Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Komplexverbindungen aus quaternären Acyloxyäthylammocarbonylmethyiamm- salzen und Jod, dadurch gekennzeichnet, dass man auf eine Verbindung der Formel

worin n eme ganze Zahl von 4 bis 20 darstellt, X ain Anion bedeutet und das quaternäre N-Atom Glied eines heterocyclischen Ringes ist, Jod einwirken lässt.
Die neuen Komplexe stellen wertvolle Keim- tötungsmittel, Baktericide und Antisepticafür Spitäler, den Haushalt und die Industrie dar.
Es ist seit langem bekannt, dass Jod wegen seiner anti, septischen Wirks, amkeit als keimt¯tungsmittel benutzt werden kann. Man hat es daher in verschicdener Form zur Desinfektion der K¯rperhaut, von Wunden und Schleimhäuten, zur Stemlisteruog von Haar, Catgut und chirurgischer Instrumente verwen- det. Es ist auch als prophylaktisches und therapeu- tisches Mittel bei Krankheiten herangezogen worden, die durch Bakterien, Viren oder Hautpilze hervor- gerufen werden sowie zur Desinfektion von Trinkwasser, Wasser in Schwimmbassins und von Essgeräten.
Eine erwünschte Eigenschaft des Jods als Keim- tötungsmittel besteht in der Tatsache, dass sein Wir kungsbereich sehr breit ist. Es reicht von den Viren bis zu den primitiven Filarien der Nematoden. Auf diese Weise übt Jod eine letale und eine schützende Wirkung gegen eine grosse Anzahl von Organismen aus, einschliesslich der Viren, Rikettsien, Sporen, B, ak- terien, Hefezellen, Schimmel, Pilze, Photozoen (Cysten) und auch gegen Trophozoiten und Helminthen (Nematoden, Trematoden und Cestoden sowie Eier und Wiirmer) aus.
Verschiedene Forscher haben festgestellt, dass Jod besonders zweckmässig als Desinfektionsmittel für chirurgische Instrumente ist. Es ist ferner ermittelt worden, dass Jod gegen Tuberkelbazillen und andere wide, rstan, dsfähige Bakterien unter Bedingungen wirk- sam ist, unter denen andere Keimt¯tungsmittel versagen. Man hat auch gezeigt, dass Jod d zur Bekämp- fung verschiedener Viruserkrankungen, einschie¯lich Influenza, infektiöser Hapatitis und Kinderlähmung verwendet werden kann.
Ausserdem hat Jod bei seiner Anwendung a ; Is Keimt¯tungsmittel noch einen anderen Vorteil. Es dient als sein eigener Indikator. Wenn die charakte ristische braune Farbe der Jodl¯sungen verschwindet, ist es ein Zeichen dafür, da¯ ihr Jodgshalt verbraucht ist. Ein weiterer Vorteil besteht darin, dal3 das Jod seine Zerstörungskraft gegenüber Mikroorganismen sehr rasch geltend macht, ohne dass umständliche Vorrichtungen, wie Autoklaven oder Ofen notwendig sind.
Trotz dieser offensichtlichen Vorz ge ist aber der Gebrauch des freien Jods als Keimtötungsmiittel und Antiseptikum weitgehend durch die erheblichen Nachteile beschränkt, die er mit sich bringt. Es ist ein starkes Reizmittel f r die tierische Haut und macht sie empfindlich. Seine Wirkung ist nicht selektiv zwit- schen dem Protein der Balcterien und dem der Säuge- tiere. Seine unbeaufsichtigte Anwendung auf der Haut führt zu tiefreichenden Geschw ren, die nur langsam heilen. Es ist sehr giftig.
Seine Wirkung kann durch Serum, Fäces, Gewebewasser, Sputum, Harn, Glyce- rin, Sirup, Eier, Milch und gewisse anorganische Stoffe, wie Natriumthiosulfat,Ammoniakwasserund verschiedene Reduktionsmittel weitgehend herabgesetzt oder vollständig aufgehoben werden.
Andere Nachteile, welche mit der Verwendung von freiem Jod verbunden sind, ergeben sich aus der Tatsache, dass es einen hohen Dampfdruck besitzt und daher rasch aus seinen Losungen oder von den Oberflächen, auf die es aufgetragen wurde, verdampft.
