Verfahren zur Herstellung von Sulfonylharnstoffen
Für die Herstellung von Sulfonylharnstoffen, insbesondere von Benzolsullonylhamstoffen, sind verschiedene Verfahren bekannt. So kann man beispielsweise Benzolsulfamid mit Alkyl- oder Phenylisocyanaten umsetzen, wobei man vorteilhaft von den Natriumsalzen der Suifonsäureamide ausgeht (vergleiche französische Patentschrift Nr. 993465). Mehrere andere Methoden sind in den Chemical Reviews 50 (1952), Band IV, Seiten 1 ff., beschrieben. Diese bekannten Verfahren sind in neuerer Zeit auch zur Herstellung von Sulfonylharnstoffen mit blutzuckersenkender Wirksamkeit herangezogen worden.
Es wurde nun ein neues, technisch vorteilhaftes und gegenüber dem bekannten Stand der Technik überraschendes Verfahren zur Herstellung von Sulfonylharnstoffen der allgemeinen Formel RO2-NH-CO-NH-R1, worin R einen gegebenenfalls ein- oder mehrfach durch Alkyl-, Alkoxygruppen oder Halogenatome substituierten Phenyl- oder Naphthylrest, einen Diphenyl-, Phenoxyphenyl-, Phenylalkyl-oder Tetrahydronaphthylrest oder einen aliphatischen bzw. cycloaliphatischen bzw.
gemischt cycloaliphatischaliphatischen Rest mit bis zu 12 Kohlenstoffatomen und Rf einen gesättigten oder ungesättigten Alkyl-, Cycloalkyl-, Cycloalkylalkyl-, Phenylalkyl- oder Phenylrest bedeuten, wobei der aliphatische Rest auch durch Sauerstoff unterbrochen sein kann, gefunden, indem man Alkalisalze von entsprechenden Sulfon säurehalogenamiden, wobei als Halogene vorzugsweise Chlor und Brom in Betracht kommen, mit Formamiden der Formel H-CO-NH :-RX umsetzt.
Das Verfahren gemäss der Erfindung verläuft beispielsweise im Falle der Verwendung von Isobutylformamid und N-(4-Methyl-benzolsulfonsäure)-chlor amidnatrium als Ausgangsstoffe entsprechend der nachstehenden Reaktionsgleichung

Abgesehen von dem überraschenden Reaktionsverlauf besteht ein besonderer Vorzug des Verfahrens gemäss der Erfindung darin, dass die benötigten Aus gangsmaterialien auf einfache Weise zugänglich sind.
Beispielsweise können Sulfonsäure-chloramid-natriumsalze aus Sulfamiden durch Umsetzung mit Chlorkalk in wässrigem Medium und Ausfällung mit Kochsalz gemäss dem in der deutschen Patentschrift Nr. 390 658 angegebenen Verfahren gewonnen werden. Sulfonsäure-bromamid-alkaliverbindungen werden beispiels weise erhalten, indem man entsprechende Sulfamide in Form ihrer Alkalisalze in wässriger Lösung in eine wässrige Hypobromitlösung einträgt, wobei gegebenenfalls durch Zusatz von wenig konzentrierter Natronlauge die Kristallisation beispielsweise des Natriumsalzes des Sulfonsäurebromamids bewirkt, bzw. vervollständigt werden kann. Die so erhaltenen Rohprodukte lassen sich nach dem Absaugen und Trocknen bei Zimmertemperatur, beispielsweise auf Ton, direkt als Ausgangsstoffe für die weitere Umsetzung verwenden.
Die erfindungsgemäss eingesetzten Formamide können auf einfache Weise in vorzüglicher Aus beute durch Erhitzen von Ameisensäureestern, beispielsweise Ameisensäuremethylester mit primären aliphatischen, cycloaliphatischen, aromatischen und aromatisch-aliphatischen Aminen bzw. aus solchen Aminen und Kohlenoxyd erhalten werden Sie stellen bequem zu handhabende Verbidung dar.
