16ä-methyl-1 , 4-pregnadien-3,20-dione-21-propioriat.
If one starts from 9a-fluoro-llss., 17a, 21-trihydroxy-16a-methyl-4-pregnen-3,20-dione; This gives 9a-fluoro-IIIss, 17a, 21-trihydroxy-16a-methyl-4-pregnen-3,20-dione-21-propionate.