Es greift Metalle stark an, so dal3 man es zur Steri- lisation vieler ärztlicher Instrumente nicht gebrauchen kann. Es lässt auf Tüchern und Geweben hässliche Flecken zurück. Aus seinen Lösungen fÏllt es aus, sobald man sie mit Wasser verd nnt. Es ist nur werig in Wasser loslich und diese Lösungen sind schwach und dringen nicht ein.
Die vorliegende Erfindung führt nun zu neuen, keimtötenden Jodverbindungen, welche alle die zahlreichen Vorz ge aufweisen, die f r das Jod charak- teristisch sind, ohne jedoch die Nachteile zu besitzen, welche dieser Substanz anhaften. Die neuen Verbindungen haben einen Phenolkoeffizienten, der h¯her ist als der des freien Jods. Sie haben einen breiten Wirkungsbereich und sind gegen ber einer gro¯en Zahl von Organismen, einschliesslich der Viren, wirksam. Diese Wirksamkeit wird nicht durch die Gegenwart fremder Stoffe, wie Serum, Albu. min und Blut, beeinträchtigt. Die neuen Verbindungen enthalten das Jod m hoher Konzentration und wirken rasch.
Sie greifen trotz dieser hohen Jodkonzentration Metalle nicht mehr an als gewöhnliches Wasser und sind daher zur Sterilis. ation aller Arten von Instrumenten geeignet. Dazu kommt, dass sie sowohl für den Azzt wie f r den Patienten nur eine geringe Toxizität aufweisen. Sie können leicht mit Wasser wegge- waschen werden und erzeugen keine bleibenden Flecken auf Tüchern oder Geweben. Sie sind auch nicht flüchtig und können, da sie gefärbt sind, ebenfalls als eigene Indikatoren betrachtet werden. Sie reizen die Haut nicht und machen sie nicht empfind- lich.
Die neuen Verbindungen bilden stabile L¯sungen.
Sie zeigen hinsichtlich der Oberflächenspannung die Eigenschaften eines Waschmittels, wodurch ihre keim- tötende Wirkung gefördert wird, da sie tief in die Haut eindringen. Schliesslich haben sie keinen unan genehmen Geruch oder Geschmack.
Der Wert der vorliegenden Erfindung beruht auf der Erkenntnis, dass die Nachteile, welche das freie Jod als s Keimt¯tungsmittel aufweist, durch seine Verbindu. ng mit den organischen quaternären Aminsalzen aufgehoben werden können, ohne e seine wertvollen Eigenschaften zu schädigen. Es s scheint sogar eine Wechselwirkung zwischen dem Jod und dem quater- nären Salz zu bestehen, da die neuen Verbindungen als Keimtötungsmittel weit wirks, amer sind als wenn man ihre Komponenten für sich allein anwendet. Die Verbindungen zeigen einen Phenolkoeffizienten, der bei manchen Organismen n gegen ber dem der Bestand- teile eine Steigerung von 200 auf 2000 aufweist.
Das Anion der vewendeten Salze ist vorzugsweise ein Halogenidion. Ein vorteilhaft angewendetes Salz ist das N-Capryloxyäthylaminocarbonyl-methylpyridin- chlorid. Andere geeignete Salze enthalten als Acylradikal die Lauroyl-, Myristoyl-, Palmitoyl- oder Stearoylgruppe. Der quaternäre Stickstoff kann dem Picolin, dem 2, 4-Lutidin, dem Pyrrolidin, dem Chinolin, dem Isochinolin oder einem Pyridinderivat ange hören. Da, s Salz kann ein Bromid, ein Jodid, ein Nitrat, Sulfat oder Phosphat sein.

Die neuen jodhaltigen Verbindungen können in Form ihrer Lösungen oder auch als feste oder halb- feste Präparate Verwendung finden. Vorzugsweise benutzt man wϯrige L¯sungen, deren Zusammensetzung aus der nachstehenden Tabelle ersichtlich ist.