Zweckmässig wird wie folgt verfahrens
Man arbeitet bei mässig erhöhten Temperaturen, vorzugsweise bei Dampfbadtemperatur ; Jedoch können auch etwas höher oder tiefer liegende Temperaturen angewendet werden. Insbesondere kommen Temperaturen zwischen 90 und 1300 in Frage. Die Erhitzungsdauer beträgt im allgemeinen etwa eine Stunde, jedoch können die Umsetzungen auch mit einer Reaktionsdauer von 15 bis 120 Minuten durchgeführt werden. Die Reaktion verläuft in besonders vorteilhafter Weise, wenn man dem Gemisch der Umsetzungskomponenten Salze zusetzt, z. B. Natriumchlorid, Kaliumcarbonat, oder insbesondere Natriumbicarbonat und Natriumcarbonat. Auch ein Zusatz von Chlorkalk hat sich als günstig erwiesen. Die Menge der beigemengten Salze kann bis zu 2 Mol bei molaren Ansätzen betragen.
Anstelle von Alkalisalzen kann man den Reaktionskomponenten auch tertiäre Amine, beispielsweise Tributylamin; zusetzen. Die eingesetzten Sulfonsäurehalogenamidsalze werden zweckmässig in Form ihrer wasserhaltigen Kristallisate oder in schwach feuchtem Zustand eingesetzt. Eine Verpuffung dieser Verbindung ist unter den angegebenen Bedingungen (Verdünnung mit Salzen, Wassergehalt, verhältnismässig niedrige Reaktionstemperatur) nicht zu befürchten.
Als Reaktionskomponenten für das Verfahren gemäss der Erfindung kommen in Frage: Halogenamid-alkali-, insbesondere -natrinmsalze aliphatischer und cycloaliphatischer Sulfonsäuren, wie beispielsweise
Heptansulfonsäure-chloramidnatrium,
Heptansulfonsäurebromamid-natrium,
Cyclohexansulfons äurechloramidnatrium, Cydohexansuffonsäurebromamid-natrium, ferner solche aromatische Sulfonsäuren, wie
Benzolsulfonsäure-chloramidnatrium,
Benzolsulfons äurebromamid-natrium, p-Toluolsulfonsäure-chloramidnatrium, p-Toluolsulfonsäurebromamid-natrium,
2-Methyl-6-chlorbenzolsulfonsäure-chloramid- natrium;.
2-Methyl-6-chlor-benzolsulfonsäure-bromamid natrium,
Naphthalin-2-sulfons äure-chloramidnatrium,
Naphthalin-2-sulfons äurebromamid-natrium, 5,6,7, 8-Tetrahydro-naphthalin-2-sulfons äure chloramid-natrium,
5,6,7, 8-Tetrahydro-naphthalin-2-sulfonsäure bromamid-natrium, w-Benzylsulfonsäure-chloramid-natrium und co-Benzylsulfonsäurebromamid-natrium.
Als Formamide kommen in Betracht: Formylverbindungen aliphatischer und cycloaliphatischer primärer Amine, beispielsweise N-Butyl- und N-Isobutylformamid, N-Cyclohexylformamid, N-Allylformamid, ferner die Formylverbindungen aromatischer und aromatisch-aliphatischer Amine, beispielsweise N-(ss- Phenyl-äthyl)-formamid und N-Phenylformamid.
Die Verfahrenserzeugnisse stellen insbesondere wertvolle Heilmittel dar, deren Eignung als oral verabreichbare Antidiabetika bereits bekannt ist oder bereits vorgeschlagen wurde.
