Erfindungsgemäss werden die neuen, in 1-Stellung durch eine basische Gruppe und in 4-Stellung durch einen heterozyklischen Rest substituierten 5-Amino- pyrazol-Derivate beispielsweise wie folgt hergestellt: Man versetzt unter Rühren bei Raumtemperatur die Suspension eines Acylessigsäurenitrils der Formel II, beispielsweise a-Propionyl-thienyl-(2)-aceto-nitril, in einem wasserfreien organischen Lösungsmittel, z. B.
Eisessig, mit der Lösung eines Hydrazin-Derivates, beispielsweise N-Methyl-piperidyl-4-hydrazin, im glei chen Lösungsmittel. Zur Vervollständigung der Reak tion wird anschliessend noch etwa 2 Stunden bei Raumtemperatur weitergerührt. Nach Entfernen des Lösungsmittels im Vakuum wird das gewünschte Endprodukt nach bekannten Methoden aufgearbeitet und z. B. durch Destillation im Hochvakuum isoliert und gegebenenfalls durch Kristallisation gereinigt.
Die erfindungsgemäss hergestellten neuen Pyrazol- Derivate sind bei Raumtemperatur kristallin oder flüssig und lassen sich im Hochvakuum unzersetzt destillieren. Es sind basische Verbindungen, die mit anorganischen und organischen Säuren beständige, bei Raumtemperatur kristallisierte wasserlösliche Salze bilden.
Die nach dem vorliegenden Verfahren hergestell ten neuen Pyrazol-Derivate sind auf Grund ihrer aus gezeichneten pharmakodynamischen Eigenschaften zur Verwendung als Heilmittel in hohem Masse ge eignet. So wirken sie bei geringer Toxizität antiphlo- gistisch, analgetisch und antipyretisch und besitzen insbesondere eine spezifische antirheumatische Wir kung.
Da die neuen Pyrazol-Derivate mit anorganischen und organischen Säuren sehr leicht in wasser lösliche Salze überführbar sind, stehen der Therapie Pyrazol-Derivate zur Verfügung, die sich durch be sonders gute Resorbierbarkeit, so auch in per os zu verabreichenden Anwendungsformen, z. B. Tabletten, auszeichnen. Ferner gestaltet sich dadurch die Her stellung hochkonzentrierter Lösungen, die in vielen Fällen für die parenterale Stosstherapie unentbehrlich sind, besonders einfach.
Darüber hinaus können die verfahrensgemäss hergestellten Pyrazol-Derivate auch als Zwischenprodukte zur Herstellung von Medika menten verwendet werden.
Die als Ausgangsmaterialien zur Verwendung gelangenden Hydrazin-Derivate werden nach den in den Schweiz. Patentschriften Nrn. 351268 und 370796 beschriebenen Verfahren hergestellt.
In den nachfolgenden Beispielen sind die Tempe raturen in Celsius-Graden angegeben. Die Schmelz- und Siedepunkte sind unkorrigiert. <I>Beispiel 1</I> 1-(N-Methyl-piperidyl-4')-3-methyl-4-(thienyl-2")- 5-amino-pyrazol Eine Lösung von 16,0 g a-Acetyl-thienyl-(2)- acetonitril und 12,5g N-Methyl-piperidyl-4-hydrazin in 225 cm3 abs. Äthanol wird 1 Stunde bei Zimmer temperatur stehengelassen und anschliessend 3 Stun den am Rückfluss zum Sieden erhitzt.
Darauf wird der Äthanol im Vakuum abgedampft und der schwarzbraune, dickflüssige Rückstand im Hoch vakuum destilliert, wobei das 1-(N-Methyl-piperidyl- 4')-3-methyl-4-(thienyl-2")-5-amino-pyrazol zwischen 179-210 bei 0,1 mm Hg überdestilliert und in der Vorlage erstarrt. Nach Umkristallisation aus Benzol schmilzt das Aminopyrazol-Derivat bei 97-99 .