Vorzugsweise verwendet man als Lösungsmittel reines Wasser. Wenn es erwünscht ist, können aber auch wasserlösliche organische Lösungsmittel zugesetzt werden. Geeignete L¯sungsmittel solcher Art sind Methanol, Athanol, Propanol, Methylacetat, ¯thylacetat, Propylacetat, Aceton, MethylÏthylketon und dergleichen. F r die blichen physiologischen Anwendungen ist es zweckmässig, sehr wenig oder besser gar keinen Zusatz von organischen Lösungsmitteln zu benutzen, da dieselben den Dampfdruck erhöhen, den Substanzverlust durch Verdunstung g steigern und freiliegendes Hautgewebe reizen können.
Bei der Herstellung der Verbindungen kann man in verschiedener Weise vorgehen. Als zweckmässig hat sich folgender Weg erwiese n: Man l¯st zunächst das quaternäre Aminsalz in Wasser auf, so dass man eine Lösung erhält, die eine Konzentration von 20-50 Gew. ío hat. Dann fügt man das elementare Jod hinzu. Die Reaktion verläuft nur langsam. Man rührt das Gemisch kräftig während 1-4 Stunden und lϯt es hernach 1-4 Tage stehen, um die vollständige Aufnahme des Jods sicherzustellen. Schlie¯lich kann man die erhaltene Lösung mit Wasser auf die gewünschte Konzentration verd nnen.
Es ist vorteilhaft, die beschriebenen Schritte bei einer Temperatur von nicht mehr als 38¯C durchzuführen. Die neuen Verbindungen neigen bei der Erhitzung zur Zersetzung. Wenn man h¯here Tem- peraturen anwendet, kann es vorkommen, dass die Lösung dunkel wird und das au. sgeschiedene Jod verdampft.
Um den Dbergang von Jod in Jodid oder Jodat zu verhindern, ist es zweckmässig, den Vorgang bei einem pH von weniger als 9, besser noch bei weniger als 7 durchzufiihren. Die Reaktion verläuft am besten bei einem pH von 1, 5-4, und bei diesem pH sind die neuen Verbindungen am wirksamsten. Man kann den günstigsten pH-Wert durch Zusatz von verschiedenen Pufferungsmitteln einstellen. Geeignete Puffer sind Mineralsäuren, wie SalzsÏure, Schwefelsäure, PhosphorsÏure, und gewisse organische Säu- ren, wie Essigsäure, Propionsäure und Benzoesäure.
Man kann auch verschiedene saure Salze benutzen, wie Natriumhydrosulfat, einbasisches Natriumphos- phat und dergleichen.
Beispiel
17 Gewichtsteile N-Capryloxyäthylaminocarbonyl methyl-pyrldin-chlorld wurden in 17 Gewichtsteilen Wasser gelöst. Darauf wurden 3 Gewichtsteile Jod hinzugefügt, und das Gemisch wurde bei Raumtemperatur während 4 Stunden kräftig gerührt. Man liess dann die Mischung 8 Tage lang stehen, bis das Jod vollständig aufgelöst war. Schliesslich wurden 63 Gewichtsteile Wasser zugesetzt. Damit war die Lösung gebrauchsfertig.
Die Beobachtung der Lösung während 30 Tagen zeigte, dass die erhaltene Verbindung stabil war und kein Jod durch Verdunstung verlor. Die Lösung griff tierische Gewebe nicht an. Sie verhielt sich auch völlig neutral gege. nüber Metallen und zeigte keinen unangenehmen Geruch oder Geschmack. Sie hatte eine braune Farbe, die so lange anhielt, als noch ent- sprechende Mengen der Verbindung zugegen waren.
Infolgedessen bildete sie während ihrer Anwendung ihren eigenen Indikator, da die braune Farbe immer heller wurde, nach Massgabe des Verbrauchs der Verbindung, und schliesslich ganz verblasste, was anzeigte, dass die Verbindung vollständig von dem behandelten Organismus aufgenommen war.