Beispiel 1 N-(4-Methyl-benzolsuljonyl)-N'-isobutyl-harnstoH
144,0 g 4 -Methyl - benzolsulfonsäure-chloramid- natrium 3 H2O, 50,5 g Ameisensäure-isobutylamid und 71,0 g Na2COs mit etwa 2 Molen Kristallwasser werden in einer Schale gut gemischt und auf dem Dampfbad eine Stunde erhitzt. Unter geringer Blasenbildung tritt Reaktipn ein. Der entstandene Reaktionskuchen wird in 750 cm3 warmem Wasser gelöst; man gibt einen Tropfen Rongalitlösung zur Reduktion des nicht umgesetzten 4-Methyl-benzolsulfonsäure-chlor- amid-natriums zu, lässt die alkalische Lösung erkalten und filtriert. Das Filtrat wird mit verdünnter Salzsäure angesäuert. Die gebildeten Kristalle werden abgesaugt und mit 1 0/obigem Ammoniak behandelt.
Nach abermaligem Filtrieren säuert man das Filtrat mit Essigsäure an und erhält eine kristalline Fällung von N-(4-Methyl-benzolsulfonyl)-N'-isobutyl-harnstoff, der nach dem Absaugen und Trocknen aus etwa 450 cm3 Methanol umkristallisiert wird. Man erhält nach Auf arbeitung der Mutterlauge 76 g N-(4-Methyl-benzol. sulfonyl)-N'-isobutyl-harnstoff (560/0 der Theorie) vom Schmelzpunkt 169 bis 171".
Beispiel 2
16,3 g 4-Methyl- benzolsulfonsäurebromamid - natrium 3 H2O, 5 g N-Isobutylformamid und 7 g etwa 2 Mol Wasser enthaltendes Natriumcarbonat werden gut gemischt. Anschliessend erhitzt man das
Gemisch auf dem Dampfbad; unter Blasenbildung tritt Reaktion ein. Nach etwa 5 Minuten lässt man erkalten und löst das Reaktionsgut in warmem Wasser auf. Man reduziert eventuell nicht verbrauchtes Toluolsulfonsäurebromamid mit Hilfe von wenig Rongalitlösung und säuert mit verdünnter Salzsäure an. Der erhaltene kristalline Niederschlag wird zur weiteren Reinigung in etwa 1 /Oigem Ammoniak ge löst. Man filtriert und säuert das Filtrat mit Salzsäure an. Der so in guter Ausbeute erhaltene Niederschlag von N-(4-Methyl-benzolsulfonyl)-N'-isobutyl-harnstoff wird abgesaugt und getrocknet.
Die Substanz ist schon weitgehend rein und schmilzt nach dem Umkristallisieren aus Methanol bei 169 bis 171".
In analoger Weise erhält man aus p-Toluol-sulfonsäurebromamid-natrium und N-Formylbutylamin den N-(4-Methyl-benzolsulfonyl)-N'-n-butyl-harnstoff, dessen Schmelzpunkt nach dem Umkristallisieren aus Essigester 125 bis 127 beträgt.
Beispiel 3
Analog Beispiel 1 erhält man durch 30 Minuten langes Erhitzen von 14 g 4-Methyl-benzolsulfonsäurechloramid-natrium, 5,0 g Ameisensäure-n-butylamid und 7,0 g Na2CO3 mit etwa 2 H20 einen Kristallkuchen, der in warmem Wasser gelöst wird. Nach Zugabe von Rongalit säuert man mit verdünnter Salzsäure an, saugt die ausgefällte Substanz ab und behandelt das Reaktionsprodukt mit 1 0/obigem Ammoniak. Man klärt mit Kohle, saugt ab und erhält durch Ansäuern mit verdünnter Salzsäure eine kristalline Fällung von N-(4-Methyl-benzolsulfonyl)-N'-n butyl-harnstoff, die abgesaugt und getrocknet wird.
Das in einer Ausbeute von 8,8 g (65 /o der Theorie) erhaltene Rohprodukt vom Schmelzpunkt 117 bis 119 ergibt nach dem Umkristallisieren aus Essigester 7,2 g (53 ovo der Theorie) N-(4-Methyl-benzolsulfonyl) N'-n-butyl-harnstoff vom Schmelzpunkt 125 bis 1270.