<I>Beispiel 2</I> 1-(N-Methyl-piperidyl-4')-3-äthyl-4-(thienyl-2")- 5-amino-pyrazol Zu einer Suspension von 15,0 g a-Propionyl- thienyl-(2)-acetonitril in 37 em3 Eisessig lässt man unter Rühren eine Lösung von 10,8g N-Methyl- piperidyl-4-hydrazin in 37 cms Eisessig so zutropfen, dass die Temperatur 30 nicht übersteigt.
Anschlie ssend wird das dunkle, graubraune Reaktionsgemisch noch 2 Stunden bei Zimmertemperatur weitergerührt, und darauf der Eisessig im Vakuum bei etwa 40 abdestilliert. Der Rückstand wird in 250 cm3 Wasser aufgenommen, und die wässerige Lösung mit Na- triumcarbonat alkalisch gestellt und gesättigt, wobei sich eine dunkelbraune, harzartige Masse ausscheidet. Man extrahiert mit total 700 cm3 Chloroform, trock net die vereinigten Chloroformextrakte über Magne- siumsulfat und entfernt das Chloroform im Vakuum.
Der kristalline Rückstand, das 1-(N-Methyl-pipe- ridyl-4')-3-äthyl-4-(thienyl-2'@-5-amino-pyrazol wird aus Benzol umkristallisiert und schmilzt dann bei 129-130 .
Das als Ausgangsmaterial zur Verwendung ge langende a-Propionyl-thienyl-(2)-acetonitril wird her gestellt, indem man Thienyl-(2)-acetonitril in äthano- lischer Lösung in Gegenwart von Natriumäthylat mit Propionsäureäthylester umsetzt. Smp. 89-91 aus Äther/Petroläther.
<I>Beispiel 3</I> 1-(N-Isopropyl-piperidyl-4')-3-äthyl-4-(thienyl-2")- 5-amino-pyrazol Zu einer Suspension von 5,4 g a-Propionyl- thienyl-(2)-acetonitril in 13 cm3 Eisessig lässt man unter Rühren eine Lösung von 4,75g N-Isopropyl- piperidyl-4-hydrazin in 13 cm3 Eisessig so zutropfen, dass die Temperatur 50 nicht übersteigt.
An schliessend wird das dunkle, graubraune Reaktions gemisch noch 2 Stunden bei Zimmertemperatur weitergerührt und darauf der Eisessig im Vakuum bei etwa 40 abdestilliert. Der Rückstand wird im Hochvakuum destilliert, wobei das 1-(N-Isopropyl- piperidyl-4') - 3 - äthyl-4-(thienyl-2")-5-amino-pyrazol zwischen 175-205 bei 0,01 mm Hg als zähflüssiges, braungelbes Öl übergeht.
Das Destillat wird nun in wenig Methanol gelöst und mit der berechneten Menge einer methanoli- schen Maleinsäurelösung versetzt. Man konzentriert die Lösung im Vakuum und gibt dann langsam Äther zu, wobei sich das 1-(N-Isopropyl-piperidyl-4')-3- äthyl - 4 - (thienyl-2") - 5 - amino-pyrazol-bis-maleinat kristallin ausscheidet.
Nach zweimaligem Umkristal- lisieren aus Methanol/Äther schmilzt das Amino- pyrazol-Derivat bei 13l-132 .
Das Ausgangsmaterial, das a-Propionyl-thienyl- (2)-aceto-nitril, wurde wie in Beispiel 2 beschrieben hergestellt.
<I>Beispiel 4</I> 1-(N-Methyl-piperidyl-4')-3-n-butyl-4-(thienyl-2")- 5-amino-pyrazo1 Zu einer Suspension von 5,1 g a-Valeroyl-thienyl- (2)-aceto-nitril in 12 cm3 Eisessig lässt man unter Rühren innert 20 Minuten eine Lösung von 3,2 g N-Methyl-piperidyl-4-hydrazin in 12 cm3 Eisessig so zutropfen, dass die Temperatur 50 nicht über steigt.