Die Wirksamkeit der neuen Verbindungen als s Keimtötungsmittel lässt sich aus folgendem Versuch erkennen : Eine Komplexverbindung von N-CapryloxyÏthylaminocarbonylmethyl-pyridinchlorid und Jod, die 3, 4 /o Jod und d 17 % des Aminsalzes enthält, wurde gegen die Sporen von Tricophyton interdigi- talis als wässrige L¯sung eingesetzt, wobei man Phenol als Vergleichsmittel benutzte. Die Ergebnisse zeigten, dass die Komplexverbindung die Sporen m 10 Minu- ten abtötete, wenn sie in einer Verdiinnung von 1 : 40000 angewendet wurde. Phenol tötete den Pilz unter den gleichen Bedingungen bei einer Verdünnung von 1 : 45.
Die Komplexverbindung hat also in der Anwendung gegen diesen Sporenbildner, der bekanntlich der Erreger der Fu¯seuche ist, einen Phenolkoeffizienten von 890.
Es wurden ferner Vergleichsversuche mit der Jod- verbindung des N - CapryloxyÏthylaminocarbonylmethyl-pyridin-chlorids und vier anderen Desinfektionsmitteln durchgeführt, wobei als Testorganismus wieder Tricophyton digitalis diente. Die Feststellung ergab, dass zur Abtötung des Organismus in 15 Se kundenfolgende Mengen an Desinfektoren erforder- lich waren : Chlor 1000 ppm, Natriumhypochlorit 1000 ppm, quaternäres Alkyl-dimethyl-benzylammonium-chlorid, ¸Roccal¯, 2500 ppm und Chloramin T¯ (Markenprodukte) 5000 ppm.
Zur Erzialung der gleichen Wirkung gen gten aber 5 ppm einer wϯrigen L¯sung der Komplexverbindung mit 3, 40/o Jod und 17 /o N-Capryloxyäthylaminocarbonylmethyl- pyridin-chlorild.
Andere Testversuche wurden ausgeführt, um die Wirksamkeit der neuen Verbindungen gegen verschiedene Mikroorganismen, nämlich M. caseolyticus, E. coli und Ps. aoruginosa, festzustellen. Diese Orga nismen wurden Aufstrichen entnommen, die 24 Stunden lang bebrütet worden waren, wob. ei Agar als Nährboden verwendet wurde. Das verwendete Keim- tötungsmittel war wieder die Verbindung von Jod und N-CapryloxyÏthylaminocarbonylmethyl-pyridinchlorid mit 3, 4 % Jod und 17 % des Aminsalzes. Man benutzte eine wässrige Lösung, die durch Pufferung mit 0, 02molarer Natriumacetatl¯sung auf einen pH Wert von 5, 0 eingestellt war. Der pH-Wert bei Be endigung der Versuche betrug 4, 9-5.
Die angewandte Methode war die Jonesmodifikatton der Weber und Black-Technik. Die erhaltenen Ergebnisse sind nachstehend zusammengefasst.

Ein weiterer Test wurde ausgeführt, um den Einfluss der neuem Verbindungen auf Hefezellen zu zeigen.
Ein NÏhrboden, der 1 % Saccharomyces ellipsoides, 1 % Glucose und 1 % Agar enthielt, wurde 18 Stunden lang bei 0¯ C bebr tet. 244 cm3 der erhaltenen Hefesuspension wurden mit 5 mg einer wässrigen Lösung der Verbindung versetzt, die 3, 4 O/o Jod und 17 % N-CapryloxyÏthylaminocarbonylmethylpyridin-chlorid enthält. Das Engebnis war die voll- ständige Abtötung der Hefezellen nach einer Berüh- rungszeit von 2 Minute.
Die beschriebenen Versuche beweisen, dass die nzuen Verbindu, ngen eine ausserordentliche Wirksam- keit gegen ber den Testorganismen entfalten, sogar in Gegenwart organischer Substanzen.
Die neuen Komplexe gemϯ der Erfindung stellen vorzügliche Keimtötungsmittel für eine grosse Anzahl der verschiedensteln Organismen dar. Sie sind wesent- lich aktiver als Jod, sie sind nicht kaustisch und besitzen nicht dessen störende Eigenschaften der Rei zung, der Fleckenbildung und der Unstabilität, welche allen Präparaten anhaften, die freines Jod enthalten.