Beispiel 4
13,3 g Benzolsulfonsäure-chloramid-natrium .3 3 H2O, 5,0 g Ameisensäure-isobutylamid und 3,5 g Kaliumcarbonat werden gut gemischt. Man erhitzt die Mischung etwa 30 Minuten auf dem Dampfbad. Nach etwa 10 Minuten entsteht eine lebhafte Gasentwicklung. Man lässt erkalten und löst das Reaktionsgut in Wasser, filtriert und säuert das Filtrat mit verdünnter Salzsäure an. Die erhaltene kristalline Fällung wird abgesaugt und aus verdünntem Äthanol umkristallisiert. Man erhält 6,3 g (500/0 der Theorie) N-Benzol sulfonyl-N'-isobutyl-harnstoff vom Schmelzpunkt 131 bis 133".
Beispiel 5 17,log 4- Methoxy - benzolsulfons äurebromamid- natrium (hergestellt aus 4-Methoxy-benzolsulfamidnatrium und Natriumhypobromit in Wasser) werden mit 6,3 g N-Formyl-cyclohexylamin und 7 g Natriumcarbonat vermischt. Man erhitzt das Gemisch 10 Minuten auf dem Dampfbad, lässt erkalten und nimmt in Wasser auf. Nach Reduktion von nicht umgesetztem 4-Methoxy-beazolsulfonsäurebromamid mit Rongalit säuert man an. Der erhaltene Niederschlag wird in etwa 1 Obigem Ammoniak aufgenommen; man filtriert und erhält durch Ansäuern des Filtrates mit verdünnter Salzsäure ein Kristallisat vonN-(4-Methoxy-benzolsulfonyl)-N'-cyclohexyl-harnstoff, das man absaugt und trocknet.
Das aus Äthanol umkristallisierte Produkt schmilzt bei 181 bis 183".
Beispiel 6
22,7 g 4- Methyl - benzolsulfonsäure - chloramid- natrium und 12,7 g Ameisensäure-cyclohexylamid werden gut gemischt und auf dem Dampfbad erhitzt; nach einigen Minuten ist eine dünnflüssige Lösung entstanden, wobei sich Gase von stechendem Geruch entwickeln. Nach insgesamt acht Minuten lässt man erkalten, versetzt das Reaktionsgut mit 1 /o wässrigem Ammoniak, filtriert von ungelösten Stoffen ab und säuert das Filtrat mit Essigsäure an. Nach nochmaligem Umiösen und Ausfällen des Reaktionsgutes mit Ammoniak bzw. Essigsäure trocknet man den erhaltenen N-(4-Methyl -benzolsulfonyl) -N'-cyclohexyl- harnstoff. Die Ausbeute beträgt 6,3 g (21 /o der Theorie). Man löst das Produkt aus verdünntem Äthanol um.
Der so erhaltene N-(4-Methyl-benzolsulfonyl)-N'-cyclohexyl-harnstoff schmilzt bei 170 bis 172".
Beispiel 7
15,8 g 2-Methyl-6-chlor-benzolsulfonsäure-chloramid-natrium, 7,0 g Na2CO3 mit etwa 2 H2O und 4,3 g Ameisensäure-allylamid werden gut gemischt und auf dem Dampfbad 30 Minuten erhitzt. Es entsteht ein dünner Brei, der nach 20 Minuten flüssig wird. Vereinzelt treten Blasen auf. Nach dem Erkalten wird das Reaktionsgut in warmem Wasser gelöst.
Durch Zusatz von einigen Tropfen Rongalitlösung wird das überschüssige Benzolsulfonsäure-chloramid- natrium reduziert und durch Versetzen mit verdünnter Salzsäure eine Fällung erhalten, die abgesaugt wird.
Die Substanz wird mit 1 6/obigem Ammoniak behandelt.
