Verfahren zur Herstellung von Sulfonylharnstoffen Gegenstand des Patents
1066 575 ist ein Verfahren zur Herstellung von Sulfonylharnstoffen der allgemeinen
Formel R-S02-NH-CO-NH-Rl, worin R einen gegebenenfalls ein- oder mehrfach durch
Alkyl-, Alkoxygruppen oder Halogenatome substituierten Phenyl- oder Naphthylrest,
einen Diphenyl-, Phenoxyphenyl-, Phenylalkyl- oder Tetrahydronaphthalinrest oder
einen aliphatischen bzw. cycloaliphatischen bzw. gemischt cycloaliphatisch- aliphatischen
Rest mit bis zu 12 Kohlenstoffatomen und R1 einen gesättigten oder ungesättigtenAlkyl-,Cycloalkylalkyl-,Cycloalkyl-,
Phenylalkyl- oder Phenylrest bedeuten, wobei der aliphatische Rest auch durch Sauerstoff
unterbrochen sein kann, dadurch gekennzeichnet, daß man Alkalisalze von Sulfonsäurechloramiden
mit Formamiden der Formel H-CO- NH-Rl, worin R1 die angegebene Bedeutung aufweist,
umsetzt. In weiterer Ausgestaltung des Verfahrens gemäß Patent 1066 575 wurde
nun gefunden, daß sich die in der Hauptanmeldung genannten Sulfonylharnstoffe vorteilhaft
auch herstellen lassen, indem man Alkalisalze entsprechender Sulfonsäurebromamide
mit Formamiden der Formel H - C O - N H - R, umsetzt. Die Umsetzung verläuft im
Falle der Verwendung von Isobutylformamid und N-(4-Methyl-benzolsulfonsäure)-bromamidnatrium
als Ausgangsstoffe entsprechend der nachstehenden Reaktionsgleichung:
Das Formelschema soll nicht nur die Darstellung des N-(4-Methyl-benzolsulfonyl)-N'-isobutyl-harnstoffs
veranschaulichen, sondern ganz allgemein den Verlauf der Reaktion demonstrieren.
Abgesehen von dem überraschenden Reaktionsverlauf besteht für das Verfahren gemäß
der Erfindung ebenso wie für das Verfahren des Hauptpatentes auch ein Vorzug darin,
daß die benötigten Ausgangsmaterialien auf einfache Weise zugänglich sind. Sie können
beispielsweise erhalten werden, indem man entsprechende Sulfamide in Form ihrer
Alkalisalze in wäßriger Lösung in eine wäßrige Hypobromitlösung einträgt. Gegebenenfalls
kann durch Zusatz von wenig konzentrierter Natronlauge die Kristallisation beispielsweise
des Natriumsalzes des Sulfonsäurebromarnids bewirkt bzw. vervollständigt werden.
Die irri allgemeinen gut kristallisierenden Alkalisalze der Sulfonsäurebromamide
scheiden sich in sehr guter Ausbeute aus und können nach dem Absaugen und Trocknen
bei Zimmertemperatur, beispielsweise auf Ton, direkt als Ausgangsstoffe für das
Verfahren gemäß der Erfindung verwendet werden (vgl. hinsichtlich der Darstellung
der Ausgangsstoffe auch die Veröffentlichung von F. D. Chattaway in J. Chem. Soc.,

Auch ein Zusatz von Chlorkalk hat sich als günstig erwiesen. Die 'Menge der beigemengten
Salze kann bis zu 2 Mol bei molaren Ansätzen betragen. An Stelle von Alkalisalzen
kann man den Reaktionskomponenten auch tertiäre Amine, beispielsweise Tributylämin,
zusetzen. Die eingesetzten Sulfonsäurechloramidsalze werden zweckmäßig in Form ihrer
wasserhaltigen Kristallisate oder in schwach feuchtem Zustand eingesetzt. Eine Verpuffung
dieser Verbindung ist unter den angegebenen Bedingungen (Verdünnung mit Salzen,

an. Der so in einer Ausbeute von 65 °/o erhaltene Niederschlag von N-(4-Methyl-benzolsulfonyl)-N'-isobutyl-harnstoff
wird abgesaugt und getrocknet. Die Substanz ist schon weitgehend rein und schmilzt

man filtriert und erhält durch Ansäuern des Filtrates mit verdünnter Salzsäure 26
°/o der Theorie eines Kristallisats von N-(4-Methoxy-benzolsulfonyl)-N'-cyclohexyl-harnstoff,
das man absaugt und trocknet. Der aus Äthanol umkristallisierte Harnstoff schmilzt
bei 181 bis 183°C. Beispiel 3 33 g 3 - Methyl - 4 - methoxy - benzolsulfonsäurebromamid-natrium
(hergestellt aus 3-Methyl-4-methoxy-benzolsulfamid-natrium und einer äquivalenten
Natriumhypobromitlösung in Wasser, wobei das genannte Salz durch Zusatz von Natronlauge

durch erneutes Ansäuern wieder ausgefällt. Die erhaltenen Kristalle von N-(3-Methyl-4-methoxy-benzolsulfonyl)-N'
ß-phenyläthyl-harnstoff werden abgesaugt und nach dem Trocknen auf Ton aus Äthanol
umkristallisiert. Man erhält 31 ()/o der Theorie des Verfahrenserzeugnisses vom