A desirable property of iodine as a germicidal agent is the fact that its range of action is very broad. It ranges from viruses to the primitive filaria of the nematodes. The anion of the salts used is preferably a halide ion. A salt that is advantageously used is N-capryloxyethylaminocarbonyl-methylpyridine chloride. Other suitable salts contain the lauroyl, myristoyl, palmitoyl or stearoyl group as acyl radical. The quaternary nitrogen can listen to picoline, 2,4-lutidine, pyrrolidine, quinoline, isoquinoline or a pyridine derivative. The salt can be a bromide, an iodide, a nitrate, a sulfate or a phosphate.

17 parts by weight of N-capryloxyethylaminocarbonyl methyl-pyrldine-chlorld were dissolved in 17 parts by weight of water. Then 3 parts by weight of iodine was added and the mixture was stirred vigorously at room temperature for 4 hours. The mixture was then allowed to stand for 8 days until the iodine was completely dissolved. Finally 63 parts by weight of water were added. The solution was then ready to use.
Observation of the solution for 30 days showed that the compound obtained was stable and did not lose iodine by evaporation. The solution did not attack animal tissues. She also behaved completely neutrally. Metals and did not show any unpleasant smell or taste. It was brown in color that lasted as long as adequate amounts of the compound were still present.
As a result, during its use, it formed its own indicator, as the brown color became lighter and lighter as the compound was consumed, and eventually faded completely, indicating that the compound was completely absorbed by the treated organism.
The effectiveness of the new compounds as germicidal agents can be seen from the following experiment: A complex compound of N-capryloxyÏthylaminocarbonylmethylpyridine chloride and iodine, which contains 3, 4 / o iodine and 17% of the amine salt, was interdigitated against the spores of Tricophyton used as an aqueous solution, using phenol as a comparison agent. The results showed that the complex compound killed the spores in 10 minutes when used at a dilution of 1: 40,000. Phenol killed the fungus under the same conditions at a 1:45 dilution.
The complex compound has a phenol coefficient of 890 when used against this spore-forming agent, which is known to be the causative agent of the foot disease.
Comparative tests were also carried out with the iodine compound of N-capryloxyethylaminocarbonylmethylpyridine chloride and four other disinfectants, with Tricophyton digitalis again serving as the test organism. The results showed that the following amounts of disinfectors were required to kill the organism in 15 seconds: chlorine 1000 ppm, sodium hypochlorite 1000 ppm, quaternary alkyl-dimethyl-benzylammonium chloride, calRoccal¯, 2500 ppm and chloramine T¯ (branded products ) 5000 ppm.
To achieve the same effect, however, 5 ppm of an aqueous solution of the complex compound with 3.40 / o iodine and 17 / o N-capryloxyethylaminocarbonylmethylpyridine chloride were sufficient.
Other tests were carried out to determine the effectiveness of the new compounds against various microorganisms, namely M. caseolyticus, E. coli and Ps. Aoruginosa. These organisms were taken from spreads which had been incubated for 24 hours, waving. agar was used as the culture medium. The germicide used was again the combination of iodine and N-capryloxylothylaminocarbonylmethylpyridine chloride with 3.4% iodine and 17% of the amine salt. An aqueous solution was used which had been adjusted to a pH of 5.0 by buffering with 0.02 molar sodium acetate solution. The pH at the end of the experiments was 4.9-5.

Another test was carried out to show the influence of the new compounds on yeast cells.
A culture medium containing 1% Saccharomyces ellipsoides, 1% glucose and 1% agar was incubated at 0ºC for 18 hours. 244 cm3 of the yeast suspension obtained were mixed with 5 mg of an aqueous solution of the compound which contains 3.4% iodine and 17% N-capryloxylothylaminocarbonylmethylpyridine chloride. The result was the complete killing of the yeast cells after a contact time of 2 minutes.
The experiments described prove that the useful compounds are extraordinarily effective against the test organisms, even in the presence of organic substances.
The new complexes according to the invention are excellent germicides for a large number of different organisms. They are much more active than iodine, they are not caustic and do not have the disruptive properties of irritation, staining and instability, all of which Adhere to preparations containing free iodine.