Man filtriert vom Ungelösten ab und säuert das Filtrat mit verdünnter Salzsäure an. Man erhält eine Fällung von 4 g N-(2-Methyl-6-chlor-benzolsulfonyl) N'-allyl-harnstoff vom Schmelzpunkt 184 bis 187 , der nach Umkristallisieren aus Essigester bei 192 bis 1940 schmilzt.
Beispiel 8
33 g 3-Methyl-4-methoxy-benzolsulfonsäurebrom- amid-natrium (hergestellt aus 3-Methyl-4-methoxybenzolsulfamid-natrium und einer äquivalenten Natriumhypobromitlösung in Wasser, wobei das genannte Salz durch Zusatz von Natronlauge zur Kristallisation gebracht wird), 14 g Natriumcarbonat und 18 g N Formyl-ss-phenyl-äthylamin werden gemischt. Das Gemisch wird in einemvRundkolben auf dem Dampfbad erhitzt. Nach etwa fünf Minuten tritt lebhafte Reaktion ein. Man lässt erkalten, löst das Reaktionsgut in Wasser und säuert mit verdünnter Salzsäure an. Der erhaltene Niederschlag wird abgesaugt, in etwa 1 0/obigem Ammoniak gelöst und nach Filtration aus dem Filtrat durch erneutes Ansäuern wieder ausgefällt.
Die erhaltenen Kristalle von N-(3-Methyl-4-methoxy-ben zolsulfonyl)-N'-ss-phenyläthyl-harnstoff werden abgesaugt und nach dem Trocknen auf Ton aus Äthanol umkristallisiert.- Die Substanz schmilzt bei 148 bis 1500.
Process for the preparation of sulfonylureas
Various processes are known for the production of sulfonylureas, in particular of benzenesulonylureas. Apart from the surprising course of the reaction, a particular advantage of the method according to the invention is that the required starting materials are easily accessible.
For example, sulfonic acid-chloramide-sodium salts can be obtained from sulfamides by reaction with chlorinated lime in an aqueous medium and precipitation with common salt according to the process specified in German Patent No. 390,658. The following procedure is appropriate
One works at moderately elevated temperatures, preferably at steam bath temperature; However, slightly higher or lower temperatures can also be used. The products of the method represent in particular valuable remedies, the suitability of which as orally administrable antidiabetic agents is already known or has already been proposed.
Example 1 N- (4-methyl-benzenesuljonyl) -N'-isobutyl-urea
144.0 g of 4-methylbenzenesulfonic acid chloramide sodium 3 H2O, 50.5 g formic acid isobutylamide and 71.0 g Na2COs with about 2 moles of water of crystallization are mixed well in a bowl and heated on the steam bath for one hour. Example 2

The substance is already largely pure and, after recrystallization from methanol, melts at 169 to 171 ".
In an analogous manner, N- (4-methylbenzenesulfonyl) -N'-n-butylurea is obtained from sodium p-toluenesulfonic acid bromamide and N-formylbutylamine, the melting point of which is 125 to 127 after recrystallization from ethyl acetate.
Example 3
Analogously to Example 1, heating 14 g of sodium 4-methylbenzenesulfonic acid chloramide, 5.0 g of n-butylamide formic acid and 7.0 g of Na2CO3 with about 2 H2O for 30 minutes gives a crystal cake which is dissolved in warm water. After the addition of Rongalit, it is acidified with dilute hydrochloric acid, the precipitated substance is filtered off with suction and the reaction product is treated with the above ammonia. It is clarified with charcoal, filtered off with suction and, by acidification with dilute hydrochloric acid, a crystalline precipitate of N- (4-methylbenzenesulfonyl) -N'-n-butylurea is obtained, which is filtered off with suction and dried.
The crude product obtained in a yield of 8.8 g (65 / o of theory) with a melting point of 117 to 119 gives, after recrystallization from ethyl acetate, 7.2 g (53 ovo of theory) of N- (4-methyl-benzenesulfonyl) N ' -n-butyl urea from melting point 125 to 1270.
Example 4