Anschliessend wird die dunkelbraune Reak tionslösung noch 2 Stunden bei Zimmertemperatur weitergerührt, und darauf der Eisessig im Vakuum bei 40-50 abdestilliert. Der Rückstand wird in 120 cm3 Wasser aufgenommen, und die wässerige Lösung unter Eiskühlung mit Pottasche alkalisch ge stellt und gesättigt, wobei sich eine dunkelbraune, harzartige Masse ausscheidet. Man extrahiert mit total 150 cm3 Chloroform, trocknet die vereinigten Chloroformextrakte über Magnesiumsulfat und ent fernt das Chloroform im Vakuum.
Der Rückstand wird destilliert, wobei das 1-(N-Methyl-piperidyl-4')- 3-n-butyl-4-(thienyl-2")-5-amino-pyrazo1 bei einer Luftbadtemperatur von 180-210 bei 0,01 mm Hg übergeht.
Zur Herstellung des Bis-maleinats wird das gelb braune, dickflüssige Destillat in 10 cm3 abs. Äthanol aufgenommen und mit einer Lösung von 4,3g Ma leinsäure in 10 cm3 abs. Äthanol versetzt. Man ent fernt das Lösungsmittel im Vakuum und kristallisiert den kristallinen Rückstand, das 1-(N-Methyl-pipe ridyl-4')-3-n-butyl-4-(thienyl-2")-5-amino-pyrazol-bis- maleinat, zweimal aus MethanoljÄther um. Smp. 128 bis 130 . Farblose Kristalle.
Das als Ausgangsmaterial zur Verwendung ge langende a-Valeroyl-thienyl-(2)-acetonitril wird her gestellt, indem man Thienyl-(2)-acetonitril in äthano- lischer Lösung in Gegenwart von Natriumäthylat mit Valeriansäure-äthylester umsetzt. Smp. 70-72 aus Äther;Petroläther.
<I>Beispiel 5</I> 1-(N-Methyl-piperidyl-4')-3-methyl-4-(pyridyl-3")- 5-amino-pyrazol 6,9 g des Natriumsalzes von a-(Pyridyl-3)-acet- essigsäurenitril in 70 cm3 Eisessig werden 15 Minu ten auf 40 erwärmt, wobei vollständige Lösung ein tritt. Nach Abkühlen auf 20 lässt man unter Rühren eine Lösung von 4,9g N-Methyl-piperidyl-4-hydrazin in 8 em3 Eisessig so zutropfen, dass die Temperatur 40 nicht übersteigt.
Anschliessend wird das Reak tionsgemisch 5 Stunden bei Zimmertemperatur wei tergerührt, über Nacht stehengelassen und darauf der Eisessig im Vakuum bei 40-50 abdestilliert. Der Rückstand wird in 100 em3 Wasser aufgenommen, mit total 120 cm3 Äther gewaschen, die wässerige Lösung mit Natriumcarbonat alkalisch gestellt und gesättigt und mit total 180 cm3 Chloroform extra hiert.
Nach Trocknen über Magnesiumsulfat wird das Chloroform im Vakuum abgedampft, und der kristalline Rückstand, das 1-(N-Methyl-piperidyl-4')- 3-methyl-4-(pyridyl-3")-5-amino-pyrazol, aus Essig ester und Chloroform/Petroläther umkristallisiert. Smp. 188-190 .
Das als Ausgangsmaterial zur Verwendung ge langende Natriumsalz des a-(Pyridyl-3)-acetessig- säurenitrils wird hergestellt, indem man Pyridyl-(3)- acetonitril in äthanolischer Lösung in Gegenwart von Natriumäthylat mit Essigsäureäthylester umsetzt.
Smp. etwa 275 (Zers.). Behandeln des Natriumsalzes mit Eisessig ergibt das a-(Pyridyl-3)-acetessigsäure- nitril. Smp. 194-196 .
